We investigated the characteristics of various Tofu coagulants extracted from sea tangle using treated deep ocean water (DOW) as the solvents. The coagulants were typical solutions extracted from sea tangle using raw DOW and strongly electrolyzed acidic (SAC) or alkaline (SAK) water. The total dissolved solids (TDS), NaCl and electric conductivity were increased in the coagulant solution extracted by SAC than the others. In the coagulant solutions extracted by acidic electrolyzed solvent, aspartic acid content (13.5 mg/100 g) was higher than others (11.2 and 12.1 mg/100 g). The Tofu obtained using coagulants extracted with treated DOW had the water contents ranging from 79.55% to 80.04%. The contents of crude protein (12.1 g/100 g) and Na, Ca, K and Mg were relatively higher than general Tofu commercially available. Therefore, coagulant solutions extracted from sea tangle using treated DOW can be used to develop natural high-grade tofu for practical use.
Some quality characteristics of tofu prepared with soyprotein isolate (SPI) were investigated to study the effects of various ratio of coagulants mixture and addition of oil, sucrose and dextrin. The tofu was prepared by addition of coagulants into the boiled SPI suspension and compression. The results showed that the mixed coagulants of CaCl $_2$-GDL(50 :50) and CaSO$_4$-GDL(25 :75) resulted the highest volume yield of 4.23g/g SPI among the various ratios of CaCl$_2$-CaSO$_4$, CaCl$_2$-GDL and CaSO$_4$-GDL. The water holding capacity(WHC) expressed as the area of water absorbed on filter paper was relatively high for those tofu coagulation with CaCl$_2$-GDL(75 : 25) and CaSO$_4$ -GDL(75 : 25). Addition of sucrose or oil-sucrose (1 : 1) decreased the yield wihle WHC was improved. Their addition also caused less hard and cohesive tofu which was coagulated by CaCl$_2$-GDL(75 : 25) and CaSO$_4$-GDL(75 : 25).
The effects of water dropwort on the physical characteristics of tofu were investigated and consumer acceptance test was conducted. pH decreased significantly with increasing water dropwort concentration from 0 to 3%, whereas titratable acidity showed the reverse trend (p<0.05). The amount of water dropwort did not significantly influence the moisture content up to 2% substitution (p>0.05). Turbidity increased significantly with increasing water dropwort concentration (p<0.05). A significantly lower moisture content in 3% water dropwort tofu may be attributed to the denser structure and hardness of that was significantly higher than those of the other samples (p<0.05). Even though $a^*$-values of 3% water dropwort tofu was lower than that of control, $a^*$-values increased significantly with increasing water dropwort content (p<0.05). On the contrary, $b^*$-values decreased significantly with increasing water dropwort content (p<0.05). 2% water dropwort tofu was found to be the best with respect to its overall acceptability score.
Okinawan Awamori is produced by fermenting steamed indica rice with black mold, yeast, and water. Awamori-pressed lees is a by-product of the Awamori production process. Tofu lees is a by-product of the Tofu production process. This research consisted of two experiments conducted to elucidate whether or not dried Awamori-pressed lees and Tofu lees can be used as a mixed feed ingredient for raising female goats. In experiment 1, digestion trials were conducted to ascertain the nutritive values of dried Awamori-pressed lees and dried Tofu lees for goats. The digestible crude protein (DCP) and total digestible nutrients (TDN) contents of dried Awamori-pressed lees and Tofu lees were 22.5%, 22.5% (DCP), and 87.2%, 94.4% (TDN) respectively. In experiment 2, 18 female goats (Japanese Saanen${\times}$Nubian, three months old, body weight $15.4{\pm}0.53$ kg) were divided into three groups of six animals (control feed group (CFG), Awamori-pressed lees mixed feed group (AMFG), Tofu lees mixed feed group (TMFG)). The CFG control used feed containing 20% soybean meal as the main protein source, while the AMFG and TMFG treatments used feed mixed with 20% dried Awamori-pressed lees or dried Tofu lees. The groups were fed mixed feed (volume to provide 100 g/d increase in body weight) twice a day (10:00, 16:00). The klein grass hay and water was given ad libitum. The hay intake was measured at 08:00 and 16:00. Body weight and size measurements were taken once a month. At the end of the experiment, a blood sample was drawn from the jugular vein of each animal. The DCP and TDN intakes in AMFG and TMFG showed no significant difference to the CFG. Cumulative measurements of growth in body weight, withers height, chest depth, chest girth, and hip width over the 10 mo period in the AMFG and TMFG were similar to the CFG. By contrast, cumulative growth in body length and hip height in the AMFG and TMFG tended to be larger than the CFG. Cumulative growth in chest width in the AMFG was significantly larger (p<0.05) than the CFG. Blood parameter values were similar to those in normal goats reported by other scientists. The coats of the AMFG and TMFG animals looked shinier than the animals in the CFG. The results demonstrate that dried Awamori-pressed lees and Tofu lees can be used as a feed ingredient for the raising female goats.
