Stable expression of N-terminal 3X-FLAG tagged human 5a-reductase type II in 293 cells: a new tool for protein purification & inhibitor screening

  • Human 5-reductase type II(5AR2) is an important target for the treatment of benign prostatic hyperplasia. In this study we describe the establishment of cell line which stably expressed 3X FLAG tagged human 5AR2. We used this cell line as a cell based assay tool and source for 5AR2 enzyme. First a plasmid (3XFLAGpCMVl0-5AR2) for the expression of 5AR2 was constructed by the use of the vector 3XFLAGpCMV10 and transfected into the HEK 293. By selection with G418 sulfate. ten HEK 293 single cell clones were obtained of which three stably exhibited high 5AR2 activity. (omitted)

공대 신입생들의 팀 활동 만족감과 관련 요인에 관한 연구 (A Study of Team Satisfaction and Associated Factors of Engineering College Freshmen)

  • 본 연구에서는 공대의 교과과정에서 흔히 요구되는 팀 활동에 대해 신입생들을 대상으로 팀 활동의 만족감과 관련 요인들을 분석하였다. 입문교과를 수강하는 신입생 64명을 대상으로 13개의 팀을 구성하여 총 5개의 개방형 과제를 통해 팀 활동을 수행하게 한 후 설문조사를 실시하였다. 팀 활동에 대한 전반적인 만족감은 팀의 성과에 긍정적인 영향을 미침을 확인할 수 있었다. 그러나 팀 만족도는 팀의 성과보다는 팀원의 활동 참여도와 팀원 간의 관계가 좋다고 인식하는 정도에 더욱 상관관계가 있음을 발견하였다. 학생들은 상호 배려하는 화목한 팀 분위기가 조성될 때 팀 활동의 만족감이 증가하며, 팀의 분위기가 성과보다 중요하다고 느끼고 있었다. 한편 팀원의 역할별 구성은 팀 활동의 만족도와 직접적인 관계가 없었으며, 또한 자율적인 팀 구성에 대한 선호도가 높음을 알 수 있었다. 이러한 결과를 바탕으로 학생들의 팀 활동 만족감이 극대화되도록 지원을 한다면 특히 최근 학제적 융복합 학습 환경에서의 팀 활동을 통한 교육적인 효과가 높아질 것으로 기대한다.

  • Many construction activities are related because they share the information of working methods and resources. Generally, the design information for coupled activities needs to be constantly collaborated in the different teams. To achieve the improvement in team collaboration, it is necessary to identify the relative coupled activities in the design teams. The activity and work partitioning arrangements are also required to accommodate the appropriate team members. This paper presents an integral method to be an evaluation in improving the collaboration for teams partitioning. A model, Team Partitioning Method (TPM) was developed to clarify the relationships between activities in a team. The results show the applicability of TPM model in team partitioning for design collaboration.

  • Education in Korean universities is rapidly expanding to online education due to COVID-19. In response to such changes, this study proposed a means of improving the learning management system of Korean universities and analyzed the effects of using the system. The important results of this study are as follows: the learning management system was composed of 'pre-class learning,' 'team activity,' and 'participation learning' to support team-based learning. The effects that the users (instructors, learners) can obtain by adopting team-based learning and using the system were analyzed. The study concludes that for instructors, teaching work may be alleviated. For learners, it was demonstrated that they could more easily access and use data required for their education.

