Experimental Assessment of Mesophilic and Thermophilic Batch Fermentative Biohydrogen Production from Palm Oil Mill Effluent Using Response Surface Methodology

  • The present work evaluated the production of biohydrogen under mesophilic and thermophilic conditions through dark fermentation of palm oil mill effluent (POME) in batch mode using the design of experiment methodology. Response surface methodology (RSM) was applied to investigate the influence of the two significant parameters, POME concentration as substrate (5, 12.5, and 20 g/l), and volumetric substrate to inoculum ratio (1:1, 1:1.5, and 1:2, v/v.%), with inoculum concentration of 14.3 g VSS/l. All the experiments were analyzed at 37 ℃ and 55 ℃ at an incubation time of 24 h. The highest chemical oxygen demand (COD) removal, hydrogen content (H2%), and hydrogen yield (HY) at a substrate concentration of 12.5 g COD/l and S:I ratio of 1:1.5 in mesophilic and thermophilic conditions were obtained (27.3, 24.2%), (57.92, 66.24%), and (6.43, 12.27 ml H2/g CODrem), respectively. The results show that thermophilic temperature in terms of COD removal was more effective for higher COD concentrations than for lower concentrations. Optimum parameters projected by RSM with S:I ratio of 1:1.6 and POME concentration of 14.3 g COD/l showed higher results in both temperatures. It is recognized how RSM and optimization processes can predict and affect the process performance under different operational conditions.

Effects of Temperature on Production of Hydrogen in Anaerobic Fermentation (혐기성 발효에서 수소 생산 시 온도의 영향에 관한 연구)

  • This study was conducted to examine temperature effects on hydrogen production in anaerobic fermentation. 18 batch reactors were operated at mesophilic ($35^{\circ}C$) and thermophilic conditions ($55^{\circ}C$) to achieve maximum hydrogen production in anaerobic fermentation. Optimum hydrogen production conditions were also investigated at each temperature. Different trends were observed regarding pH effects on hydrogen production. This effect was not significant for mesophilic fermentation ($35^{\circ}C$). In this case, pH may not drop to interfere hydrogen production during the test. However, hydrogen production decreased without pH control for thermophilic condition ($55^{\circ}C$). Effects of heat treatment were observed for both fermentation process. Hydrogen production with heat treatment was higher than hydrogen production without heat treatment for both fermentation processes. The amount of produced hydrogen for each substrate concentration with temperature changes showed that more hydrogen was produced at $35^{\circ}C$ than at $55^{\circ}C$.

Membrane fouling in thermophilic membrane bioreactor with different sludge retention times

  • As membrane fouling is based on various factors, it is a complex phenomenon that is hard to estimate. This study investigated membrane fouling in a thermophilic jet loop membrane bioreactor (JLMBR). With this purpose, four different empirical membrane fouling models with different sludge retention times were applied on the flow data obtained in the system. As a result of the model implementation, it was found for all sludge retention times that, standard blocking is effective in the first 1.5 hours of filtration, while cake filtration was dominant in the remaining duration. Additionally, it was observed that as the sludge retention time increases, membrane fouling rate decreases.

Effects of Iron-Reducing Bacteria on Carbon Steel Corrosion Induced by Thermophilic Sulfate-Reducing Consortia

  • Four thermophilic bacterial species, including the iron-reducing bacterium Geobacillus sp. G2 and the sulfate-reducing bacterium Desulfotomaculum sp. SRB-M, were employed to integrate a bacterial consortium. A second consortium was integrated with the same bacteria, except for Geobacillus sp. G2. Carbon steel coupons were subjected to batch cultures of both consortia. The corrosion induced by the complete consortium was 10 times higher than that induced by the second consortium, and the ferrous ion concentration was consistently higher in iron-reducing consortia. Scanning electronic microscopy analysis of the carbon steel surface showed mineral films colonized by bacteria. The complete consortium caused profuse fracturing of the mineral film, whereas the non-iron-reducing consortium did not generate fractures. These data show that the iron-reducing activity of Geobacillus sp. G2 promotes fracturing of mineral films, thereby increasing steel corrosion.

  • 1. The preparation and some enzymatic properties of crude alkaline protease from a thermophilic mold, Myriococcum sp. was investigated. 2. Optimum pH for the hydrolysis of casein was 9.0 at $50^{\circ}C$ for 10 minutes. Optimum temperature was $55^{\circ}C$ at pH 9.0 for 10 minutes. The enzyme was highly stable at the range of pH 6.0 to 11.0 at $30^{\circ}C$ 3. The alkaline protease in the culture filterate was isolated two fractions by elution column chromatography on DEAE-Cellulose.