김치양념과 두부를 혼합한 유산발효기법을 이용한 발효두부의 항산화 활성 및 유리아미노산의 변화를 조사하였다. 유산균을 접종하지 않은 김치양념에 침지한 두부(control), 유산균을 접종(2%, v/v)한 김치양념에 침지한 두부(TL, tofu and kimchi ingredients with lactic acid bacteria)와 유산균를 접종하여 $37^{\circ}C$에서 24시간 발효시킨 김치양념에 침지한 두부(TPL, tofu in pre-fermented kimchi ingredients with lactic acid bacteria)의 발효기간 중 total polyphenol 함량은 모든 처리구에서 증가하였고, 그중 TPL구가 가장 높았다. DPPH radical 소거능은 TPL구가 발효 12주에 84.11%로 가장 높게 나타났으며, 아질산염 소거능, SOD 유사활성 역시 TPL구가 각각 57.40%, 26.05%로 가장 높은 활성을 나타내었다. 총 polyphenol 함량, 전자공여능, 아질산염소거능, SOD 유사활성이 가장 우수한 TPL구의 isoflavone과 유리아미노산의 함량을 측정한 결과 isoflavone은 발효 전에 비해 30% 증가하여 2.34 mg/g이었다. 유리아미노산 역시 발효 전 1.51 mg/g에서 발효 14주에 TPL구의 유리아미노산 함량은 20.81 mg/g으로 발효 전에 비해 92% 증가하였다. 필수아미노산의 함량은 5.55 mg/g이었으며, 전체 아미노산의 27% 수준이었다. 필수아미노산의 함량은 lysine (1.24 mg/g)> leucine(1.20 mg/g)> valine(0.92 mg/g)> phenylalanine(0.78 mg/g)> isoleucine(0.65 mg/g)> threonine(0.50 mg/g)> methionin(0.26 mg/g) 순으로 나타났다.
분리대두단백(SPI)을 주원료로 마쇄와 여과, 압착 과정을 생략한 두부의 연속적 제조 방법을 위한 기초자료로서 분리대두단백의 보수력과 보유력, 가열시간, 단일응고제 소요량, 혼합응고제에 따른 두부의 텍스쳐 및 수율변화 그리고 관능적 특성을 조사하였다. SPI 분산액의 $100^{\circ}C$에서 가열시간이 길어짐에 따라 $CaCl_{2}$와 GDL응고제 양은 감소하다가 증가하였고, $MgCl_2$는 감소하는 경향이었으며, $CaSO_{4}$는 대조구를 제외하고 큰 차이가 없었다. 응고에 필요한 응고제의 양은 $CaSO_{4}>GDL>MgCl_{2}>CaCl_{2}>$순이었다. 압착두부 제조방법에 의한 두부의 수율은 $CaSO_{4}$로 응고한 두부로 가열시간 6분일 때 가장 높았으며, GDL로 응고한 두부의 수율은 다른 응고제에 비하여 낮았으나 6분에서 약간 높은 수율을 보였고, 이외의 응고제에 의한 두부수율은 가열시간에 큰 영향을 받지 않았다. SPI의 보수력은 $100^{\circ}C$에서 가열시간이 증가함에 따라 크게 감소하였고, 보유력은 9분까지 증가하다가 감소하였다. 또 혼합응고제$(CaSO_{4}-GDL,\;CaSO_{4}-CaCl_{2},\;CaCl_{2}-GDL)$로 응고한 압착두부의 물리적 특성은 $CaCl_{2}$와 GDL 비율이 많아짐에 따라 경두부의 특성을 나타내었다. 최고의 수율은 $CaSO_{4}$ 100% 응고두부였으며, 최소의 수율은 GDL로 응고한 두부였다. 관능검사 결과 $CaSO_{4}$ 비율이 증가함에 따라 부드럽고, 균일한 조직을 얻었으며, GDL과 $CaCl_{2}$의 비율이 많은 혼합응고제로 제조한 두부는 신맛과 쓴맛의 강도가 컸다.
