분석조건별 담수어류의 환경 DNA 메타바코딩 효율 비교: 필터, 추출 키트, 프라이머 조합 및 PCR 방법 (Efficiency Comparison of Environmental DNA Metabarcoding of Freshwater Fishes according to Filters, Extraction Kits, Primer Sets and PCR Methods)

  • 메타바코딩을 이용한 환경 DNA 분석은 검출 감도가 높아 어류의 생물다양성 평가 및 멸종위기종의 검출에 유용한 기술이다. 이번 연구는 메타바코딩을 이용해 우리나라 담수어류를 대상으로 높은 검출 효율을 보일 수 있는 적합한 분석방법을 확인하기 위해 4가지 분석조건별, 즉 필터(cellulose nitrate filter, glass fiber filter), 추출 키트(DNeasy® Blood & Tissue Kit, DNeasy® PowerWater Kit), 프라이머 조합(12S rDNA, 16S rDNA) 그리고 PCR 방법(conventional PCR, touchdown PCR)로 나타나는 Operational Taxonomic Units(OTUs) 수와 종 조성을 비교하였다. Glass fiber filter와 DNeasy® Tissue & Blood Kit를 이용해 추출한 시료는 12S rDNA와 16S rDNA 프라이머 조합에서 담수어류 OTUs가 가장 많이 검출되었다. 모든 분석조건 중 프라이머 조합에서만 조기어강(Class Actinopterygii) 평균 OTUs 수에서 통계적으로 유의한 차이를 보였고(Non-parametric Wilcoxon Signed Ranks Test, p=0.005), 담수어류 평균 OTUs 수는 유의하지 않았다. 종 조성 비교 결과 역시 프라이머 조합에서 유의한 차이를 보였고(PERMANOVA, Pseudo-F=6.9489, p=0.006), 나머지 분석조건에서는 유의한 차이를 보이지 않았다. NMDS 분석 결과 종 조성은 유사도 65% 기준에서 프라이머 조합에 따라 묶였고, 16S rDNA 프라이머 세트는 주로 멸종위기종인 모래주사(Microphysogobio koreensis), 꼬치동자개(Pseudogobio brevicorpus)가 기여하였고, 12S rDNA 프라이머 세트는 주로 일반종인 피라미(Zacco platypus), 꺽지(Coreoperca herzi) 등이 기여한 것으로 나타났다. 본 연구는 국내 하천에서 채취한 시료에 대한 메타바코딩을 이용한 종 다양성 분석의 기초정보를 제공한다.

역혈행 후골간 도상피판을 이용한 전기아크손상으로 인한 손목부 결손의 재건 (The Reverse Posterior Interosseous Island Flap for the Reconstruction of Soft Tissue Defects in the Wrist Injured by Electrical Arc)

  • Purpose: High tension electrical injuries result in major tissue(eg. bones, tendons, vessels and nerves) destruction. Therefore, the management of mutilating wrist caused by electrical injuries still represents a challenge. There are various approaches to this problem including local and regional flaps as well as pedicled distant flaps and microsurgical free tissue transfer. Although it has not gained wide acceptance, because of the technically demanding dissection of the pedicle, posterior interosseous flap is now well accepted for the reconstruction of hand and wrist in hand surgery. The principal advantages of this flap are minimal donor site morbidity, minimal vascular compromise, one stage operation. This flap also offers the advantages of ideal color match and composition. In this report, we describe our experience with the reverse posterior interosseous island flap for reconstruction of mutilating wrist with main vessel injuries. Methods: From October, 2004 to June, 2006, we treated 11 patients with soft tissue defects and main vessel injuries on the wrist that were covered with reverse posterior interosseous island flap. Results: These 11 patients were all male. The ages ranged from 27 to 67 years(mean age 41.75) and the follow-up period varied from 4 to 19 months. Complete healing of the reverse posterior interosseous island flaps were observed in 11 patients(12 flaps). The majority of these flaps showed a certain degree of venous congestion, which in a flap was treated with medical leech. 1 flap has partial necrosis owing to sustained venous congestion, requiring secondary skin graft. flap size varied from $3.5{\times}8cm$ to $10{\times}12cm$(mean size $6.4{\times}8.9m$). The donor site defect was closed directly in 5 flaps, and by skin graft in 7 flaps. Conclusion: We found that the reverse posterior interosseous island flap is reliable and very useful for reconstruction of mutilating wrist and we recommend it as first choice in coverage of soft tissue defects in the wrist with electrical arc injuries.

