The study was attempted to observe effects of voluntary exercise on energy expenditure and on storage of excessive energy into body fat in adult mice. Mice, body weight 25-30g, were divided into two groups, exercise and sedentary group. Voluntary exercize by running wheel was allowed for former group but not for the mice of the latter group which were restricted by limiting space allowance 13.5$\times$11.5$\times$15.0cm per mouse. During a period of 4 weeks of feeding trial, they were fed ad libitum starch-casein based diet added with $^3\textrm{H}$-glucose (D-[1-$^3\textrm{H}$(N)]-glucose) at a level of 20 nCi per g of diet. Measurements were made to study hematology, lipase activity in epididymal adipose tissue, total contents of body waste and fat, and radiactivity of $^3\textrm{H}$-glucose incoporated into body fat. Dietary intake, body weight gain and amount of voluntary physical activities were also measured. The results obtained ard summarized as follows; 1) Amount of metabolizable energy intake, body weight gain and body fat were not statistically different between the two group. However, mice of the exercise group tended to show lower body weight gain body fat contants, but higher energy expenditure than those of the sedentary group. 2) Radioactivity of $^3\textrm{H}$-glucose incoporated into body fat appeared lower for exercise group expressd on a whole body fat basis. The activity was, however, higher for exercise group when expressed based on per g of fat compared to that of sedentary group. 3) Exercise group showed also higher activity of lipase in epididymal adipose tissue than the other group. 4) Mice whose physical activities were restricted appeared to have lower levels of hemoglobin and hematocrit values than of the exercise group. These results seem to support the theory that turn-over rate of body fat is activated by exercise and to suggest that consumed energy is to be converted primarily into body fat before its use as energy source by oxidation even during a period of continuous energy expenditure by exercise.

This study aimed to evaluate the inhibitory effect of micro-current stimulation(MCS) on adipogenesis regarding with Wnt/β-catenin pathway using the ob/ob mouse and 3T3-L1 cell line. 6-week old ob/ob male mice were equally assigned to four groups: obese group(ob), obese with MCS groups(50 μA, 200 μA, and 400 μA). 6-week old C57BL/6J male mice were assigned to the control group(CON). We analyzed abdominal adipose tissue volume by using in vivo micro-CT and measured the body weight, feed intake, liver weight and triglycerides in serum. All the MCS groups showed that significantly reduced body weight and triglycerides in serum. In the case of liver weight and abdominal adipose tissue volume, the inhibitory effect of adipogenesis was shown in the 200 μA and 400 μA groups. To elucidate the anti-obesity effect of MCS, β-catenin, C/EBPα and FAS protein expressions were analyzed by western blotting. β-catenin expression was upregulated, C/EBPα and FAS expression were down-regulated in the relatively high-intensity groups(200 μA and 400 μA). Thus, the 200 μA and 400 μA for the intensity of MCS were chosen for cell experiments. In the 3T3-L1 cell line, Wnt/β-catenin pathway including Wnt10b, Wnt3a, β-catenin and Cyclin D1 was activated in all MCS groups. Accordingly, the expression level of C/EBPα was decreased during the differentiation and lipid droplet was significantly reduced in Oil red O staining results. These results suggest that the Wnt/β-catenin signaling might be activated by MCS with current intensities between 200-400 μA and it may lead to anti-obesity effects.
Objectives : The researcher investigated the anti-obesity effect of Alismatis Orientale Rhizoma(AP) water extract in mice fed a high fat diet and focused on the analysis of local area adipose tissue. Methods : Male ICR mice were divided into three groups, which were fed either a normal AIN diet, a 45% high fat diet (CT group), or a high fat diet and orally administration with a concentrations of 100 mg/kg (AP100 group) and 300 mg/kg body weight (AP300 group) for eight weeks. Results : As compared with CT group, AP100 group showed significant reductions in absolute weight of liver. As compared with CT group, AP100 group and AP300 group showed significant reductions in weight gain and relative weight of total fat. AST, triglyceride, total-cholesterol levels from the AP 100 group and 300 group were significantly lower than those of the CT, and ALT, LDL-cholesterol levels from the AP 100 group was significantly lower than those of the CT. But serum HDL-cholesterol levels from the AP 100 group and 300 group were significantly higher than those of the CT. And serum adiponectin levels from the AP 100 group was significantly higher than those of the CT. In result of real time PCR, all mRNA expression(PEPCK, G6Pase, PGC-$1{\alpha}$ and $ERR{\gamma}$) of two experimental groups were significantly decreased compared to those of CT group. The treatment with AP on local abdominal area made a fat cell size lessen on the fat tissue in the abdominal cavity and subcutaneous area. Conclusions : These results suggest that AP has an anti-obesity effect and the effect is mediated by inhibition of fat gain.
