The G23 and G25 Genes of Temperate Mycobacteriophage L1 Are Essential for The Transcription of Its Late Genes

  • Two lysis-defective but DNA synthesis non-defective temperature-sensitive (ts) mutants of mycobacteriophage L1, L1G23ts23 and L1G25ts889 were found to be defective also in phage-specific RNA synthesis in the late period of their growth at 42$^{\circ}C$each to the extent of 50% of that at 32$^{\circ}C$The double mutant, L1G23ts23G25ts889 showed the ts defect in phage RNA synthesis that was nearly additive of those shown individually by the two single-mutant parents. Both G23 and G25 were shown to start functioning sometimes between 30 and 45 min after infection but the former gene might be dispensable after 45 min, while the latter was not. Northern analysis also shows that at 42$^{\circ}C$>, L1G23ts23 affects RNA synthesis more strongly than L1G25ts889 from L1 DNA segments that serve as the template for late gene transcription. Among the 21 virion and 12 non-virion late proteins synthesized by L1, L1G23ts23 is defective in the synthesis of at least 9 virion and all of non-virion proteins at 42$^{\circ}C$>. In contrast, L1G25ts889 is completely defective in synthesis of all the 33 late proteins. Possible roles of G23 and G25 in the positive regulation of transcription of different sets of late genes of L1 have been discussed.

  • To investigate the association between Korean red ginseng (KRG) intake in HIV-1 infected patients and occurrence of grossly deleted nef genes ($g{\Delta}nef$), we characterized nef genes in 10 long-term slow progressors (LTSP) infected with HIV-1 subtype B and 34 control patients. LTSP was defined whose the annual decrease in CD4 T cells was less than $20/{\mu}l$ over 10 years in the absence of antiretroviral therapy. They were treated with KRG for a prolonged period. Nef genes were amplified from peripheral blood mononuclear cells (PBMC) using nested PCR and products were sequenced directly. Patient CD4 T cell counts decreased from $444{\pm}207/{\mu}l$ to $294{\pm}177/{\mu}l$ over $136{\pm}23$ months of KRG intake. This corresponds to an annual decrease in the level of CD4 T cells of $13.3/{\mu}l$. A total of 479 nef genes were amplified from 137 PBMC samples. Nine out of the 10 patients, 47 (34.3%) out of the 137 samples, and 92 out of the 479 genes revealed $g{\Delta}nef$. The deletion extended outside the nef gene in 25 $g{\Delta}nef$ obtained from 6 patients. The proportion of samples with $g{\Delta}nef$ (34.3%) was significantly higher than 4.8% in control patients (P<0.001). In addition, it significantly increased as the duration of KRG intake prolongs (P<0.01). These data suggest the possibility that occurrence of $g{\Delta}nef$ might be associated with long-term intake of KRG.

해수 및 수산물에서 분리한 Vibrio vulnificus의 병원성 유전자 및 항균제 내성 (Profiles of Virulence Genes and Antimicrobial Resistance of Vibrio vulnificus Strains Isolated from Seawater and Fisheries)

  • We isolated 28 Vibrio vulnificus strains from seawater and fisheries and investigated the positive rate of eight virulence genes. Additionally, we evaluated the susceptibility of these strains to 25 antimicrobials. The positive rates of fur, vvhA, tcp, rtxA, vcgC, viuB, vvp, and acfA were 100, 92.9, 92.9, 67.9, 64.3, 25.0, 14.3, and 7.1%, respectively. A disk diffusion susceptibility test revealed that, all the investigated strains had the highest resistance to amoxicillin and oxacillin, followed by that to streptomycin (96.4%), cefoxitin (92.9%), clindamycin (82.1%), amikacin (67.9%), vancomycin (46.4%), nalidixic acid (7.1%), penicillin G (7.1%), and ampicillin (3.6%). Moreover, they were susceptible to 10 other antimicrobials, including cefotaxime, chloramphenicol, erythromycin, gentamicin, and rifampicin. Notably, amoxicillin, oxacillin, and streptomycin had average minimum inhibitory concentrations of 132.6, 603.4, and 23.1 ㎍/mL against V. vulnificus, respectively. These observations provide new insights regarding the necessity for sanitation of commercial fisheries and can potentially, help reduce the risk posed by fisheries contaminated with bacteria resistant to antimicrobials.