Effects of Flaxseed Diets on Fattening Response of Hanwoo Cattle : 2. Fatty Acid Composition of Serum and Adipose Tissues

  • Two separate trials were designed to determine effects of dietary level of whole flaxseed (WFS) on fatty acid composition of serum, and subcutaneous, perirenal, intermuscular, and intramuscular adipose tissues of Korean Hanwoo cattle. Twentyone bulls (trial 1) and 15 cows (trial 2) were assigned to diets containing 0, 10 or 15% WFS. Relative treatment effects were similar between bulls and cows. The proportion of C18:3 in serum and to a lesser extent in adipose tissues were increased by dietary inclusion of WFS, reflecting supplemented lipid composition of WFS that escaped ruminal biohydrogenation. Animals fed WFS had a lower proportion of saturated fatty acids in serum and adipose tissues than animals fed diets without WFS, while the opposite trend was observed in unsaturated fatty acids with little differences between two WFS groups. WFS-fed animals had higher proportions of C18:1, 18:2, 18:3, 20:3, and 22:3 and lower proportions of C12:0, 14:0, 16:0 and 18:0 in intramuscular fat than animals fed diets without WFS. Furthermore, feeding WFS increased proportions of both $\omega$-3 and $\omega$-6 fatty acids but decreased the ratio of $\omega$-6/$\omega$-3 substantially. In conclusion, feeding WFS can be an effective method of increasing absorption of unsaturated fatty acids, and subsequent deposition in adipose tissues.

Effects of Rumen Protected Oleic Acid in the Diet on Animal Performances, Carcass Quality and Fatty Acid Composition of Hanwoo Steers

  • The effects of different rumen protected forms, oleamide, Ca oleate, of dietary oleic acid on the carcass quality and fatty acid composition in intramuscular and subcutaneous fat tissues of Hanwoo steer were examined. Sixty, 25 month old Hanwoo steers divided into three groups were fed no supplement (Control), 2% of oleamide (Oleamide) or Ca-oleate (Ca-Oleate) in their diet for 45 or 90 days. Disappearance rates of oleic acid supplements in digestive tracts (Rumen bypass, abomasal and intestinal disappearance rate) were 48.5, 68.4 for oleamide and Ca oleate, respectively. Both oleic acid supplements affected feed intake, growth rate, cold carcass weight and carcass fatness. Live weight gain, carcass weight, backfat thickness and marbling score were higher in the oleic acid supplemented steers compared with those from the control. Oleic acid supplements increased marbling score and ether extract in Hanwoo steer m. logissi thoracicmus. Rumen protected oleic acid increased not only the level of oleic acid but also polyunsaturated fatty acids in intramuscular and subcutaneous fat tissue. Total saturated fatty acid contents in both fat tissues were decreased whereas total unsaturated fatty acid content was increased compared with those from control. Linoleic acid, linolenic acid and polyunsaturated fatty acid contents were significantly higher in Ca oleate than any other steers. Lipid metabolites in blood were increased in rumen protected oleic acid treatments. HDL content in blood was increased in Ca-oleate supplemented steers whereas LDL was decreased compared with control. The changes of fatty acid compositions in the rumen protected oleic acid supplemented steers suggest that the oleic acid and unsaturated fatty acid were protected from rumen biohydrogenation and can be deposited in the fat tissues.

