Distribution of Pathogenic Vibrios in the Aquatic Environment Adjacent to Coastal Areas of South Korea and Analysis of the Environmental Factors Affecting Their Occurrence (2016년도 국내 해양환경내 병원성 비브리오균의 분포 및 해양환경인자간의 상관성 분석)

  • Objectives: The pathogenic Vibrios genus denotes halophilic bacteria that are distributed in aquatic environments, including both sea and freshwater. Vibrio cholerae, Vibrio vulnificus, and Vibrio parahaemolyticus are the most important species since they can be potent human pathogens and leading causes of septicemia, wound infections, and seafood borne gastroenteritis. The recent emergence of a potential pandemic clone, V. cholera serotype O1 and the cholera outbreak in South Korea in 2016 indicates the importance of consistent surveillance of pathogenic Vibrio genus within coastal areas. Methods: The present study was undertaken to determine where and how vibrios live in the aquatic environment adjacent to coastal areas of South Korea. For this survey, a total of 838 samples were obtained at 35 different sites in South Korean coastal areas during the period from January 2016 to December 2016. Pathogenic vibrios was determined using the real-time PCR method, and its clones were isolated using three selective plating media. We also monitored changes in seawater and atmospheric temperature, salinity, turbidity, and hydrogen ion concentration at the collection points. Results: The total isolation rates of V. vulnificus, V. cholera (non-pathogenic, non-O1, non-O139 serogroups), and V. parahaemolyticus from seawater specimens in 2016 were 14.2, 13.48, and 67.06%, respectively. Conclusions: The isolation rates of pathogenic vibrios genus showed a positive correlation with temperature of seawater and atmosphere but were negatively correlated with salinity and turbidity.

Effective Water Pollution Management using Reservoir Tank Automatic Classification (저수조 자동 분류를 이용한 효과적인 수질 오염 관리)

  • With the development of IT convergence technology and the construction of master plan for the four rivers restoration of the government, the importance of the eco-friendly water pollution management is being spotlighted. In this paper, we proposed the effective water pollution management using the reservoir tank automatic classification for improving the water quality and on-line managing efforts of ceo-friendly reservoir tanks. The proposed method defined the seven factors of water pollution evaluation and managed the water pollution according to hydrogen ion concentration(pH), chemical oxygen demand(COD), suspend solid(SS), dissolved oxygen(DO), count of coliform group(MPN), total phosphorus(T-P), and total nitrogen(T-N) using the sensors. We measured the values for the seven factors from the reservoir tank and normalized to ranging from 1 to 9. To evaluate the performance of the water pollution management using the reservoir tank automatic classification, we conducted F-measure so as to verify usefulness. This evaluation found that the difference of satisfaction by the traditional system was statistically meaningful.

The Compositional Differences of Sipjeondaebo-tang(Siquandabu-tang) Decoctions Extracted by Different Extraction Method and Extraction Time (전탕 방법 및 전탕 시간에 따른 십전대보탕 전탕액 비교 연구)

  • Objectives: This study was performed to compare the differences between decoctions extracted by different extraction method and extraction time. Methods: Decoctions were prepared with pressurized or non-pressurized extraction for 60, 120 and 180min. The yield of extract, total soluble solid content, hydrogen ion concentration(pH) and the content of reference compound in Sipjeondaebo-tang (Siquandabu-tang) were investigated. Results: While yields and the total soluble solid of decoction were higher in pressurized method proportional to extraction time, pH values were lower in pressurized method and showed decreasing values with increasing extraction time. Albilflorin, ferulic acid, nodakenin, coumarin, cinnamaldehyde and glycyrrhizin were contained in decoctions by pressurized extraction method more than non-pressurized method whereas the content of coumarin was higher in decoctions by non-pressurized extraction method. In addition, coumarin was extracted increasingly with extraction time in decoctions by pressurized method, however, nine compounds except cinnamaldehyde showed the tendency of increasing proportional to extraction time. Conclusions: The results show that extraction methods including pressurized or non-pressurized extraction, and extraction time could affect the physicochemical characteristic or composition of Sipjeondaebo-tang(Siquandabu-tang) decoction.

