Blood Lead and ZPP Concentrations of Underground Parking Lot Workers (지하주차장 근무자의 혈중 연 및 Zinc Protoporphyrin 농도)

  • A study was conducted to investigate the blood lead and zinc protoporphyrin(ZPP) concentrations of the underground parking lot workers who exposed to vehicular exhaust aerosols which contained lead. The blood samples were collected from 25 study subjects of underground parking lot and from 33 controls in Taegu, from July to October, 1997. And also the related factors to the blood lead and ZPP concentrations were investigated. Blood lead concentration and ZPP concentration were measured by flameless furnace atomic absorption spectrophotometer (1L.551) and hemetofluorometer, respectively. The mean blood lead concentrations of study subjects and controls were $23.10{\pm}20.77{\mu}g/dl$ and $12.99{\pm}12.71{\mu}g/dl$, respectively(p<0.05), and the mean blood ZPP concentrations of study subjects and controls were $40.72{\pm}9.46{\mu}g/dl$ and $38.21{\pm}10.97{\mu}g/dl$, respectively. No significant correlations were observed between the blood lead concentration of the study subjects and their job duration and between blood ZPP concentration of the study subjects and their job duration. There were no statistically significant differences in the blood lead concentration and the blood ZPP concentration of smokers and nonsmokers. In multiple regression analysis on blood lead concentration of the study subjects and controls, occupation was a only significant dependent variable. In case of bood ZPP concentration, age was a only significant dependent variable in the study subjects and controls.

Analysis of hybridity of Asplenium castaneo-viride Baker (거미꼬리고사리의 잡종성 분석)

  • To verify hybridity of Asplenium castaneo-viride, external morphology, spore morphology, anatomy and chromosomes of the species and of the two presumed parental species, A. incisum and A. ruprechtii, were examined. A. castaneo-viride usually had 1-pinnately divided frond. However, some individuals had almost simple fronds with pinnatisect basal parts similar to A. ruprechtii, while others had fronds similar to A. incisum in having oblanceolate blades and basal pinnae with triangular, 2-3 lobed apices. On the surface of the spores, sculpturing consisted of folds that were usually prominent; forming long wings, and irregular or incomplete reticulation. However, reticulation patterns varied among species. A. castaneo-viride showed a wide range of variation from sparse to dense patterns, whereas A. incisum showed only from sparse to intermediate patterns. A. ruprechtii showed from intermediate to dense patterns. The spore size of A. castaneo-viride was $54.63{\mu}m$, larger than other two species ($47.81{\mu}m$ in A. incisum and $44.22{\mu}m$ in A. ruprechtii). The level of undulation of epidermal cell wall was also different. A. incisum had the most shallowly undulated wall, and A. castaneo-viride had a pattern intermediate between the two presumed parental species. This same patterns was recognized in the density of stomata. The density of $45.91/mm^2$ in A. castaneo-viride was intermediate between the two presumed parental species ($67.00/mm^2$ in A. incisum, and $37.86/mm^2$ in A. ruprechtii). Chromosome number was constant (2x =2n = 72) as in A. incisum and A. ruprechtii. However, A. castaneo-viride showed a different ploidy level. The populations of Mt. Mai (Jeonbuk province) and Mt. Duryun (Jeonnam province) were diploid (2n = 72) which is a new record for this taxon, whereas the population of Mt. Buram (Seoul) was tetraploid (2n = 144). Conclusively, A. castaneo-viride was revealed to be a hybrid of A. ruprechtii and A. incisum based on evidence involving leaves, spores, epidermal cells, stomata and chromosome number.

Development and Validation of an Analytical Method for the Insecticide Sulfoxaflor in Agricultural Commodities using HPLC-UVD (HPLC-UVD를 이용한 농산물 중 살충제 sulfoxaflor의 시험법 개발 및 검증)

  • Sulfoxaflor is a new active ingredient within the sulfoximine insecticide class that acts via a unique interaction with the nicotinic receptor. The MRLs (maximun residue limit) of sulfoxaflor in apple and pear are set at 0.4 mg/kg and that in pepper is set at 0.5 mg/kg. The purpose of this study was to develop an analytical method for the determination of sulfoxaflor residues in agricultural commodities using HPLC-UVD and LC-MS. The analysis of sulfoxaflor was performed by reverse phase-HPLC using an UV detector. Acetone and methanol were used for the extraction and aminopropyl ($NH_2$) cartridge was used for the clean-up in the samples. Recovery experiments were conducted on 7 representative agricultural products to validate the analytical method. The recoveries of the proposed method ranged from 82.8% to 108.2% and relative standard deviations were less than 10%. Finally, LC-MS with selected ion monitoring was also applied to confirm the suspected residues of sulfoxaflor in agricultural commodities.

