Isolation and Characterization of a New Hydrogen Sulfide-Oxidizing Bacterium Thiobacillus Sp. (황화수소 산화세균인 새로운 Thiobacillus sp.의 분리 및 특성)

  • A new hydrogen sulfide-oxidation bacterium, Thiobacillus sp. was isolated from waste coal mine water around Hawsun in Chunnam province. The isolate was motile gram-negative rod shape, formed spore and grew up to be aerobically facultative chemolithotroph by using energy released from the oxidation of reduced inorganic sulfur compounds. It could assimilate various kinds of organic compounds and grew well upon thiosulfate-supplemented basal medium. To the lelvel of 32 mM in thiosulfate concentration, thiosulfate in itself was utilized as energy source for growth. However, from those of the higher concentration than 32 mM, thiosulfate functioned specifically as the substrate inhibitor rather than as the energy source. It was found that the optimum thiosulfate concentration for growth was 32 mM. The G+C content of the DNA was 65.0 mol%. The isolate had 16 : 1 + 17$_{cyc}$, 16 : 0 as their major non-hydroxylated cellular fatty acids, 3-OH 12 : 0 as a hydroxylated fatty acid and also contained unidentified $C_{18}$ branched fatty acid. The ubiquinone system in the respiratory chain was Q-9. Based on the physiological and biochemical characteristics, the isolate was assigned to a novel species of the genus Thiobacillus sp. iw.

Variation of Characteristics on the Surface of Pyrite as Microbial Leaching by Thiobacillus ferrooxidans Progresses (Thiobacillus ferrooxidans에 의한 Pyrite의 생물학적 침출에 따른 기질 표면 특성변화)

  • The leaching effect of Thiobacillus ferrooxidance (ATCC 19859) upon polished pyrite ore in 9K medium at $30^{circ}C$ for 30 days was investigated. The surface atomic ratios for Fe, S, Al, Si, and Cu were analyzed by EPMA using fresh and leached samples. The atomic ratio of Fe and S were changed to Fe rich phase as leaching progressed over 13 days but the Fe/S ratio became constant between 13 and 30 days. SEM imaging showed that $10\mum$ oblong shapes formed on the surface after 13 days and that these further developed until 23 days. Fe, S and K atomic ratios were analyzed by SEM-EDS.

Immunological Identification of Thiobacillus ferro-oxidans and Thiobacillus thiooxidans (Thiobacillus ferrooxidans와 Thiobacillus thiooxidans의 면역학적 동정)

  • Detergent soluble fractions were obtained from T. ferrooxidans ATCC 13598 and the T. thiooxidans ATCC 8085 which were treated with 3% of Tween 20. The detergent soluble antigen(crude antigen) fractions of the T.ferrooxidans and the T.thiooxidans were subjected to hydroxyapatite. In the case of T.thiooxidans, further purification was carried out on the DEAE-cellulose column chromatography. The antigen fractions, such as the hydroxyapatite peak-1(Tf, HA-1) and peak-2 from T.ferrooxidans(Tf. HA-2) and hydroxyapatite peak-1(Tt, HA-1), DEAE-cellulose peak-1(Tt, DP-1) and peak-2(Tt, DP-2) from T. thiooxidans wre compared each other with the homologous and the heterologous and the heterologous antisera against to the Thiobacillus species. The hydroxyapatite peak-2 fraction from the T.ferrooxidans(Tf, HA-2) and DEAE-cellulose peak-2 fraction from the T.thiooxidans(Tt, DP-2) were represented the type-specific immuno-reactivities between the T.ferrooxidans and the T.thiooxidans on the several sets of double gel diffusioin analysis. The type-specific antigen fractions from both of the baceteria were mainly composed of protein with entierly different electrophoretic mobility on the SDS-polyacrylamide gel electrophoresis. However, the PAS positive banding patterns on the electrophorogram showed wide range of common antigenic properties in the T. ferrooxidans and the T.thiooxidans, respectively.

  • Thiobacillus neopolitanus R-10, which produces a active thiosulfate oxidase was isolated from nightsoil. The optimal culture conditions of Thiobacillus neopolitanus R-10 for the production of enzyme was determined as followed: 0.8% $Na_2S_2O_3$, 0.2% $KH_2PO_4$, 0.2% $K_2HPO_4$, 0.04% $Na_2CO_3$, 0.02% $MgSO_4$.$7H_2O$, 2ml trace elements solution, ann PH 6.5 at 3$0^{\circ}C$ and 72hr cultivation. The oxidase was successively purified 83 folds yield by ${(NH_2)}_2SO_4$ fractionation, DEAE-Cellulose, Sephadex A-50 column chromatogrophy and gel Sephadex G-150 gel filteration with yield of 5.9%. The molecular weight of purified enzyme was estimated to be 43.000 dalton by SDS-polyacrylamide gel electrophoresis and gel filteration column chromatography The enzyme activity was highest at 40t and PH 7.0 The enzyme activity was relatively high by $\beta$-mercaptoethanol but strongly inhibited by cysteine.