Formation and Behavior of Sedimentary Inorganic Sulfides in Banweol Intertidal Flat, Kyoung-gi Bay, West Coast of Korea (황해 경기만 반월조간대 퇴적물 내의 황화물 형성과 행동에 관한 연구)

  • This study investigated the behaviour of sulfur species after the early diegenetic reduction of sulfate from pore solution in an anoxic intertidal flat deposit in the Banweol area of Kyeong-gi Bay, west coast of Korea. A total of seven sediment cores were collected during 1990∼1992 and were analyzed for their solid-phase sulfur species (acid-volatile sulfur, element sulfur, pyrite sulfur) as well as for chemical components in the pore solution, such as sulfate, ammonium, hydrogen sulfide, phosphate and Fe ion. The pore water sulfate oncentration was found to decrease rapidly downward from the sediment surface, while that of hydrogen sulfide, ammonium and phosphate showed and increase. The dissolved iron concentration in pore water, on the other hand, was found high in the surface layer of sediment, but fell sharply below this layer. these characteristic profiles of pore water sulfide and iron concentrations suggest that some reaction occurs between dissolved iron and sulfide ions, leading to the formation of various sulfide minerals in the sedimentary phase. The amount of inorganic sulfur species in the sediment increased downward, and showed a maximum of up to 7.9 mg/g. among the three species analyzed, acid-volatile sulfur (AVS) was dominant comprising more than 50% of the total. The amount of pyrite sulfur was greater than that of element sulfur. This implies that the formation of pyrite was restricted in this environment. the limited amount of element sulfur in this deposit may have discouraged the active formation of pyrite.

Changes in pH values in the oral cavity according to the intake method of powdered probiotics (분말형 프로바이오틱스 섭취방법에 따른 구강 내 pH 변화)

  • Objectives: The purpose of this study was to investigate the changes in pH in the oral cavity using the probiotic intake method. Methods: A total of 109 participants were enrolled and randomly assigned to three groups. Participants in the control group did not ingest powdered probiotics, those in experimental group 1 ingested powdered probiotics by dissolving them on the tongue, and those in experimental group 2 dissolved powdered probiotics on the tongue and rinsed with water. pH values were measured 5 times in all. The significance of each group was examined by the Kruskal-Wallis test. The trend over time was expressed as a graph with groupwise means and confidence intervals, considering repeated measurement data. Results: A significant difference was found between the control group and experimental group 1 at two time-points, i.e., immediately after intake and 3 min after ingestion. As a result of the time trend, the pH value of experimental group 2 was smaller than that of experimental group 1, compared to the control group. Conclusions: Studies have shown that taking probiotics with water may help reduce changes in oral pH. Probiotics should be aware of live bacteria and provide consumers with more detailed information on proper dosage and precautions.

Low-Temperature Si and SiGe Epitaxial Growth by Ultrahigh Vacuum Electron Cyclotron Resonance Chemical Vapor Deposition (UHV-ECRCVD)

  • Low-temperature epitaxial growth of Si and SiGe layers of Si is one of the important processes for the fabrication of the high-speed Si-based heterostructure devices such as heterojunction bipolar transistors. Low-temperature growth ensures the abrupt compositional and doping concentration profiles for future novel devices. Especially in SiGe epitaxy, low-temperature growth is a prerequisite for two-dimensional growth mode for the growth of thin, uniform layers. UHV-ECRCVD is a new growth technique for Si and SiGe epilayers and it is possible to grow epilayers at even lower temperatures than conventional CVD's. SiH and GeH and dopant gases are dissociated by an ECR plasma in an ultrahigh vacuum growth chamber. In situ hydrogen plasma cleaning of the Si native oxide before the epitaxial growth is successfully developed in UHV-ECRCVD. Structural quality of the epilayers are examined by reflection high energy electron diffraction, transmission electron microscopy, Nomarski microscope and atomic force microscope. Device-quality Si and SiGe epilayers are successfully grown at temperatures lower than 600℃ after proper optimization of process parameters such as temperature, total pressure, partial pressures of input gases, plasma power, and substrate dc bias. Dopant incorporation and activation for B in Si and SiGe are studied by secondary ion mass spectrometry and spreading resistance profilometry. Silicon p-n homojunction diodes are fabricated from in situ doped Si layers. I-V characteristics of the diodes shows that the ideality factor is 1.2, implying that the low-temperature silicon epilayers grown by UHV-ECRCVD is truly of device-quality.