Mycoplasma pneumoniae의 분리 및 항생제 감수성 검사(III) (Isolation of Mycoplasma pneumoniae and Antimicrobial Susceptibilities of the Isolates(III))

  • 2002년 2월부터 2005년 2월까지 성인 및 소아 호홉기질환자 994명의 상기도 도말물에서 M. pneumoniae 균주를 분리하고, 분리 균주의 ciprofloxacin, ofloxacin, minecline, tetracycline, sparfloxacin, josamycin, erythromycin에 대한 감수성 검사를 실시하였으며, 분리된 균주의 235 rRNA domain II와 V에서 erythromycin저항성 변이가 일어났는가를 PCR과 유전자 염기서열분석으로 erythromycin에 감수성인 M. pneumonine균주의 염기서열과 비교분석하여 확인하였다. 호흡기질환자에서 M. pneumoniae의 분리율은 123/994$(12.4\%)$이었으며, 분리된 M. pneumoniae 균주의 minocycline, sparfloxacin, tetracycline, ciprofloxacin, ofloxacin, josamycin, erythromycin MIC 범위는 각각 $0.015\~0.25,\;0.06\~0.5,\;0.06\~0.5,\;0.25\~0.5,\;0.25\~0.5,\;0.015\~8.0,\;0.015\~8.0{\mu}m$이었다. 분리 동정된 M. pneumoniae 균주 중에서 erythromycin에 저항성인 균주가 60주$(48.8\%)$였으며, 모두가 ribosomal protein L4 영역과 23S rRNA domain V에 내성변이가 일어났으며, 이 중 2균주는 23S rRNA domain II에도 변이가 일어난 균주도 있었다. 국내에서 분리되는 M. pneumoniae균주의 $48\%$가 erytomycin에 저항성인 균주이므로 앞으로 이 균에 의한 폐렴의 치료에 주의가 요구된다.

  • Objectives : Hemistepta lyrata Bunge (Bunge) is a wild herb that has been used for managing fever and wound in Korean Traditional Medicine. The present study explored the effects of H. lyrata extract on liver X receptor (LXR) α-dependent lipogenic genes in hepatocyte-derived cells. Methods : After HepG2 cells or Huh7 cells were pre-treated with 1-10 ㎍/mL of H. lyrata extract or its fractionated extract for 0.5 h, the cells were subsequently exposed to LXR ligand for 6-24 h. Cell viability, LXR response element (LXRE)-driven luciferase activity, sterol regulatory element binding protein-response element (SREBP-RE)-driven luciferase activity, SREBP-1c expression, and mRNA levels of LXRα and its-dependent target genes were determined. In addition, LC-MS/MS analysis was conducted to explore major compounds in H. lyrata-chloroform fractionated extract #4 (HL-CF4). Results : Of various H. lyrata extracts tested, chloroform extract and its fractionated extract #4, HL-CF4, significantly decreased T0901317-mediated SREBP-1c expression. In addition, HL-CF4 significantly reduced LXRE atransactivation and LXRα mRNA expression without any cytotoxicity. Moreover, HL-CF4 prevented the SREBP-RE-driven luciferase activity and mRNA levels of fatty acid synthase and stearoyl-CoA desaturase-1 induced by T0901317. Results from LC-MS/MS analysis at positive/negative mode indicated that HL-CF4 contained several compounds showing m/z 197.1176 (C11H17O3), 693.2913/227.1069 (C38H45O12/C15H15O2), 203.1797 (C15H23), 181.1225 (C11H17O2), 591.2957 (C35H43O8), 379.1040 (C18H19O9), 409.1509 (C20H25O9), 309.1348 (C16H21O6), 391.1404 (C20H23O8), and 669.2924/389.1248 (C36H45O12/C20H21O8). Conclusion : Based on its inhibition of the LXRα-dependent signaling pathway, H. lyrata chloroform extract and HL-CF4 have prophylactic potentials for managing non-alcoholic fatty liver.

