A Lab-Made Wound Maker for Analysis of Cell Migration in a 96-Well Plate (세포 이동능력 분석을 위한 96-Well Plate 전용 Lab-Made Wound Maker)

  • Cell migration is a central process for recovering from wounds triggered by physical distress besides embryogenesis and cancer metastasis. Wound healing assay is widely used as a fundamental research technique for investigation of two-dimensional cell migration in vitro. The most common approach for imitating physical wound in vitro is mechanical scratching on the surface of the confluent monolayer by using sharp materials. The iron metal pin with a suspension spring for fine adjustment of the orthogonal contact surface between the scratching point and the individual bottom of multi-well plate with planar curvatures were adopted for the creative invention of a 96-well plate wound maker. While classic tips drew diverse and zigzag scratching patterns on the confluent monolayer, our wound maker displayed synchronized linear wounds in the middle of each well of a 96-well plate that was seeded with several cell lines. Given that several types of multi-well plates commercially available are compatible with our lab-made wound maker for creating uniform scratches on the confluent monolayer for the collective cell migration in wound healing assay, it is certain that the application of this wound maker to the real-time wound healing assay in high content screening (HCS) is superior than utilization of typical polypropylene pipette tips.

Introduction of Water Quality Management in Korean Pond Ecosystems (국내 연못생태계의 수질관리)

  • This study was conducted from July to December 2004, in order to find measures to improve the water quality and the function of pond ecosystems which are often negatively affected by phytoplankton and suspended particle matters. Most of the time, the management of ponds in Korea does not consider the ecological concepts such as self-purification and nutrient recycling. Instead, conventional methods depend on other factors such as fish farming and the function of fountains and waterfalls. Sustainability of pond ecosystems must be studied with the highest emphasis given to water quality. Water discharges of inflow and outflow as well as balance adjustment for different factors must be thoroughly studied. There is a great need for studies on sustainability because it can be realized through the horizontal or vertical balances of an ecosystem. Our current research offers useful information to the academia and public on maintaining sustainability in terms of structure and function of the pond.

First Remote Operation of the High Voltage Electron Microscope Newly Installed in KBSI (초고전압 투과전자현미경의 원격시범운영)

  • The high voltage electron microscope (HVEM) newly installed in KBSI is an advanced transmission electron microscope capable of atomic resolution (${\leq}1.2{\AA}$ point-to-point resolution) together with high titling function (${\pm}60^{\circ}$), which are suitable to do 3-dimensional atomic imaging of a specimen. In addition, the instrument can be controlled by remote operation system, named as 'FasTEM' for the HVEM, which is favorable to overcome some environmental obstacles resulting from the direct operation. The FasTEM remote operation system has been established between the headquarter of KBSI in Daejeon and the Seoul branch. The server system in the headquarter has been connected with a portable client console system in the Seoul branch using an advanced internet resource, 'KOREN' of 155 Mbps grade. Most of the HVEM functions essential to do remote operation are available on the portable client console. The experiment to acquire the high resolution image of [001] Au has been achieved by excellent transmission of control signals and communication with the HVEM. Real-time reaction like direct operation, such as controls of the illumination and projection parameters, acquisition and adjustment of each detector signal, and electrical steering of each motor-driven system has been realized in remote site. It is positively anticipated that the first remote operation of HVEM in conjunction with IT infraengineering plays a important role in constructing the network based e-Science Grid in Korea for national user s facilities.

Transplanted and many other machines were developed in the early 1990s. In ginseng breeding only pure line selection was of practical significance several verities were at the stage of seed propagation at ginseng plantations. Mutation breeding (${\gamma}$-ray. X-ray chemicals) was not successful. The research on plantlet formation through tissue culture was a little progressed but still far behind to vegetative propagation. Disease control research was concentrated in the isolation and identification of pathogans. their ecological charactelistics and biological control and soil humigation. Potato root rot nematodes was found and control method was established. Insect and small animal control research was greatly progresses in identification, ecological investigation, and ecological and physical control. Weed control was less important due to the development of mulching method of ridge and ditch. Quality factors of raw ginseng in relation to red ginseng process were extensively studied. Traditional quality measures were elucidated in accordance with modern analytical chemistry resulting in the importance of peptides in the centrat part rather than ginsenosides For large root production growth promoting rootzone micrcorganisms (PGPRM) were isolated and active compounds were identified. Field test on PGPRM was on going. Varictus methods formality improvement through cultivation were developed. Management research of ginseng production was rare Extension was active throuch official and private organizations and through workshop for the extension specialists, and direct lectures to grower's. Extension services made the researcher to understand the existing problems at grower's fields. Research environment for ginseng production was in prime time only for three years when Korea Ginseng Research Institute was established then gradually aggravated.

