인체지방유래 간질세포의 부착 및 연골분화유도를 위한 PLGA 지지체의 플라즈마 처리 효과 (The Effect of the Plasma Treatment on PLGA Scaffold for Adhesion and Chondrogenic Differentiation of Human Adipose-derived Stromal Cells)

  • High-density micromass culture was needed to take three dimensions culture with ASCs(adipose derived stromal cells) and chondrogenesis. However, the synthetic polymer has hydrophobic character and low affinity to cells and other biomolecules. Therefore, the surface modification without changes of physical and chemical properties is necessary for more suitable condition to cells and biomolecules. This study was performed to investigate the effect of surface modification of poly (lactic-co-glycolic acid)(PLGA) scaffold by plasma treatment (P(+)) on the adhesion, proliferation and chondrogenesis of ASCs, and not plasma treatment (P(-)). ASCs were isolated from human subcutaneous adipose tissue obtained by lipectomy and liposuction. At 1 hour 30 minutes and 3days after cell seeding onto the P(-) group and the P(+) group, total DNA amount of attached and proliferated ASCs markedly increased in the P(+) group (p < 0.05). The changes of the actin under confocal microscope were done for evaluation of cellular affinity, at 1 hour 30 minutes, the shape of the cells was spherical form in all group. At 3rd day, the shape of the cells was fiber network form and finely arranged in P(+) group rather than in P(-) group. RT-PCR analysis of cartilage-specific type II collagen and link protein were expressed in 1, 2 weeks of induction. Amount of Glycoaminoglycan (GAG) markedly increased in P(+) group(p < 0.05). In a week, extracellular matrix was not observed in the Alcian blue and Safranin O staining. However in 2 weeks, it was observed that sulfated proteoglycan increased in P(+) group rather than in P(-) group. In conclusion, we recognized that plasma treatment of PLGA scaffold could increase the hydrophilic property of cells, and provide suitable environment for high-density micromass culture to chondrogenesis

5-Fluorouracil 및 Mitomycin이 가슴샘의 미세구조에 미치는 영향 (Ultrastructural Study on the Thymus following the Administration of 5-Fluoruracil or Mitomycin)

  • The experiment was performed to study the morphological responses of the thymus of the mice, to antitumour agents (5-Fluorouracil or mitomycin C). Healthy adult mice weighing 25 gm each were divided into normal and experimental groups. 5-Fluorouracil (60 mg/kg) or Mitomycin-C $(400{\mu}g/kg)$ were injected subcutaneously to the animals every other day. Animals were sacrificed at 4 days and 7 days following the first injection. Pieces of the tissue taken from the thymus were prefixed with 2.5% paraformaldehyde-l.5% glutaraldehyde, followed by post-fixation with 1% osmium tetroxide. Ultrathin sections stained with uranyl acetate and lead citrate were observed with a JEM 100 CX-II electron microscope. The observed results were as follow: 1. Apoptoses of T-lymphocytes were observed more frequently in the thymus of the experimental groups than in those of a normal group. 2. In the experimental group, the plasma cells with distended cisternae of the granular endoplasmic reticulum and the eosinophile leukocytes were observed frequently. 3. In the experimental group, newly forming Hassall's corpurscles were observed frequently. 4. In the mitomycin-treated group, the epithelial reticular cells containing distended perinuclear cisternae, distended the granular endoplasmic reticula and pyknotic nuclei were observed in the cortico-medullary junctional area. 5. In the mitomycin-treated group, nuclear bodies with medium electron dense materials were often observed in the T lymphocyte. 6. In the 5-fluorouracil-treated groups, fused and dissolved tonofilament bundles and apoptotic bodies were observed in the some epithelial reticular cells in the medullary area. 7. In the 5-fluorouracil-treated groups, some elongated and bar-shaped lysosomes with electron lucent gap were often observed in the macrophages. 8. In the 5-fluorouracil-treated group, membrane complex of the smooth endoplasmic reticulum were ofen observed in the macrophage. From the above results, it was suggested that 5-fluorouracil or mitomycin could induce rapid involution of the thymus, and disturb maturation and differentiation of T lymphocytes, and, in turn, supress immunity.

