Effects study of Aconiti Lateralis Radix Preparata extract on the regulation of heat and cold in PTU-induced hypothyroidism rats (포부자추출물의 갑상선기능저하증 흰쥐모델에서의 한열조절작용에 의한 개선효능 연구)

  • Objective : To suggest a scientific evidence of Aconitum carmichaeli Debx. (Aconiti Lateralis Radix Preparata: ALRP) as one of cooling and heating medicines on the regulation of body temperature, we investigated the effects of ALRP water extract on hypothyroidism. Methods : Hypothyroidism was induced by intradermal injection with PTU for 4 weeks in SD rats. ALRP extract or L-thyroxine as a control drug was orally administrated for 2 weeks with PTU injection in rats. The physiological and serological parameters were measured in rats. The histological change of thyroid tissues was observed by H&E staining, and also the expression of thermo-regulating proteins was determined by Western blot in dorsal root ganglia and brain tissues of rats. Results : The administration of ALRP extract in PTU-induced hypothyroidism rats was significantly increased body temperature, but did not changes on body weight, food and water intake. ALRP extract did not effect on the levels of TSH and T4 in the hypothyroidism rats. ALRP extract significantly decreased the levels of GPT, glucose, HDL-cholesterol, LDL-cholesterol, and total cholesterol in the hypothyroidism rats. In histological observation, the enlarged epithelium and atrophic follicles with higher concentration of follicular cells on hypothyroidism were improved by ALRP extract. In addition, ALRP extract increased the expression of TRPV1 and TRPM8 ion channel proteins in hypothyroidism rats. Conclusion : These results indicate that ALRP extract can improve PTU-induced hypothyroidism through regulation of body temperature and lipid accumulation. The action mechanism of ALRP extract is related with body temperature control by thermoregulation with TRP ion channels.

Automatic Fire Detector Spacing Calculation for Performance Based Design (성능위주설계를 위한 화재감지기배치의 공학적연구)

  • Placement method for fire detectors prescribed in current fire safety regulation is just about placing a prescribed number of detectors according to the areas. However, this regulation has no scientific basis and standards from foreign countries are just introduced and fire detectors are installed complying with them. There are two standards in designing fire protection systems; Prescriptive-Based Design that follows stipulated regulations like fire safety standards and Performance-Based Design based on engineering knowledge such as fire dynamics, structural dynamics, mechanics of materials, fluid mechanics, and thermo dynamics. Recently, Fire Protection System Construction Business Act was revised so that fire protection systems can be designed using Performance-Based Design method ('05. 8. 4), though the method has not activated until now. In addition, the enforcement decree defines the range for specific objects of fire protection to which Performance-Based Design is applied ('07, 1. 24). At the moment, by manufacturing simulator so that formulas can be introduced and calculated with software in order to install fire detector of automatic fire detection systems keeping optimized distance, comparing the results with the state of fire detector placed according to Performance-Based Design and analyzing them, this study was intended to settle Performance-Based Design method in the future.

Diagnostic Utility of p63 (Ab-1) and (Ab-4) Tumor Markers in the Squamous Cell Carcinomas of Head and Neck

  • P63 is a gene product required in cell cycle regulation which plays vital roles in tumor differentiation. Aims of the present study were to assess the frequency, pattern, sensitivity and specificity of two p63 protein clones P63 4A4 and P63 4A4+Y4A3 in squamous cell carcinomas (SCCs). Thirty cases of head and neck region SCC diagnosed on the basis of H&E staining were examined along with 60 cases of head and neck region biopsies other than squamous cell carcinoma, negative on H&E staining, were taken as control. Immunostaining was performed on slides according to the Thermo Scientific UltraVision LP detection System. P63 4A4+Y4A3 clone is more sensitive 96.6% in comparison to 86% in P63 4A4 with having greater NPV of 98.3%. The results signify the importance of P63 4A4+Y4A3 marker over the old markers and may be used as a confirmatory marker of squamous cell carcinoma.

