The antibiotic chemotherapy produces sometimes side effects and fails to eliminate bacterial infection. The occurrence of strains resistant to antibiotics would be expected to increase, and it is nowadays important to search for non-antibiotic substances. We are aimed to evaluate the effects of Torilis fructus extracts against enteropathogenic Escherichia coli (E. coli) in Piglets. The piglets were divided with three groups; Negative control group, E. coli-infected positive control group, and the Torilis fructus extracts treated group with E. coli infection. During the study period, we compared clinical signs, weight increase rate, fecal scores, gross findings between the treated group and non treated group. After necropsy, necropsy findings and histopathological findings were conducted with the comparison between the groups. As the results of this study, the Torilis fructus extracts additive showed the effects on the suppression of E. coli-induced lesions. On the basis of this study results, our data suggest that the Torilis fructus extracts additive have the antimicrobial effects. The Torilis fructus extracts additive could be used as the alternative material for antimicrobial feed additives.

Immunopotentiating activity of water extract of Lycii Fructus, Rubus Fructus, Torilis Fructus, Schizandrae Fructus and Cuscutae Semen were investigated using mouse fed with normal diet and protein deficient diet. Body weight, ratio of spleen or thymus to body weight, number of total peritoneal exudate cell, content of total serum protein and albumin content were decreased in the mouse fed with protein deficient diets as compared to the mouse fed with normal diet. Regardless of the nutritional condition, the supplement of the water extract of Lycii, Rubus Torilis, Schizandrae Fructus or Cuscutae Semen increased the ratio of spleen to body weight. The increment of this ratio was significant by the supplement of water extracts of Lycii and Torilis Fructus in mouse fed with protein deficient diet. The supplement of water extracts of Torilis Fructus to the mouse fed with protein deficient diet increased the content of total serum protein and albumin content significantly. Similar results were obtained with Lycii, Rubus, Schizandrae Fructus and Cuscutae Similar. The electrophorogram shows the difference in the concentration of some serum protein constituents between the mouse fed with normal diet and protein deficient diet.
Skin wrinkle formations are associated with collagen synthesis and matrix metalloproteinase-I(MMP-1) activity. This study was carried out to find out skin wrinkle reducing components in Torilis Fructus. Torilis Fructus were extracted with 70% ethanol and the ethanol extracts were systematically fractionated with n-hexane, ethylacetate, n-butanol and distilled water. Among them, antiwrinkle component from n-hexane fraction was purified by several column chromatographies and HPLC, which identified as torilin by $^1H-NMR,\;^{13}C-NMR$ and ESI-MS. To determine cell viability, collagen biosynthesis and MMP-1 activity, human dermal fibroblast was treated with 1-5 ppm concentrations of Torilis Fructus extract fraction and torilin. Cell viability was showed 84-102% at all group treated with 1-5 ppm. Collagen synthesis was increased in all group, especially torilin-treated group was highest amount. Active forms of MMP-1 were decreased in all group. From these results, we consider that Torilis Fructus have several antiwrinkle components and torilin may be one of the effective components.
For the propose of the quality control of Torilis Fructus, one major compound was isolated by several column chromatograpies and identified by its spectral data as torilin (8,11-dihydroxy-8-angeloyl-11-acetyl-4-guaien-3-one). A quantitative analysis of torilin using HPLC method showed that the average contents at 230 and 254 nm were $7.167{\pm}0.168mg/g\;and\;7.052{\pm}0.160mg/g$, respectively, in 18 samples collected throughout the regions of Korea and China. This method provided a tool for standardization of the commercial Torilis Fructus.
The methanol extract of Torilis Fructus showed potent anti-edematous effect in carrageenan-induced paw edema in rats. Systematic fractionation of the extract with hexane, chloroform, ethyl acetate and butanol resulted in potent anti-edematous action only in hexane fraction at an oral dose of 237 mg/kg which is corresponding to 4.8 g of raw Fructus. This hexane fraction showed inhibitory effects at 77 mg/kg p.o on vascular permeability in mice, at 8mg/pouch on leucocyte emigration in rats, and at 120 mg/kg p.o. on adjuvant arthritis model in rats. These results demonstrate that hexane fraction of the extract possesses potent anti-inflammatory effect which may support its traditional uses.
The acetone precipitates of the hot water extract of Torilis ftuctus showed strong hemostatic activity which was not inhibited by aspirin. This activity was not through platelet activation but possibly through activating some coagulation factors in plasma. The hemostatic action of the precipitates was not active by oral adminitration and no behavioral toxicity was appeared in treated mice. However, mice treated with the acetone precipitates through tail vein showed serious tremor and then were killed probably by the thrombus produced in the body. The hemostatic activity was still remained after treatment with $\beta$-glucosidase, $\beta$-alactosidase, $\alpha$-amylase, subtilisin BPN', or trypsin but completely lost by acid hydrolysis. The active components seemed to be a complex of unidentified macromolecules to which some phenolic compounds were strongly bound.
UV-irradiation is a major factor of photo-aged skin, by which pigmentation, wrinkles and laxity are increased. In addition, the epidermal barrier is disrupted, ultimately causing dryness in photo-aged skin. As an effort to search dietary sources for improving the dryness of UV irradiated skin, the dietary effect of red ginseng based functional foods on the epidermal level of ceramides, a major lipid maintaining epidermal barrier, was determined in this study. Albino hairless mice were fed either a control diet [group UV (UV-irradiated control)] or diets with 0.5% (group M0.5) or 1% (group M1.0) of red ginseng extracts mixed with Torilis fructus and Corni fructus (66.7% red ginseng) in parallel with UV irradiation for 5 wks. A normal control group (group C) was fed a control diet without UV irradiation for 5 wks. The epidermal level of ceramides in group UV was significantly lower than that in group C, in which ceramidase, an enzyme involved in ceramide degradation, was highly expressed. In group M0.5, the epidermal level of ceramide was significantly increased to the level even higher than in group C. In addition, protein expression of serine palmitoyl transferase (SPT), a key enzyme involved in de novo ceramide synthesis, was increased in group M0.5. However the epidermal levels of ceramides as well as of ceramidase protein expression in group M1.0 did not differ from those in group UV. In conclusion, we demonstrate that dietary supplementation of red-ginseng extracts mixed with Torilis fructus and Corni fructus at a level of 0.5% level in diet increased the epidermal level of ceramides coupled with the elevated expression of SPT protein.

