Nitric Oxide Prevents the Bovine Cerebral Endothelial Cell Death Induced by Serum-Deprivation

  • Endothelial cells play a central role in the inflammatory processes, and activation of nuclear factor kappa B ($NF-_{\kappa}B$) is a key component in that inflammatory processes. Previously, we reported that tumor necrosis factor alpha($TNF{\alpha}$) had protective effect of cell death induced by serum deprivation and this protection was related to $NF-_{\kappa}B$ activation. Inducible nitric oxide synthase (iNOS) is a member of the molecules which transcription is regulated mainly by $NF-_{\kappa}B$. And the role of nitric oxide (NO) generated by iNOS on cell viability is still controversial. To elucidate the mechanism of $TNF{\alpha}$ and $NF-_{\kappa}B$ activation on cell death protection, we investigate the effect of NO on the cell death induced by serum- deprivation in bovine cerebral endothelial cells in this study. Addition of $TNF{\alpha}$, which are inducer of iNOS, prevented serum-deprivation induced cell death. Increased expression of iNOS was confirmed indirectly by nitrite measurement. When selective iNOS inhibitors were treated, the protective effect of $TNF{\alpha}$ on cell death was partially blocked, suggesting that iNOS expression was involved in controlling cell death. Exogenously added NO substrate (L-arginine) and NO donors (sodium nitroprusside and S-nitroso-N-acetylpenicillamine) also inhibited the cell death induced by serum deprivation. These results suggest that NO has protective effect on bovine cerebral endothelial cell death induced by serum-deprivation and that iNOS is one of the possible target molecules by which $NF-_{\kappa}B$ exerts its cytoprotective effect.

탄소방전등과 저강도 레이저 조사가 화상 쥐의 Cytokine 변화에 미치는 영향 (An Effect of Carbon Arc Lamp and Low Level Laser Therapy on the Changes of Burned Mice's Cytokine)

  • Purpose: To provide the basic method for physical therapy to contribute to early stage treatment of a burned patients. We investigated whether or not carbon arc lamp treatment and low level laser one among various laser treatments have the effect of reducing Cytokine, and to elucidate the effect of carbon arc lamp treatment and laser one. Methods: The 6-week old BALB/c types of 92 mice were used for the experimental test, and they were burned with $100^{\circ}C$ water, they were divided into 4 groups in accordance with the method of treatment respectively. And the blood and the tissue from the subject of each group were extracted and analyzed each time after they were cured for 3, 6, 9 days. For the analysis of the results, SPSS statistical program was used in this study. Results: The quantity of $TNF-{\alpha}$ and $IL-1{\beta}$ within the blood of a burned mouse increased more than that of a normal mouse(p<0.01). In only a burned mouse, carbon arc lamp treatment gave the effect on the decrease of $TNF-{\alpha}$ thickness the 6th day to the 9th day(p<0.01). $IL-1{\beta}$ quantity was more decreased than that of control group around the 6th day. In comparison with only a control group, low level laser treatment has more significant effect in decreasing the quantity of $TNF-{\alpha}$ a and $IL-1{\beta}$ than the two different methods(p<0.01). In case that the two treatment methods, carbon arc lamp treatment and low level laser, were executed together, there was the effect of decreasing $TNF-{\alpha}$ until the 6th day(p<0.01). Conclusion: Both low level laser treatment and carbon arc lamp one would be able to have an effect on the inflammation inhibition of burned patients and tissues reproduction. However, it must be also considered for the two treatments to be done at the same time.

  • The effect of prolonged treatment with Mori Folium column fraction(MFCF) on intestinal ${\alpha}$-glycosidase activity has been studied in mice made hyperglycemic and hyperinsulinemic by feeding a high carbohydrate-containing diet. Mice were treated for 10 week with or without MFCF, added to the high carbohydrate-containing diet at 50mg/100g food. While MFCF had no effect on body weight, it prevented the rise in glycemia and insulinemia. Maltase, sucrase and lactase activities were measured in intestinal homogenates of proximal, middle and distal segments of jejunoileum. Following 10 week of MFCF administration, MFCF significantly induced all three enzyme activities especially at middle and distal segments. The mechanisms responsible for these changes and their potential biochemical implications remain to be determined.

석이에서 분리한 GE974의 혈당상승억제효과 (Hypoglycemic Effect of GE974 isolated from Gyrophora esculenta in Normal and Diabetic mice)

  • GE974, isolated from Gyrophora esculenta, showed significant inhibitory effect on several ${\alpha}-glucosidases$ in vitro in previous study. In the present study, GE974 showed significant inhibitory effect on blood glucose elevation in mice loaded with maltose, sucrose, starch and lactose. Also, it exhibited similar effect in alloxan and streptozotocin induced diabetic mice, and genetic diabetic mice(db/db mice) loaded with maltose in dose dependent manner. These results suggest that GE974 may possess hypoglycemic effect that inhibit competitively ${\alpha}-glucosidase$ activities.

  • Lf는 cytokines의 생성 및 면역반응 등의 생체 방어적 기능을 하는 물질로 알려져 있다. 본 연구에서는 Lf과 Lf-h에 의해 macrophage에서 TNF-$\alpha$와 NO 생성에 미치는 효과를 조사하였다. K-Lf 및 K-Lf-h는 단독으로 TNF-$\alpha$의 생성을 증가시켰으며, Lf의 농도에 따라 그 생성량이 증가하였다. LPS와 함께 작용시켰을 경우에는 큰 효과가 없는 것으로 나타났으나, 세포 성장률은 증가시켰다. 그러나 B-Lf, H-Lf, 그리고 이들의 가수분해물들은 단독으로는, RAW264.7 cell을 자극하여 TNF-$\alpha$나 NO의 생성을 증가시키지 못하였다. 또한, K-Lf는 그 자체만으로 TNF-$\alpha$에서 보여준 것처럼 NO생성에 영향을 미치며 농도가 높아짐에 따라 NO 생성을 증가시키는 경향을 보여주었다.