The Effect of Green Oak Acorn (Quercus ilex) Based Diet on Growth Performance and Meat Fatty Acid Composition of Broilers

  • This experiment was conducted to compare the effects of oak acorns and corn on broilers growth performance, carcass characteristics and meat fatty acid (FA) composition according to different ages of broilers. Two separate groups of 400 male broilers, reared for six weeks, were fed diets containing 33.5% green oak acorns (GO) and 67% of corn (C), respectively. At day 35, the body weight of the chickens fed the GO diet was 12% lower (p<0.05) compared with controls (C). However, at day 56 birds reached similar final weights. Abdominal adipose tissue (AAT) was 78% higher in the controls compared to those fed the oak acorn-based diet. The total lipids (TL) of thigh muscle were higher in the controls compared to dietary treatment GO (3.1 against 2.5 g/100 g of muscle). There were significant differences in fatty acid (FA) composition of the muscle between two groups. The broilers fed oak acorn exhibited more C18:2, as well as a higher proportion of polyunsaturated fatty acid (PUFA) and PUFA:SFA (saturated fatty acid) ratio than those on the control diet, but no significant differences were observed among groups for the age of birds. There was a higher proportion of linolenic acid in broilers fed the oak acorn compared to the controls (0.8 vs. 0.6%). Furthermore, linolenic acid content decreased with age (p<0.001) in both groups. The results indicated no significant differences in FA with the age of birds. In conclusion, the meat fatty acids provided by the GO diet appeared to improve the nutritional quality preferred by the consumer.

Characterization of Chicken By-products by Mean of Proximate and Nutritional Compositions

  • Though a great amount of chicken by-products are consumed everyday in many countries worldwide, however, no attention has been paid to the investigation of nutritional composition of these by-products. In the present work, the basic information regarding the aspects of nutritional composition of chicken by-products such as; liver, gizzard, heart, lung, crop, small intestines, cecum and duodenum was studied. Our results revealed that the approximate composition range (minimum to maximum) of these by-products was found as such: moisture 76.68-83.23%; fat 0.81-4.53%, protein 10.96-17.70% and calories 983.20-1,426.0 cal/g tissue, in which liver and gizzard had the highest protein content. Liver had higher (p<0.05) vitamin A, B1, B2, B3, B5 and B6 contents in comparison to other remaining byproducts. Total saturated fatty acids (SFA), unsaturated fatty acids (UFA), polyunsaturated fatty acids (PUFA) levels ranged between the by-products from 31.82% to 43.96%, 56.04% to 68.19%, and 18.27% to 32.05%, respectively. Remarkably, all of by-products showed desirable PUFA/SFA ratios. Furthermore, all of chicken by-products, especially liver, contained higher levels of trace elements (e.g., Fe, Mn and Zn) in comparison with those from muscle tissues published in literature. Overall, the study indicated that most of chicken byproducts examined are good sources of essential nutrients and these obtained results will be the useful information to consumers and meat processors.

Effects of Flaxseed Diets on Performance, Carcass Characteristics and Fatty Acid Composition of Hanwoo Steers

  • This study was conducted to determine the effects of dietary level of whole flaxseed (WFS; 0, 10 and 15%) on performance, carcass characteristics and fatty acid composition of serum and subcutaneous, perirenal, and intramuscular adipose tissues of Korean Hanwoo cattle. The daily gains were not different among treatments. Dietary inclusion of WFS decreased (p<0.05) feed intake but improved (p<0.05) feed conversion ratio (feed/gain). Backfat thickness and marbling score were increased (p<0.05) by dietary WFS. Carcass weight, dressing percentage, loin-eye area, and carcass yield and quality were not different among treatments. The proportion of C18:3 in serum and, to a lesser extent, in adipose tissues were increased (p<0.01) by dietary WFS, indicating that lipids from WFS escaped ruminal biohydrogenation. Animals fed WFS had lower proportions of saturated fatty acid (SFA; C14:0 and 16:0) and higher proportions of polyunsaturated fatty acids (PUFA; C18:2. 18:3, 20:2, 20:4, 20:5 and 22:6) in perirenal and intramuscular fat than animals fed diets without WFS, resulting in an increased PUFA/SFA ratio. Furthermore, feeding WFS increased (p<0.01) proportions of $\omega$-3 and $\omega$-6 fatty acids in intramuscular fat but decreased (p<0.05) the $\omega$-6/$\omega$-3 ratio. Relative treatment effects were similar between 10 and 15% WFS. Feeding WFS can effectively alter composition of adipose tissues with enhanced feed conversion ratio.

