  • Background and Objective : Cyperus rotundus L. (CR) is a commonly used herbal medicine in Asian countries such as Korea, China and Japan. The present study was designated to evaluate the direct effects of CR on helper T cell activities and on Th1/Th2 lineage development in vitro. Materials and Methods : Spleen cells from 8 week BALB/c mice were cultured in CR extracts containing medium without activation for 24 hours and with activation for 48 hours. CD4+ T cells were isolated and analyzed for mRNA expression levels of INF-$\gamma$, IL-4, T-bet and GATA-3 by RT-PCR and secretion cytokines levels of INF-$\gamma$, IL-4, IL-5 and IL-10 by ELISA. Results : The results demonstrated that CR had no mitogenic effects on unstimulated CD4+ T cells, but augmented CD4+ T-cell proliferation upon activation with anti-CD3/anti-CD28 antibodies in a dose-dependent manner. CR treatment significantly increased CD4+ T cell population and the IFN-$\gamma$ expression was significantly enhanced, while IL-4 expression was significantly decreased. In addition, in vitro Th1/Th2 polarization experiments revealed that CR enhanced IFN-$\gamma$ secretion in Th1 cells, but reduced the IL-4 in Th2 cells in a dose-dependent manner. Conclusion : These results suggest that CR treatment could be a desirable alternative therapy for the prevention or correction of Th2 dominant pathological disorders, such as allergy and asthma.

  • The commitment of T helper (Th) cells to Thl or Th2 cells is of crucial importance with respective to susceptibility or resistance to particular infections, or to autoimmune diseases and allergic diseases. The nature of Thl or Th2 polarizing signals is not yet fully understood. However, the cytokines that are present in the environment of the $CD4^{+}$ T cell at the time it encounters the antigen significantly regulate the differentiation of Th cells into either Thl or Th2 subsets. • Transcription factor GATA-3 is the critical transcription factor for Th2 cell differentiation. In spite of its importance in Th2 cell differentiation, the molecular mechanism for its action in Th2 differentiation is poorly understood. Previous studies have suggested that GATA-3 may be involved in the chromatin remodeling in the Th2 cytokine locus. To determine whether GATA-3 exerts its effect on its target sites in the extrachromosomal status, cell transfection assay was performed. In this assay, 800 bp IL4 promoter-luciferase constructs linked with GATA-3 target sites were transfected into the M12 B cell line, D10 mouse Th2 cell lines, and human T lymphoma Jurkat cell lines with or without the GATA-3 expression vector. The GATA-3 effects on its target sites were minimal in the extrachromosomal status, supporting the previous propositions that GATA-3 functions at the chromatin level by remodeling chromatin structure.

  • The aim of this study is to investigate the wound healing effects of the Opuntia ficus-indica. Four full-thickness skin wounds(2.5 cm × 2.5 cm) were made in different areas on the back in each of five dogs. Two wounds were used for wound size measurement and two for skin biopsy. The ten full-thickness wounds were treated with the extract of Opuntia ficus-indica, and the ten full-thickness wounds were treated with normal saline. The wound areas were measured every other day for three weeks after wounding. Statistical analysis was performed with the paired t-test. The wound healing rates were 32.9±4.0%, 28.2±7.1%, 42.2±4.1%, 55.0±4.3%, 76.6±2.8%, 86.8±2.3%, 94.0±1.l%, 97.3±0.8%, 99.3±0.3%, 99.8±0.2%, 100.0±0.0% in the experimental group, and 18.0± 8.2%, 13.S± 11.6%, 15.2±6.1 %, 36.9±8.8%, 62.4±4.1%, 80.0±2.3%, 86.0±1.4%, 94.0±0.9%, 96.9±0.8%, 97.8±0.3%, 99.7±0.1% in the control group on the 1/sup st/, 3/sup rd/, 5/sup th/, 7/sup th/, 9/sup th/, 11/sup th/, 13/sup th/, 15/sup th/, 17/sup th/, 19/sup th/, 21/sup st/ day. On the 17/sup th/ day, the wound closed above 99.9% in 1 dog of the experimental group. On the 19/sup th/ day, the wound closed above 99.9% in 3 dogs of the experimental group. On the 21/sup st/ day, the wound closed above 99.9% in 5 dogs of the experimental group, and 1 dog of the control group. The wound healing period was shortened by 2 to 4 days in the experimental group as compared with the control group. The tensile strength was 4,508±513 g/㎠ in the experimental group and 2,772±400 g/㎠ in the control group on the 21/sup st/ day after wounding. The tensile strength was significantly higher in the experimental group than in the control group(p< 0.05). Inflammation rapidly spread and disappeared in the experimental group but not in the control group. Fibroblast and vascularization were found on the 5/sup th/ day in both group and increased only in the experimental group on the 9/sup th/ day. The firs finding of epithelialization was on the 13/sup th/ day in the experimental group and the 21/sup st/ day in the control group. A significantly higher wound healing rate(p< 0.05), tensile strength(p< 0.05) and better histopathological findings were observed in the experimental group than in the control group throughout the experimental period. These results suggest that topical application of Opuntia ficus-indica extract can promote wound healing in dogs.

  • Purpose : Catecholamines are the neuro-transmitters in the sympathetic nervous system (SNS) and are activated by stress stimulus. Tyrosine hydroxylase (TH) and Dopamine-${\beta}$-Hydroxylase (DBH) are very important enzymes in the catecholamine synthesis. Corticotropin releasing hormone (CRH) is released in the process of reacting to stresses. The aim of this study is to find out what effects immobilization stresses have on the expression of TH, BDH and CRH mRNA in a rat's brains. Methods : We compare expression levels in rat's brains of TH, DBH and CRH mRNA induced by immobilization stresses between the test group and controled group. The expression levels of TH, DBH and CRH mRNA are measured by RT-PCR and the Western Blotting Analysis (WBA). Results : In brains and adrenal glands of the immobilization stress group, the expression levels of TH and DBH mRNAs are significantly two to three times higher (P<0.01), and CRH mRNAs are approximately one and a half times higher (P<0.05) than those of controlled group. Conclusion : This study suggest that the expression levels of TH, DBH and CRH mRNAs are activated by stress stimulus in a rat's brains and adrenal glands.