Renal protective effects of zingerone in a mouse model of sepsis

  • Zingerone (ZGR), a phenolic alkanone isolated from ginger, has been reported to possess pharmacological activities such as anti-inflammatory and anti-apoptotic effects. This study was initiated to determine whether ZGR could modulate renal functional damage in a mouse model of sepsis and to elucidate the underlying mechanisms. The potential of ZGR treatment to reduce renal damage induced by cecal ligation and puncture (CLP) surgery in mice was measured by assessment of serum creatinine, blood urea nitrogen (BUN), lipid peroxidation, total glutathione, glutathione peroxidase activity, catalase activity, and superoxide dismutase activity. Treatment with ZGR resulted in elevated plasma levels of BUN and creatinine, and of protein in urine in mice with CLP-induced renal damage. Moreover, ZGR inhibited nuclear $factor-{\kappa}B$ activation and reduced the induction of nitric oxide synthase and excessive production of nitric acid. ZGR treatment also reduced the plasma levels of interleukin-6 and tumor necrosis $factor-{\alpha}$, reduced lethality due to CLP-induced sepsis, increased lipid peroxidation, and markedly enhanced the antioxidant defense system by restoring the levels of superoxide dismutase, glutathione peroxidase, and catalase in kidney tissues. Our study showed renal suppressive effects of zingerone in a mouse model of sepsis, suggesting that ZGR protects mice against sepsis-triggered renal injury.

Suggestion and Application of Emergency Simulation Educatin using Real-time Video Observation for Inactive Nurses

  • This study is a pilot study to confirm the effectiveness of training after applying emergency simulation training for inactive nurses and to present a new model of simulation training operation method. In this study, the control group is a group that directly participates in the simulation activity, and the experimental group is the group that observes the control group's simulation activity. Experimental group and control group were matched 1:1 to experience all the roles of the resuscitation team. The study participants were 5 inactive nurses in the experimental group and 5 inactive nurses in the control group, and the total training time was 5 hours. The emergency simulation operation composition consists of theory education, skill education, and simulation. The interview was conducted. The educational satisfaction of the participants was 4.65 points for theory education and 4.70 points for practical education based on 5 points. Participants' performance confidence improved from 3.60 points before operation to 7.20 points after operation. Emergency simulation operation consisted of pre-test, theory education, skill education, simulation implementation, debriefing, and post-test. Participants expressed that the choice of group greatly reduced the burden and anxiety about performing the role of the resuscitation team. However, difficulties and inexperience in the operation of the defibrillator were reported in the experimental group. The control group reported that the simulation activity of the experimental group was not significantly different from theirs. Through the results of this study, it was confirmed that emergency simulation education not only reduced the burden and anxiety of inactive nurses, but also had an effect of education. Based on the research results, it is proposed to expand the participants and verify the effectiveness of education through specific variables such as learning commitment, learner confidence, simulation satisfaction, and team effectiveness.

Maqui Berry Extract Activates Dendritic Cells Maturation by Increasing the Levels of Co-stimulatory Molecules and IL-12 Production

  • Dendritic cells play a very important role in the immune response as antigen-presenting cells that are critical for initiating both innate and acquired immunity. They recognize, process and present foreign antigens to other key immune cells to trigger and regulate the immune response. The ability to activate these dendritic cells can be used as a treatment for various immune diseases. Maqui berry has been reported to have anticancer, antibacterial and anti-inflammatory properties. However, its effect on the activity of dendritic cells has not been studied. In this study, we investigated the efficacy of maqui berry extract in modulating dendritic cell activity. Treatment of dendritic cells with maqui berry extract induced the costimulatory molecules CD80, CD86, and MHC class I and II in a concentration-dependent manner. Furthermore, the antigen-presenting capacity of dendritic cells was inhibited, which confirms their ability to present antigens, and the production of Interleukin (IL)-12, which is important for dendritic cell activity, was increased. These results indicated that Maqui berry extract activates dendritic cells maturation by inducing the production of co-stimulatory molecules and IL-12. These results suggest that maqui berry extract may act as an effective adjuvant to enhance dendritic cell-based immune responses.