두부의 기능성 및 저장성을 향상시킬 목적으로 큰느타리버섯 균사체를 이용한 발효두부를 제조하여 물과 메탄올로 추출하여 면역세포 활성에 미치는 효과를 조사하였다. 큰느타리버섯 균사체를 배양하기 위한 최적 배지는 PD broth 배지인 것을 확인하였으며, 큰타리버섯 균사체를 이용한 두부의 최적 발효기간은 7일 정도가 적당하였다. 큰느타리버섯 균사체를 이용하여 발효한 두부의 물 및 메탄올추출물은 $0.01 {\mu}g/mL$ 농도 이상에서 비장세포의 증식을 유도하였으며, 이들 추출물은 IL-6, IFN-$\gamma$ 분비를 유도하는 것으로 나타났다. 발효두부 물 추출물은 대조군에 비해 대식세포의 일산화질소 생산을 $1 {\mu}g/mL$ 농도 이상에서 유의적으로 증가시키는 것을 알 수 있었으며, 메탄올 추출물은 $10 {\mu}g/mL$ 농도 이상에서 그 생산을 증가시켰다. 발효두부 추출물들은 대식세포가 분비하는 IL-6, TNF-$\alpha$, IL-1$\beta$ 및 GM-CSF 분비량을 유의하게 증가시켰다. 따라서 큰느타리버섯 균사체로 발효한 두부는 기능성 두부로 개발이 가능하리라 생각된다.
LOX-1,2,3 결핍과 비소화성 당류인 raffinose와 stachyose 함량이 낮은 품종(LRS)인 진양콩, 하영콩 및 대복콩을 원료로 하여 제조한 두부의 품질특성을 태광콩 두부와 비교하였다. 태광콩과 하영콩의 백립중은 여타 시료에 비해 유의적으로 높았다. 조단백질 함량은 태광콩에 비해 진양콩 및 하영콩에서 유의적으로 많았다. 두부의 수율은 태광콩, 하영콩 및 대복콩으로 만든 두부가 비슷한 수준이었다. 두부의 경도는 태광콩 및 대복콩으로 만든 두부가 가장 낮았으며, 하영콩으로 만든 두부에서 유의적으로 높았다. 콩 및 두부의 이소플라본 함량은 대조구에 비해 LRS 콩은 약 1.4~1.6배, 두부는 약 1.8~3.4배 높은 함량이었다. 콩의 총 페놀 함량은 대조구에 비해 하영콩 및 대복콩에서 유의적으로 높았다. 두부의 총 페놀 및 플라보노이드 함량은 대조구에 비해 하영콩 및 대복콩으로 만든 두부에서 유의적으로 높았다. 콩의 항산화 활성은 태광콩이 LRS 콩에 비해 높은 활성이었으나, 두부에서는 하영콩 및 대복콩으모 만든 두부가 대조구에 비해 유의적으로 높았다. 따라서 LRS 콩은 두부 제조에 적합하나 특히 하영콩이 콩의 단백질, 이소플라본 함량 및 두부의 항산화 활성 측면에서 가장 우수한 것으로 판단된다.
