송사리 태아를 이용한 농약기형독성에 관한 연구 (Teratological test of pesticide using medaka embryo)

  • This study was performed to examine the availability of using medaka (Oryzias latipes) in teratological test. Medaka embryos were collected within 2 hours post-fertilization and cultured in petri dishes containing buffered saline until hatching. The embryos were treated with 0.56 mg/l chlorpyrifos-methyl and 10 mM methyl methanesulfonate at 20 stages (about 35 hours post-fertilization). Eleven developmental features were selected and observed from 33 stages (about 9 days post-fertilization). Scoring system was developed and applicated for the measurement of potential teratological effects by the test compound. Chlorpyrifos-methyl did not induce teratological effect in medaka embryos. However, we found teratological test using medaka embryo reduced the cost, labors, period and space of experiment significantly compared with teratological study using rodents. Above findings strongly suggest that medaka embryo can be used as a lab animal model for teratogenicity test instead of rodents.

Teratological Studies of Ginkgo biloba Extract(EGb 761) in Rabbits

  • A teratological study was performed using New Zealand White rabbits to examine the teratological potential of Ginkgo biloba extract(EGb 761), which is a known strong platelet activating factor antagonist. Ginkgo biloba extract(EGb 761) was administered per intravenously during the organogenesis period (day 6th to 18th of gestation) of rabbits at dose levels of 7.5, 15, and 30 mg/kg/day. All pregnant females were sacrificed on day 29 of gestation and teratological abnormalities of their fetuses was examined. No statistically significant difference of body weight change between control and treated groups during experimental periods was noted. There was no statistically signifiant difference of numbers of corpus lutes and implantations, fetal death ratio, fetal sex ratio, and placental weight between control and rabbits exposed to three different concentration ranges of Ginkgo biloba extract (EGb 761). No marked external, visceral and skeletal abnormalities related to Ginkgo biloba extract(EGb 761) were observed in the fetuses. In conclusion Ginkgo biloba extract(EGb 761) does not show any effect on implantation or embryonic development.

병아리 태자에 대한 엔로플록사신의 최기형 독성 연구 (Teratological Effects of Enrofloxacin on the Embryos and Fetus During the Hatching of Chicken Eggs)

  • The purpose of this study was to investigate the teratological effects of enrofloxacin, a veterinary antibiotic, on the embryos and fetus of hatching chicken eggs. A control group and four experimental fertilized egg groups were set up. The four experimental groups were injected with 0.05 ml, 0.1 ml, 0.2 ml, and 0.3 ml enrofloxacin ($50{\mu}g/ml$) respectively. During the hatching period, the weights of total eggs, inside material(yolk and white) and embryos or fetuses, embryo growth, and teratological effects were investigated. In the experimental groups (fertilized eggs injected with 0.05 ml, 0.1 ml, 0.2 ml and 0.3 ml enrofloxacin), the weights of total eggs were decreased, but the yolk and white weights were increased by the higher amount of antibiotics. The weights of embryos (fetuses) in experimental groups were 86.1%, 78.6%, 65.6% and 61.4% of the control, respectively. Retarded growth, deformity and embryo loss were observed in experimental groups. Teratological effects such as undeveloped eyes, wings and legs, and deformed head and bill were also detected. In conclusion, we found that veterinary antibiotics, enrofloxacin have made teratological effects on the embryos and fetus of hatching eggs.

Teratological Study of LBD-001, a Recombinant Human Interferon $\gamma$, in Rabbits

  • LBD-001, a recombinant human interferon $\gamma$ produced by genetically engineered yeast as a host system, was intravenously administered to pregnant female rabbits (New Zealand White strain) from day 6 to 18 of gestation at dose levels of $0.35 \times 10^6$, $0. 69 \times 10^6$, and $1.38 \times 10^6$ I.U./kg/day. Hydrocortisone sodium succinate (0.3 mg/kg/day) was also given in the same way. Teratological effects of the test agents on the organogenesis of fetuses and the development of offsprings (F1 rabbits) were investigated. The results were as followings: (1) No significant changes by the treatment of LBD-001 or hydrocortisone sodium succinate were observed in the body weights, the food and water consumption, the lactating or nurshing behaviors, and the autopsy of the pregnant rabbits. (2) No significant changes in the resorption rate, the fetal organogenesis, and the normal develpoment of offsprings (F1) by the treatment of LBD-001 or hydrocortisone sodium succinate were detected. The results show that LBD-001 at the dose of $1.38 \times 10^6$ I.U./kg/day or less and hydrocortisone sodium succinate at the dose of 0.3 mg/kg/day are neither teratogenic in the organogensis of the fetuses and the development of the offsprings (F1) nor toxic to the mother rabbits.

Teratological Study of LBD-001, a Recombinant Human Interferon $\gamma$, in Rats

  • LBD-001, a recombinant human interferon ${\gamma}$ produced by genetically engineered yeast as a host system, was intravenously administered to pregnant female rats (Sprague-Dawley) from day 7 to 17 of gestation at dose levels of 0.35$\times$10$^{6}$ , 0.69$\times$10$^{6}$ , and 1.38$\times$10$^{6}$ I.U./kg/day. As the control groups, hydrocortisone sodium succinate (5 or 10 mg/kg/day) was also similarly administered. Teratological effects of the test agents on fetuses and development of offsprings (F1 rats) were investigated. (1) No significant changes by the treatment of LBD-001 were observed in body weight, food and water consumption, feeding and nursing behaviors, and autopsy of pregnant or lactating mother rats. However, in hydrocortisone sodium succinate (10 mg/kg/day)-treated group, significant decreases of body weight on day 16, 18, and 20 of gestation and food consumption on day 20 of gestation and outstanding atrophy of thymus and adrenals were observed in two rats autopsied on day 20 of gestation. (2) No significant changes in resorption rate, skeletal or visceral development of fetuses, and physical or sensory development of offsprings (Fl) by the treatment of LBD-001 were detected. In hydrocortisone sodium succinate (10 mg/kg/day)-treated group, however, there were significant decreases of body weight of fetuses, delay of ossification, temporary delay of body weights of offsprings (F1) on day 1 and 3 of lactation, and increased tendency of stillborn rate and malformation rate of bone. The results show that LBD-001 at the dose of 1.38$\times$10$^{6}$ I.U./kg/day or less is not teratogenic in organogenesis of fetuses and the development of offsprings (F1). Meanwhile, hydrocortisone sodium succinate (10 mg/kg/day) seems to delay ossification of fetuses and temporarily retard the development of offsprings (Fl).

