The aim of this study was to develop a composite human skin equivalent for wound healing. Collagen type1 and acellular dermal matrix powder were utilized as the scaffold with dermal fibroblasts and keratinocytes for the development of a composite human skin equivalent. Fibroblast maintained the volume of composite skin equivalent and also induced keratinocytes to attach and proliferate on the surface of composite skin equivalent. The composite human skin equivalent had a structure and curvature similar to those of real skin. Balb-C nu/nu mice were used for the evaluation of full-thickness wound healing effect of the composite human skin equivalent. Graft of composite skin equivalent on full-thickness wound promoted re-epithelialization and granulation tissue formation at 9 days. Given the average wound-healing time (14 days), the wound in the developed composite skin equivalent healed quickly. The overall results indicated that this three-dimensional composite human skin equivalent can be used to effectively enhance wound healing.

BACKGROUND: Because three-dimensional (3D) models more closely mimic native tissues, one of the goals of 3D in vitro tissue models is to aid in the development and toxicity screening of new drug therapies. In this study, a 3D skin wound healing model comprising of a collagen type I construct with fibrin-filled defects was developed. METHODS: Optical imaging was used to measure keratinocyte migration in the presence of fibroblasts over 7 days onto the fibrin-filled defects. Additionally, cell viability and growth of fibroblasts and keratinocytes was measured using the $alamarBlue^{(R)}$ assay and changes in the mechanical stiffness of the 3D construct was monitored using compressive indentation testing. RESULTS: Keratinocyte migration rate was significantly increased in the presence of fibroblasts with the cells reaching the center of the defect as early as day 3 in the co-culture constructs compared to day 7 for the control keratinocyte monoculture constructs. Additionally, constructs with the greatest rate of keratinocyte migration had reduced cell growth. When fibroblasts were cultured alone in the wound healing construct, there was a 1.3 to 3.4-fold increase in cell growth and a 1.2 to 1.4-fold increase in cell growth for keratinocyte monocultures. However, co-culture constructs exhibited no significant growth over 7 days. Finally, mechanical testing showed that fibroblasts and keratinocytes had varying effects on matrix stiffness with fibroblasts degrading the constructs while keratinocytes increased the construct's stiffness. CONCLUSION: This 3D in vitro wound healing model is a step towards developing a mimetic construct that recapitulates the complex microenvironment of healing wounds and could aid in the early studies of novel therapeutics that promote migration and proliferation of epithelial cells.

Slow Feature Discriminant Analysis (SFDA) is a supervised feature extraction method inspired by biological mechanism. In this paper, a novel method called Two Dimensional Slow Feature Discriminant Analysis via $L_{2,1}$ norm minimization ($2DSFDA-L_{2,1}$) is proposed. $2DSFDA-L_{2,1}$ integrates $L_{2,1}$ norm regularization and 2D statically uncorrelated constraint to extract discriminant feature. First, $L_{2,1}$ norm regularization can promote the projection matrix row-sparsity, which makes the feature selection and subspace learning simultaneously. Second, uncorrelated features of minimum redundancy are effective for classification. We define 2D statistically uncorrelated model that each row (or column) are independent. Third, we provide a feasible solution by transforming the proposed $L_{2,1}$ nonlinear model into a linear regression type. Additionally, $2DSFDA-L_{2,1}$ is extended to a bilateral projection version called $BSFDA-L_{2,1}$. The advantage of $BSFDA-L_{2,1}$ is that an image can be represented with much less coefficients. Experimental results on three face databases demonstrate that the proposed $2DSFDA-L_{2,1}/BSFDA-L_{2,1}$ can obtain competitive performance.
This paper develops a 3D homogenization based continuum damage mechanics (HCDM) model for fiber reinforced composites undergoing micromechanical damage under monotonic and cyclic loading. Micromechanical damage in a representative volume element (RVE) of the material occurs by fiber-matrix interfacial debonding, which is incorporated in the model through a hysteretic bilinear cohesive zone model. The proposed model expresses a damage evolution surface in the strain space in the principal damage coordinate system or PDCS. PDCS enables the model to account for the effect of non-proportional load history. The loading/unloading criterion during cyclic loading is based on the scalar product of the strain increment and the normal to the damage surface in strain space. The material constitutive law involves a fourth order orthotropic tensor with stiffness characterized as a macroscopic internal variable. Three dimensional damage in composites is accounted for through functional forms of the fourth order damage tensor in terms of components of macroscopic strain and elastic stiffness tensors. The HCDM model parameters are calibrated from homogenization of micromechanical solutions of the RVE for a few representative strain histories. The proposed model is validated by comparing results of the HCDM model with pure micromechanical analysis results followed by homogenization. Finally, the potential of HCDM model as a design tool is demonstrated through macro-micro analysis of monotonic and cyclic damage progression in composite structures.
This paper suggests generalize mathematica mode for the benefit of volumetric error analysis of a multi-axis machine tool machining a sculptured surfaces. The volumetric error, in this paper, is defined as a three dimensional error at the cutting point, which is caused by the geometric errors and the kinematic errors of each axis and alignment errors of the cutting tool. The actual cutting position is analyzed based on the form shaping model including a geometric error of the moving carriage, where a form shaping model is derived from the homogeneous transformation matrix. Then the volumetric error is obtained by calculating the position difference between the actual cutting position and the ideal one calculated from a Nonuniform Rational B-Spline named as NURES. The simulation study shows the effectiveness for predicting the behavior of machining error and for the method of error compensation.
In the present analysis, the spline finite strip with higher-order shear deformation is formulated for the static analysis of piezoelectric composite plates. The proposed method incorporates Reddy's third-order shear deformation theory, Touratier's "Sine" model, Afaq's exponential model, Cho's higher-order zigzag laminate theory, as well as the classic plate theory and the first-order plate theory. Thus, the analysis can be conducted based on any of the above-mentioned theories. The selection of a specific method is done by simply changing a few terms in a 2 by 2 square matrix and the results, obtained according to different plate theories, can be compared to each other. Numerical examples are presented for piezoelectric composite plates subjected to mechanical loading. The results based on different shear deformation theories are compared with the three-dimensional solutions. The behaviours of piezoelectric composite plates with different length-to-thickness ratios, fibre orientations, and boundary conditions are also investigated in these examples.
A new model of generalized thermoelasticity equations for isotropic media with temperature-dependent mechanical properties is established. The modulus of elasticity is taken as a linear function of reference temperature. The present model is described both generalizations, Lord Shulman (L-S) theory with one relaxation time and Green-Lindsay (G-L) with two relaxation times, as well as the coupled theory, instantaneously. The method of the matrix exponential, which constitutes the basis of the state space approach of modern control theory, applied to two-dimensional equations. Laplace and Fourier integral transforms are used. The resulting formulation is applied to a problem of a thick plate subject to heating on parts of the upper and lower surfaces of the plate that varies exponentially with time. Numerical results are given and illustrated graphically for the problem considered. A comparison was made with the results obtained in case of temperature-independent modulus of elasticity in each theory.

The Gaussian geostatistical model has been widely used for modeling spatial data. However, this model suffers from a severe difficulty in computation because inference requires to invert a large covariance matrix in evaluating log-likelihood. In addressing this computational challenge, three strategies have been employed: likelihood approximation, lower dimensional space approximation, and Markov random field approximation. In this paper, we reviewed statistical approaches attacking the computational challenge. As an illustration, we also applied integrated nested Laplace approximation (INLA) technology, one of Markov approximation approach, to real data to provide an example of its use in practice dealing with large spatial data.
