The Dynamic Basement Amplification Characteristics of a Dam Site using a Reference Site Method (기준관측소 방법을 이용한 댐체 기반암의 동적 지반증폭특성)

  • Observed ground motions are composed of three factors such as, seismic source, attenuation, and site amplification effect. Among them, the site amplification characteristics should be considered significantly when estimating seismic source and attenuation characteristics with more confidence. The site effect is also necessary when estimating not only seismic hazard in seismic design engineering but also rock mechanical properties. This study uses the method of H/V spectral ratio of observed ground motion between target site and reference site called a reference site method. In addition to using the vertical Fourier spectrum of the reference site, we try out the horizontal Fourier spectrum as a new method in this study. We analyze H/V spectral ratio of six ground motions respectively, observed at four sites close to Yedang Reservoir. We then compare site amplification effects at each site using 3 kinds of seismic energies including S waves, Coda waves energy, and background noise. The results suggest that each site showed similar site amplification patterns in S waves and Coda waves energy. However, the site amplification of background noise shows much different characteristics from those of S waves and Coda wave energy, which suggests that the background noises at each site have their own developing mechanism. Each station shows its own characteristics of specific resonance frequency and site amplification properties in low, high and specific resonance frequency ranges. Comparison of the method used in this study to the others that used different methods can provide us with more information about the dynamic amplification of a site characteristics and site classification.

Method for Screening and Confirming Meldonium in Human Urine by High-Resolution Mass Spectrometry and Identification of Endogenous Interferences for Anti-Doping Testing

  • Meldonium is a drug for treating ischemia by expanding the arteries but it can also enhance the performance of sports players. The World Anti-Doping Agency (WADA) has included it in the list of prohibited substances since 2016. Meldonium is one of the challenging substances for anti-doping testing because it is difficult to recover by general liquid-liquid or solid phase extraction due to its permanent charge and high polarity. Therefore, high-performance liquid chromatography (HPLC) is currently used by injecting a diluted urine sample (known as the "dilute-and-shoot" strategy). There is no loss of target compounds in the extraction/cleanup procedure but its high matrix effect could interfere in their separation or detection from the endogenous urinary compounds. We report a single method using high-resolution mass spectrometry that can be used for both screening and confirmation, which follows the "dilute-and-shoot" strategy. In this method, the endogenous compounds' interfering peaks in the mass spectrum are separated at a high resolution of FWHM 140,000, and the results are suitable for substance detection following the WADA guidelines. The interferences in the obtained mass spectrum of the urine matrix are identified as acetylcholine, lysine, and glutamine by further analysis and database searching. Validation of the method is performed in routine anti-doping testing, and the limit of detection is 50 ng/mL. This method uses simple sample preparation and a general reverse phase HPLC column, and it can be easily applied to other substances.

  • Hydrogen analysis by elastic recoil detection has been performed utilizing polyimide film as a reference sample of known hydrogen content assuming the soundness of ion beam current integration. However beam current integration at higher incidence angle is not reliable. Scattering yield per unit fluence by current integration which is normalized per unit path length decreases as the sample tilt angle is getting higher. Moreover because beam current integration at high tilt angle is incomplete, hydrogen evaluation is very risky by direct comparison of sequentially collected recoil spectra between reference and target sample. In this study, primary ion beam dose is determined by backscattering spectrum that is collected simultaneously with recoil spectrum instead of ion beam current integration in order to reduce uncertainty arising in the process of current integration and to enhance the reliability of quantitative analysis. Three test samples are selected $-7.6{\mu}m$ polyimide film, hydrogen implanted silicondioxide and Au deposited carbon wafer- and analyzed by two methods and compared.

Fabrication and Characteristics of High Brightness White Emission Electroluminescent Device (고휘도 백색방출 전계발광소자의 제작 및 특성)

  • White emission thin film electroluminescent device was fabricated using ZnS for phosphor layer and BST ferroelectric thin film for insulating layer. For fabrication conditions of BST thin film, stoichiometry of target was $Ba_{0.5}Sr_{0.5}TiO_3$, substrate temperature was $400^{\circ}C$, working pressure was 30 mTorr, and A:$O_2$ ratio was 9:1. At this time, dielectric constant was 209 at 1kHz frequency. For phosphor layer ZnS:Mn, ZnS:Tb, and ZnS:Ag were used. Mixing rates of activators were respectively 0.8, 0.8, and 1 wt%. Total thickness of phosphor tapers was 500 nm, thickness of lower insulating layer was 200 nm, and thickness of upper insulating layer was 400 nm. In this conditions, luminescence threshold voltage of thin film electroluminescent device was $95\;V_{rms}$, maximum brightness was $3,000\;cd/m^2$ at $150\;V_{rms}$. Luminescence spectrum peak was observed at region of blue(450 nm), green(550 nm), and red(600 nm).

