Analysis of the Changes in Metabolic Diversity of Microbial Community in pH-gradient Microcosm

  • The Biolog redox technology was carried out for evaluation of acidification effect on microbial communities at each stage of pH gradient microcosm. While the number of heterotrophic bacterial population and activities of extracellular enzyme decreased as the pH decreased, the number of total bacteria in the microcosm was not affected. The average color development of sample at each pH-gradient showed a sigmoidal curve, and at higher pH, more overall color development appeared in Biolog plates. Average color development value in Biolog plates was stabilized at 50 hours as an optimum incubation time. The color production in the Biolog plates was caused by cell density at above pH 5.0, but by cell activity below pH 4.0. Principal component analysis of color responses revealed distinctive patterns among the pH-gradient microcosm samples.

Microwave를 이용한 칫솔 살균효과 (Toothbrush sterilizing effects of using microwave)

  • Objectives : The purpose of this study is to analyze the sterilizing effects of toothbrushes by administering bacteria into toothbrushes and reproducing the antibacterial effects using a microwave oven. Methods : The heads of four-row mid-strength toothbrushes were cut, put in a bacterial solution ($3{\times}10^9cells/ml$) for vortexing, and sterilized with microwaves for 0, 30, and 60 seconds. They were then moved into four tubes containing DW 10 ml and suspended in a vortex mixer for two minutes to separate bacteria from them. DW 9ml was added by 1ml of bacteria for dilution of $10{\sim}10^6$ times. It was then administered to the BHI agar plate by 0.1ml and cultured at $37^{\circ}C$ for 24 hours. Total number of bacteria adhered to a toothbrush was obtained by multiplying the number of colonies by the dilution factor. The experiment was done in the first, second, and third step, being repeated in a normal temperature drier ($23^{\circ}C$) after 5, 9 and 24 hours. Results : The results of the experiment revealed that the sterilizing effects were 95% or over. When toothbrushes were sterilized for 60 seconds, the number of colonies is about 11 after drying for 5 hours, 7 after drying for 9 hours and 2 after drying for 24 hours. The sterilizing effects reached 98% when the bacteria-administered toothbrush was sterilized for 1 minute after drying for 24 hours. Conclusions : The results demonstrated that toothbrush sterilizing by using microwave is a suitable way to prevent cross-contamination of toothbrushes by oral bacterial infection and thus easy to use at home. However, this study suggests that toothbrush sterilizing by using microwave should be limited within two times a week because the physical properties of toothbrush might be changed.

낙동강 상·중·하 수역에서의 질화세균군의 변화 (Changes of Nitrifying Bacteria in the Different Zone (Upper·Mid·Lower Part) of the Nak-Dong River)

  • Nitrifying bacteria were detected by fluorescent in situ hybridization (FISH) method at 6 sampling sites with different eutrophication degree in the Nak-Dong River and their tributaries. And conventional physico-chemical parameters including $NH_4-N$, $NO_3-N$, and TN were determined concurrently. In rainy period (July), there was no noticeable difference between the number of ammonia/nitrite-oxidizing bacteria detected at each site except Sang-Ju and the ratio of nitrifying bacteria to total counts stained by DAPI varied in 6~33%. By contrast, in the dry period (October), both of bacterial population was increased differently and the ratio of nitrifying bacteria to total counts ranged more widely from 6% in heavily polluted water zone, Hwa-Won to 60% in upper tributary with high agricultural land use. Byung-Sung-Chun. In January, the numbers of ammonia-oxidizing bacteria was reduced up to one tenth, while those of nitrite-oxidizing bacteria was apparently increased maybe due to high DO and low DOC.

경남 북부지역 납유원유의 미생물학적 품질에 관한 연구 (A Study on the Microbiological Quality of Raw Milk in the North of Kyeongnam Area)

  • This study was carried out to obtain basic data for the Improvement of microbiological quality of raw milk. Total bacterial, psychrotrophic, thermoduric and spore counts of raw milk samples taken from milk tankers in the nothern part of kyeongnam were investigated for one year from March, 1989. The result obtained were summarized as follow 1. The number of total bacteria in raw milk averaged $4.0{\times}10^6$ CFU / ml and was not affected by seasons 2. The psychrotrophic counts of raw milk averaged $1.5{\times}10^6$ CFU / ml and it was higher in winter than in summer 3. The thermoduric counts of raw milk averaged $5.8{\times}10^4$ CFU / ml and was the lowest in winter 4. The spore counts of raw milk ranging from 3-1, 880/ ml averaged 306/ ml and was the lowest in summer

