Mitochondrial DNA Sequence Variation of the Oriental Mole Cricket, Gryllotalpa orientalis (Orthoptera: Gryllotalpidae) in Korea

  • The mole crickets, Gryllotalpa, are insect pest distributed in the world. In Korea, G. orientalis was reported to occur, but previous ecological studies suggested the presence of two ecological types. To test this hypothesis, we sequenced a portion of mitochondrial (mt) genome from 48 G. orientali individuals collected over five Korean localities: Busan, Suwon, Okchon, Wonju, and Gangneung. From the sequence analysis, only two haplotypes were obtained, but the sequence divergence between the two haplotypes was 11 %, suggesting the presence of two distinct genetic groups in Korea. Although the population of Busan, Okchon, Wonju, and Gangneung was identified as a single haplotype, but that of Suwon was occupied by both hapotypes. Considering sequence divergence of other insect species occurring in Korea, the divergence estimate found between the two haplotypes seems to be too large to be considered as identical species. This result may suggest that the two differentiated haplotypes found in this study may reflect the previously reported two ecological types found in Suwon, Korea. To further understand the genetic divergence of the two phylogenetic groups, analysis of more variable regions of G. orientalis genome is required.

  • To understand the molecular structure and pathogenesis mechanism of Korean garlic viruses, we have isolate cDNA clones for garlic viruses. The partial nucleotide sequences of 24 cDNA clones were determined and that of six clones containing poly (A) tail were compared with those of other plant viruses. One of those clones, V9 has 81.8% similarity in nucleotide sequence and 93.0% in deduced amino acid sequence, respectively, to the coat protein gene for garlic mosaic virus (GMV). Northern blot analysis with the clone V9 demonstrated that the genome of GMV is 7.8 kb long and has poly (A) tail. The anti-coat protein antibody for GMV recognizes 35 kDa polypeptide which could be the coat protein of GMV from infected garlic leaf extract or virus preparation. Clone G7 has about 62% of deduced amino acid sequence identity with the members of potyvirus group. Northern blot analysis with the clone G7 demonstrated that the genome of the potyvirus I garlic is 9.0 kb long and has poly (A) tail. The third clone, S81, shows 42% amino acid identity to the potexvirus. The other clones are under the characterization. To test the possibility of producing garlic virus resistant plant, we have designed a hairpin type ribozyme to cleave V9 RNA at the middle of the coat protein gene. From the cleavage reactions in vitro with two different sizes of RNA substrates, V9SUB (144 nucleotides) and V9 RNA (1,361 nucleotides), the ribozyme can cleave V9 sequence effectively at the predicted site. To study the activity of the ribozyme in vivo, plant transformation is in progress. Further possibilities to produce garlic virus resistant plant will be discussed.

  • This report summarized the static test of the rudder system for the KTX-1 basic trainer. The test loads are applied up to the limit and ultimate loads in a stepping sequence. Test loads and test results matt the strength and stiffness requirements of the rudder control system.. Using #004 full scale structure test airframe.

  • The Propulsion System Test Complex(PSTC) was constructed for the verification test of each stage's propulsion system of KSLV-II. Main Control System(MCS) is the system to operate the onboard equipment and the ground equipment of the PSTC simultaneously. This paper describes the critical design, the development status, and test results of Main Control System. The MCS will be used for the interface connection between ground control systems and onboard equipment. Test sequence and operation process of the Work Manager will be conducted by MCS.

Image Segmentation based on Statistics of Sequential Frame Imagery of a Static Scene (정지장면의 연속 프레임 영상 간 통계에 기반한 영상분할)

  • This study presents a method to segment an image, employing the statistics observed at each pixel location across sequential frame images. In the acquisition and analysis of spatial information, utilization of digital image processing technique has very important implications. Various image segmentation techniques have been presented to distinguish the area of digital images. In this study, based on the analysis of the spectroscopic characteristics of sequential frame images that had been previously researched, an image segmentation method was proposed by using the randomness occurring among a sequence of frame images for a same scene. First of all, we computed the mean and standard deviation values at each pixel and found reliable pixels to determine seed points using their standard deviation value. For segmenting an image into individual regions, we conducted region growing based on a T-test between reference and candidate sample sets. A comparative analysis was conducted to assure the performance of the proposed method with reference to a previous method. From a set of experimental results, it is confirmed that the proposed method using a sequence of frame images segments a scene better than a method using a single frame image.