  • The synthesis of well-defined dendrons based on aliphatic polyether dendritic cores and glassy polystyrene peripheries is described. The synthetic route involves a combination of living anionic polymerization and a stepwise convergent method consisting of iterative Williamson etherification and hydroboration/oxidation reactions. On the basis of molecular weight, as characterized by gel permeation chromatography (GPC), the first generation dendron (Generation-1) shows a random coil conformation like a linear polystyrene, while higher generations (Generation-2 and 3) reveal globular forms in solution.

  • In order to recover valuable metals from the waste electronic scrap, bioleaching of Cu, Zn, Al, Co, Ni, Sn and Pb was carried out using Thiobacillus ferrooxidans as a leaching microorganism in a shaking flask. In a preliminary study, to obtain the data on the leaching of Cu, Zn, Al, Co and Ni, the metal leaching was accomplished using metal powers instead of electronic scrap. The leaching percentaga of Cu, Zn, Co, Al and Ni powers was reduced with the increase of metal power concentration in solution. Below the metal concentration of 0.5 g/L, more than 85% of Cu, Co and Zn powers was leached out. At the electronic scrap concentration of 100 g/L, Thiobacillus ferrooxidans were able to leach more than 90% of the available Cu and Co while Al, Zn and Ni were able to leach less than 40%. Pb and Sn were not detected in the leachate. Pb was precipitated as PbSO$_4$, whereas Sn precipitated probably as SnO.

Simultaneous Removal of Hydrogen Sulfide and Ammonia Using Thiobacillus sp. IW in a Three-Phase Fluidized-Bed Bioreactor

  • A three-phase fluidized-bed bioreactor including Thiobacillus sp. IW was tested to remove H_2S and $NH_3$ simultaneously. The inlet $H_2S$ was oxidized to $SO_4^{2-}$ by Thiobacillus sp. IW, and the $NH_3$ reacted with the $SO_4^{2-}$ to form $(NH_4)_2SO_4$. The removal efficiency of $H_2S$ was 98.4-99.9% for an inlet concentration of 36-730 ppm and that of $NH_3$ was 60.2-99.2% for an inlet concentration of 45-412 ppm. The removal efficiency of $NH_3$ was reduced when the inlet loading rate of $NH_3$ was increased above 10 mg/l/h. When the bioreactor was operated for 25 days with a lower inlet concentration of $NH_3$ compared with the of $H_2S$, the bioreactor exhibited an excellent performance with a stable pH, dissolved oxygen content, and cell concentration.

  • Methyl mercaptan oxidase was isolated and purified from Thiobacillus thiooxidans KCTC2505 for the detection of mercaptans. T The produre of purification involved DEAE-Sephacel and Superose 12 column chromatographies with recovery yields of 40 a and 6.3%, and specific activity of 19.7 and 80.1 units/mg-protein, respectively. The molecular weight of purified methyl m mercaptan oxidase was determined to be 68.1 kDa by SDS-PAGE. The extract from DEAE-Sephacel column chromatography h had a high activity in oxidizing methyl mercaptan to produce formaldehyde which can be easily detected by purpald-coloring m method. Optimum temperature for activity was observed at $43^{\circ}C$. This enzyme was activated by $NH_4CI and (NH_4)_2S0_4$, and | inhibited by KCI and NaC!.

  • To reducing offensive odor form compost UIW-6 mutant obtained by UV treatment from sulfur-oxidizing bacteria, Thiobacillus sp. IW. The UIW-6 mutant was found 1.6 times faster in growth than the parent strain on thiosulfate medium (TM) at 36 h after incubation. Initial pH, temperature and agitation for the optimum growth of UIW-6 were 6.5, $35^{\circ}C$ and 200 rpm, respectively. The UIW-6 mutant growth was two times higher than parent strain at 6 h culture in TM liquid medium containing 50 mM sodium thiosulfate. The UIW-6 mutant used fructose and sucrose as carbon sources and yeast extract> tryptone> peptone as nitrogen ones. It was found that the growth of UIW-6 was increased in addition of 0.2% yeast extract.

Effect of Sludge Concentration on Removal of Heavy Metals from Digested Sludge by Thiobacillus ferrooxidans (Thiobacillus ferrooxidans를 이용한 소화 슬러지의 중금속 제거에 미치는 슬러지 농도의 영향)

  • To investigate the feasibility of the microbial process for removal of heavy metals from the high solid content sludge, the effect of sludge concentration on the solubilization of heavy metals by an iron oxidizing bacterium Thiolbacillus ferrooxidans was examined. With increasing the sludge concentration, the removal efficiency of heavy metals and the oxidation rate of iron were inhibited. Especially, when the sludge concentration is over 5% (w/v), the activity of T. ferrooxidans was remarkably inhibited. This inhibition is considered to occur due to the dissolved inhibitory materials such as organic compounds, heavy metals, and others which were extracted from the sludge during incubation period. In conclusion, the microbial process by T. ferrooxidans is only effectively used in ranges of 1.3 to 4.0% (w/v) sludge concentration.