Effect of Irradiated Red Pepper Powder on Kimchi Quality during Fermentation

  • Irradiated red pepper powder (IRPP) was tested for its ability to retard fermentation and to maintain a high quality of Kimchi by the reduction of the initial microbial load. Kimchi containing IRPP at the doses of 0, 5, 10, 15, or 20 kGy was prepared. Quality indices for Kimchi in this study were pH, titratable acidity, reducing sugar content, total microbial count, lactic acid bacterial load, and sensory evaluation. Based on the pH and titratable acidity, the Kimchi with IRPP showed a retarded fermentation until 15 days. The number of the total aerobes and lactic acid bacteria of the Kimchi with IRPP were lower by about 1 log CFU/mL compared to control at day 0, however, the counts increased to 8.5 log CFU/mL after 10 days, which was similar to the control group. Kimchi that was fermented with 5 kGy IRPP was better than control and other treatments in odor and color, whereas the control scored highest in taste. Addition of IRPP showed a limited retardation of Kimchi fermentation without other quality deterioration.

낙동강 하구의 세균분포와 활성에 미치는 환경요인 (The Environmental Factors Affecting the Distribution and Activity of Bacteria in the Estuary of Naktong River)

  • From July 1985 to December 1986, 28 variables of phycal-chemical factors, bacteria and heterotrophic activity were investigated 17 times at 3 stations in the estuary of Naktong River and the influences of environmental factors to bacterial population and heterotrophic activity were analyzed through multiple regression. The results of multiple regression were as follows. At station 1, total bacteria and heterotrophic bacteria(Z-25) could explain 57% of the variation of maximum uptake velocity for glucose and 54% of turnover time for glucose was explained by total coliform bacteria and MBOD, Sixty four percent of the variation of Kt+SN was accounted for salinity, MBOD-N and inorganic phosphate. Turnover rate for acetate was also accounted for the change of MBOD-P by 56%. At station 2 maximum uptake velocity for glucose depends on MBOD-N by 81%; turnover time on bacteria by 50%; Kt+Sn on avilable nutrient by 61%. More than 50% of maximum uptake velocity and turnover time for glucose were influenced by bacteria and that of Kt+Sn by the change of nutrient in the surface water of station 3. In the bottom water of station 3, the change of maximumuptake velocity, turnover time and Kt+Sn for glucose was controlled by total bacteria and available nutrient, bacteria, the change of nutrient salts respectively. On the whole, more than 50% of maximum uptake velocity and turnover time for glucose could be due to the change in the number of bacetria and the value of Kt+Sn was affected by the change of nutrient salts. Turnover rate for acetate was controlled by available phosphate at station 1 and by bacteria at station 2 and 3, which showed a distinct difference between the environmental factors which govern the rate of glucose and acetate uptake in the Naktong esturine ecosystem. And bacterial communities were controlled by available nutrients at station 1, by nutrient salts and salinity at station 2 and in the surface water of station 3 and by salinity in the bottom water of station 3.

젓갈류의 원료에 따른 세균학적 안전성 평가 (Evaluation of Microbiological Safety of Commercially Salt-fermented Fishery Products by Raw Materials)

  • Eighty-nine different types of commercially salt-fermented fishery products comprising various raw materials were analyzed for total aerobic bacteria, number of coliform bacteria, fecal coliform, and Escherichia coli. The food-poisoning bacterial content of the samples was investigated using next-generation sequencing. The mean mass of total aerobic bacteria in Jeotgal was 6-1.8×109 CFU/g, and that in Aekjeot and Sikhae was 4-2.2×105 CFU/mL and 1.9×105-8.4×108 CFU/g, respectively. Coliform bacteria were detected in 9 (28.1%) of 32 Jeotgal samples; 15 (46.8%) of 32 seasonal Jeotgal samples; and in 5 (55.5%) of 9 Sikhae samples. Fecal coliform and E. coli were not detected in 86 of the 89 samples. Yersinia enterocolitica was detected only in Galchi jeot (salt-fermented hairtail) (1 type) and not in other Jeotgal samples. These results contribute to our knowledge regarding the bacterial stability of salt-fermented fishery products.

