LPS로 유도한 RAW 264.7 세포의 염증반응에서 흰민들레의 항염증 효과 (The anti-inflammatory effect of Taraxacum coreanum on lipopolysaccharide induced inflammatory response on RAW 264.7 cells)

  • Objectives : Taraxacum coreanum (TC) have been used as a traditional medicine to treat inflammatory diseases and anti-oxidant effect in Korea. However, the anti-inflammatory effect of TC water extract on lipopolysaccharide (LPS)-induced inflammation is not well-known. Therefore, this study was performed to identify the anti-inflammatory effect of TC on LPS induced inflammatory. Methods : RAW 264.7 cells were treated with 500 ng/mL of LPS. Water extracts of TC (0.1, 0.25, 0.5 mg/ml) was treated 1 h prior to LPS. Cell viability was measured by MTT assay. Levels of nitric oxide (NO) were measured with Griess reagent and pro-inflammatory cytokines were measured by enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (real-time PCR). We also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor-B ($NF-{\kappa}B$) activation by western blot. Results : Water Extract from TC itself did not have any cytotoxic effect in RAW 264.7 cells. TC treatment inhibited the production of NO production, and pro-inflamamtory cytokines such as interleukin (IL)-6 and $IL-1{\beta}$ on protein and mRNA levels. In addition, TC treatment inhibited the LPS-induced activation of MAPKs such as extracellular signal-regulated kinase1/2 (ERK1/2), p38 kinases (p38), c-Jun $NH_2$-terminal kinase (JNK) and $NF-{\kappa}B$. Conclusions : In summary, our result suggest that treatment of TC could reduce the LPS-induced inflammation. Thereby, TC could be used as a protective agent against inflammation. Also, this study could give a clinical basis that TC could be a drug or agent to prevent inflammation.

Carthamus tinctorius seeds-Taraxacum coreanum combination attenuates scopolamine-induced memory deficit through regulation of inflammatory response and cholinergic function

  • BACKGROUND/OBJECTIVES: There is growing interest in herbal medicines for managing age-related diseases, such as Alzheimer's and Parkinson's. Safflower seeds (Carthamus tinctorius L. seeds, CTS) and dandelions (Taraxacum coreanum, TC) are widely used to treat bone- or inflammation-related diseases in Oriental countries. This study investigated the protective effect of the CTS-TC combination on scopolamine (Sco)-induced memory deficits through inflammatory response and cholinergic function. Moreover, marker components such as serotonin, N-(p-coumaroyl) serotonin, N-feruloylserotonin, chlorogenic acid, and chicoric acid in the CTS-TC combination were analyzed for their potential benefits on memory function. MATERIALS/METHODS: Water extracts of CTS, TC, and the CTS-TC combination at various ratios (4:1, 1:1, and 1:4) (100 mg/kg) were orally administered to mice for 14 days. Sco (1 mg/kg) was intraperitoneally injected into the mice before each behavioral test. T-maze and novel object recognition tests were conducted to monitor behavioral changes after the treatment. Western blotting was performed to detect protein expression. In addition, the presence of 5 biomarkers, serotonin, N-(p-coumaroyl) serotonin, N-feruloylserotonin, chlorogenic acid, and chicoric acid, was analyzed using high-performance liquid chromatography (HPLC). RESULTS: Behavioral tests showed that the CTS-TC combination enhanced memory function in Sco-injected mice. Inflammation-related proteins (inducible nitric oxide synthase, cyclooxygenase-2, and glial fibrillary acidic protein) were downregulated after treatment with the CTS-TC combination. The acetylcholinesterase protein expression was also downregulated. HPLC analysis revealed that N-feruloylserotonin and chicoric acid were the predominant components, followed by N-(p-coumaroyl) serotonin, chlorogenic acid, and serotonin. CONCLUSION: These findings suggest that the CTS-TC combination protects against Sco-induced memory deficits by inhibiting inflammatory responses and cholinergic dysfunction. N-feruloylserotonin and chicoric acid, along with N-(p-coumaroyl) serotonin, chlorogenic acid, and serotonin, might be biomarkers for the CTS-TC combination, and their effects on memory protection warrant further study.

Remedy effects of dandelion and milk thistle on fatty liver hemorrhagic syndrome in laying hens

  • Fatty liver hemorrhagic syndrome (FLHS) is a metabolic disorder found in caged layer hens and causes reduced egg production and sudden death. Dandelion (Taraxacum coreanum, TC) and milk thistle (Cirsium japonicum var. ussuriense, CJ) are well known wild herbs inhabiting Korean peninsula and presenting antioxidative effects. This study investigated alleviate effects of these herbal mixture (6:4, w/w) composed of dried powder of TC and CJ against fatty liver in laying hens. The herbs mixture 5.0, 10.0, 20.0 or 40.0 g/kg feed was provided via feed admixture for 3 weeks to laying hens having FLHS. FLHS was induced by intramuscular injection of β-estradiol (2 mg/kg bw) 2 times per week for 3 weeks and supply with high caloric feed. Egg production rate was reduced from 76.2% at pre-treatment to 49.4% at 1 week and further decreased according to β-estradiol treatment. Increment of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), total cholesterol (T-Chol) and total bilirubin (T-Bil) and decrement of serum superoxide dismutase (SOD) and glutathione peroxidase (GPX) with fatty liver were found by the treatment of β-estradiol. Supplementation of TC and CJ mixture via feed admixture recovered the reduction of egg production and attenuated serological changes and gross and pathological lesions of fatty liver with the best amelioration effects at 5 and 10 g TC and CJ mixture per kg feed. In conclusion, TC and CJ mixture attenuates FLHS by means of antioxidative effects. Further mechanistic study is required to explain TC and CJ's amelioration effects against FLHS in laying hens.

