Factors Affecting the Healing of Radial Fractures Using Acrylic Pin External Fixation in Small Breed Dogs

  • Healing of 48 cases of radial fractures in small breed dogs treated with acrylic pin external fixation was evaluated retrospectively to investigate the factors affecting the healing of radial fractures. The mean age of the subjects was 15.02 months, the mean body weight was 3.48 kg, and the mean maximum length of the radius (MLR) was 90.77 mm. External skeletal fixation frames type 1a, 2a, and 3 were used; among them type 2a was used most frequently (75%). The mean time to clinical union (TCU) was 67.17 days, and the success rate was 95.8%. Age, bodyweight, MLR, and occurrence of complications increased TCU significantly. Among complications, pin loosening, swelling of the operated forelimb tissue for more than 2 weeks, and coexistence of two or more complications increased TCU significantly (p < 0.05). There was no relationship between location of the fracture and TCU. The results of this study may be useful to predict the prognosis of radial fractures in small dogs.

Relationship between Condition Index Values and Expression Levels of Gene and Protein in the Adductor Muscle of Diploid and Triploid Oysters Crassostrea gigas

  • Three proteins [myosin heavy chain (MHC), filamin-C fragment (FIL-C), and actin 2 (ACT2)] were identified in adductor muscle from diploid and triploid Pacific oysters (Crassostrea gigas) and the relationship between the condition index (CI) and mRNA expression of these genes was investigated, together with the mRNA expression of molluscan insulin-related peptide (MIP), C. gigas insulin receptor-related receptor (CIR), and insulin-like growth factor binding protein complex acid labile subunit (IGFBP-ALS). Monthly changes in the CI were similar to the changes in the tissue weight rate in both groups. ACT2 and MHC mRNA expression was statistically higher in the triploid than the diploid, while FIL-C mRNA expression was significantly higher in the diploid (p<0.05). The MIP, CIR, and IGFBP-ALS mRNA expression of the diploid oysters were all significantly higher in July than in other months (p<0.05). The MIP, CIR, and IGFBP-ALS mRNA expression in the triploid oysters was high in July, but there were no significant differences (p>0.05). Changes in the expression levels of the genes investigated in this study could be used as intrinsic indicators of the annual growth, maturity, and spawning period of cultured diploid and triploid C. gigas in Tongyeong, Korea.

Effects of Tea Fungus/Kombucha Beverage on Growth, Serum Protein Level and Enzyme Activities in Male Rats (발효홍차가 흰쥐의 성장률, 혈청 단백질농도 및 효소활성에 미치는 영향)

  • The current study was undertaken to determine the prolinged effects of fasting-refecding on the lipoprotein lipase (LPL) activity and lipogenesis of adipose tissus in male Sprague-Dawley rals and to investigate the effects of various degrecs of food restriction during refeeding on the LPL activity and lipogenesis. The control group (n=5) was fed ad libitum and killed in the fed state at the beginning of the experiment. All rats except conteol group were fasted for 2 days (n=50). Five rats were killed at the end of fasting and others (n=45) were refed either ad libitum (ad libitum group) or mildly restricted (20% food restricted group), or esverely restricted diet (40% food restricted group). Rats were killed on the day of 7th, 14th, and 21st of refeeding. Lipogenesis was determined by the amount of glucose converted to the total lipid. Body weight and epididymal adipose tissue weight returned to control states by 5 days in ad libitym group and by 14 days in 20% food restricted group. As expected, in 40% food restriction during refeeding weight and epididymal adipose tissue weight did not rcturn to control states until day 21. On day 21 after refeeding, the serum total cholesterol concentration of ad libitum group was significantly (p<0.05) higher than that of control group. The serum HDL-C concentration of 40% food restricted group during refeeding was significantly (p<0.05) higher than that of control group. However, there were no significant dif-ferences in serum HDL-C/total cholesterol (TC) ratio and trigluceride concentration among the groups. Fasting for 2 days decreased lipogenesis and LPL activity (p<0.05). On day 21 after refecding, the lipogencsis of ad libitum group was significantly (p<0.05) lower than that of control group. The lipogenesis of 40% food restricted group during refeeding was significantly (p<0.05) higher than that of conrtol group. Ad libitum group and 20% food restricted group during refecding allowed heparin-releasable (HR) LPL or total extractable (TE) LPL activity to return to control states. 40% food restricted group during refeeding delayed the return of HR-LOL or TE-LPL activity to return to control states until day 21 of refeeding. These results suggest that food restriction during refeeding can partially or completely prevent the overshoot of LPL activity, and this may influence the rate of lipid accumulation in adipose tissue during refeeding.