소양호에서 활성세균수의 계절적.수직적 변화 (The Seasonal Variation of Active Bacterial Abundance in Lake Soyang)

  • 소양호에서 총세균수와 활성세균수의 계절적.수직적 변화를 측정하였다. 총세균수는 AODC방법을, 활성세균수 측정은 전자전달계를 이용한 호흡활성도 측정인 CTC 방법을 이용하였다. 조사기간 동안 총세균수는 $2.1{\times}10^5 ~ 3.1{\times}10^6 $ cells.$ml^{-1}$이었으며 활성세균수는 $1.8{\times}10^4 ~ 8.0{\times}10^5 $ cells..$ml^{-1}$로 나타났고, 총세균수에 대한 활성세균수의 비율은 3.7~44.2%이였다. 수직 분포를 보면 총세균수는 수온약층에 따라 그 수가 변화하였으나, 활성세균의 수는 큰 변화가 없었다. 총세균수의 값과 $\beta$-glucosidase 활성도의 간이 유사한 1999년 8월과 9월의 조사결과에서 $\beta$-glucosidase specific activity를 활성세균의 값에서 구한 결과, 9월의 활성이 8월보다 약 6.7배 높았다. 이 활성세균의 측정으로 수중생태계에서 세균의 역할에 대한 새로운 정보를 얻을 수 있다.

산림 토착 미생물 군집에 미치는 유류 첨가제 노출 농도 및 시간의 영향 (Effect of Exposure Concentration and Time of Fuel Additives on the Indigenous Microbial Community in Forests)

  • The toxicity of methyl tert-butyl ether (MTBE), tert-butyl alcohol (TBA) and formaldehyde (FA) on the indigenous microbial community in forest soil was studied. MTBE, TBA and FA with different concentrations were added into microcosms containing forest soil samples. After 10 and 30 days, total viable cell number and dehydrogenase activity in the microcosms were evaluated. Bacterial communities in the microcosms were also analyzed using a denaturing gradient gel electrophoresis (DGGE). Dehydrogenase activity and total viable cell number were decreased according to the increase of MTBE, TBA and FA concentrations (P<0.05). FA toxicity was the highest, but TBA toxicity was the lowest. The results of principal component analysis using DGGE fingerprints showed that the microbial communities contaminated MTBE, TBA and FA were grouped by exposure time not exposure concentration. Dominant species in the microcosms were as follows: Photobacterium damselae sub sp. and Bacillus sp. KAR28 for MTBE; Mycobacterium sp. and Uncultured Clostridium sp. for TBA; and Uncultured Paenibacillaceae bacterium and Anxynobacillus, Flavithermus for FA.

DGGE 기법을 이용한 생물활성탄 공정의 부착 박테리아 군집분석 (Analysis of Attached Bacterial Communities of Biological Activated Carbon Process Using DGGE Method)

  • BAC 공정 운전초기부터 부착 박테리아들의 생체량이 정상상태(steady-state)에 도달한 이후까지 $BDOC_{total/rapid/slow}$ 제거율의 변화와 DGGE와 ATP 분석을 통하여 부착 박테리아들의 군집과 생체량을 평가하였다. 용존 유기물질 제거율 평가에 따른 BAC 공정의 정상상태 도달 여부 평가결과를 보면 DOC의 경우 운전 bed volume 27,500 부근에서 BAC 공정이 정상상태에 도달하였고, $BDOC_{rapid}$$BDOC_{total/slow}$의 경우는 각각 운전 bed volume 15,000 부근과 32,000 부근에서 정상상태에 도달하였다. BAC 공정의 운전기간 증가에 따른 HPC 및 ATP 농도 분석을 통한 부착 박테리아들의 생체량 평가결과 bed volume 22,500 이후로 거의 일정한 생체량을 유지하였으며, 이 때 HPC와 ATP 농도는 각각 $3.3{\times}10^8$ cells/g 및 $2.14{\mu}g/g$ 정도로 나타났다. DGGE를 이용하여 운전기간 증가에 따른 BAC 부착 박테리아들의 군집분석 결과 운전초기(bed volume 8,916)의 경우 분석가능한 DGGE band 개수가 5개였으나 운전기간 증가에 따라 분석가능한 DGGE band 개수는 최대 11개로 증가하였다. 또한, DGGE를 이용한 박테리아 군집분석 결과 BAC 운전기간의 증가에 따라 다양한 박테리아 그룹들이 존재하였고, Pseudomonas fluorescens, Acinetobacteria와 유사한 uncultured bacterium, uncultured Novosphingobium sp. 및 Flavobacterium frigidarium은 운전초기 단계부터 지속적으로 부착 박테리아 군집을 형성하였고, 전체적으로 Proteobacteria 그룹이 비교적 높은 비율로 우점하였다.