Deduction of Data Quality Control Strategy for High Density Rain Gauge Network in Seoul Area (서울시 고밀도 지상강우자료 품질관리방안 도출)

  • This study used high density network of integrated meteorological sensor, which are operated by SK Planet, with KMA weather stations to estimate the quantitative precipitation field in Seoul area. We introduced SK Planet network and analyzed quality of the observed data for 3 months data from 1 July to 30 September 2013. As the quality analysis result, we checked most SK Planet stations observed similar with previous KMA stations. We developed the real-time quality check and adjustment method to reduce the error effect for hydrological application by missing and outlier value and we confirmed the developed method can be corrected the missing and outlier value. Through this method, we used the 190 stations(KMA 34 stations, SK Planet 156 stations) that missing ratio is less than 20% and the effect of the outlier was the smallest for quantitative precipitation estimation. Moreover, we evaluated reproducibility of rainfall field high density rain gauge network has $3km^2$/gauge. As the result, the spatial relative frequency of rainfall field using SK Planet and KMA stations is similar with radar rainfall field. And, it supplement the blank of KMA observation network. Especially, through this research we will take advantage of the density of the network to estimate rainfall field which can be considered as a very good approximation of the true value.

Exploring for Impact of Learning Strategies on Participation Level in Online Collaborative Learning Process (온라인 협력학습 과정의 참여 수준에 대한 학습전략의 영향 탐색)

  • This study was conducted to explore the impact of learning strategies on the level of participation in the online collaborative learning process. To do this, We studied 91 students who took professorship courses at A university in the Seoul metropolitan area. At the beginning of the semester, the learning strategies were measured through MSLQ, rehearsal, elaboration, organizing, critical thinking, metacognition, learning management, effort control, peer learning, and seeking help. Next, cooperative tasks were carried out to measure the interaction, and group composition consisted of 4-5 persons. The level of participation was measured by scores given to the messages created for interaction. The process of collaborative learning was divided into the steps of identifying learning goals, learning plans, performing individual learning, sharing learning results, and writing reports. The effects of learning strategies on participation level were analyzed through multiple regression analysis (stepwise selection method). As a result, the learning goal step influenced the highest level of metacognition, and the learning plan is the management of the learning time, the demonstration of the learning execution, the adjustment of the effort, the acquisition of help, the collegial learning, Writing was influenced by organization, elaboration, critical thinking, and critical thinking, metacognition, and elaboration.

Multi-Modal Controller Usability for Smart TV Control

  • Objective: The objective of this study was to suggest a multi-modal controller type for Smart TV Control. Background: Recently, many issues regarding the Smart TV are arising due to the rising complexity of features in a Smart TV. One of the specific issues involves what type of controller must be utilized in order to perform regulated tasks. This study examines the ongoing trend of the controller. Method: The selected participants had experiences with the Smart TV and were 20 to 30 years of age. A pre-survey determined the first independent variable of five tasks(Live TV, Record, Share, Web, App Store). The second independent variable was the type of controllers(Conventional, Mouse, Voice-Based Remote Controllers). The dependent variables were preference, task completion time, and error rate. The experiment consist a series of three experiments. The first experiment utilized a uni-modal Controller for tasks; the second experiment utilized a dual-modal Controller, while the third experiment utilized a triple-modal Controller. Results: The first experiment revealed that the uni-modal Controller (Conventional, Voice Controller) showed the best results for the Live TV task. The second experiment revealed that the dual-modal Controller(Conventional-Voice, Conventional-Mouse combinations) showed the best results for the Share, Web, App Store tasks. The third experiment revealed that the triple-modal Controller among all the level had not effective compared with dual-modal Controller. Conclusion: In order to control simple tasks in a smart TV, our results showed that a uni-modal Controller was more effective than a dual-modal controller. However, the control of complex tasks was better suited to the dual-modal Controller. User preference for a controller differs according the Smart TV functions. For instance, there was a high user preference for the uni-Controller for simple functions while high user preference appeared for Dual-Controllers when the task was complex. Additionally, in accordance with task characteristics, there was a high user preference for the Voice Controller for channel and volume adjustment. Furthermore, there was a high user preference for the Conventional Controller for menu selection. In situations where the user had to input text, the Voice Controller had the highest preference among users while the Mouse Type, Voice Controller had the highest user preference for performing a search or selecting items on the menu. Application: The results of this study may be utilized in the design of a controller which can effectively carry out the various tasks of the Smart TV.