[Purpose] The purpose of this study was to determine whether different types of carbohydrate diets with or without exercise changes energy metabolism at rest and during exercise. [Methods] To minimize differences in food and energy intake between experimental groups, mice were pairfed. After 1 week of adaptation, 40 male ICR mice (6 weeks old) were randomly divided into four groups: Sta. (high fat + high starch), Scu. (high fat + high sucrose), StaEX. (high fat + high starch + exercise), and SucEX. (high fat + high sucrose + exercise). StaEX. and ScuEX. groups underwent training by running on a treadmill five times a week. After 10 weeks of training, energy metabolism was measured for 24 h and during a 1 h exercise period. [Results] The final body weight showed no significant difference between the groups. However, the weight of abdominal tissues (epididymal, perirenal, and mesenteric adipose tissue) in training groups was markedly decreased following 10 weeks of training. Results of all energy metabolism (24 h at rest and during 1 h of exercise) showed no significant interactions between diet and exercise. A brief summary of the results of the energy metabolism is that the metabolism related indicators over 24 h were more affected by the dietary pattern than the exercise but during the 1 h of exercise, training had more effect on energy metabolism than diet. [Conclusion] Our findings confirm that: (a) the type of carbohydrates included in the diet influence the metabolic responses over 24 h, (b) training had more effect on energy metabolism than diet during 1 h of exercise, (c) both results; abdominal adipose tissue weight and fat oxidation during exercise are suggestive for a beneficial effect of moderate physical activity on weight maintenance.
Purpose: The carbohydrate-restricted diet has been recognized to be effective into preventing and alleviating lifestyle-related diseases, such as obesity and type 2 diabetes. The rare sugar ᴅ-allulose is a functional monosaccharide with anti-obesity effects. In the present study, we examined the effects of dietary ᴅ-allulose on body fat accumulation in rats fed severely carbohydrate-restricted diets containing high concentrations of different fats, beef tallow, or soybean oil. Methods: Male Wistar rats (n = 35, 3-week-old) were divided into 5 groups: One chow-fed control (C) group, and four carbohydrate-restricted groups, namely, beef tallow (B), beef tallow + ᴅ-allulose (BA), soybean oil (S), and soybean oil + ᴅ-allulose (SA), with free access to the diet and water for 8 weeks. The B and BA diets contained 23% beef tallow and 2% soybean oil, whereas the S and SA diets contained 25% soybean oil. Furthermore, the BA and SA diets contained 5% ᴅ-allulose. Results: The final body weight, weight gain, and food intake were significantly higher, and food efficiency was significantly lower in the control group compared to the other carbohydrate-restricted groups. Intra-abdominal adipose tissue, carcass fat, and total body fat weights were not influenced by dietary fat type or ᴅ-allulose supplementation, except for the epididymal adipose tissue weight. In contrast, carbohydrate restriction suppressed body weight gain in rats, but remarkably increased body fat accumulation. Conclusion: Under carbohydrate-restricted conditions, no anti-obesity effects of dietary ᴅ-allulose were observed, regardless of the dietary fat type. The causes of these effects are unknown. However, they may be influenced by a very low carbohydrate and high protein diet. Further research is required to elucidate the effects of ᴅ-allulose under various nutrient compositions with different fat, carbohydrate, and protein energy ratios.
This study investigated whether increased adiposity is prevented by estrogen replacement in female ovariectomized (OVX) C57BL/6J mice, an animal model of human menopause and whether these metabolic changes reflect the inhibitory action of estrogen on peroxisome proliferator-activated receptor $\gamma$ ($PPAR{\gamma}$)-regulated gene expression. Treatment of $17{\beta}$-estradiol for the last one week of the experiment decreased high fat diet-induced body weight gain and white adipose tissue mass compared to OVX control mice. Histological analysis showed that administration of $17{\beta}$-estradiol to mice decreased the size of adipocytes in parametrial adipose tissue versus OVX control mice. In addition, $17{\beta}$-estradiol reduced the adipose expression of $PPAR{\gamma}$ as well as $PPAR{\gamma}$ target genes such as adipocyte fatty acid binding protein and tumor necrosis factor $\alpha$. These results suggest that $17{\beta}$-estradiol may inhibit adiposity through reducing the $PPAR{\gamma}$ activities in female OVX mice.
