인삼 육성계통 캘러스부터 Polyacetylene의 분석 및 생합성에 미치는 배양조건 (Analysis and Culture Conditions for Biosynthesis of Polyacetylene from Callus of Ginseng Superior lines)

  • 인삼 callus의 기내배양에 의해서 항암물질을 대량생산하기 위한 연구의 일환으로 인삼 우수계통으로부터 기내에서 callus를 유기하여 polyacetylene 고함유 세포주를 선발하고, 인삼 callus의 polyacetylene 생산에 미치는 식물호르몬 및 배지의 영향을 조사하였다. 식물호르몬 CPA가 첨가된 MS배지에서 유도된 인삼 우수계통 callus에서는 polyacetylene 중에서 panaxynol은 TLC와 GC에 의해서 전혀 검출할 수 없었으나, callus에 따라서는 panaxydol이 형성됨을 확인할 수 있었다. 특히, 강력한 항암효과를 가지고 있는 panaxydol은 우수계통 callus 18종 중에서 5종이 형성하였으며, 30201계통 callus에서 가장 많이 검출되었다. 또한 인삼 10301계통 callus는 CPA가 단독으로 첨가된 배지에서 panaxydol을 생성하지 못하였으나, CPA 2 mg/L와 BA 0.05 mg/L 첨가구에서는 callus의 생장도 양호하였으며, panaxydol도 생성하였다. 인삼 callus의 생장과 panaxydol의 합성에는 MS배지보다 SH 배지가 더 양호하였으며, NAA와 sucrose는 전혀 영향을 미치지 않았다.

Anticonvulsant potential of callus cultures of Convolvulus microphyllus Sieb.

  • Callus cultures of Convolvulus microphyllus Sieb. was induced on Murashige and Skoog's medium supplemented with 2,4-dichloro phenoxy acetic acid, 6-benzyl adenine, indole acetic acid and kinetin (1 ppm each). Methanolic extracts of whole plant, leaf, stem and leaf and stem calli were tested for anticonvulsant activity against standard drug phenytoin using maximal electroshock model on mice. It was observed that the animals treated with methanolic extracts of stem callus, leaf callus and whole plant (200 mg/kg, oral) showed significant protection against tonic convulsions induced by transcorneal electroshock. Anticonvulsant activity of methanolic extract of stem callus was comparable to that of standard drug phenytoin.

조직배양(組織培養)한 시호근(柴胡根)의 Saikosaponin 함량(含量) (Saikosaponin Contents in Bupleurum falcatum Root Produced by Tissue Culture)

  • The study for the quantitative analysis of saikosaponin of Bupleurum falcatum root produced by tissue culture was attempted. The optimal concentration of 2,4-D was 0.1 mg/l for inducing the callus. The induction and growth of the adventitious root from the callus was obtained in the suspension culture containing MS basal medium supplemented with no plant growth regulators. The results of the quantitative analysis of saikosaponins by high performance liquid chromatography (HPLC) showed that the content of saikosaponin a and c was high in the one-year-old root from somatic embryos and that of saikosaponin d was remarkably high in three-months-old adventitious root from callus.

  • The occurrence of TTX in ciliated protozoa was investigated in order to clarify tetrodotoxin (TTX) accumulation mechanisms in marine organisms. Tissue culture bioassay, HPLC, and GC-MS analyses confirmed the occurrence of TTX in Euplotes mutabilis and also in bacteria isolated from the culture medium. Fluorescently labeled bacteria (FLB) were prepared with those bacteria, and predation by E. mutabilis was observed. The results indicated that TTX in bacteria can be transferred to higher trophic levels through the food chain.

In vitro Formation of Tuberous Roots from Root Ends in the Rooted Tuberous stem without shoots in Cyclamen persicum MILL.

  • In Japan, propagation of cyclamen is mainly from seedlings. However, seeds are expensive and germination is slow and non..uniform. Therefore, to achieve genetically uniform propagation, multiplication must be vegetative. The rooted tuberous stems without shoots as sources of explants were cultured on the media containing BA and sucrose. After 30 days cultivation, tuberous roots were produced from the root ends attached to a tuberous stem and its capability was dependent on the type of media. The highest percentage of tuberous root formation was observed in Culture on the medium of 1/3 MS containing 0.05mgL$^{-1}$ NAA, 0.5mg L$^{-1}$ BA and 5% sucrose. Growth rates of the tuberous roots were greatly influenced by the cutting positions of a root in explants. The highest growth of was observed if small amount of root end was cut at initiation of tissue culture.

  • In hypoxic tissue conditions, pyruvate can not enter the Krebs cycle and lactic acid, produced from pyruvate, accumulates to induce lactic acidosis. Pyruvate, However, can also be converted to alanine by glutamate pyruvate transaminase, that could be enhanced by glutamate. Therefore, it would be a fundamental measure to treat the lactic acidosis in tissue hypoxic conditions when one can convert the accumulated lactic acid, through pyruvate, to alanine. To test the above hypothesis, we induced a lactic acidosis in cats and the effect of glutamate on recovery of acid base state and removal of the lactic acid from blood were assessed and the results were compared with those of bicarbonate administration, which is one of the most frequently used conventional measure for correction of the acid base state during lactic acidosis. The results were that glutamate and combined glutamate bicarbonate solutions not only restored the acid base status completely from the lactic acidosis in an hour or two, but also restored the blood level of lactate partially. We concluded that administration of glutamate solution to convert pyruvate into alanine is effective in preventing lactic acid accumulation and treating lactic acidosis.