Manufacturing process and food safety analysis of sous-vide production for small and medium sized manufacturing companies: Focusing on the Korean HMR market (중소규모 생산업체의 수비드 제품 생산을 위한 공정 및 안전성 분석: 한국 HMR 시장 중심으로)

  • The present study identified the restrictions on the use of sous-vide products in the Korean HMR market for small and medium-sized manufacturing companies. A detailed literature review revealed that the HMR market in Korea is close to saturation. Notably, the technologically advanced products produced using sous-vide seem to display significant potential to overcome market saturation. The sous-vide method differs from conventional cooking techniques and is characterized by maintenance of food texture along with flavor enhancement. However, due to the unfamiliarity of the manufacturers with this method and the unclear food safety regulations, mass food manufacturing companies do not agree on using this method; hence, sous-vide production is usually undertaken by small/medium sized companies catering primarily through online marketing portals. This study highlights the various restrictions to the implementation of sous-vide production, and discusses several practical implications of sous-vide production that would help users of this technique enter the HMR market.

Coexpression of $P2X_3$ with TRPV1 in the Rat Trigeminal Sensory Nuclei (흰쥐 삼차신경감각핵에서 $P2X_3$와 TRPV1의 공존에 관한 연구)

  • Trigeminal primary afferents expressing $P2X_3$ or transient receptor potential vanilloid 1 (TRPV1) are involved in the transmission of nociceptive information. In order to characterize $P2X_3$- and TRPV1-immunopositive neurons in the trigeminal ganglion (TG) and trigeminal caudal nucleus (Vc), we performed immunofluorescence experiments using anti-$P2X_3$ and anti-TRPV1 antisera and a morphometric analysis. 77.4% (1,401/1.801) of all the $P2X_3$-postive neurons coexpressed TRPV1 and 51.9% (1,401/2,698) of all the THFV1-immunopositive neurons also costained for $P2X_3$ in the TG. Immunoreactivity for both $P2X_3$ and TRPV1 were present in medium-sized neurons but not in small- and large-sized neurons. $P2X_3$ and/or TRPV1-immunopositive fibers were observed in the primary afferents and their associated axons in the Vc. These fibers and terminals were distributed in the superficial lamina of Vc: $P2X_3$-immunopositive fibers and terminals were distributed in the lamina I and II, expecially in the inner part of lamina II (lamina IIi), whereas TRPV1-immunopositive ones were densely detected in the lamina I and outer part of lamina II (lamina IIo). Immunopositive fibers and terminals for both $P2X_3$ and TRPV1 were observed on the border between lamina IIi and IIo. These results suggest that terminals coexpressing $P2X_3$ and TRPV1 are involved in specific roles in the transmission and processing of orofacial nociceptive information.

Studies on the Estimation of the Genetic Parameters on All Traits in Korean Native Ogol Fowl IV. Estimations of the Heritabilities and Genetic Correlations on the Body Conformations (한국재래오골계의 제형질에 대한 유전모수 추정에 관한 연구 IV. 체형에 대한 유전력 및 유전상알 추정)