In order to define the effect of Torilis Fructus(TF) extract which has been used for the treatment of erectile dysfunction, experiments were carried out by organ bath study, histochemical and immunohistochemical methods. First, in the organ bath study, when TF extract was administered to the maxillary contracted corpus cavernosum by PE ($10^{-6}M$), there was a significant relaxation effect on corpus cavernosum at concentration of 1, $3mg/m{\ell}$. Compared with the absence of $\text\tiny{L}$-NNA pretreatmen, pretreatment of $\text\tiny{L}$-NNA was inhibited the relaxation effect of penile corpus cavernosum. In the immunohistochemical study, the eNOS positive reaction was significantly increased, and the PDE5 positive reaction was significantly decreased due to the administration of TF extract. Therefore, it show that the TF enhances the production of eNOS and NO, inhibits PDE5 which blocks the action of increased cGMP, relaxes the corpus cavernosum. So TF relaxes the corpus cavernosum and it can be used as a safer erectile dysfunction treatment.

Objectives : The purpose of this study was to estimate the antioxidant activity of water extract of Cnidii Fructus (CF) and Torilis Fructus (TF) in Leydig cells. Methods : Free radical scavenging activity of CF and TF against 2,2-diphenyl-1-picrylhydrazyl (DPPH) was determined spectrophotometrically. We investigated the effect of CF and TF in Leydig cells by MTT assay. The protective effects of CF and TF against hydrogen peroxide-induced oxidative stress in Leydig cells. Superoxide dismutase (SOD), and catalase activity assays were performed in Leydig cells. Results : The results showed that CF scavenged DPPH radical in a dose-dependent manner by up to 81.2%, TF scavenged DPPH radical in a dose-dependent manner by up to 63.8%. CF showed cell viability as 121.0, 132.7, 126.6% in 5, 10, $100{\mu}g/ml$ concentrations. TF showed cell viability as 127.5, 111.8% in 5, $100{\mu}g/ml$ concentraions, respectively. The hydrogen peroxide-induced cytotoxicity of Leydig cells were protected to 86.3% by CF at concentration of $10{\mu}g/ml$ and protected to 83.5% by TF at concentration of $100{\mu}g/ml$. Both CF and TF at all concentrations, SOD activity was not significantly changed. Catalase activity was significantly increased at 10, $100{\mu}g/ml$ concentrations of CF, respectively. TF's catalase activity showed no significant difference from that of the control. Conclusions : These results suggest that CF, as an antioxidant, protects Leydig cells in hydrogen peroxide-induced oxidative stress. know that "Kwangjebikeup" played a role in settlement and spreading of foreign knowledge to civilians.

We reported that the hexane fraction of Torilis Fructus have an anti-inflammatory and analgesic effect. Therefore, in order to isolate the active compound, the hexan fraction of Torilis Fructus was chromatographed on silica gel column. The subfraction of hexane fraction was crystallized as colorless stout needles. The chemical structure of this compound was verified to be torilin through m.p., UV, IR, GC-MS, and NMR spectral data. In pharmacological tests, torilin exhibited strong anticarrageenan activity at the dose of 90 and 270 mg/kg, p.o. in rats, and it had inhibitory effect on the vascular permeability at the dose of 30 and 90 mg/kg, p.o. in mice. Torilin showed potent inhibition of leucocyte emigration in CMC-pouch at the dose of 3 and 9 mg/rat, s.c. Torilin have the analgesic effect at the dose of 30, 90 and 270 mg/kg, p.o. in both of the acetic acid- and phenyl-p-benzoquinone-induced writhing syndrome. It also increased the pain threshold at the dose of 30, 90 and 270 mg/kg, p.o. in the tail pressure method and the Randall-Selitto method. Torilin did not show a hypothermic action at the dose of 30 and 90 mg/kg, p.o. in mice. The acute toxicity of torilin was very weak: the LD$\_$50/ value was more than 5000 mg/kg, p.o. and 2000 mg/kg, Lp. in mice. From the above mentioned results, it was suggested that torilin had potent anti-inflammatory and analgesic activities in animals.