흰 민들레 뿌리의 항염증 및 산화 스트레스 개선 효과 (Effects of Root of Taraxacum coreanum Nakai on the Inhibition of Inflammation and Oxidative Stress Induced by Lipopolysaccharide in ICR Mice)

  • 한방에서 염증 치료 목적으로 사용하는 흰 민들레 뿌리의 열수 추출물을 제조하여 항염증 효과 및 산화 스트레스 개선효과를 확인하고자 하였다. ICR mice에 lipopolysaccharide(LPS)를 1회 복강 투여하여(10 mg/kg bw) 염증 및 산화스트레스를 유발하고, 한국에만 자생하는 흰 민들레 뿌리 열수 추출물을 1.4 g/kg bw 농도로 2회 경구 투여하였다. LPS에 의한 염증 유발은 체중 감소 현상으로 확인하였고, 민들레 뿌리 열수 추출물 투여군은 유의적으로 체중이 증가하여 염증 억제 효과가 관찰되었다. LPS 처리는 혈액의 중성지방질, 혈액 및 간의 reactive oxygen species(ROS), peroxynitrite 농도 그리고 간의 지질과산화 수준을 증가시켰다(P<0.05). LPS를 처리한 대조군에 비해 흰 민들레 뿌리 열수 추출물 투여군은 혈중 및 간의 중성지방질, ROS, peroxynitrite 농도 그리고 간의 지질과산화물 수준이 유의적으로 감소하였다(P<0.05). 이는 흰 민들레 뿌리 열수 추출물이 간의 $NF-{\kappa}B$ 전사인자 및 이의 조절을 받는 COX-2의 단백질 발현을 억제함으로써 염증 관련 매개체 및 유리기 생성을 유의적으로 억제하였기 때문으로 생각된다. LPS 처리군에서 간의 중성지방질 농도가 감소한 것은 염증에 의해 간 조직이 손상되어 중성지방질의 합성이 억제되었기 때문으로 생각된다. 본 연구는 한방에서 사용하여 온 한국 토종 흰 민들레 뿌리가 $NF-{\kappa}B$ 전사인자의 발현을 억제함으로써 염증 및 산화 스트레스를 개선하는 효과가 있음을 확인하였다.

흰민들레, 고들빼기, 씀바귀 추출물의 암세포 증식 억제 효과 (Induction of Apoptosis in Human Cancer Cells with Extracts of Taraxacum coreanum, Youngia sonchifolia and Ixeris dentate)

  • 본 연구는 인체 유방암세포 SK-BR-3, MDA-MB-231과 위암세포 AGS를 대상으로 국화과 식물 중 흰민들레(Taraxacum coreanum, TC), 고들빼기(Youngia sonchifolia, YS), 씀바귀(Ixeris dentate, ID) 추출물에 의한 증식 억제효과를 비교 한 후, 가장 억제효율이 높은 추출물을 선택하여 apoptosis 유도 효과를 조사하였다. 암세포의 증식 억제효과는 TC 추출물에 의한 영향은 약한 반면, YS와 ID 추출물에 의한 영향은 농도 의존적으로 증가하는 것을 확인하였다. 세 가지 추출물 중 가장 효과가 뛰어난 ID 추출물을 이용하여 추가적으로 농도 설정을 진행한 다음 이후 실험을 진행하였다. ID 추출물에 의한 apoptosis 양성 세포를 확인하기 위해 DAPI assay를 진행한 결과, ID 추출물을 처치한 군에서 apoptotic body와 세포질 응축을 확인하였다. MTT assay와 DAPI staining의 결과를 바탕으로 ID 추출물이 SK-BR-3, MDA-MB-231, AGS 세포에서 apoptosis와 관련한 단백질 발현 양상에 미치는 영향을 확인하기 위해 western blot을 실시하였다. ID 추출물에 의해 SK-BR-3, MDA-MB-231, AGS 세포에서 pro-apoptosis인 Bax 단백질 발현은 농도 의존적으로 증가하였고, anti-apoptosis인 Bcl-2 단백질 발현은 SK-BR-3 세포에서는 발현의 변화가 거의 없었지만, MDA-MB-231과 AGS 세포에서는 감소하였다. 세포사멸의 주요임상지표인 Bax/Bcl-2 ratio는 MDA-MB-231세포에서 가장 유의적으로 증가하였다. 또한, 손상된 DNA를 복구하는 단백질인 PARP의 발현은 감소하면서, cleaved-PARP이 증가하였다. 이러한 결과들을 종합하였을 때, YS와 ID 추출물은 TC 추출물에 비해 암세포의 생존을 효과적으로 억제하였고, 특히 ID 추출물은 낮은 농도에서도 암세포의 생존 억제효과가 우수한 것을 확인하였다. 또한, ID 추출물에 의한 유방암세포 SK-BR-3, MDA-MB-231와 위암세포 AGS의 생존율 억제는 apoptosis 유도를 통해 이루어 지는 것으로 사료되며, 그 중 유방암세포 MDA-MB-231에서 apoptosis 유도 효과가 뛰어난 것을 미루어 보아, 유방암의 종류와 그 특징에 따른 차이를 보이지만 ID는 향후 유방암 예방 및 치료제로 개발 가능성을 제시하며, 추후 지속적인 연구를 통하여 in vivo에서의 ID 추출물의 항암효과에 대해 심도 있는 연구가 이루어 져야 할 것으로 사료된다.