Study on the Adequacy and Improvement of the Threshold Speed of Expressway Congestion (고속도로 정체 기준 속도의 적정성 검토 및 개선 연구)

  • Much time has passed since Korea's expressway congestion-threshold speed was revised in 2011. In the meantime, various expressway environments have changed owing to improved performance of vehicles, expanded operations of transport competition (i.e., the KTX), and increased speed limits along some expressway sections. In addition, the speed that expressway users expect to travel at is also increasing. Therefore, through a survey, this study investigates expressway users' perceptions of congestion, and reviews the adjustment of the expressway speed congestion threshold by analyzing expressway traffic flow. One result of the survey confirms that the threshold speed expressway users consider to be congestion has slightly increased. Analyzing traffic and speed data through a K-means algorithm found that the threshold speed for congestion is 60 km/h. In addition, assuming the congestion threshold speed increase from 40 km/h to 50 km/h and 60 km/h, frequently congested expressway sections are identified, determining that 50 km/h is appropriate as a congestion threshold for proper expressway mobility management.

Analysis and Development of Experimental Method of Charle's Law Applicable to School (학교 현장에 적용 가능한 '샤를의 법칙' 실험방법의 분석 및 개발)

  • In this study, we developed an experimental method of the Charles' law applicable to school. Science textbooks and literatures on this principle were analyzed to extract factors utilized in organizing the experimental setup and method. A combined structure such as with a vial and a glass tube, the former of which is for deciding the total volume and the latter of which is for easy measurement of volume, was better in measurement of volume with temperature rather than a simple structure such as syringe. Use of graduated cylinder as a water bath to control the temperature showed advantage in cooling time than using other bath of larger volume such as a beaker. A liquid drop was used as a plug in the glass tube. This plug has little resistance with the glass wall when the gas volume changes. Water as a liquid drop in the glass tube had a significant effect in volume change of gas due to evaporation, especially in the beginning of the measurement. Glycerol showing negligible effect in volume change was used. This method took about one hour and produced a good linear relationship between the temperature and volume of gas with $R^2$ = 0.999 and absolute zero temperature = $-216.7\;{^{\circ}C}$. The Charles' law experiment developed in this study can be performed with appropriate adjustment of procedure considering the purpose of the curriculum of science and chemistry subject at each school level.

Comparative study of working conditions of Korea and Europe (우리나라와 유럽의 근로환경조사 비교연구)

  • Global competition and unstable economic situation in recent years have brought in adverse working conditions such as proliferation of temporary or part-time workers and new shift system. In this study we use exploratory data analysis method to find the relationship between some of the important variables which are related to working hours in KWCS (Korean Working Conditions Survey) and EWCS (European Working Conditions Survey). Next, working hours are explored in terms of sex, employment type, occupation and industry using 2006, 2010, 2011, 2014 KWCS data as well as 2010 EWCS data. Lastly, respondents are divided into two groups of healthy and nonhealthy workers and their working hours are explored in terms of the same categories as above. Finally, simple regression analysis is used to find the effect of health on weekly working hours and adjustment of the effect of health on weekly working hours is made using multiple regression analysis with some other independent variable such as sex, age and employment type included in the model.