  • 객체 지향 소프트웨어의 확장과 더불어 소프트웨어의 재사용성의 중요도가 부각되면서 소프트웨어를 콤포넌트 단위로 구현하는 추세이다. 따라서 콤포넌트 기반의 소프트웨어 개발에서의 통합 테스트가 중요한 이슈로 떠오르고 있다. 그러나 콤포넌트들의 통합 테스트에 대한 연구는 미흡한 상태이다. 본 논문에서는 UML을 기반으로 체계적인 통합 테스트 모형을 제안하고. 나아가 본 논문에서는 "멀티미디어 정보처리 시스템"의 사례를 본 논문의 콤포넌트 통합 테스트 모형에 적용한 결과를 분석하여 기술한다. 콤포넌트 통합 테스트 모형은 UML(Unified Modeling Language)의 순서도(sequence diagram)와 협력도(collaboration diagram)를 이용하여 전체 시스템에서 UML의 사건흐름을 구성하는 콤포넌트들 사이의 인터페이스 영역에 존재하는 오류들을 추출한다. 그리고 UML을 기반으로 통합 테스트를 수행함으로써, 테스트 준비 작업을 줄이고, 기존의 UML 지원 도구들과 연계하여 테스트 자동화 도구의 구현을 앞당길 수 있다. 또한 시스템의 순차적 흐름 뿐 아니라, 동시에 수행되는 흐름에 대한 정보까지 모두 수용하여 테스트함으로써, 콤포넌트 기반의 분산 환경의 특성에 적합하다. Abstract As the object-oriented approach to software development becomes more mature, software development from pre-existing, independently developed components becomes an important aim of software engineering. Therefore, integration testing becomes an important aspect of component-based software development. However, there has been little work done in the area of the component-based integration testing. In this paper, we propose the "component integration test model" which is based on UML. Furthermore, we describe a case study on "Multimedia Information Processing System" conducted to analyse the result from which our model is applied. Our model extracts the faults, which exist in interfaces of components, using sequence diagram and collaboration diagram of UML(Unified Modeling Language). As our model is based on UML, the preparation effort for testing is reduced and its test-tools can be implemented more easily through linking existing UML tool. And our model accepts the information of concurrent flow represented by collaboration diagram as well as sequential flow, so it is more suitable to component-based distributed environment.based distributed environment.

High-level Modeling and Test Generation With VHDL for Sequential Circuits (상위레벨에서의 VHDL에 의한 순차회로 모델링과 테스트생성)

  • In this paper, we propose a modeling method for the flip-flops and test generation algorithms to detect the faults in the sequential circuits using VHDL in the high-level design environment. RS, JK, D and T flip-flops are modeled using data flow types. The sequence of micro-operation which is the basic structure of a chip-level leads to a control point where varnishing occurs to one of two micro- operation sequence. In order to model the fault of one micro-operation(FMOP) that perturb another micro-operation effectively, the concept of goal trees and some heuristic rules are used. Given a faulty FMOP or fault of control point (FCON), a test pattern is generated by fault sensitization, path sensitization and determination of the imput combinations that will justify the path sensitization. The fault models are restricted to the data flow model in the ARCHITECTURE statement of VHDL. The proposed algorithm is implemented in the C language and its efficiency is confirmed by some examples.

OSL Age Determination of the Hearths in a Bronze Age Dwelling Site by using Bayesian Statistics (베이지안 통계학을 이용한 청동기시대 주거지내 화덕자리들의 광자극발광(OSL) 연대 결정)

  • OSL dating for three hearths having the sequence of use and discard in No. 29 and 29-1 dwelling sites at Sogol cultural site was carried out. Resulting from the deconvolution of natural CW-OSL decay curve and thermal zeroing test, it was turned out that OSL signal was entirely composed of the heat- and light-sensitive fast component with high photoionization cross-section and all quartz OSL signals were thermally bleached under $300^{\circ}C$ which is the minimum temperature related to heating and cooking in Bronze age. After dose recovery test and plateau test, paleodose of each hearth sample was evaluated by using SAR method, and OSL age was determined from the ratio of paleodose to annual dose rate. For the purpose of the precision improvement of OSL age, Bayesian statistics was applied to each hearth's age and the archaeological sequence information. Finally, it could be concluded to the accurate use period of each hearth from the resultant OSL ages.

A Study on the Evaluation of Tension-Compression Fatigue Characteristics of Glass Fiber/Epoxy 4-Harness Satin Woven Laminate Composite for the Railway Bogie Application (철도차량 대차 적용 유리섬유/에폭시 4-매 주자직 적층 복합재의 인장-압축 피로특성 평가 연구)

  • This paper describes the evaluations of tension-compression fatigue characteristics and life for glass fiber/epoxy laminate composite applied to railway bogie to reduce weight. Test samples of tension-compression fatigue were composed of glass fiber/epoxy 4-harness woven laminate composites with different stacking sequence of warp-direction, fill-direction and ${\pm}45^{\circ}$-direction. The tension-compression fatigue test was conducted with stress ratio (R) of -1 and frequency of 5Hz. Goodman diagram were used to evaluate the fatigue characteristics and life of glass fiber/epoxy 4-harness satin woven laminate composite. Anti-buckling jig was designed to prevent buckling of specimen under compression load. The test results showed that the fatigue characteristics of glass fiber/epoxy 4-harness satin woven laminate composite with stacking sequence of warp-direction had a good performance in comparison with that of SM490 used to conventional metal railway bogie.

Purification and Properties of Osteopontin from Bovine Milk (우유로부터 Osteopontin의 분리.정제 및 특성에 관한 연구)

  • The purpose of this study is to observe purification and properties of osteopontin(OPN) from bovine milk. The purification of osteopontin from bovine milk was performed by using ion-exchange and hydrophobic chromatography. SDS-PAGE analysis revealed that the protein migrated at Mw. 60,000. NH2-terminal sequence analysis of the first seven amio acids revealed the protein to be identical to that previously reported for bovine OPN. 35-wk-old chickens, including 3 Single Comb White Leghorn (SCWL), were used to produce egg yolk antibody(IgY) against OPNas a antigen. However, the anti-OPN antibody activities determined by ELISA. Immunological assy of OPN in milk was performed using radial immunodiffusion test based on the standard curve of pure OPN. The radial precipitation lines of four different milk samples indicated that the concentrations of OPN in the milk samples were within the range of 31.7 to 39.7${\mu}g$/ml. On inhibition with OPN on precipitation of calcium phosphate, OPN was slightly higher than casein phosphopeptide(CPP) and poly-glutamic acid.