The Influence of Pressure and Time on the Preparation of Gumiganghwal-tang Decoctions

  • Objectives: This study compared Gumiganghwal-tang (GGT) decoctions produced using different pressure levels for various extraction times to determine the optimal extraction conditions through hydrogen ion concentration (pH), total soluble solids content (TSSC), extraction yield, and content of chemical compounds. Methods: Decoctions were prepared by the pressure levels of 0 or $1kgf/cm^2$ for 30-180 min. The pH and TSSC were measured, the extraction yield was calculated, and the amounts of the chemical compounds were determined using high performance liquid chromatography. Results: The higher pressure and longer extraction time decreased the pH value, while those conditions increased TSSC and extraction yield: the decoction produced in 180 min by pressurized method showed the minimum value of pH, but maximum values of TSSC and extraction yield. The chemical compounds showed higher amounts in decoctions produced by non-pressurized methods than pressurized methods and their amounts were decreased over the peak extraction time in both pressurized and non-pressurized methods. The results of regression analysis confirmed the correlative influences of the pressure and extraction time on pH, TSSC, and extraction yield. Conclusions: This study suggests that pressure and extraction time influence the compositional constituents in GGT decoctions, and the non-pressurized method for 120 min should be chosen as the optimal extraction condition for the preparation of GGT decoction.

  • Objectives : Gwakhyangjeonggi-san (GJS) which consists of 13 herbal medicines has been used to treat gastrointestinal disorders caused by common cold. This study was performed to compare GJS decoctions produced using different pressure levels for various extraction times. Methods : Decoctions were prepared by the pressure levels of $0kgf/cm^2$ (non-pressurized) or $1kgf/cm^2$ (pressurized) for 30-180 min. The extraction yield, total soluble solid content (TSSC), and hydrogen ion concentration (pH) were measured, and the contents of the nine marker compounds were determined using high performance liquid chromatography. Results : The higher pressure and longer extraction time significantly increased TSSC value, while decreased the pH value. However, only extraction time affected the extraction yield of pressurized decoction. Variation of the amounts of chemical compounds was shown in pressurized and non-pressurized decoctions during extraction time. The result of regression analysis showed that pressure and extraction time can influence to extraction yield, TSSC, pH, and the content of chemical compounds. Conclusions : This study suggests that the pressure and extraction time can significantly affect the extraction efficiency of components from GJS decoctions.

Effects of Operating Parameters on Phenol Degradation by Pulsed Corona Discharges in Aqueous Solutions (펄스 코로나 방전에 의한 페놀 분해에 미치는 운전변수의 영향)

  • Effects of operating parameters such as applied voltage, solution conductivity, ferrous ion concentration, electrode material on phenol degradation by pulsed corona discharges were investigated in laboratory scale experiments. The increase of applied voltage enhanced the phenol degradation by generating more energetic electrons. The solution conductivity inversely affected phenol removal rate in the tested ranges because the increase of conductivity decreased the electric field strength through the liquid phase. The addition of ferrous sulfate promoted the phenol degradation through the OH radical production by the Fentonlike reactions between ferrous ion and hydrogen peroxide generated by pulsed corona discharges. Catechol and hydroquinone were detected as primary intermediates of phenol degradation and the decrease of pH and the increase of conductivity were observed probably due to the generation of organic acids. Almost all of the initial phenol was disappeared and 29% of total organic corbon (TOC) was removed in the condition of 0.5 mM of ferrous sulfate after approximately 230 kJ of discharge energy transferred to the reactor.

Measurement of dental caries activity of commercial frozen desserts in Korea (국내 시판 빙과류의 치아우식 활성 위험도 측정)

  • Objectives: This study was conducted to analyze the sugar content, acidity, and viscosity of the Korean frozen desert. Methods: A total of 60 types of frozen desserts marketed in Korea were randomly selected and classified into frozen desserts, milk-fat ice cream group, and non-fat ice cream group according to the classification criteria for frozen desserts. Each product was treated under the same conditions to investigate the component of each product, and sugar content, acidity, and viscosity were measured. Frequency analysis and one-way ANOVA were performed. Results: As a result of analyzing the characteristics related to dental caries activity by group, the frozen dessert had the lowest average pH of 3.67, and the sugar content was significantly higher in the milk-fat group (33.22) and non-fat group (32.89) than in the frozen dessert. The viscosity was also the highest in the milk-fat group at 32.62, and the frozen dessert was significantly lower at 9.42. Conclusions: Due to the abnormal temperature and spread of the coronavirus, consumption of frozen desserts at home is on the rise. To prevent dental caries in children, education is needed for children and their guardians for proper oral care after eating frozen desserts.
