  • Fusarium oxysporum trypsin (FOT) is a fungal serine protease similar to mammal trypsin. The FOT could be successfully expressed in Pichiapastoris by engineering the natural propeptide APQEIPN. In this study, we constructed two recombinant enzymes with engineered amino acid sequences added to the N-terminus of FOT and expressed in P. pastoris. The N-terminal residues had various effects on the structural and functional properties of trypsin. The FOT, and the recombinants TE (with peptide YVEF) and TS (with peptide YV) displayed the same optimum temperature ($40^{\circ}C$) and pH (8.0). However, the combinants TE and TS showed significantly increased thermal stability at $40^{\circ}C$ and $50^{\circ}C$. Moreover, the combinants TE and TS also showed enhanced tolerance of alkaline pH conditions. Compared with those of wild-type FOT, the intramolecular hydrogen bonds and the cation ${\pi}$-interactions of the recombinants TE and TS were significantly increased. The recombinants TE and TS also had significantly increased catalytic efficiencies (referring to the specificity constant, $k_{cat}/K_m$), 1.75-fold and 1.23-fold than wild-type FOT. In silico modeling analysis uncovered that the introduction of the peptides YVEF and YV resulted in shorter distances between the substrate binding pocket (D174, G198, and G208) and catalytic triad (His42, Asp102, and Ser180), which would improve the electron transfer rate and catalytic efficiency. In addition, N-terminal residues modification described here may be a useful approach for improving the catalytic efficiencies and characteristics of other target enzymes.

  • Thermally stable $TiO_2$/Pt/$SiO_2$ core-shell nanocatalyst has been synthesized by chemical processes. Citrated capped Pt nanoparticles were deposited on amine functionalized silica produced by Stober process. Ultrathin layer of titania was coated on Pt/$SiO_2$ for preventing sintering of the metal nanoparticles at high temperatures. Thermal stability of the metal-oxide hybrid catalyst was demonstrated heating the sample up to $600^{\circ}C$ in air and by investigating the morphology and integrity of the structure by transmission electron spectroscopy. The surface analysis of the constituent elements was performed by X-ray photoemission spectroscopy. The catalytic activity of the hybrid catalysts was investigated by CO oxidation reaction with oxygen as a model reaction.

  • In this study, investigated the synthesis method of p-Phenylenediamine (PPD) by amination of p-Diiodobenzene (PDIB) under supercritical ammonia and CuI catalyst conditions. We examined the effects of various process variables (e.g., reaction temperature, pressure, amount of ammonia inserted, amount of catalyst inserted, and reaction time) on the production yield of PPD by analyzing the Gas Chromatography (GC). The experimental results demonstrated that PPD was not produced under non-catalyst conditions, and PPD production yield increased with increasing temperature, pressure, amount of catalyst inserted, and reaction time. However, for the reaction temperature case, it was found that $200^{\circ}C$ was the optimal temperature, because thermal degradation of PPD occurred above $250^{\circ}C$. In addition, we confirmed the structure of PPD and the bonding characteristics of the amine group via FT-IR and H-NMR analysis.

  • To preserve atmospheric environment of urban areas, it needs to create urban space considering air pollution sources and natural and geographical properties such as wind circulation. According to this study could examine climate and environmental characteristics of Daegu and accordingly suggest a climate map in urban environment and an "advice map" in urban planning. The urban area(area paved with asphalt and concrete) of Daegu has increased by more than five times since 1960. In addition, the analysis of thermal environment through satellite data shows that the surface temperature between a place paved with artificial structures and a farmland shows $10{\sim}20^{\circ}C$ difference during the daytime in the summer. Regarding the parks inhibiting the heat island of a city have the small area of trees, and the road paved with concrete is wide so that they hardly serve as the source of heat absorption. As Apsan is located to the south of Daegu and Palgonsan to the north and Daegu has east high west low type, mountain wind from mountains in the south and north passes a city and delivers heat and air pollutions at night. In the west of Daegue, there is the poorest environment and industrial facilities and environmental basic facilities are mostly located, so large residential complexes that are being built around the industrial facilities as if they set up a folding screen and therefore the poor environment is increasingly worse.