  • This study was conducted to estimate heritabilities and genetic correlations on body shape components in Korean Native Ogol fowl , The data analysis were record of 450 pullets producted from 150 dams and 20 sires of Korean Native Ogol fowl raised at Chungnam National University from June 18, 1987 to April 6, 1989. The results obtained are sammarized as follows ; 1. On the body components, shank length at 8, 24 weeks and 300 days of age were $8.717\pm$0.363, $9.576\pm$0.390 and $9.646\pm$0.389cm, the breast width were 3.469$\pm$0.234, $4.310\pm$0.345 and $4.752\pm$0.343cm; the breast girth at 8, 24weeks and 300 days of age were $22.819\pm$1.082, $36.719\pm$1.793 and $39.008\pm$1.709cm; the tibia length at 8, 24weeks and 300days of age were $9.941\pm$0.888, $13.183\pm$0.499 and $13.560\pm$0.552cm. 2. The heritability estimates of body shape components based on the variances of sires, dams and combined components from 8, 24 weeks and 300 days of age were 0.235-0.290, 0.589-0.890 and 0.437-0.565 for shank length ; 0.185-0.237, 0.444-0.536 and 0.314-0.392 for breast width ; 0.218-0.552, 0.499-0.746, 0.486-0.558 for breast girth ; 0.339-0.432, 0.281-0.543 and 0.340-0.446 for tibia length, respectively. 3. The genetic correlation coefficient of body shape components were as follows ; between shank length and breast width, breast girth, tibia length were 0.237-0.836, 0.277-0,729 and 0.378-0.915;between breast width and breast girth, tibia length were 0.637-0.889 and 0.384-0.903;between breast girth and tibia length was 0.905-0.995, respectively.

Analysis of Stress Response of Domestic Chicken Breeds for the Development of a New Synthetic Parent Stock (국산 종계 개발을 위한 토종 계통들의 스트레스 반응 정도 분석)

  • We compared the degrees of stress response of 12 domestic purebred chicken strains that have been bred at National Institute of Animal Science, RDA, Korea since 1980. As a physiological marker of stress response, the expression levels of heat shock protein (HSP)-70, HSP-$90{\alpha}$, HSP-$90{\beta}$, hydroxyl-3-methyl-glutaryl coenzyme A reductase (HMGCR) genes and telomere length were measured by quantitative real-time polymerase chain reaction using the lymphocytes of 1,101 chickens. There was significant difference in HSP-70, HSP-$90{\alpha}$, HMGCR expression and telomere length among the strains. There was also significant difference in HSP-$90{\alpha}$, HSP-$90{\beta}$, and HMGCR expression between male and female chickens. Different age groups of chicken exhibited different expression levels of HSP-70, HSP-$90{\alpha}$ and telomere length. The results of the HSPs expression level suggested that, the strains of R, L and Y were highly resistant to stress, whereas the strains of S, O and W were susceptible to stress. Although the statistical differences in some of HSPs gene expression existed between genders, the HSP expression results varied in different strains that some opposed to the others, and there might be interaction between strains and genders, which conclude that there was no difference in stress response between male and female chickens. Moreover, despite of significant difference in some of HSPs expression level, it was considered that there was no difference in stress response between ages due to the inconsistent trends among HSP markers.

Effects of Gamma-Irradiation on the Sterilization of Primordial Germ Cells in Quail (메추리 원시생식세포 감소를 위한 감마선 조사의 효과)

  • Quail is a very useful animal model for studying vertebrate development because of its small body size and unique reproductive traits. This species is also ideal model for producing germline chimeras via transferring exogenous primordial germ cells (PGCs) into the recipient embryo. To increase the contribution efficiency of donor PGCs into recipients' tissues, decreasing the population of endogenous PGCs has been rate-limiting factor. We therefore conducted this study to investigate if gamma ($\gamma$)-irradiation depletes endogenous PGCs in developing quail embryo. Firstly, freshly laid stage X quail embryos were irradiated with various output of $\gamma$-irradiation and its teratogenic effect on the embryo was evaluated. Although a dose-dependent increase in the number of embryo showing malformation was found as the output increased (0, 250, 500, 750, and 1,000 rads), only a maximum of 10.1% of embryos were abnormal in 1,000 rads. Immunocytochemical analysis using the QCR1 antibody, which is specific marker for quail PGCs, was conducted to analyze the effect of sterilization. As results, $\gamma$-rays at a dose-rate of 500 rads/73 sec onto undeveloped stage X embryo significantly reduced the number of germ cells to an average of 75.55 % and 82.03 % in male and female embryos, respectively. We conclude that $\gamma$-ray selectively targets PGCs while affects minimally to the somatic development in quail embryo. Our results will not only provide important data for germline chimera production but can be used for analyzing the effect of ionized rays on the differentiating germ cells in various stages during animal development.