한우와 흑한우 CCAAT/Enhancer Binding Protein β(C/EBPβ) 유전자의 발현과 다형분석 (Polymorphism Analysis and Expression of the CCAAT/Enhancer Binding Protein β(C/EBPβ) in the Korean Native Cattle and Black Cattle Storage)

  • 지방세포분화 초기에 중요한 역할을 수행하는 것으로 보고되고 있는 C/EBPβ 유전자를 cloning하기 위하여 한우 26개월령 등심조직을 이용하여 Total RNA를 추출하고 RT-PCR을 수행한 결과 한우 및 흑한우 C/EBPβ는 1047bp, 348 아미노산 서열을 가지고 있었다. 한우 및 흑한우 C/EBPβ의 아미노산 서열을 NCBI에 등록된 Japanese black cattle C/EBPβ(NCBI accession No. NM_176788) 비교한 결과 약 97%의 상동성을 나타내었다. 또한 인간, 닭, 생쥐, 쥐의 C/EBPβ 아미노산 서열을 비교한 결과 각각 96%, 58%, 74%, 75%의 상동성을 나타내었다. 그러나 각 종간의 leucine zipper domain간의 상동성은 매우 높게 나타났다. 한우 C/EBPβ의 발현은 폐, 등심, 지방조직에서 나타나고 있으나, 지방조직에서 좀 더 많은 발현을 보이고 있었다. 그리고 C/EBPβ 유전자의 bZIP 영역에서 polymorphism이 나타나지 않음을 확인하였다.

고려엉겅퀴 주정 추출물을 함유하는 임상시험제품의 항비만 활성 평가 (Anti-obesity Activities of Cirsium setidens Nakai Ethanolic Extract)

  • 본 연구는 고려엉겅퀴 주정추출물을 가지고 제작된 임상시험용제품(CNTM)이 3T3-L1 지방세포 및 고지방식이로 유도된 비만 쥐에 미치는 항비만 효능 관찰을 통하여 체지방 개선 기능성식품을 개발하기위하여 인체적용시험 시료를 제작한 후, 인체적용시험전에 그 효력이 유지됨을 확인하기위해 수행되었다. 본 연구에 사용된 시료 CNTM은 $80{\sim}320{\mu}g/mL$ 농도에서 세포독성이 관찰되지 않았으며, 지방 축적억제 효능 및 지방세포 분화, 지질대사 관련 유전인자들을 유의적으로 변화시키는 것으로 확인되었다. 또한 동물실험에서 CNTM 처리에 의하여 체중의 감소를 확인하였으며 혈중지질 성분 가운데 HDL-C/TC의 비율은 유의적으로 증가하며 LDL-cholesterol (LDL-C)과 non-esterified fatty acid (NEFA)의 비율은 유의적으로 감소하여 혈중 유리지방산 농도를 감소시키며 혈중 콜레스테롤 상승을 억제시키고 혈당을 감소시키는 효과가 있는 것으로 평가되었다. 따라서 고려엉겅퀴 주정추출물이 함유된 인체적용시험시료(CNTM)는 체지방 개선에 우수한 효과가 있는 것으로 확인되었다.

인핸서 RNA에 의한 유전자 전사 조절 (Transcriptional Regulation of Genes by Enhancer RNAs)