  • Teratological study on pranoprofen, as antiinflammatory agent, was conducted by oral intubation in Sprague-Dawley rats. Pranoprofen was administered doses of 1.0, 2.5 and 5.0mg/kg/day and doze of 0.5mg/kg/day of Indomethacin was used as positive control. The rats were dosed from day 7 to 17 of gestation. At necropsy on day 20 of gestation, pathologically changes of gastroin-testinal system, liver and adrenal gland were examined at the high dose administered group. There were no differences between control and treated group on the number of implantations, the number of alive and dead fetuses, tail length, and external visceral and skeletal malformations.

의약후보물질의 생식독성평가 원칙 및 방법 (Principles and Methods for the Reproductive-toxicological Evaluation of New Drug Candidates)

  • The purpose of reproductive toxicity studies is to evaluate all effects resulting from paternal or maternal exposure that interfere with conception, development, birth, and maturation of offspring. In 1966, the US Food and Drug Administration (US FDA) published guidelines for a three-segment study for drug testing to examine adverse effects on fertility and pregnancy. Three segments were proposed: Segment I, Study of Fertility and General Reproductive Performance, to provide information on breeding, fertility, nidation, parturition, neonatal effects and lactation: Segment II, Teratological study, to provide information on embryo toxicity and teratogenicity: and Segment III. perinatal and Postnatal Study, to provide information on late fetal development, labour and delivery, neonatal viability, and growth and lactation. The classic guideline is still used to this day with only monor modification throughout the world. In the present review, the principles and methods of reproductive toxicity studies are discussed with special attention given to scientific issues.

한국산 목본식물에 대한 계통분류학적 연구 -녹나무과의 외부 형태- (Systematic Studies on Some Korean Woody Plants -External Morphology of Lauraceae-)

  • Floral structures of 11 korean lauraceous species belonging to six genera have been studied. Machilus is a primitive genus having relatively large bisexual flower, six tepals and staminodes in fourth whorl of stamen. The flowers of Neolitsea are highly reduced state as unisexual flower, six stamens arranged to three whorls and no staminode in the flower, so regared as more advanced group. Intrafamilial classification systems of Nakai (1939) and Pax (1891) are more reasonable but still have some discrepancies with results. The origin of tepals of this group has been a long controversial problem; whether the differentiation of tepals occur. Teratological study of Lindera erythrocarpa, inner tepals of this group must be originated from the staminode and outer tepals from leafy tepals. Although morphological differentiation does not sitll occur, inner and outer whorls of tepals are originated through different way.

  • A teratogenicity study was carried out on Sprague-Dawley rats which have been given the intravenously or intraperitonealy injections of rHuIFN-${\alpha}$A, an available therapeutic agent, at dose levels of $1{\times}10^5$, $4{\times}10^5$ and $1.2{\times}10^6$ I.U/kg/day for a period of 11 days from day 7 to day 17 of gestation. Two-thirds of the pregnant females in each group were sacrificed on day 20 of gestation and their fetuses were examined. The remaining dams were allowed to litter naturally, and the postnatal development of the off springs was observed. No changes were observed in all aspects of parameters between the treated and the control dams. The incidence of external, internal, and skeletal anomalies were not significantly increased in the fetuses of any treated groups. The rHuIFN-${\alpha}A$ caused no effects on parturition, lactation, and postnatal growth.

The Effects of Multi-minerals on Susceptibility to Lead Toxicity in Rats

  • Female Wistar rats were randomly divided into 5 groups: Control, received distilled water; Low lead, received 0.5 g/ιlead (as acetate) in drinking water; High lead, received 2.0 g/ιlead; Low lead + Minerals, received 0.5 g/ιlead in drinking water and received minerals (Ca$^{2+}$, 25 mg/kg/day; Fe$^{3+}$, 0.47 mg/ kg/day; Zn$^{2+}$, 0.33 mg/kg/day; Se, 0.83 $\mu\textrm{g}$/kg/day) by gavage; High lead + Minerals, received 2.0 g/ιlead and received the same minerals. Animals exposure to lead was from 10 days before mating till postnatal day 21; and the minerals was administered from the first day of pregnancy and during lactation. No statistical difference was found either in body weights or in blood lead levels between the pups received minerals and those only exposed to lead at the same dose. The developmental and behavioral teratological effects of lead on pups, such as time-lag of eye opening, pinna detachment, fur developing, incisor eruption, ear unfolding, and surface righting were observed in this study; and the minerals decreased the toxicity of lead either in low or in high lead exposure pups. The numbers of step-down were significantly increased in lead exposed animals, and the effect of intervention by the minerals was appeared only in the pups exposed to low lead. The ChAT activity and levels of glutamate and aspartate in hippocampus decreased in treated animals compared to control animals, no effect of intervention by the minerals was found. The results of this study indicate that the applied multi-minerals can alter the outcome of develop-mental lead poisoning in rats.s.s.s.