The α-Effect in SNAr Reaction of 1-Fluoro-2,4-dinitrobenzene with Hydrazine: Ground-State Destabilization versus Transition-State Stabilization

  • A kinetic study is reported on SNAr reaction of 1-fluoro-2,4-dinitrobenzene with a series of primary amines including hydrazine in $H_2O$ at $25.0^{\circ}C$. The plots of $k_{obsd}$ vs. [amine] are linear and pass through the origin, indicating that general-base catalysis by a second amine molecule is absent. The Br${\o}$nsted-type plot exhibits an excellent linear correlation with ${\beta}_{nuc}$ = 0.46 when hydrazine is excluded from the correlation. The reaction has been suggested to proceed through a stepwise mechanism, in which expulsion of the leaving group occurs after the rate-determining step (RDS). Hydrazine is ca. 10 times more reactive than similarly basic glycylglycine (i.e., the ${\alpha}$-effect). A five-membered cyclic intermediate has been suggested for the reaction with hydrazine, in which intramolecular H-bonding interactions would facilitate expulsion of the leaving group. However, the enhanced leaving-group ability is not responsible for the ${\alpha}$-effect shown by hydrazine because expulsion of the leaving group occurs after RDS. Destabilization of the ground-state of hydrazine through the electronic repulsion between the nonbonding electron pairs is responsible for the ${\alpha}$-effect found in the current $S_NAr$ reaction.

  • The effects of salts which was used as a catalysis in formation of $\alpha$-calciumusulfate hemihydrate from dicalcium sulfate hydrate were investigated on the hydration of $\alpha$-calciumsulfate hemihydrate. The hydration of $\alpha$-calciumsulfate hemihydrate was studied by the measurements of crystalline water, heat evolution. Also the hydrates were analyzed by XRD, DSC and SEM. The promotive effect each salts on the hydration was as follows: NaCl>NH4Cl>NaNO3>NH4NO3, and the hydration rate was accelerated with concentration of salts. The effect of Al2(SO4)3 and potassium sodium tartrate on the hydration was slmilar to water, whereas sodium succinate and gelatin retarded the hydration in comparision with water. These salts affected the hydration time but total heat evoution was similar.

Effect of Titanium Ion and Resistance Encoding Plasmid of Pseudomonas aeruginosa ATCC 10145

  • Titanium and its alloys are technically superior and cost-effective materials, with a wide variety of aerospace, industrial, marine, and commercial applications. In this study, the effects of titanium ions on bacterial growth were evaluated. Six strains of bacteria known to be resistant to both metal ions and antibiotics were used in this study. Two strains, Escherichia coli ATCC 15489, and Pseudomonas aeruginosa ATCC 10145, proved to be resistant to titanium ions. Plasmid-cured p. aeruginosa resulted in the loss of one or move resistance markers, indicating plasmid-encoded resistance. The plasmid profile of p. aeruginosa revealed the presence of a 23-kb plasmid. The plasmid was isolated and transformed into $DH5{\alpha}$. Interestingly, the untransformed $DH5{\alpha}$ did not grow in 300 mg/l titanium ions, but the transformed $DH5{\alpha}$ grew quite well under such conditions. The survival rate of the transformed $DH5{\alpha}$ also increased more than 3-fold compared to that of untransformed $DH5{\alpha}$.

  • Rhizopus nigricans에 의한 progesterone의 $11{\alpha}-hydroxylation$${5\alpha}-reduction$반응에 미치는 이온의 효과를 조사하였다. $Cu^{2+},\;Cd^{2+},\;Co^{2+},\;Mn^{2+},\;Zn^{2+},\;Fe^{2+},\;Mg^{2+},\;Fe^{3+}$$Na^+$$11{\alpha}-hydroxylation$ 반응의 활성을 저하시켰으나 $K^+$은 이 반응을 촉진시켰다. $11{\alpha}-hydroxyprogesterone$$5{\alpha}-reduction$시키는 효소는 $Fe^{2+},\;Mn^{2+},\;Mg^{2+},\;Co^{2+},\;Zn^{2+},\;Fe^{2+},\;K^+,\;Na^+$에 의하여 그 활성이 증가되었고 $Cd^{2+}$$Cu^{2+}$은 이 반응을 억제하였다. $11{\alpha}-hydroxylation$ 반응을 촉진하는데는 $10^{-3}\;M$$K^+$이 효과적이었으며 $5{\alpha}-reduction$반응을 엑제하기 위하여는 $10^{-4}\;M$$Cd^{2+}$이 적합하였다. R. nigricans는 세포내의 다효소체계에 의하여 progesterone이 $11\{alpha}-hydroxyprogesterone$으로 전환된 후 $11{\alpha}-hydroxy-allopregnane-3, 20-dione으로 $5{\alpha}-reduction$되므로 $10^{-3}\;M$$K^+$을 먼저 첨가하여 2시간 동안 progesterone 전환 반응을 실시한 후에 $10^{-4}\;M$$Cd^{2+}$을 첨가하였을 때 가장 높은 $11\{alpha}-hydroxyprogesterone$의 수득률을 얻을 수 있었다.