The Inhibitory Effect of L. plantarum Q180 on Adipocyte Differentiation in 3T3-L1 and Reduction of Adipocyte Size in Mice Fed High-fat Diet

  • In this study, we examined the inhibitory effect of L. plantarum Q180 on adipocyte differentiation in 3T3-L1 and reduction of adipocyte size in mice fed high-fat diet. L. plantarum Q180 inhibited the adipocyte differentiation of 3T3-L1 cells ($18.47{\pm}0.32%$) at a concentration of $400{\mu}g/mL$ ($10^8CFU/g$). As a result of western blot analysis, the expression of $C/EBP{\alpha}$ and $PPAR{\gamma}$ in 3T3-L1 adipocyte treated with $400{\mu}g/mL$ of L. plantarum Q180 decreased 35.16% and 40.07%, respectively, compared with the control. To examine the effects, mice were fed three different diets as follows: ND (n=6) was fed ND and orally administered saline solution; HFD (n=6), HFD and orally administered saline solution; and HFD+Q180 (n=6), HFD and orally administered L. plantarum Q180. After six weeks, the rate of increase of body weight was 13.7% lower in the HFD+Q180 group compared to the HFD group. In addition, the epididymal fat weights of the HFD+Q180 group were lower than that of the HFD group. The change of adipocyte size was measured in diet-induced obese mice. Consequently, the number of large-size adipose tissue was less distributed in the ND and HFD+Q180 groups than in the HFD group. L. plantarum Q180 has an effect on the inhibition of 3T3-L1 adipocyte differentiation, fat absorption and reduction of adipocyte size. L. plantarum Q180 could be applied to functional food products that help improve obesity.

Effect of Exogenous Administration of Tamoxifen on Hormonal Profile and Sexual Maturity in Indian Native Kadaknath Fowl

  • The effect of exogenous administration of tamoxifen (TAM) on hormonal profile and sexual maturity in Indian native Kadaknath (KN) fowl was investigated. Day-old chicks from the same hatch were randomly divided into 15 groups with 20 chicks in each group (5 treatments${\times}$3 replicates). The chicks were placed in battery brooders with wire-mesh floors and reared under uniform husbandry conditions (14 h light/d, 25-32${^{\circ}C}$) on a standard basal diet. At the age of two weeks (wk), birds from the control group ($T_{1}$) were injected with maize oil intramuscularly (I/M), whereas the other four experimental groups $T_{2}$, $T_{3}$, $T_{4}$ and $T_{5}$ were given tamoxifen (I/M) dissolved in maize oil at the rate of 0.5 mg (0.5 TAM), 1.0 mg (1.0 TAM), 2.5 mg (2.5 TAM) and 5.0 mg (5.0 TAM)/kg body weight, respectively, up to 30 wks on every alternate day. At every 6-wk interval, blood samples were collected from nine birds of each treatment group for estimation of estrogen and progesterone. The same birds were sacrificed for determination of the weight of ovary, oviduct, liver and adipose tissue. There was no significant difference in egg production traits except onset of lay and egg number. Low doses of TAM ($T_{3}$) advanced the onset of egg laying by 15 days over the control. Tamoxifen influenced the hormonal profile (estrogen and progesterone) in a dose dependent manner. However, higher doses of TAM suppressed ovary and oviductal growth. From this study, it may be concluded that lower doses of TAM enhanced sexual maturity while higher doses suppressed ovary and oviductal growth.