Weathering Sensitivity Characterization for Rock Slope, Considering Time Dependent Strength Changes (시간에 따른 강도변화를 고려한 암반사면의 풍화민감특성 분석)

  • Rocks undergo weathering processes influenced by changing in pressure-temperature condition, atmosphere, underground water, and rainfall. The weathering processes change physical and chemical characteristics of the rocks. Once the rocks are weathered, the characteristics of them are changed and, because of the changing, several disadvantages such as rock slope failures and underground water spouts are can occur. Before we cut a large rock slope, therefore, we must analyze current weathering conditions of rocks and predict weathering processes in the future. Through the results of such analyses, we can judge reinforcement works. In order to comply with such requests, chemical weathering sensitivity analysis which was analyzed from chemical weathering velocities and other characteristics of rocks has been applied in several prior construction works in Korea. But, It is defective to use directly in engineering fields because it was developed for soils(not rocks), it has too mny factors must be considered and the relationships between the factors are not clear, and it is hard to explain the weathering processes in engineering time range. Besides above, because it has been used for isotropic rocks, this method is hard to apply to anisotropic rocks such as sedimentary rocks. Acceding to studies from morphologists (e.g. Oguchi et al., 1994; Sunamura, 1996; Norwick and Dexter, 2002), time dependent strength reduction influenced by weathering shows a negative exponential function form. Appling this relation, one can synthesize the factors which influence the weathering processes to the strength reduction, and get meaningful estimates in engineering viewpoint. We suggest this weathering sensitivity characterization method as a technique that can explain time dependent weathering sensitivity characteristics through strength changes and can directly applied the rock slope design.

Evaluation of Average CT to Reduce the Artifact in PET/CT (PET/CT 검사에서 호흡에 따른 인공산물을 줄이기 위한 Average CT의 유용성)

  • Purpose: The usefulness of Positron Emission Tomography (PET) images in diagnosis, staging, recurrent and treatment response evaluation has already been known. However, tumors which are small size, located in lower lobe of lung or upper lobe of liver are shown misalignment, distortion and different Standard Uptake Value (SUV) by respiration in PET images. Therefore, if radiotherapy based on normal respiration, it may cause low treatment response or more side effects because targets which had to treat, out of treat range or over dose to normal tissue. The purpose of this study is to evaluate attenuation-correction with Average CT (ACT) for more accuracy SUV measurement and minimize artifact by respiration. Materials and Methods: 13 patients, who had tumors which are around the diaphragm, underwent ACT scan after Helical CT (HCT) scan with PET/CT (Discovery DSTE 8; GE Healthcare). We quantified the differences between attenuation corrected image with HCT and attenuation corrected image with ACT in artifact size and maximum SUV ($SUV_{max}$). Artifacts were evaluated by measurement of the curved photogenic area in the lower thorax of the PET images for all patients. $SUV_{max}$ was measured separately at the primary tumors. Analysis program was Advantage Workstation v4.3 (GE Healthcare). Patients were injected with 7.4 MBq (0.2 $mC_i$) per kg of $^{18}F$-FDG and scanned 1 hour after injection. The PET acquisition was 3 minute per bed. Results: Significantly lower artifact were observed in PET/ACT images than in PET/HCT images (below-thoracic artifacts caused by under corrected $1.5{\pm}3.5$ cm vs. $13.4{\pm}4.2$ cm). Significantly higher $SUV_{max}$ were noted in PET/ACT images than in PET/HCT images in the primary tumor. Compared with PET/HCT images, $SUV_{max}$ in PET/ACT images were higher by $5.3{\pm}3.9%$ (mean value) tumor. The highest difference was observed in Lower lobe of lung (7.7 to 8.7; 13%). Conclusion: Due to its significantly reduced artifacts in lower thoracic, attenuation corrected image with ACT images provided more reliable $SUV_{max}$ and may be helpful in monitoring treatment response. Moreover, ACT can separate upper lobe of liver and lower lobe of lung, it may be helpful in interpretation. ACT will be clinically useful, considering increased dose caused by ACT scan and adapt.