A rapid and economical method of simultaneous extraction of DNA and RNA from seaweeds has been developed by the use of lithium chloride. Lithium chloride facilitates the softening of cell walls resulting in a decrease in both compressive and tensile modulus of elasticity. The DNA was characterized by high molecular weight larger than 27 kb and a relative lack of carbohydrate and protein contamination. The DNA and RNA extracted by the method from many seaweeds were of sufficient quality to be used as a template for per amplification with a plant intergenic gene primer set, for RAPD analysis with arbitrary primers, and for differential display with arbitrary primers in the morphologically distinct regions of the matured Porphyra thallus. The cDNA polymorphism indicated that the reproductive tissue types (male, female, patch) had a relatively high degree of similarity; the vegetative tissue types (dividing, non-dividing) also showed a similar pattern with respect to each other. Holdfast tissue had very low similarity with the other tissues, but appeared most similar to vegetative non-dividing tissue type.
The fatty acid composition of a rapeseed oil being on the market was analyzed and the effect on gain of the body weight and lipid levels in serum and liver tissue of male rats of Sprague-Dawely strain fed the diet containing the rapeseed oil were studied. The fatty acid components of marketed rapeseed oil was oleic acid 29.4%, erucic acid 26.52%, linoleic acid 20.39% and linolenic acid 8.68%. The contents of total lipid in serum W3S Significantly higher in RSO20 group than Contr01 group(P< 0.01) . But that in the liver tissue did 001 show significant differences. The contents of triglyceride in serum was control group 84.14mg/dll, RSO15 group 100.33mg 141 and RSO20 group 122.00mg 141 and showed significant difference between each group, but that in the livertissue did not show significant differences. The contents of phospholipid in serum did not show significant differences. But that in the liver tissue showed significant difference between the control group 8.42mg /g and Rs02o group 7.34mg /g(p<0.001). The contents of total-cholesterol and free-cholesterol in serum and liver tissue of the RSO20 group showed the highest levels compared with control group, but there did not show significant differences. The contents of ester-cholesterol in serum showed significant.
In the present study, we investigated temporal variation in the reproductive effort and biochemical contents in adult Manila clam Ruditapes philippinarum at Shi-Heung-Ri beach on the east coast of Jeju Island. Gonad-somatic index (GSI), a ratio of the egg mass to the total biomass determined using ELISA increased dramatically from late May to June (7.1% to 19.5%). In mid-July, GSI dropped to 15.1%, indicating spawning on a small scale during this period. GSI also declined dramatically from late August (15.5%) to September (4.3%), suggesting that massive spawning occurred during this period. A positive correlation was observed between clam size and potential fecundity, while potential fecundity ranged between 1.19-8.40 million eggs/clam. Total proteins and lipids in the tissue increased from late spring to mid-summer, coinciding with an increase in GSI. Protein, lipid and carbohydrate contents in the tissue were lowest in January, indicating that clams were suffering from poor nutritional circumstances, which may be associated with poor food supply from the environment. Monthly increase and/or decrease in the body weight were closely related to gonad maturation and subsequent spawning, which was also linked to temporal changes in the contents of protein, lipid and carbohydrates in the tissue.

This study was observed that liver, kidney, and stomach were altered in mouse by the effect of chronic alcohol administration. The mouse were sacrificed to obtain the tissue after mouse were orally injected with 25 % ethanol $18m{\ell}/kg/day$ for 120days. The tissue were stained by hematoxylin and eosin and then observed by light microscope. The results of this study were as follows : 1. The congestion was appeared in liver after 120days alcohol admistration. 2. The destruction of glomerulus were increased and the parietal cell of Bowman's capsule were swelled such as cuboidal cell after 120days alcohol administration. The congestion was appeared in alcohol administrated group. 3. The mucosa and gastric pit were destructed and the ulceration was appeared in stomach after 120days administration. The parietal cells and chief cells were damaged. Above results were shown that the tissue were damaged by chronic alcohol administration.
