Effects of Onion (Allium cepa) Skin Extract on Pancreatic Lipase and Body Weight-related Parameters

  • The aim of the present study was to assess the effects of onion (Allium cepa) skin extract (OSE) on pancreatic lipase (PL), the key enzyme of the digestion and absorption of dietary fat in the small intestine, and to evaluate its potential for the inhibition of body-weight gain. OSE inhibited PL with an $IC_{50}$ of 53.70 mg/mL, which means as potent as 0.07635% of the activity of orlistat. At 3 and 4 hr after administration of OSE, the plasma triacylglycerol concentration was significantly lower in the OSE-treated rats than control. Body-weight gain and parametrial adipose tissue weights were significantly lower in mice fed the high-fat diet (fat comprises 31% of total calories) with 5%(w/w) OSE than in control. The results suggest that OSE may be an effective nutraceutical for the inhibition of body-weight gain.

Reconstruction for the Soft Tissue Defect of Heel using Free Lateral Arm Neurosensory Flap (유리 외측 상박 감각신경 피판술을 이용한 종부 연부조직 결손의 재건)

  • Soft tissue defect on heel area of the foot present difficult problems particularly because of anatomic property of plantar surface of the foot. There is a paucity of available local tissue in the foot for coverage. In addition to having little expandable tissue, the foot's plantar surface has a unique structure, making its replacement especially challenging. Plantar skin is attached to the underlying bone by fibrous septa, preventing shear of the soft-tissue surfaces from the underlying skeleton. Plantar surface of foot is in constant contact with the environment. Protective sensibility also would be maintained or restored in the ideal reconstruction. So the ideal flap for reconstruction of the heel should include thin, durable hairless skin with potential for reinnervation. The aim of this article is to present a clinical experience of free lateral arm neurosensory flap for reconstruction of the heel. From March 1995 to December 1997, a total 16 lateral arm free flaps were performed to soft tissue defects on the weight-bearing area of the hindfoot. we used tibial nerve as recepient nerve in 11 and calcaneal branch of tibial nerve in 5 for restoration of sensibility of flap. All cases survived completely. A static two-point discrimination of 14 to 34mm was detected in the flap. Radial nerve palsy which was caused by hematoma in donor site occured in one case, but recorverd in 3 weeks later completely. In conclusion, the lateral arm free flaps are versatile, reliable and sensible cutaneous flap and especially indicated for soft tissue defect on plantar surface of the hindfoot which are not good indications for other better-known flaps.

The effects of genistein and exercise on obesity and adipose tissue inflammation in ovariectomized mice (난소절제 쥐에서 비만과 지방조직 염증에 대한 제니스테인과 운동의 영향)

  • This study investigated the effects of exercise on genistein-induced adipose tissue reduction and anti-inflammation of adipose tissue in ovariectomized mice fed a high-fat diet. Compared to mice fed a high-fat diet (C), both genistein (G) or exercise only (Ex) not only decreased adipose tissue weight and adipocyte size, but also glucose and free fatty acid levels in serum, which were more effectively reduced when genistein and exercise were treated simultaneously (G/Ex). The expression of lipogenesis and inflammatory cytokine genes of both G and Ex in adipose tissue was decreased compared to C. In particular, the decrease of these genes expression in G/Ex was more effective than each treatment alone. Thus, this study revealed that simultaneous treatment of genistein and exercise resulted in more effective improvement of obesity and adipose tissue inflammation than genistein single treatment in ovariectomized mice fed a high-fat diet, and it is a result of positive regulation of lipogenesis genes. This study suggests that the exercise has a beneficial effect on anti-obesity and adipose tissue anti-inflammation of genistein.

Impact of Pre-planting NO3:NH4 Ratios in Root Media on the Growth of Tomato Plug Seedlings (혼합상토에 기비로 혼합된 NO3:NH4 비율이 토마토 플러그 묘 생장에 미치는 영향)

  • This research was conducted to investigate the influence of various pre-planting $NO_3:NH_4$ ratios in the coir dust+peatmoss+perlite (3.5:3.5:3, v/v/v) medium on the growth of tomato plug seedlings (cv. Dotaerang Dia), changes in chemical properties of root media, and tissue nutrient contents. With the fixation of N concentration to $300mg{\cdot}L^{-1}$, the $NO_3:NH_4$ ratios were adjusted to 100:0, 73:27, 50:50, 27:73, 0:100. Then, root media containing various $NO_3:NH_4$ ratios and equal concentration of other essential nutrients were filled into 50-cell plug trays and the seeds of 'Dotaerang Die' tomato were sown. The investigation of seedling growth and tissue analysis for mineral nutrient contents based on the dry weight of above-ground tissue were conducted 6 weeks after sowing. As seedlings grew, the EC decreased quickly and pH increased gradually in the all treatment media. The plant height, fresh weight and dry weight of seedlings in the treatment of 50:50 ($NO_3:NH_4$) were 29.0 cm, 13.7 g, and 1.21 g, respectively, which were the highest among treatments tested. However, the seedlings in the treatment of 0:100 ($NO_3:NH_4$) had 26.5 cm, 11.2 g, and 0.92 g in plant height, fresh weight and dry weight, respectively. These were the poorest among the treatments tested. The tissue contents of N were 2.77 to 3.22% in all the treatments. The treatment of 27:73 ($NO_3:NH_4$) had the highest contents of Fe, Mn and Zn and that of 0:100 ($NO_3:NH_4$) had the lowest contents of Mg, Na, Cu, Mn and Zn among the treatments tested. The results indicate that $NH_4$ ratio should be lower than 50% in the coir dust+peatmoss+perlite (3.5:3.5:3, v/v/v) medium for seedling growth of tomato and the optimum ratio will be used to draw up guide lines for plug seedling production.