  • 다세포 생물의 유전자들은 발생 및 분화 그리고 조직 특이적으로 전사되며, 이러한 유전자 전사는 게놈 상에서 멀리 떨어져 존재하는 인핸서(enhancer) 부위에 의해 조절된다. 최근의 연구들은 활성화된 인핸서에서 RNA Polymerase II (Pol II)에 의해 noncoding RNA가 전사된다고 보고하고 있으며, 이들은 인핸서 RNA (eRNA)라 불리고 있다. eRNA는 인핸서 중심으로부터 양방향으로 합성되며, 5’ capping은 일어나지만, splicing이나 3’ tailing은 되지 않는다. eRNA의 전사는 전사 활성자의 결합에 의해 일어나며, 표적 유전자의 전사 수준과 비례하게 일어난다. 인위적으로 eRNA의 전사를 억제하거나 합성된 eRNA를 제거하면 표적 유전자의 전사는 억제된다. eRNA의 전사 과정은 인핸서 부분의 활성 히스톤 변형을 유도하며, 합성된 eRNA는 인핸서와 프로모터 사이의 크로마틴 고리 구조 형성을 매개한다. 또한 표적 유전자의 프로모터에 RNA Pol II를 모집하고 이들의 신장을 촉진하는 것도 eRNA의 역할로 보인다. 본 총설은 인핸서 유래 eRNA의 특징에 대해 살펴보고, eRNA의 합성 기작 및 표적 유전자의 전사 조절을 위한 eRNA의 역할을 정리해보고자 한다.

Use of Real-Time Quantitative PCR to Identify High Expressed Genes in Head and Neck Squamous Cell Carcinoma Cell Lines

  • Head and neck squamous cell carcinoma(HNSCC) is the sixth most common cancer among men in the developed world affecting the tongue, pharynx, larynx and oral cavity. HNSCC is thought to represent a multistep process whereby carcinogen exposure leads to genetic instability in the tissue and accumulation of specific genetic events, which result in dysregulation of proliferation, differentiation, and cell loss and the acquisition of invasive capacity. Despite therapeutic and diagnostic progress in oncology during the past decades, the prognosis of HNSCC remains poor. Thus it seems that finding a biological tumor markers which will increase the early diagnosis and treatment monitoring rates, is of paramount importance in respect to improving prognosis. In an effort to identify gene expression signatures that may serve as biomarkers, this study several genes were selected, such as H3,3A, S100A7, UCHL1, GSTP1, PAI-2, PLK, TGF${\beta}$1 and bFGF, and used 7 HNSCC cell lines that were established various anatomical sites, and also 17 other cancer cell lines were used for control group using real-time quantitative RT-PCR and immunocytochemical analysis with a monoclonal antibody. In this study, S100A7 showed a clearly restricted occurrence in tongue originated cell line, and GSTP1 expression level in the pharynx originated cell line was very increased, relative to corresponding other cell lines. These results suggest that S100A7 and GSTP1 genes' expression can occur during tongue and pharynx originated head and neck tumorigenesis and that genetic change is an important driving force in the carcinogenesis process. This data indicate that S100A7 and GSTP1 expression pattern in HNSCC reflect both diagnostic clue and biological marker. And this is provides a foundation for the development of site-specific diagnostic strategies and treatments for HNSCC.

반복유산을 경험한 환자에서 임신중 태반항원과 동종항원에 노출된 모체 림프구면역반응은 언제부터 소실되나? (When Dose Losses of Maternal Lymphocytes Response to Trophoblast Antigen or Alloantigen Occur in Women with a History of Recurrent Spontaneous Abortion?)