A Study on the Thermo-Mechanical Fatigue Loading for Time Reduction in Fabricating an Artificial Cracked Specimen (열-기계적 피로하중을 받는 균열시편 제작시간 단축에 관한 연구)

  • In the nuclear power plant, early detection of fatigue crack by non-destructive test (NDT) equipment due to the thermal cyclic load is very important in terms of strict safety regulation. To this end, many efforts are exerted to the fabrication of artificial cracked specimen for practicing engineers in the NDT company. The crack of this kind, however, cannot be made by conventional machining, but should be made under thermal cyclic load that is close to the in-situ condition, which takes tremendous time due to the repetition. In this study, thermal loading condition is investigated to minimize the time for fabricating the cracked specimen using simulation technique which predicts the crack initiation and propagation behavior. Simulation and experiment are conducted under an initial assumed condition for validation purpose. A number of simulations are conducted next under a variety of heating and cooling conditions, from which the best solution to achieve minimum time for crack with wanted size is found. In the simulation, general purpose software ANSYS is used for the stress analysis, MATLAB is used to compute crack initiation life, and ZENCRACK, which is special purpose software for crack growth prediction, is used to compute crack propagation life. As a result of the study, the time for the crack to reach the size of 1mm is predicted from the 418 hours at the initial condition to the 319 hours at the optimum condition, which is about 24% reduction.

A Study on Quantitative Wear Training for the Improvement of Heat Tolerance in Summer (Part I) -20's Females' Optimal Wearing: Focus on Inside Clothing Temperatures in Pre-summer- (여름철 내열성 증진을 위한 정량적 착의훈련의 효과 (제1보) -20대 여성의 적정착의 온도 설정: 의복내 온도를 중심으로-)

  • This study suggests quantitative guidelines for inside clothing temperatures to improve the heat tolerance of 20's females in summer. First, the inside clothing temperatures ($T_{cl}$) of each subject was measured in daily use. The subjects were asked to record subjective thermal sensations, clothing items worn, clothing weight, and activities during an experiment designed to determine the comfort zone of $T_{cl}$. In a thermally neutral state, the comfort zone of $T_{cl}$ was decided on a mean value $T_{cl}{\pm}1{\sigma}$. Second, the subjects were asked to wear clothing that would enable them to feel 'slightly warm but still comfortable'. The rest of the processes were the same as previous steps that were designed to understand the way and degree of clothing control. The comfort zone of $T_{cl}$ was decided in the same manner as the previous step. The two comfort zones were combined and named the combined comfort zone of the definitive comfort zone. The results were as follows: 1. Thermally comfortable $T_{cl}$, Hcl were $34.0{\pm}1.1^{\circ}C$, $40{\pm}9%%RH$ and the thermally comfortable ambient climate was $25.0{\pm}1.6^{\circ}C$, $53{\pm}7%$RH. 2. When subjects were asked to wear 'slightly warm but still comfortable', there were difference in thermally comfortable $T_{cl}$, clothing weight and clothing layer by subject. 3. In this study, the optimal $T_{cl}$ was decided on the mid-point of the definitive comfort zone of $T_{cl}$.

Thermal Deformation Induced Preload Changein the Tilting Pad Journal Bearing (열변형으로 인한 틸팅패드 저널베어링의 예압 변화)

  • This paper focuses on the thermal deformation induced preload change in the tilting pad journal bearing, using a three-dimensional (3D) thermo-hydro-dynamic (THD) approach. Preload is considered as a critical factor in designing the tilting pad journal bearing. The initial preload measured under nil external load and nil thermal gradient is influenced by two factors, namely, the thermal deformation and elastic deformation. Thermal deformation is due to a temperature distribution in the bearing pads, whereas the elastic deformation is due to fluid forces acting on the pads. This study focuses on the changes induced in preload and film clearance due to thermal deformation. The generalized Reynolds equation is used to evaluate the force of the fluid and the 3D energy equation is used to calculate the temperature of the lubricant. The abovementioned equations are combined by establishing a relationship between viscosity and temperature. The heat transfer within the bearing pads, the lubricant, and the spinning journal is calculated using the heat flux boundary condition. The 3D Finite Element Method (FEM) is used in modeling the (1) heat conduction in the spinning journal and bearing pads, (2) thermal gradient induced thermal distortion of the spinning journal and pads, and (3) viscous shearing, and heat conduction and convection in a thin film. This evaluation method has an increased fidelity, and it can prove to be a cost-effective tool that can be used by designers to predict the dynamic behavior of a bearing.