Initial Culture Conditions for Primary Cell Populations Derived from Radula Tissue in Abalone Haliotis discus hannai

  • Abalone immortal cell lines can be used to study the physiological properties and disease mechanisms of abalone at the cellular and molecular level. As a first step for the final goal to establish abalone immortal cell lines, we examined various initial culture conditions for primary cell populations derived from Haliotis discus hannai radula tissue. The survival rate after cell isolation procedures using the enzymatic method was as low as $9.95{\pm}2.37%$. Based on three different experimental conditions for H. discus hannai radula-derived cell culture, we found that the salinity of the media and the presence of growth-promoting factors were important to support radula-derived primary cell populations during the initial culture. The growth factor-containing media adjusted to 35 psu salinity could induce 100% (8 out of 8 trials) initial cell attachment, and the rate of cell attachment reached 50-70%. The data obtained from this study will provide useful information for developing immortal cell lines from abalone species.

흡수성 차폐막에 배양된 구개관세포의 증식양상의 비교 (Comparison of the Proliferation pattern of Cultured Rat Calvaria Cell on the Resorbable Barrier Membrane)

  • The purpose of this study is to evaluate the phenomenon of attachment and spreading of the cultured rat calvarial cell inoculated on their surface of different kinds of biodegradable membrane which had been used on tissue regeneration on periodontal defects by using scanning electron microscope. In this experiment 30 Sprague-Dawley male rats (mean BW 150gm) were used to harvest abundant number of cell in the short period. The rats were sacrificed by decapitatioan to obtain the calvaria for bone cell culture. Calvarial cells were cultured with Dulbecco's Modified Essential Medium contained with 10% Fetal Bovine Serum under the conventional conditions. Biodegradable barrier membrane were collected with collagen type, and were divided into 3 different kind of surface such as scattered, polarized and fine-net type as their surface texture. Microcover plate which usually used for cell culture was used as control for smooth surface. All the membrane were seeded with cultured calvarial cell on their surface. The number of cell inoculated on the membrane were $1{\times}10^6$ Cells/ml. After the culture as designed time, all the membrane were washed with 0.1 M Phosphate Buffered saline and fuxed with 2.5% Glutaraldehyde. And all specimen were treated with $OsO_4$, and Tannic acid before drying the cell for coating the cell with gold. Scanning Electron Microscope was used to observation. The following results were obtained. I. During the whole period of experiment, the phenomenon of cell attachment and spreading were revealed similar pattern to compare with smooth surface culture plate and ordinary culture dish. 2. The shape of cell attachment and spreading on the surface of barrier membrane were observed no remarked difference pattern between smooth surface culture plate and ordinary culture dish. 3. The cytoplasmic process of cultured calvaria cell extent to the deep portion of barrier membrane like as their own proper shape. 4. There were no remarkable relationships between the degree of cultured cell spreading and surface structure of barrier membrane. 5. Slight starified layer of cultured calvaria cell were observed on the scattered type of resorbable membrane, Conclusively, this study thus suggest that cultured bone cell inoculated onto the biodegradable barrier membrane may have an important role of carrier for many cell which could be used as new tissue regeneration, and those tissue engeering technique may become an new method in the approach to the repair of bone defects.

조직배양에 의한 네리네 대량증식 (Bulb Propagation on Nerine by Tissue Culture)

  • 조직배양에 의한 네리네 기내 대량증식 조건을 확립하고자 대량증식 배지선발, 자구비대 및 출현율 향상을 위한 첨가물질, 배양부위별 증식효율 등에 관한 연구를 수행하였다. 네리네 기내 대량증식을 위한 생장조정제 처리결과 자구형성수는 단용처리보다 혼용처리에서 효과적이었다. 생장조정제 처리별 자구수에서 Nerine bowdenii ‘Favorite’ 와 Nerine sarniensis ‘Red’ 모두 NAA $0{\sim}0.5$ + BA $0.5{\sim}2.0mg\;{\cdot}\;L^{-1}$에서 무처리 1.2개보다 $66%{\sim}100%$향상된 $1.8{\sim}2.4$개로 효과적이었다. 자구비대 및 출현율 향상을 위한 최적 배지원과 농도는 Glucose 7%로 가장 양호한 자구비대 및 순화시기에 관련 없는 높은 출현율을 보였다. 또한 질소원으로는 질산태와 암모니아태 질소 혼용 40mM에서 토양내 자구 출현율이 가장 높았다. 배양부위별 증식효율은 재료 확보가 제한적인 생장점 배양보다는 인편번식 배양이 54배 높은 증식효율을 보였으며, 인편번식 배양을 위한 부위로는 중인편이 양호한 생장 및 가장 높은 자구 형성(1.8개/절편)이 가능하였다.

  • The phytoremediation of trinitrotoluene, nitroglycerine, pentaerytritoltetranitrate in plant tissue cultures of Solanum aviculare, Rheum palmatum and Populus simonii were studied. All above mentioned explosives were degradated to to less toxic products and finally mineralized or bound to the cell wall.