  • The maintenance of a viable pregnancy has long been viewed as an immunological paradox. The deveolping embryo and trophoblast are immunologically foreign to the maternal immune system due to their maternally inherited genes products and tissue-specific differentiation antigens (Hill & Anderson, 1988). Therefore, speculation has arisen that spontaneous abortion may be caused by impaired maternal immune tolerance to the semiallogenic conceptus (Hill, 1990). Loss of recall antigen has been reported in immunosuppressed transplant recipients and is associated with graft survival (Muluk et al., 1991; Schulik et al., 1994). Progesterone $(10^{-5}M)$ has immunosuppressive capabilities (Szekeres-Bartho et al., 1985). Previous study showed that fertile women, but not women with unexplained recurrent abortion (URA), lose their immune response to recall antigens when pregnant (Bermas & Hill, 1997). Therefore, we hypothesized that immunosuppressive doses of progesterone may affect proliferative response of lymphocytes to trophoblast antigen and alloantigen. Proliferative responses using $^3H$-thymidine ($^3H$-TdR) incorporation of peripheral blood mononuclear cells (PBMCs) to the irradiated allogeneic periperal blood mononuclear cells as alloantigen, trophoblast extract and Flu as recall antigen, and PHA as mitogen were serially checked in 9 women who had experienced unexplained recurrent miscarriage. Progesterone vaginal suppositories (100mg b.i.d; Utrogestan, Organon) beginning 3 days after ovulation were given to 9 women with unexplained RSA who had prior evidence of Th1 immunity to trophoblast. We checked proliferation responses to conception cycle before and after progesterone supplementation once a week through the first 7 weeks of pregnancy. All patients of alloantigen and PHA had a positive proliferation response that occmed in the baseline phase. But 4 out of 9 patients (44.4%) of trophoblast antigen and Flu antigen had a positive proliferative response. The suppression of proliferation response to each antigen were started after proliferative phase and during pregnancy cycles. Our data demonstrated that since in vivo progesterone treated PBMCs suppressed more T-lymphocyte activation and $^3H$-TdR incorporation compare to PBMCs, which are not influenced by progesterone. This data suggested that it might be influenced by immunosuppressive effect of progesterone. In conclusion, progesterone may play an important immunological role in regulating local immune response in the fetal-placental unit. Furthermore, in the 9 women given progesterone during a conception cycle, Only two (22%) repeat pregnancy losses occured in these 9 women despite loss of antigen responsiveness (one chemical pregnancy loss and one loss at 8 weeks of growth which was karyotyped as a Trisomy 4). These finding suggested that pregnancy loss due to fetal aneuploidy is not associated with immunological phenomena.

Morphological and Genetic Characteristics of Colletotrichum gloeosporioides Isolated from Newly Emerging Static-Symptom Anthracnose in Apple

  • Filamentous fungi of the genus Colletotrichum (teleomorph, Glomerella) are considered major plant pathogens worldwide. Cereals, legumes, vegetables, and fruit trees may be seriously affected by this pathogen (1). Colletotrichum species cause typical disease symptoms known as anthracnoses, characterized by sunken necrotic tissue, where orange conidial masses are produced. Anthracnose appears in both developing and mature plant tissues (2). We investigated disease occurrence in apple orchards from 2013 to 2014 in northern Gyeongbuk province, Korea. Typical anthracnose with advanced symptoms was observed in all apple orchards studied. Of late, static fruit spot symptoms are being observed in apple orchards. A small lesion, which does not expand further and remains static until the harvesting season, is observed at the beginning of fruit growth period. In our study, static symptoms, together with the typical symptoms, were observed on apples. The isolated fungus was tested for pathogenicity on cv. 'Fuji apple' (fully ripe fruits, unripe fruits, and cross-section of fruits) by inoculating the fruits with a conidial suspension ($10^5$ conidia/ml). In apple inoculated with typical anthracnose fungus, the anthracnose symptoms progressed, and dark lesions with salmon-colored masses of conidia were observed on fruit, which were also soft and sunken. However, in apple inoculated with fungi causing static symptoms, the size of the spots did not increase. Interestingly, the shape and size of the conidia and the shape of the appressoria of both types of fungi were found to be similar. The conidia of the two types of fungi were straight and cylindrical, with an obtuse apex. The culture and morphological characteristics of the conidia were similar to those of C. gloeosporioides (5). The conidia of C. gloeosporioides germinate and form appressoria in response to chemical signals such as host surface wax and the fruitripening hormone ethylene (3). In this study, the spores started to germinate 4 h after incubation with an ethephon suspension. Then, the germ tubes began to swell, and subsequently, differentiation into appressoria with dark thick walls was completed by 8 h. In advanced symptoms, fungal spores of virtually all the appressoria formed primary hyphae within 16 h. However, in the static-symptom fungus spores, no primary hyphae formed by 16 h. The two types of isolates exhibited different growth rates on medium containing apple pectin, Na polypectate, or glucose as the sole carbon. Static-symptom fungi had a >10% reduction in growth (apple pectin, 14.9%; Na polypectate, 27.7%; glucose, 10.4%). The fungal isolates were also genetically characterized by sequencing. ITS regions of rDNA, chitin synthase 1 (CHS1), actin (ACT), and ${\beta}$-tubulin (${\beta}t$) were amplified from isolates using primer pairs ITS 1 and ITS 4 (4), CHS-79F and CHS-354R, ACT-512F and ACT-783R, and T1 and ${\beta}t2$ (5), respectively. The resulting sequences showed 100% identity with sequences of C. gloeosporioides at KC493156, and the sequence of the ${\beta}$t gene showed 100% identity with C. gloeosporioides at JX009557.1. Therefore, sequence data from the four loci studied proves that the isolated pathogen is C. gloeosporioides. We also performed random amplified polymorphic DNA-PCR, which showed clearly differentiated subgroups of C. gloeosporioides genotypes. The clustering of these groups was highly related to the symptom types of the individual strains.