Low Noise Vacuum Cleaner Design (저소음 청소기 개발)

  • Vacuum cleaner is a close life product that can remove various dusts from our surroundings. However well vacuum cleaner clean our environments, many people are looking away from it, due to its loud noise. Its noise causes a big trouble in the usual life, for example, catch calls, TV watching and discussing etc. To reduce these inconveniences, noise reduction methods and systematic design of low noise vacuum cleaner are studied in this paper. At first, sound quality investigation is performed to get the noise level and quality that make people TV watching and catch calls available. Based on the European and domestic customer SQ survey result, sound power, peak noise level and target sound spectrum guideline are studied and introduced. As a second, precise product sound spectrums are designed into each part based on the sound quality result. Fan-motor, brush, mainbody, cyclone spectrums are decided to get the final target sound based on the contribution level. Fan-motor is the major noise source of vacuum cleaner. Specially, its peak sound, RPM peak and BPF Peak, cause the people nervous. To reduce these peak sounds, high rotating impeller and diffuser are focused due to its interaction. A lot of experimental and numerical tests, operation points are investigated and optimization of flow path area between diffusers is performed. As a bagless device, cyclones are one of the major noise sources of vacuum cleaner. To reduce its noise, previous research is used and adopted well. Brush is the most difficult part to reduce noise. Its noise sources are all comes from aero-acoustic phenomena. Numerical analysis helps the understanding of flow structure and pattern, and a lot of experimental test are performed to reduce the noise. Gaps between the carpet and brush are optimized and flow paths are re-designed to lower the noise. Reduction is performed with keeping the cleaning efficiency and handling power together and much reduction of noise is acquired. With all above parts, main-body design is studied. To do a systematic design, configuration design developments technique is introduced from airplane design and evolved with each component design. As a first configuration, fan-motor installation position is investigated and 10 configuration ideas are developed and tested. As a second step, reduced size and compressed configuration candidates are tested and evaluated by a lot of major factor. Noise, power, mass production availability, size, flow path are evaluated together. If noise reduction configuration results in other performance degrade, the noise reduction configuration is ineffective. As a third configuration, cyclones are introduced and the size is reduced one more time and fourth, fifth, sixth, seventh configuration are evolved with size and design image with noise and other performance indexes. Finally we can get a overall much noise level reduction configuration. All above investigations are adopted into vacuum cleaner design and final customer satisfaction tests in Europe are performed. 1st grade sound quality and lowest noise level of bagless vacuum cleaner are achieved.

Overexpression and Periplasmic Transport of 5-Enolpyruvylshikimate 3-Phosphate Synthase in E. coli (대장균에서 5-Enolpyruvylshikimate 3-Phosphate Synthase의 대량 발현 및 Periplasmic Space로의 Transport)

  • 5-Enolpyruvylshikimate 3-phosphate(EPSP) synthase is the sixth enzyme of the shikimate pathway that synthesizes aromatic amino acids. The enzyme is a primary target for the glyphos'lte which is a broad-spectrum and environmetally safe herbicide. As a first step toward development of glyphpsate-resistant EPSP synthase, the EPSP synthase gene(aroA) was amplified by polymerase chain reaction and cluned into pET-25b vector. In this construct. designated pET-aro, the aroA gene is expressed under control of strong T7 promoter. and the EPSP synthase is produced as a fusion protein with pelB leader at N-terminus and HSV-tag and His-tag at C-terminus. When the pET-aro clone was induced to produce the enzyme, it was found that the EPSP synthase was successfully exported to peri plasmic space. The periplasmic transport was greatly dependent on the induction temperatures. Among the induction temperatures examined($25^{\circ}C$, $30^{\circ}C$, $34^{\circ}C$ and $37^{\circ}C$). induction at $34^{\circ}C$ gave rise to maximal periplasmic transport. The recomhinant EPSP synthase could have been purified hy $Ni^{2+}$ -affinity chromatography using the His-tag. and detected hy anti-HSV -tag antibody. The recombinant EPSP synthase also hound to phosphocellulose resin and was eluted hy shikimate 3-phosphate and phosphoenolpyruvate. as expected. The recombinant EPSP synthase purified from phosphocellulose resin showed typical EPSP synthase activity.