  • The conditions for the $\beta$-fructofuranosidase production from alkalophilic, thermophilic Bacillus sp. TA-11 were investigated. The maximal enzyme production was obtained when the strain was cultured at 50$\circ$C for 36 hrs with batch culture in the optimal medium containing 1.0% sucrose, 0.6% yeast extract, 0.1% each of KH$_{2}$PO$_{4}$, K$_{2}$HP0$_{4}$, NaH$_{2}$PO$_{4}$, and initial pH 9.5, and the final enzyme activity under the above condition was 102 units per ml of cell free extract.

Fluorescence in situ hybridization using 16S rRNA-targeted oligonucleotides reveals abundance and spatial organization of methanogens in thermophilic sludge granules

  • In situ hybridization with fluorescent oligonucleotides(FISH) was used to detect and localize microorganisms in the granules of lab-scale upflow anaerobic sludge blanket(UASB) reactors. An UASB reactor was seeded with mesophilically-grown($35^{\circ}\;C$) granular sludge, and thermophilically($55^{\circ}\;C$) operated by feeding with a synthetic wastewater. Sections of the granules were hybridized with 16S rRNA-targeted oligonucleotide probes for Eubacteria, Archaeabacteria, and specific phylogenetic groups of methanogens. FISH clearly showed the layed structure of thermophilic granules, which was consisted of outer bacterial cells and inner archaeal cells. Methanoseata-, Methanosarcina-like cells were also found to be localized inside the granules. These results demonstrated FISH was useful in studying the spatial organizations of methanogens and in situ morphologies and metabolic functions in thermophilic granular sludges.

Crystal Structure of TTC0263, a Thermophilic TPR Protein from Thermus thermophilus HB27

  • The hypothetical protein TTC0263 of Thermus thermophilus HB27 is a thermophilic tetratricopeptide repeat (TPR)-containing protein. In the present study, the TPR region (residues 26-230) was resolved at $2.5{\AA}$ with R-factors of $R/R_{free}$ = 23.6%/28.6% $R/R_{free}=23.6%/28.6%$. TTC0263 consists of 11 helices that form five TPR units. Uniquely, it contains one atypical "extended" TPR (eTPR) unit. This comprises extended helical residues near the loop region of TTC0263, such that the helical length of eTPR is longer than that of the canonical TPR sequence. In addition, the hybrid TPR domain of TTC0263 possesses oligomer-forming characteristics. TPR domains are generally involved in forming multi-subunit complexes by interacting with each other or with other subunit proteins. The dynamic structure of TTC0263 described here goes some way to explaining how TPR domains mediate the formation of multi-subunit complexes.

Degradation of Dibenzothiophene, and Desulfurization of Crude Oil and Bunker C Oil by Sulfate Reducing Bacteria (황산염 환원세균에 의한 Dibenzothiophene, 원유 및 Bunker C 유의 탈황)

  • Dibenzothiophene, crude oil and bunker C oil were used in the microbial desulfurization experiments using thermophilic and mesophilic strains of Desulfovibrio and Desulfotomaculum. Mesophilic Desulforvibrio desulfuricans M6 showed the degrees of sulfur removal about 42% and 17% from dibenzothiophene and crude oil, respectively. Thermophilic Desulfovibrio thermophilus showed the degrees of sulfur removal about 68% and 33% from dibenzothiophene and bunker C oil. The strains of Desulfotomaculum were much less efficient than strains of Desulfovibrio. The latter have more complex and stronger gydrogen metabolism. These results showed that desulfurization is closely related to the hydrogen metabolism of the sulfate reducing bacteria.

  • In both untreated and conventionally stabilized swine manures antibiotic resistant (AR) microorganisms, Staphylococcus-like and Salmonella-like microorganisms were detected. Also pathogens with MAR phynotype were detected. Presence of such microorganisms suggest high level of pathogen-related health risk to farmers who may be in direct contact with the manure and its conventionally stabilized product In contrast the autothermal thermophilic aerobic digestion (ATAD) treatment have efficiently reduced AR and pathogenicity from the swine manure. When soil was fertilized using swine manure and its stabilized products, despite no detection of MAR-exhibiting pathogen-like microorganisms in fertilized soil, potential pathogen-related health risk could not be ruled out from the fertilized soil since the organic fertilization led to increase in AR and pathogenicity in the soil microbial communities. As conclusion, this microbiological study demonstrated that an ATAD process is applicable in control of pathogen-related health risk in livestock manure.