본 연구에서는 홍국쌀에서 분리한 M. purpureus KCCM 60570을 찐 쌀에 1.0% (w/w) 접종 후 $25^{\circ}C$에서 배양하면서 홍국쌀 제조 조건 확립 및 품질 특성을 확인하였다. 또한 기존의 두부 셰이크에 홍국쌀을 첨가한 두부 셰이크의 기호도 조사를 통해 적정한 혼합비율을 결정하고, 품질 특성을 확인하였다. M. purpureus KCCM 60570으로 제조한 홍국쌀의 monacolin K와 곰팡이수를 확인한 결과 배양 8일차까지 급격히 증가하다가 이후 완만히 증가하여 홍국쌀의 적정제조온도 및 시간은 $25^{\circ}C$에서 8일로 나타났다. 기존의 두부 셰이크에 M. purpureus KCCM 6057으로 $25^{\circ}C$에서 8일간 제조한 홍국쌀을 서로 다른 농도로 첨가한 두부 셰이크의 관능검사 결과 맛, 향, 조직감 및 전체적인 기호도에서 0.5%(w/w) 홍국쌀을 첨가한 두부 셰이크의 기호도가 가장 높은 것으로 나타나 두부 셰이크의 적정 홍국쌀 혼합 비율은 0.5% (w/w)인 것으로 나타났다. 홍국쌀을 0.5% (w/w) 첨가한 두부 셰이크와 기존 시판 두부 셰이크의 품질 특성을 비교한 결과 홍국쌀 첨가 두부 셰이크의 환원당과 총당함량, 붉은 색소인 monacolin K와 색도 a 값이 월등히 높은 것으로 나타났다. 또한 기존 두부 셰이크와 0.5% (w/w) 홍국쌀을 첨가한 두부 셰이크와의 관능검사를 비교한 결과 색과 조직감이 좋은 것으로 나타나 두부 셰이크 제조에 홍국쌀을 첨가할 경우 두부 셰이크의 품질을 개선할 수 있는 가능성을 확인할 수 있었다.
매생이 분말을 첨가한 두부를 제조한 후 이화학적, 관능적 품질 특성을 평가한 결과는 다음과 같다. 매생이 두부의 수율은 대조구(매생이 가루 무첨가 두부)에 비해 높았으며, 첨가비율이 증가할수록 수율은 높게 나타났다. 매생이 두부의 무기질함량은 칼륨이 가장 많았으며, 다음으로 마그네슘으로 나타났다. 두부의 pH는 매생이 두부가 대조구에 비해 다소 낮은 값을 나타내었다. 매생이를 첨가한 두부의 색도는 대조구에 비해 L값(명도)과 a값(적색도)이 유의적으로(p<0.05) 낮게 나타났다. 매생이 두부의 texture 측정에서 두부의 견고성은 대조구에 비해 2% 첨가구가 가장 높게 나타났으며, 첨가량이 증가할수록 견고성은 감소되는 경향을 나타냈다. 부착성은 대조구에 비해 4% 매생이 첨가구가 유의적으로 높게 나타났으나 매생이 첨가군 간에는 차이가 없었다. 응집성은 2% 첨가구가 가장 높게 나타났으며, 4%, 6% 첨가구는 응집성이 대조구와 2% 첨가구에 비해 유의적으로 감소하는 것으로 나타났다. 탄성, 검성 및 부서짐성은 2% 첨가구가 다른 군에 비해 유의적으로 높게 나타났다. 두부의 미생물 측정에서 저장 12일까지의 총균수는 매생이 가루가 첨가된 두부가 대조구에 비해 미생물 증식을 지연시키는 결과를 나타냈으며, 특히 2%, 6% 첨가군에 비해 4% 첨가군에서 지연효과가 가장 우수한 것으로 나타났다. 관능평가 결과에서 전반적으로 4% 첨가구가 좋은 결과를 나타내 매생이 두부 제품으로서의 가능성을 알 수 있었다.