Studies on the effects of Bujaleejungtang (부자이중탕(附子理中湯)의 효능(效能)에 관(關)한 실험적(實驗的) 연구(硏究))

  • In order to investigate the effect of Bujaleejungtang, by means of oral medication to rats and mice, to isolated intestine and stomach, and the effect to pyloric ulcer, indomethacin-induced ulcer, secretion of gastric juice, and to transport ability of intestine content were studied as the action to G-I tract. The effect to normal rats and resperpine-treated rats were studied as the action to thermo-regulation. The results were as follows: 1. Bujaleejungtang showed the inhibitory effect on the smooth muscle contraction induced by acethylcholine chloride and barium chloride in the isolated mice ileum. 2. Bujaleejungtang showed inhibitory effect on the contraction induced by acetylcholine chloride and barium chloride in the rat fundus-strip. 3. Inhibitory effect of Bujaleejungtang on pyloric ulcer, indomethacin-induced gastric ulcer in rats was statistical recognized(p<0.05). 4. No inhibitory effect of Bujaleejungtang on gastric juice secretion in Shay rats was recognized. 5. Inhibitory effect of Bujaleejungtang on gastric free acidity and total acidity in Shay rats was recognized only when Bujaleejungtang was medicated in high thickness(2000mg/kg) (p<0.001). 6. Inhibitory effect of Bujaleejungtang on pepsin output in Shay rats was recognized only when Bujaleejungtang was medicated in high thickness(2000mg/kg)(p<0.001). 7. Inhibitory effect of Bujaleejungtang on barium sulfate transport in the small intestine of mice was recognized only when Bujaleejungtang was medicated in high thickness(2000mg/kg)(p<0.05). 8. Inhibitory effect of Bujaleejungtang on barium sulfate transport in the large intestine of mice was recognized(p<0.05). 9. Inhibitory effect of Bujaleejungtang on rectal temperature in normal rats was recognized. 10. Inhibitory effect of Bujaleejungtang on rectal temperature in reserpine-treated rats was recognized only when Bujaleejungtang was medicated in high thickness(2000mg/kg)(p<0.05).

Internal Changes of Blood Compartment and Heat Distribution in Swamp Buffaloes under Hot Conditions : Comparative Study of Thermo-Regulation in Buffaloes and Friesian Cows

  • From previous studies, there is a strong possibility in buffaloes that the marked increase in blood volume (BV) under hot conditions contributes to heat transportation from the rectum to the skin. The present study was done to clarify changes with environmental temperature on water-shift between blood and extracellular fluid (ECF), heat distribution between the rectum and the skin, and blood flow rates (BFR) at the hind legs (reflecting the skin surface). Four buffaloes and four Friesian cows were successively exposed to three different temperatures of $20^{\circ}C$, $30^{\circ}C$ and $35^{\circ}C$. BV and ECF volume were measured with Evans' blue and sodium-thiocyanate dilution methods, respectively. Rectal and subcutaneous (as the skin) temperatures were measured by copper-constantan thermocouples. BFR were measured by a supersonic blood flow meter. With an increase in environmental temperature, skin temperature in buffaloes increased significantly than cows, but rectal temperature was not significantly different between two species. BV, especially plasma compartment, increased significantly in only buffaloes, while ECF volume did not change in both species. BFR increased significantly in buffaloes, but not in cows. From these results, the increased of BV may be caused by water flowing from ECF compartment. The water-shift may induce the increase of BFR and skin temperature. It is suggested in the present study that internal changes of blood compartment in buffaloes contribute to transfer of heat to the skin surface.