The Effects of a Mineral Supplement (Aquamin $F^{(R)}$) and Its Combination with Multi-Species Lactic Acid Bacteria (LAB) on Bone Accretion in an Ovariectomized Rat Model

  • Although an adequete intake of calcium (Ca) is recommended for the treatment and prevention of osteoporosis, the intake of Ca should be restricted because of its low rate of intestinal absorption. The purpose of this experiment was to identity the effect of the combined administration of Aquamin F (AQF) (a calcium agent) and lactic acid bacteria (LAB) on osteoporosis. Thirty ovariectomized (OVX) rats and six control rats were assigned to the following six groups, with six animals per group: sham Ca-deficient diet (Ca-D), OVX, LAB, AQF, and LAB-AQF. During the experiment, the body weight was measured; and after the experiment was completed, the serum biochemical analysis, the alkaline phosphatase, calcium, and inorganic phosphorus leves were measured. The tissue of the femur was stained and then scanned via CT. The body weight of the OVX group increased more significantly than that of the control group. The results of the bone mineral content (BMC), Bone mineral density (BMD), serum biochemical analysis and histological test on the femur epiphysis showed no difference between the OVX group and the LAB group, whereas the results of the AQF group were more significant than those of the OVX group. In particular, the LAB+AQF group showed more significant increases in the aforementioned results than the AQF group. This experiment showed that the combined administration of AQF and LAB in ovariectomized rats more significantly increased bone density than did a single administration of either AQF or LAB.

A Study on the Subchronic Inhalation Toxicity of 2-Butanethiol Using SD Rats (2-부탄티올의 SD Rats를 이용한 아만성 흡입독성연구)

  • With the 2-Butanethiol, which is an unidentified inhalation toxic material, acute inhalation toxicity was tested with SD rats. The $LC_{50}$ was evaluated to be 2,500 ppm (9.22 mg/L) or higher which falls under the criteria of acute toxicity Category 3 (500<$LC_{50}$<2,500 ppm) in the Industrial Safety and Health Act. In the subchronical inhalation toxicity test by 0, 25, 100, and 400 ppm, 6 hours a day, 5 days a week, for 13 weeks repeated exposure, though no death or particular clinical presentation was observed, in the female 25 and 400 ppm group, including weight change, and in each concentration group including 400 ppm, change of feed rate, eye stimulation, motility change in male group, and lesions in blood and blood biochemical were observed. In the internal organs weight, 25, 100, and 400 ppm groups in male and 400 ppm group in female showed significant (p<0.05) changes in kidney, liver, thymus, and lung. In the pathological tissue test, severe cortical tubular hyaline droplets were observed in the male 400 ppm group, and all male rats of 400 ppm group and 2 female individuals showed tubular degeneration/regeneration accompanied with pigmentation, showing that the target organs of inhalation exposure of 2-Butanethiol are spleen, kidney, nasal cavity, and adrenal. Through the tests, the NOEL of 2-Butanethiol was evaluated to be 25 ppm (0.092 mg/L) or less for both male and female.

Effects of Wekyungtang and Kami-Wekyungtang on Pulmonary Tumor Cells and the Changes of Tissues (위경탕(葦莖湯).가미위경탕(加味葦莖湯)의 B16-Fo에 대(對)한 항종양(抗腫瘍) 효과(效果)와 조직(組織) 변화(變化))

  • In order to prove the antitumor effect of Wekyungtang(WKT) that was originated in Bigeubchunkeumyobang(備急千金要方), Wekyungtang with Houttuyniae Herba(WKT-I) and Wekyungtang with Oldenlandiae diffusae Herba(WKT-II) experimentally, the studies were done, We evaluated the cytotoxic activity against B16- Fo as well as the synergistic effects with anticancer drugs such as cyclophophamide (CPM), cisplatin(CPT) and 5-fluorouracil (5-FU) in vitro and measured body weight, survival time, hematological changes, changes of tissues in G57BL/6 implanted with B16-Fo. The results were obtained as follows: 1. In vitro cytotoxic effect against B16-Fo was shown in all groups as compared with control group, but the concentrations showing inhibitory growth rate below 55% of control was recognized in all concentrations of Wekyungtang(WKT) against B16-Fo and also concentration of $10^4$g/ml above in all group with cyclophophamide (CPM), concentration of $10^3$g/ml in Wekyungtang(WKT-l) with cisplatin(CPT) in synergistic effect, 2. In vivo body and tumor weight were significantly suppressed in all groups as compared with control group 3. The number of platelet, WBC, RBC were significantly increased in all groups, platelet aggregation was significantly increased in WKT-I and WKT-II as compared with control group. 4. In changes of tissues heavy infiltration oh cancer was shown in portal vein, pulmonary tissue, vein, peribronchiole, aveoli, while WKT-I was effective in antihepatic metastasis and WKT-II in pulmonary matastasis. From above results it was concluded that wekyungtang(WKT), wekyungtang with Houttuyniae Herba(WKT-I) and wekyungtang with Oldenlandiae diffusae Herba(WKT -II) had antitumor effect, and also wekyungtang combined with Houttuyniae Herba or Oldenlandiae diffusae Herba were more effective than wekyungtang only and also cyclophophamide (CPM), cisplatin(CPT) showed the more synergistic effect which suggests the necessity of continuous study on the mechanism of antitumor action of Houttuyniae Herba or Oldenlandiae diffusae Herba.