가미소요산의 새로운 제형에 대한 항산화 활성 및 항염증 효능평가 (Evaluation on Anti-oxidant Activity and Anti-inflammatory Effects for the New Formulation of Gamisoyosan)

  • Objectives : Gamisoyosan (GMS) is a useful prescription for treating insomnia, dysmenorrhea and infertility induced by a stress. Also, GMS has been used traditionally to improve systemic circulation and biological energy production. The purpose of this study was to assess the anti-oxidant activity and anti-inflammatory effects of Gamisoyosan Formulation (Soft extract, GMS-SE). Methods : The biological activities such as anti-oxidant and anti-inflammatory effects were measured through cell line-based in vitro assay. We investigated the anti-oxidant properties of GMS-SE on the 1,1-diphenyl-2-picryhydrazyl (DPPH) radical, contents of total flavonoid and polyphenol. GMS-SE compared to butyl hydroxy anizole (BHA). Furthermore, based on this result the anti-inflammatory effects of GMS-SE have verified by mechanism from LPS- treated Raw264.7 macrophages. Results : The anti-oxidant activities of GMS-SE increased markedly, in a dose-dependent manner. The GMS-SE showed significant scavenging activity (GMS-SE $500{\mu}g/m{\ell}$ : $32.77{\pm}1.65%$, GMS-SE $1000{\mu}g/m{\ell}$ : $45.06{\pm}1.04%$ and BHA $100{\mu}g/m{\ell}$ : $39.25{\pm}2.41%$ for DPPH assay). and, The total phenolic compound and flavonoids contents of GMS-SE were $73.93{\pm}6.87{\mu}g/mg$ and $698.75{\pm}6.78{\mu}g/mg$. GMS-SE which is LPS has diminished in the LPS-induced release of inflammatory mediators (NO, iNOS, COX2 and PGE2) and pro-inflammatory cytokines (TNF-${\alpha}$, IL-6 and IL-$1{\beta}$) from the RAW264.7 macrophages. Moreover, GMS-SE inhibited the activation of phosphorylation of p38 and ERK MAPKs by induced LPS. Conclusion : The present results indicate that GMS-SE has an anti-oxidant and anti-inflammatory properties, therefore may be beneficial in diseases which related to oxidative stress-mediated inflammatory disorders.

  • General contractors' overhead costs are increasing relative to direct costs. However, it is difficult to apply the traditional activity-based costing directly to the construction site overhead costing because the resource consumption rate per each activity is varied depending on the attributes of activities. The research develops a methodology of hybrid cost allocation system when resources are assigned to cost objects unlike the traditional activity-based costing. The study also develops a database program and demonstrates how it can be applied to the construction projects using a case study.

현미 발효 슬러리의 항균활성 (Antimicrobial Efficiency in the Fermented Slurry of Unpolished Rice)

  • 현미는 도정한 백미보다 이로운 영양분을 더 많이 함유하고 있다. 본 연구에서는 현미를 이용하여 만든 현미 발효 슬러리의 이화학적 특성과 항균활성에 대해 시험하였으며, 현미발효슬러리의 항균활성은 paper disc-agar diffusion 방법을 이용하여 6가지 병원성 균주(Staphylococcus aureus, Escherichia coli, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella typhimurium and Yersinia enterocolitica)와 2가지 발효균주(Gluconacetobacter intermedius and Lodderomyces elongisporus)에 대해 항균성을 조사하였다. 특히, Staphylococcus aureus, E. coli, Listeria monocytogenes, P. aeruginosa, Salmonella typhimurium, Y. enterocolitica 그리고 Lodderomyces elongisporus에 대해서는 시판 항생제인 카베니실린과 테트라사이클린보다 더 높은 항균활성을 보였다. 항산화활성은 2,2-diphenyl-1-picrylhydrazyl (DPPH) 라디칼 소거능을 이용하여 측정하였을 때, 대표되는 항산화제인 아스코르빅 산과 비슷한 활성을 나타내었다. 현미발효슬러리의 발효중에 나타나는 균주를 동정하기 위해 TSB 고체배지와 YPD 고체배지에 현미발효슬러리를 도포하였을 때, 분리된 콜로니를 16S rDNA sequence 분석을 통하여, 네가지 균주를 분리하였으며, phylogenic tree 분석법을 이용하여 조사하였을 때, 각각 G. intermedius, Lactobacillus casei, Lactobacillus plantarum 그리고 Acetobacter peroxydans와 유사하였다.