임상에서 분리된 CTX-M형 Extended-Spectrum $\beta$-Lactamases를 생산하는 Escherichia coli와 Klebsiella pneumoniae의 유행 (Prevalence of CTX-M-type Extended-Spectrum $\beta$-Lactamases Producing Escherichia coli and Klebsieilla pneumoniae Isolates in General Hospitals in 2005)

  • 병원내 항생제 다제 내성을 일으키는 CTX-M형 ESBL을 생성하는 E. coli와 Klebsielia pneumoniae의 생성현황을 조사하고 이들 균주로 인한 감염증치료와 역학적 조사연구에 도움이 되고자 효소의 유전형을 규명하였다. 2005년 7월-12월에 부산에 소재하고 있는 2개의 종합병원에서 분리된 E. coli와 K. pneumoniae 각각 153주, 52주를 수집하였다. 그 중에서 ESBL을 생성 하는 균주를 검출하기 위해 Double disk synergy test를 시행하여서 E. coli 23주와 K. pneumoniae 13주를 분리하였다. 균주의 동정은 Vitek system GNI card(bioMerieux Vitek Inc., Hazelwood, Mo., U.S.A.)로 확인하였고, 항생제감수성시험은 disk diffusion method 와 agar dilution method를 사용하였다. 분리된 균주들의 내성을 일으키는 ESBL유전형을 규명하기 위하여 Isoelectric focusing(IEF), polymerase chain reaction test, DNA sequencing을 시행하였다. A병원의 13주와 B병원의 10주로 총 23주의 E. coli(15.0%)와 A병원의 7주와 B병원의 6주로 K. pneumoniae 13주(25.0%)가 double disk synergy test 양성으로 ESBL 생성균주로 판정하였다. ESBL 생성 36균주를 대상으로 bla$_{TEM}$, bla$_{SHV}$, bla$_{CTX-M}$ 유전자 검출을 위한 PCR을 시행한 결과 bla$_{TEM}$ 유전자는 13주(36.1%), bla$_{SHV}$ 유전자는 13주(36.1%), bla$_{CTX-M}$ 유전자는 32주(88.9%)가 양성반응을 보여서 bla$_{CTX-M}$ 유전자를 가진 균주가 가장 많이 나타났다. 그리고, bla$_{TME}$, bla$_{SHV}$ 두 가지 유전자를 가지고 있는 균주는 1주(2.8%)만 나타났고 bla$_{TEM}$, bla$_{CTX-M}$두 가지 유전자를 가지고 있는 균주는 9주(25.0%), bla$_{SHV}$, bla$_{CTX-M}$ 두 가지 유전자를 가지고 있는 균주가 10주(27.8%)로 나타나 bla$_{CTX-M}$을 포함하는 복합유전자가 많이 증가함을 알 수 있었다. 또한 CTX-M형 ESBL을 생성하는 E. coli와 K. pneumoniae에 대한 cefutaxime의 MIC는 256 $\mu$g/m1 이상으로 ceftazidime의 16-256 $\mu$g/mL 이상보다 높은 분포를 보였다. 즉, CTX-M형 ESBL 유전자를 지닌 균주에 대한 cefotaxim의 MIC는 ceftazidime의 MIC에 비해서 상대적으로 높은 양상을 보였다. 이러한 결과는 국내의 대학병원 뿐 만 아니라 일반종합병원에서도 CTX-M형 ESBL 생성 E. coli와 K. pneumoniae가 존재하며 확산 중임을 시사한다. 앞으로 CTX-M형 ESBL의 만연과 변종 CTX-M형 ESBL의 출연을 감시하기 위한 정기적인 연구와 조사가 필요한 것으로 생각한다.