Enhanced Pathogenicity of Baculovirus Using Immunosuppressive Genes Derived From Cotesia plutellae Bracovirus (폴리드나바이러스(CpBV) 유래 면역억제 유전자를 이용한 베큘로바이러스 병원력 제고 기술)

  • Baculoviruses have been used to control some serious lepidopteran pests. However, their narrow target insect spectrum and slow efficacy are main limitations to be used in various applications. This study introduces a technique to overcome these limitations by inhibiting insect immune defence to enhance the viral pathogenicity. Polydnaviruses are an insect DNA virus group and symbiotic to some ichneumonid and braconid endoparasitoids. Cotesia plutellae bracovirus (CpBV) is a braconid polydnavirus and encodes several immunosuppressive genes. We selected seven CpBV genes and recombined them to wild type Autographa California multiple nucleopolyhedrovirus (AcNPV). A bioassay of these seven recombinants indicated that most recombinants had similar or superior efficacy to wild type AcNPV against beet armyworm, Spodoptera exigua, and diamondback moth, Plutella xylostella. Recombinant AcNPV with CpBV-ELP was the most potent in terms of lethal time by shortening more than 2 days compared to wild type AcNPV. This recombinant was further proved in its dose-dependent pathogenicity and its efficacy by spray application on S. exigua infesting cabbage cultivated in pots. We discussed the efficacy of CpBV-ELP recombinant AcNPV in terms of suppressing antiviral activity of target insects.

Evaluation of Image Quality using Monte Carlo Simulation in Digital Mammography System (디지털유방영상시스템에서 몬테카를로 시뮬레이션을 이용한 영상평가)

  • For the purpose of early diagnosis of the breast cancer, the attention on the screening mammography has been increasing-with supply of digital mammography day by day. Image quality is decided by target materials and filter combinations. Optimized selection by a glandular density and a thickness is needed, because these combinations change x-ray spectrum and effect to image quality. The purpose of this study is to find out optimized target and filter combinations through the evaluation of quantitative image quality and to suggest means which minimize patient dose through MCNPX. In results, spatial frequency resolution evaluation which is quantitative image quality evaluation method, MTF, NPS, DQE shows that we have to select Mo/Mo combinations or Mo/Rh combinations when compressed breast is thin. but in case of that when compressed breast is thick, we have to select Rh/Rh combinations or W /Rh combinations. In a comprehensive evaluation, W!Rh combinations which are not used in thin breasts in practice was superior to all image quality evaluation. This result is somewhat different-with clinical examination results. Secondary end point was organ dose evaluation, radiation dose of opposite breast was approximately 47 ~73% effectiveness when selecting standard breast. In conculsion, the most important point is that we have to select the optimal combinations-with considering dose evaluation and various thickness.

A Case Study on the Professional Education Using SAFMEDS Teaching Strategy (SAFMEDS 교수전략을 적용한 전문가 교육 사례연구)

  • This study reported a case study that showed educational usefulness of SAFMEDS (Say All Fast a Minute Every Day Shuffled) on the improvement of Fluency. The participants were 3 experts with special teacher and speech and pathology, who enrolled a graduate level course, Research in Children with Autism Spectrum Disorder. The SAFMEDS strategy was employed as a study tool for the participants to acquire fluent verbal repertoires related to the key terminologies of Skinner's (1957) analysis of verbal behavior, list 60 pairs of terms. The participants developed 60 term flash cards which presented a target term on the front of the card, and its definition on the back. During the intervention, the participants were required to see the definition and says its term. The results of this study indicated that the SAFMEDS was effective to improve participants' fluent verbal repertoires in terms of both accuracy and fluency. The results of this study would be able to contribute for education professionals to improve certain target operant's accuracy and fluency.

3D Fusion Imaging based on Spectral Computed Tomography Using K-edge Images (K-각 영상을 이용한 스펙트럼 전산화단층촬영 기반 3차원 융합진단영상화에 관한 연구)

  • The purpose of this study was to obtain the K-edge images using a spectral CT system based on a photon-counting detector and implement the 3D fusion imaging using the conventional and spectral CT images. Also, we evaluated the clinical feasibility of the 3D fusion images though the quantitative analysis of image quality. A spectral CT system based on a CdTe photon-counting detector was used to obtain K-edge images. A pork phantom was manufactured with the six tubes including diluted iodine and gadolinium solutions. The K-edge images were obtained by the low-energy thresholds of 35 and 52 keV for iodine and gadolinium imaging with the X-ray spectrum, which was generated at a tube voltage of 100 kVp with a tube current of $500{\mu}A$. We implemented 3D fusion imaging by combining the iodine and gadolinium K-edge images with the conventional CT images. The results showed that the CNRs of the 3D fusion images were 6.76-14.9 times higher than those of the conventional CT images. Also, the 3D fusion images was able to provide the maps of target materials. Therefore, the technique proposed in this study can improve the quality of CT images and the diagnostic efficiency through the additional information of target materials.