Effects of Gamigwichulpajing-Tang on the Development of Experimentally-induced Endometriosis in Rats (가미귀출파징탕(加味歸朮破?湯)이 실험적으로 유발된 흰쥐의 자궁내막증에 미치는 영향)

  • Purpose : This study was carried out to investigate the effects of Gamigwichulpajing-Tang(GGT) on the development of experimentally-induced endometriosis in rats. Materials and Methods : Endometriosis was induced in rats by auotransplanting uterine tissue to the peritoneum and devided them into three groups: (1) sham-operated group(n=8), (2) surgically induced endometriosis and untreated control group(n=8), (3)surgically induced endometriosis and GGT treated group. GGT(700mg/head) was orally administrated for 15days after operation. Then we measured the body weight, the volumes of endometriotic implants, the weight of uterus and ovary, and investigated the content of cytokines(MCP-1, $TNF-{\alpha}$, $IL-l{\beta}$) in serum and ascites. Histopathology, immunohistochemistry for COX-2, and histochemistry for mast cell in transplanted uterine tissue were performed. Results :- The $volume(mm^3)$ of endometriotic implants in GGT-treated group$(53.50\;{\pm}18.63)$ was significantly decreased(p<<0.01) compared with control group$(404.50{\pm}317.68)$. - The content(pg/ml) of MCP-1 in ascites in GGT-treated group$(4265{\pm}108)$ was significantly decreased(p<<0.001) compared with control group$(8632{\pm}1245)$. - The content(pg/ml) of $TNF-{\alpha}$ in serum in GGT-treated group$(64.5{\pm}21.6)$ was significantly decreased(p<<0.05) compared with control group$(147.1{\pm}78.2)$. - The content(pg/ml)- of $TNF-{\alpha}$ in ascites in GGT-treated group$(738.3{\pm}502.4)$ was significantly decreased(p<<0.05) compared with control group$(1245.2{\pm}362.2)$. - The percentage(%) of positive epithelial layers for COX-2 in GGT-treated group$(22.9{\pm}9.3)$ was significantly decreased(p<<0.001) compared with control group$(50.2{\pm}8.2)$. - The number of mast cells in adjacent tissue of transplanted uterine tissue in GGT-treated group$(61.4{\pm}13.9)$ was significantly decreased(p<<0.001) compared with control group$(109.3{\pm}30.2)$. - The number of mast cells in stroma of transplanted uterine tissue in GGT-treated group$(9.4{\pm}2.7)$ was significantly decreased(p<<0.001) compared with control group$(26.0{\pm}7.7)$. - Histopathologically, proliferation of endomeuiotic epithelia and stroma, and infiltration of inflammatory cells in transplanted uterine tissue of GGT-treated group were weakly observed than those of control group. Conclusion :On the basis of these results, we concluded that Gamigwichulpajing-Tang have inhibiting effects on the development of transplanted uterine tissue. And these effects may be related with decreased Production of MCP-1 and $TNF-{\alpha}$, decreased expression of COX-2, and decreased infiltration of mast cells by administration of Gamigwichulpajing-Tang.

Effects of Keukhachukeo-Tang on the Development of Experimentally-induced Endometriosis in Rats (격하축동탕(膈下逐疼湯)이 실험적으로 유발된 흰쥐의 자궁내막증에 미치는 영향)