중간엽줄기세포의 노화에 따른 후생유전학적 변화 (Epigenomic Alteration in Replicative Senescent-mesenchymal Stem Cells)

  • 중간엽줄기세포는 성체줄기세포의 한 종류로, 자기재생산능력(self-renwal)과 다분화능(multipotency)을 가지고 있고, 다양한 자양인자(trophic factors)들을 분비한다. 뿐만 아니라, 중간엽줄기세포는 골수, 지방, 탯줄과 같은 조직에서 쉽게 얻을 수 있기 때문에 줄기세포치료에 좋은 도구로 이용되고 있다. 하지만, 줄기세포치료의 효율성을 높이기 위해 추출한 세포의 개체 수를 늘리는 과정에서 중간엽줄기세포는 점차적인 노화를 겪게 되고, 이는 줄기세포 자체의 기능적인 감소를 야기한다. 인체 내에서, 노화된 줄기세포는 조직 내의 항상성 유지에 부정적인 영향 을 미치게 되고, 이러한 상태가 지속되면 대표적인 노인성 질환인 퇴행성 질환의 원인이 된다. 최근 연구들에 의하면 중간엽줄기세포가 노화를 겪을 때, 노화 관련된 DNA 메틸화 패턴의 변화와 히스톤의 변형이 일어남을 확인하였다. 또한, 중간엽줄기세포의 노화에 있어서 DNA 메틸화효소(DNA methyltransferase) 억제제와 히스톤 아세틸화효소(histone deacetylase) 억제제가 부분적으로 노화를 개선하는 효과를 관찰한 연구사례들이 있다. 본 총설에서는, 노화에 따른 후생유전학적인 변화에 의해, 조절되는 노화 관련 유전자들과 중간엽줄기세포의 노화에 대한 연구사례들을 분석하여 서술하고자 한다.

나노구조 표면에 관한 문헌고찰 (A Literature Review on Nano-Modified Implant Surfaces)

  • 티타늄 산화막을 나노단위에서 변형시키는 방법은 다공성 표면을 강화하는 내부적 접근과 나노입자를 피복하는 외부적 접근으로 나눌 수 있다. 나노표면은 나노튜브, 나노피트, 나노노듈 및 다형구조 등 다양한 형태를 지닌다. 형성방법 및 형성재료에 따라 다른 표면이 생성되지만, 현재까지 표준화된 형성방법은 없다. 나노표면을 분석해 보면 마이크론 단위의 표면구조에는 영향을 미치지 않으며 전기화학적으로 안정적이다. 나노표면은 세포독성이 거의 없으며 조골세포의 증식과 분화를 모두 촉진하고, 섬유모세포의 증식을 저해하여 연조직 개재를 감소시키는 효과를 가진다. 또한 세포 및 단백질과 유사한 크기 및 형태를 가지기 때문에 조직과의 친화성이 우수하여 골유착을 증진시킨다. 하지만 그 작용이 미치는 범위는 극히 제한되어 있기 때문에 골조직과의 거리가 있는 경우에는 효과가 미미하다. 마이크론 단위의 표면과는 달리 나노표면은 광촉매효과로 인한 항균작용을 가지지만 지속시간이 짧아 실제 임상에서의 적용효과는 의문시 된다. 하지만 마이크론 단위의 표면거칠기가 가지는 단점을 배제할 수 있어 다양한 가능성을 가지기 때문에 더 많은 연구가 필요하다.
