A study on the Detection of Artificial Dyes in the Commercial Jellys by Use of Paper Chromatography and Thin-Layer Chromatography. (Paper chromatography와 Thin-layer chromatography에 의한 시판 Jelly류의 착색료에 관한 고찰)

  • A study was carried out to detect of illegal artificial dyes and to confirm the used rate of illegal dyes in the production process of commercial jellys by thindayer chromatography and paper chromatography, from March, 1976 to July, 1976. The following conclusions were obtained 1. Seperated dyes were amaranth, erythrosin, tatrazin, sunset yellow FCF, fight green SF yellowish, fast green FCF and the most frequent use of amaranth. 2. Used rate of legal dyes were 94.12% (96 samples) and illegal dyes were 5.88% (6 samples) with samples 102. 3. The average Rf value of T.L.C. were amaranth (0.92), erythrosin (0.48), tatrazin (0.83), sunset yellow FCF(0.86), fast green FCF (0.63), light green SF yellowish (0.23) and paper chromatography were amaranth (0.76), erythrosin (0.44), tatrazin(0.75), sunset yellow FCF (0.76), fast green FCF (0.86), light green SF yellowish(0.52).

A Study of Glutathione S-transferase Inhibitors obtained from Allium cepa var. cepa Extract (양파 추출물에서의 글루타티온 전달효소 활성 저해제에 관한 연구)

  • In this study, an ethanol extract being obtained from Allium cepa var. cepa examins the inhibitory effects on the glutathione S-transferase and the separation had been done by silica-gel column chromatography using various eluents, such as ethyl acetate, methanol, and 50% methanol. A volume of column fraction was 50ml and evaporation has been performde by the rotary evaporator under reduced pressure. Each fraction is being examined by thin layer chromatography and the UV spectrum at 365 nm was used to investigate separation patterns of spots on thin layer chromatography. When the eluent was changed, the spot patterns showed another different pattern on thin layer chromatography, so on. Fractions showing similar pattern are combined and eventually, three fractions are obtained. Each fraction is testified to examine the inhibition effects on glutathione S-transferase. All of these showed inhibition effects on glutathione S-transferase. The GC-MS shows that each fraction contains more than 2 compounds.

  • Objective: The purpose of this study is to expatiate on high performance thin layer chromatography (HPTLC) as a simple, easy and scientific method for evaluation and standardization of herbal formulae. Methods: Through retrieving HPTLC application for herbal formulae in the literatures, the current situation of HPTLC application, and potential as well as limitation of HPTLC as a standardization method for multi-herbal drug was studied. Results: HPTLC is a speedy, inexpensive and well-operable tool for possessing multi-capability on component identification, separation, quantification and purification compared to other methods, such as high performance liquid chromatography (HPLC), gas chromatography (GC). Conclusions: HPTLC is considered as an available and convenient method for quality control and standardization of multi-herbal drugs. Thereby, this method could be recommended to widely applicate in the traditional Korean medicine.

High Purity Extraction and Simultaneous High-performance Liquid Chromatography Analysis of Curcuminoids in Turmeric (강황에서 curcuminoids의 고순도 추출 및 고성능 액체 크로마토그래피 동시분석)

  • Three major curcuminoids in turmeric (Curcuma longa), curcumin, demethoxycurcumin, and bisdemethoxycurcumin, were efficiently extracted by optimizing extraction condition and simultaneously analyzed and identified using reverse phase high-performance liquid chromatography, thin-layer chromatography, and liquid chromatography-mass spectrometry method. The highest yield of extraction amount 0.279 g, 9.30% was obtained by dipping method with extraction time of 7 h.

  • Tissue extracts were prepared from liquid-cultured Arabidopsis and reacted with UDP-[$^3H$]-GlcNAc. Phospholipid fractions were then extracted by butanol partitioning. Consecutive thin-layer chromatography identified two glycolipids sensitive to PI-specific phospholipase C, known as early intermediates in glycosylphosphatidylinositol anchor biosynthesis; phosphatidylinositol N-acetylglucosamine and phosphatidylinositol glucosamine.

  • As the projects of this institute, 280 species of plants which are currently used as herb drugs in Korea were screened on the presence of alkaloids, phenolic compounds, flavonoids, chalcones, lactones, glycosides, carbohydrates, terpenoids, steroids, proteins, polypeptides, saponins, and organic acids, and the most reliable presence of alkaloids detected by thin layer chromatography is added by screening of 75 species.

  • The resolving capacities of xanthophyll pigments on thin-layers of Silica Gel, Hyflo super-Cel, and Micro-Cel C with varying concentrations of acetone in petroleum ether as the developing solvent were compared. The results showed that the resolving capacity of Micro-Cel C thin-layer was superior to others and satisfactory for the separation of leaf carotenoids in clearly separated six bands; carotenes, lutein-zeaxanthin, antheraxanthin, violaxanthin, an unidentified band, and neoxanthin, when it was developed with $13\%$ acetone-petroleum ether solution for 15 to 20minutes in an unsaturated chamber. Adhension of Micro-Cel C to glass was adequate without binder. Calcium sulfate used as a binder appeared to inactivate the resolving capacity of Micro-Cel C. Removing an about 0.2cm-wide layer on bo side of thin-layer slide helped to prevent 'edge effect' which gave tailing and faster solvent ascending along the side than the center. An adequate thickness of thin-layer was obtained when a 3 ml aliquot of the suspension in which l0g powdered Micro-Cel C was suspended in 75 ml distilled water was coated on a $2\times20cm$ glass slide.

  • This study was carried out to confirm the agent responsible for the antibacterial activity in milk culture or Lactobacillus bulgaricus to extract and purify it. The following results were summarized as followings : The antibacterial agent was extracted from the cultured skim milk with methanol and acetone and was purified by Sephadex G-50 gel filteration and thin layer chromatography on silica gel. The antibacterial substance other than lactic acid was confirmed by turbidimetric technique using the neutralized culture filtrate which inhibited the growth of Bacillus subtilis. The purified agent showed inhibitory activity against Bacillus subtilis, Escherichica coli, Pseudomonas fluorescens Staphylococcus aureus, Proteus vulgaris and Shigells $dysenteria^2$. The agent obtained from thin layer chromatography was free from $H_2O_2$ or lactic acid.

  • This study was identified 6 species of the genus Acer in Korea by the method of thin layer chromatography on lipids in heart wood, and researched affinity among the species for the characteristics of color reaction. The results were summarized as follows; 1) Total-lipid in heart wood of the six species was 4.39%, mean free-lipid was 2.85% and mean bound-lipid was 1.54%. 2) All the six species; A. ginnala, A. mono, A. negundo, A. palmatum, A. saccharinum and A, triflorum were identified by the characteristics of color reaction. Judging from the color reactions, A. palmatum and A. triflorum seem to be closer than other species.