Effects of Dietary Xylooligosaccharides on Hepatic HMG-CoA Reductase Activity and Morphological Exchange of liver in Rats Fed High Fat Diets (고지방 식이를 섭취한 흰쥐에 있어서 Xylooligo당이 간의 HMG-CoA Reductase 활성 및 간조직의 형태학적 변화에 미치는 영향)

  • This study was conducted to examine the effects of dietary xylooligosaccharides on hepatic HMG-CoA reductase activity and morphological exchange of liver in rats fed high fat diet. Sprague-Dawley male rats weighing 100 $\pm$ 10 g were randomly divided into four groups, two normal diets and two high fat diets containing 1% cholesterol and 10% lard. Two normal diets were classified into a basal diet (normal group) and 10% xylooligosaccharide diet (NX group). The high fat diet groups were classified into a HF group without xylooligosaccharides diet and HFX group supplemented 10% xylooligosacchride diet. Experimental diets were fed ad libidum to the rats for 4 weeks and then they were sacrificed. The body weight of high fat diet (HF group) was increased more than that of normal group, but it was significantly decreased by xylooligosacchrides supplementation. The food intake was not significantly different among the all groups. The weight of liver, small intestine and cecum of all xylooligosaccharide supplemented groups were significantly heavier than those of normal and HF groups. The activity of hepatic HMG-CoA reductase, a rate limiting enzyme of cholesterol biosynthesis, in xylooligosaccharide supplemented groups was higher than that of HF group. Light micrographs revealed that the structures of hepatocytes in xylooligosaccharide supplemented groups were preserved well, compared to HF group. The xylooligosaccharide supplementation exerted a lipid-lowering action by decreasing cholesterol and triglycerides contents in hepatic tissue. In conclusion, the activity of hepatic HMG-CoA reductase and damage of liver in rats fed high fat diets were improved by dietary xylooligosaccharides.

The Inhibitory Effect of Fermented Kalopanax pictus by Bioconversion on Endotoxemia and the Competitive Inhibitor Activity on LPS (생물전환을 통한 음나무발효물의 LPS에 대한 경쟁적 억제제 효과 및 내독소혈증 억제 효과)

  • The objective of this study was to evaluate the effect of fermented Kalopanax pictus (KP-F) on macrophage activation and its effect as a competitive inhibitor of LPS and inhibitory effect on endotoxemia. The results showed that KP-F could activate macrophage in a dose-dependent manner, and KP-F was confirmed to act as a ligand for TLR4. Also, it was found that KP-F did not exhibit the same biotoxicity as LPS in intraperitoneal injection, and that it could suppress the neutrophil migration induced by LPS administration. In normal mice, the body weight, tissue weight, and amount of nitrite and pro-inflammatory cytokines in serum showed no significant changes with KP-F diet for 2 weeks, confirming that administration of KP-F in normal mice did not lead to over activation of immune response and biotoxicity. In the mouse model of endotoxemia induced by LPS and D-galactosamine(D-GalN) in sub-lethal dose, the diet of KP-F effectively inhibited the amount of nitrite and cytokines in the blood, and thus was found to be able to relieve the hepatic and kidney injury. In addition, in the endotoxemia mouse model induced by LPS and D-GalN of lethal dose, the survival rate was increased by KP-F diet in a dose-dependent manner.