  • A sensor array with 10 discrete sensors integrated on a substrate was developed for discriminating the kinds and quantities of explosive gases. The sensor array consisted of 10 oxide semiconductor gas sensors with $SnO_2$ as base material and had broad sensitivity to specific gas. The sensor array was designed with uniform thermal distribution and had also high sensitivity and reproductivity to low gas concentration through nano-sized sensing materials with different additives. By using the sensitivity signal of the sensor array at $400^{\circ}C$, we could reliably discriminate the kinds and quantities of explosive gases like butane, propane and methane under the lower explosion limit through the principal component analysis (PCA) method.

  • Dwelling environments that can help elderly farmers to live more safely, independently, and conveniently are becoming more and more important. Many rural houses are built without any particular architectural or energy-related criteria, so most of them have poor insulation. The construction technology used is also not precise, which increases the loads for heating and cooling. Therefore, rural houses need to be improved. Also, there is more and more need for plans to realize eco-friendly dwellings, so the principle of nature-oriented plans related to the direction, insulation, or landscaping of a house is being emphasized. Insulation is one of the most effective ways to save energy for heating and cooling. This preliminary study to improve the insulation of rural houses examined three regions in South Korea: the central region, the southern region, and the Jeju Island. A field investigation was conducted on a total of 18 houses, including six from each town in the selected regions. The information was used to figure out the current status of rural houses and the characteristics of the buildings. The main living spaces are the living room for the central region and the main room in the southern region and Jeju Island. The southern regions are plane shapes surrounded by rooms, and all ventilation is accomplished by windows. The studied houses were mostly masonry structures with slate rooftops. Additions and improvements included room expansions and bathroom interior installations.

  • The study on the velocity distribution and the pressure drop characteristic of the nuclear fuel assembly is of importance for the thermal hydraulic design and safety analysis. The purpose of this experimental study is to investigate the hydraulic mixing behind the different kinds of spacer grids in the now or rod bundles. In this study, the detailed hydraulic characteristics in subchannels of 5$\times$5 PWR(Pressurized Water Reactor) rod bundles were measured using one-component He-Ne LDV(Laser Doppler Velocimeter). Measurements of the axial velocity, turbulent intensities and pressure drops were peformed Lateral velocity, turbulent intensities and Reynolds shear stress were also measured by adjust-ing LDV alignment. Friction factors in rod bundles and loss coefficients for spacer grids were evaluated from the measured pressure drops. Hydraulic mixing performance for different kinds of spacer grids could be investigated by estimating the turbulent cross-flow mixing rates between neighboring subchannels.

  • In order to dispose radioactive wastes safely, it is needed to understand the mechanical, thermal, fluid behavior of rockmass and physico-chemical interactions between rockmass and water. Also, the knowledge about mechanical and hydraulic properties of rocks is required to predict and to model many conditions of geological structure, underground in-situ stress, folding, hot water interaction, intrusion of magma, plate tectonics etc. This study is based on researches about rock mechanics issues associated with a waste disposal in deep rockmass. This paper includes the mechanical and hydraulic behavior of rocks in varying temperature conditions, thermo-hydro-mechanical coupling analysis in rock mass and deformation behavior of discontinuous rocks. The mechanical properties were measured with Interaken rock mechanics testing systems and hydraulic properties were measured with transient pulse permeability measuring systems. In all results, rock properties were sensitive to temperature variation.

  • There are many factors affecting the results of CMP (Chemical Mechanical Polishing). Among them, the temperature is related to the removal rate and WIWNU (Within Wafer Non-Uniformity). In other words, the removal rate is proportional to the temperature and the variation of temperature distribution on a pad affects the non-uniformity within a wafer. In the former case, the active chemistry improves the rate of chemical reaction and the removal rate becomes better. But, there are not many advanced studies. In the latter case, a kinematical analysis between work-piece and pad can be obtained. And such result analysed from the mechanical aspect can be directly related to the temperature distribution on a pad affecting WIWNU. Meanwhile, the temperature change affects the quantities of both slurry and pad. The change of a pH value of the slurry chemistry due to a temperature variation affects the surface state of an abrasive particle and hence the agglomeration of abrasives happens above the certain temperature. And the pH alteration also affects the zeta potential of a pad surface and therefore the electrical force between pad and abrasive changes. Such results could affect the removal rate and etc. Moreover, the temperature changes the 1st and 2nd elastic moduli of a pad which are closely related to the removal rate and the WIWNU.