  • Purpose: This study was performed to investigate the effects of Keukhachukeo-Tang (KCT) on the development of experimentally-induced endometriosis in rats. Methods : Endometriosis was induced in rats by autotransplanting uterine tissue to the peritoneum and devided them into three groups: (1) sham-operated group (n=8), (2) surgically induced endometriosis and untreated control group (n =8), (3) surgically induced endometriosis and KCT treated group. KCT (1,200 mg/head) was orally administrated for 15 days after operation. Then we measured the body weight, the volumes of endometriotic implants. The weight (body, left uterus and ovaries) and concentrations of cytokines (MCP-1, TNF-${\alpha}$, IL-l${\beta}$) in serum and peritoneal fluid were also measured. Histopathology, immunohistochemistry for COX-2, and histochemistry for mast cells in transplanted uterine tissue were performed. Results : - The volumes(mm$^3$) of endometriotic implants in KCT-treated group (107${\pm}$66) were significantly decreased (p<0.05) compared with control group (405${\pm}$318). - The contents(pg/ml) of MCP-1 in peritoneal fluid in KCT-treated group (6,940${\pm}$893) were significantly decreased (p<0.01) compared with control group (8,632${\pm}$1,245). - The contents(pg/ml) of TNF-${\alpha}$ in peritoneal fluid in KCT-treated group (847${\pm}$330) were significantly decreased (p<0.05) compared with control group (1,245${\pm}$362). - The percentages(%) of positive epithelial layers for COX-2 in KCT-treated group (31${\pm}$10) were significantly decreased (p<0.01) compared with control group (50${\pm}$8). - The numbers of mast cells in adjacent tissue of transplanted uterine tissue in KCT-treated group (69${\pm}$18) were significantly decreased (p<0.01) compared with control group (109${\pm}$30). - The numbers of mast cells in stroma of transplanted uterine tissue in KCT-treated group(16${\pm}$5) were significantly decreased (p<0.01) compared with control group (26${\pm}$8). - Histopathologically, proliferation of endometriotic epithelia and stroma, and infiltration of inflammatory cells in transplanted uterine tissue of KCT-treated group were weakly observed than those of control group. Conclusion : From the above results, Keukhachukeo-Tang (KCT) have inhibiting effects on the development of transplanted uterine tissue. And these effects are related to the decreased concentration of MCP-1 and TNF-${\alpha}$, decreased expression of COX-2, and decreased infiltration of mast cells by administration of Keukhachukeo-Tang.

Effect of aerobic exercise on peroxisome proliferator-activated receptor α anti-inflammatory in white adipose tissue (백색지방조직에서 peroxisome proliferator-activated receptor α 항염증에 대한 유산소 운동의 영향)

  • It was investigated whether a combination of PPARα activator fenofibrate and swimming exercise (H/F/S) would have a beneficial synergistic effect on improving inflammation of white adipose tissue compared to the single prescription of fenofibrate (H/F) and swimming exercise (H/S) in male mice that fed high fat diet. The body weight, weight of white adipose tissue and total cholesterol levels in the serum increased in mice-fed high fat diets (H) compared to mice-fed low fat diets (L). Compared to H, both H/F and H/S decreased these. These levels reduced by fenofibrate were more effectively reduced by the combination of fenofibrate and swimming exercise (H/F/S). As a result of examining the expression of inflammatory cytokines genes and fatty acid oxidation genes in white adipose tissue, H increased compared to L, both H/F and H/S decreased compared to H, and H/F/S decreased further compared to H/F. Thus, this study revealed that the combination of fenofibrate and swimming exercise in male mice fed high-fat diet suppresses inflammation of white adipose tissue caused by obesity through promoted fatty acid oxidation more effectively than the fenofibrate alone, and suggested a practical way to improve inflammation of adipose tissue caused by obesity.

성장발육에 따른 골 백서경조직의 무기질함량의 변화

  • This present investigation was concerned with the content of ash, Mg, Ca, K and N'a of bone, cartilage, incisor teeth and molar teeth in rats of different weights. The results obtained were summarized as follows. 1. Cartilage is the most adequate model for the study in vivo of mechanisms concerned with normal calcification. 2. The percentage of inorganic material in cartilage rose from approx 13% of the dry weight in 5g rat to 46% in the tissue from rats weighing 134g 3. There was no marked change in the content of ash, Ca, Mg, K and Na after eruptions due to the increasing weight.

Hair Tissue Mineral Analysis(HTMA) of Children and Teenagers with Short Stature & Low Weight (저신장저체중 소아청소년의 모발미네랄분석)

  • Objectives The study was designed to find out the relationships between short stature and low weight among children and teenagers. Methods This research was performed in 21 children and teenagers from August 2006 to June 2007. We measured height, weight, body mass index, obesity rare, bone age, and HTMA. Moreover, we analyzed correlation with height, weight, mineral concentrations and ratios obtained from HTMA. Results There were significant correlations between HTMA and short stature and low weight. 1. Most subjects(76%) had slow metabolic types. 2. Ca, Mg, Zn, Ca/P, and Cr had significant differences between high bone age group and high chronological age group. 3. Fe/Cu had significant differences between a group that recent height is more smaller than inherited height and a group that recent height is more taller than inherited height. 4. Ca, Mg, Ca/P had significant differences between a low weight group and a normal weight group classified by obesity rate. Conclusions Children and teenagers with short stature and low weight have specific mineral characters.

In vitro and in vivo antibacterial activities of root extract of tissue cultured Pluchea indica (L.) Less.

  • The methanolic root extract of tissue cultured Pluchea indica (L.) Less. was tested for its antibacterial potentiality against 102 different strains of bacteria belonging to both Gram positive and Gram negative groups. The bacteria could be arranged according to their decreasing order of sensitivity as follows: Staphylococcus aureus, Vibrio cholerae, Bacillus spp. Vibrio parahaemolyticus, shigellae, Salmonella spp., and Escherichia coli. The extract was found to be bacteriostatic in nature against Salmonella typhimurium NCTC 74. When administered to Swiss strain of white mice at the doses of 0.5 and 1.0 mg/kg body weight, the extract could significantly protect the animals challenged with 50 MLD of S. typhimurium NCTC 74. According to the chi-square test, the in vivo data is highly significant (P < 0.001).