  • The cellularity and composition of the spleen lymph node thymus and peripheral blood and tempo of regeneration were studied at various time points after syngeneic bone marrow transplantation(BMT) in C3H/Hen mice. Significant depression of absolute lymphocyte count was noted on week 1 after lethal whole-body irradiation and BMT. In comparison to the lymph node thymus and spleen had an rapid regeneration of cellularity. The distinct cell populations($CD4^+,\;CD8^+,\;CD28^+,\;B220^+) have determined in the lymphoid tissue of mice subjected to irradiation. The relative representation of these subpopulations was significantly different from that in nonirradiated control. $CD4^+\;and\;CD8^+$ cells were present in very low numbers whereas the $B220^+$ cells reached more than normal range at 2 weeks after BMT. The number of $CD4^+$ cells returned to normal relatively soon than $CD8^+$ cell. At week 4 after BMT, the cellularity and composition of spleen lymph node and peripheral blood lymphocyte reached about 50% of the normal range therefore we can choose this time point for the other tests of immune function after BMT.

Excessive Dietary Conjugated Linoleic Acid Affects Hepatic Lipid Content and Muscular Fatty Acid Composition in Young Chicks

  • The effects of dietary conjugated linoleic acid (CLA) on lipid concentrations and fatty acid composition of various tissues were studied in young chicks. From 7 days of age, a total of 160 chicks were divided into 4 groups, placed into 4 pens per group (10 birds per pen) and fed one of four experimental diets containing 6% tallow (TO 6%), 4% tallow plus 2% CLA (TO 4%-CLA 2%), 2% tallow plus 4% CLA (TO 2%-CLA 4%) or 6% CLA (CLA 6%) for 3 weeks. There were no significant differences in growth performances and the relative weights of various organs, but relative liver weight of chicks fed dietary CLA at 4 and 6% levels was significantly higher (p<0.05) than that of TO 6% group. The chemical compositions of leg muscle were not affected by CLA feeding. However, hepatic total lipid of chicks fed 6% CLA diet was significantly higher (p<0.05) than those of TO 6% and TO 4%-CLA 2% groups. The concentrations of various lipid fractions in serum were not affected by CLA feeding. With the increase in dietary CLA levels, cis 9-trans 11 CLA, trans 10-cis 12 CLA and total CLA of leg muscle increased linearly. The relative proportions of C18:1 $\omega$ -9 and C20:4 $\omega$-6 fatty acids in the leg muscles of chicks fed the CLA containing diets were significantly lower (p<0.05) than those of TO 6% group. These results indicate that the levels of CLA isomers were increased linearly in dose-dependent manner after feeding of synthetic CLA source. But it was also observed that excessive amount of dietary CLA resulted in the possible adversely effects, such as increase of liver weight, hepatic lipid accumulation and serum GOT level.

Changes in the metabolic profile and nutritional composition of rice in response to NaCl stress

  • Salinity is a major abiotic stress that adversely affects crop productivity and quality. In this study, the metabolic profile and nutritional composition of rice in response to NaCl were analyzed. The plants were exposed to stressed or unstressed conditions, and their metabolic changes were examined in the shoots, roots, and grains collected at different growth stages. The levels of nutrients and anti-nutrients, including proximates, amino acids, fatty acids, minerals, vitamins, and phytic acid, were also determined for the grains. Application of NaCl significantly decreased the shoot and root growth and induced metabolic alterations at the tillering stage. During the heading stage, only the root metabolites were influenced by NaCl, and no metabolic variations related to salinity were found in the shoot, roots, and grains at the ripening stage. Nutritional analysis of the grain samples revealed that the amounts of linolenic acid and tricosanoic acid were significantly reduced while those of copper, sodium, and phytic acid were enhanced in response to stress. However, except for sodium, those differences were not great. Our results suggest that although NaCl-salinity influences the phenotypic and metabolic profiles of rice shoots and roots at the tillering stage, this impact becomes negligible as tissue development proceeds. This is especially true for the grains. Compositional analysis of the grains indicated that salinity induces some changes in fatty acids, minerals, and anti-nutrients.
