태음조위탕(太陰調胃湯)의 경구 투여에 의한 SD계 흰쥐의 아급성 독성시험 연구 (Subacute Oral Toxicity of Taeumjowi-tang in SD Rats)

  • Taeumjowi-tang is originated in Donguisusebowon edited by Lee Je-Ma. The objective of this study was to investigate the subacute toxicity of Taeumjowi-tang in rats. Several doses(0, 125, 250, 500, 1,000, and 2,000 mg/kg) of Taeumjowi-tang were administered to rats for 4 weeks. The mortality, clinical signs, body weights and gross findings were examined for experimental period. No dead animals were found during the experimental period. In addition, any differences were not found between control and treated groups in clinical signs, hematology, serum biochemistry, and other findings. In conclusion, the no observed adverse effect level(NOAEL) for Taeumjowi-tang was over 2,000 mg/kg/day in rats.

  • 1. Objectives The purpose of this study is to find out effects of Choeongsimyeonja-tang and Taeumjowi-tang on the against decline of physical function as aging. 2. Methods In Wistar rat 10, 30, 50 week and 40 week Choeongsimyeonja-tang & Taeumjowi-tang between 10 weeks form condition change of weight, change of intestine weight, hematology, blood chemistry, research result about serum content following conclusion get. 3. Results and Conclusions 1. Observed gain in weight than control group form of Cheongsimyeonja-tang & Taeumjowi-tang to aged Wistar rat. 2. Is thought to promote activation of living body action gaining intestine weight along with gain in weight. 3. Displayed decrease of MDA's content of serum than control group form of Cheongsimyeonja-tang & Taeumjowi-tang to aged Wistar rat. 4. Change that is Wistar rat's hematological value by aging according to 10, 30, 50 week WBC, RBC, Hgb, monocyces, eosinophil ere. increase, and HCT, PLT ere. showed tendency that decrease accorcling to old-week, and observed improvement that is hematological value than control group form of Cheongsimyeonja-tang & Taeumjowi-tang.5. Change that is Wistar rat's biochemical value by aging was measured highest in 50 week because ALT, AST, BUN, CRN, T-bili., T-chol., TG, TP, ALB, A/G. P ete. increase according to 10, 30, 50 week, and observed improvement that is biochemical value than control group form of Cheongsimyeonja-tang & Taeumjowi-tang. Is considered by being effect that Cheongsimyeonja-tang & Taeumjowi-tang living body function decline by aging by this result.

양격산화탕과 태음조위탕 추출물의 in vitro CYP450 효소 활성 억제 효과 (In vitro inhibitory effects of Yangguksanhwa-tang and Taeumjowi-tang on CYP450 isozymes)

  • Objective : Herb-drug interactions have become an important issue because of the consumption of herbal remedies has increased in the world. Yangguksanhaw-tang (Liang ge san huo-tang) and Taeumjowi-tang (Tai yin tiao wei-tang) are typical herbal formulas on Sasang constitution medicine (four-constitution medicine). This study was aimed at evaluating the effects of Yangguksanhaw-tang and Taeumjowi-tang on drug metabolizing enzymes, cytochrome P450 (CYP450) isozymes. Methods : Vivid$^{(R)}$ CYP450 Screening Kits were used to measure of CYP3A4, CYP2C19, CYP2D6 and CYP2E1 activities. This method is based on the use of fluorescent CYP450 substrates that are efficiently metabolized by specific CYP450 isozymes to yield a product with altered fluorescent properties. The percent inhibitions of CYP450s by herbal formulas were calculated. Results : Yangguksanhaw-tang inhibited CYP2C19 and CYP2E1 activities higher than that other CYP450 isozymes. The $IC_{50}$ values of CYP2C19 and CYP2E1 were 159.83 ${\mu}g/mL$ and 261.40 ${\mu}g/mL$, respectively. The CYP2E1 activity was inhibited ($IC_{50}=215.17{\mu}g/mL$) higher than that other CYP450 isozymes by Taeumjowi-tang. Conclusions : These results suggest that Yangguksanhaw-tang may inhibit the metabolism of co-administered drugs whose primary route of metabolism is via CYP2C19 or CYP2E1. Taeumjowi-tang could inhibit the metabolism of co-administered drugs, which are substrates for CYP2E1. Therefore, co-administration of the herbal formulas and other conventional drugs should be undertaken with care.

  • The objective of this study was to evaluate the acute toxicity of Taeumjowi-tang in rats. SPF Sprague-Dawley male and female rats were administered orally with Taeumjowi-tang extract of 2,500 mg/kg(low dosage group), and 5,000 mg/kg(high dosage group). We daily examined number of deaths, clinical signs, body weights and gross findings for 14 days. No dead animal and no significant changes of body weights were found during the experimental period. In addition, no differences were found between control and treated groups in clinical signs, hematology, serum biochemistry, and other findings. In conclusion, Taeumjowi-tang extract did not show any toxic effects, and oral LD50 value was over 5,000 mg/kg in SD rats.

시스플라틴으로 유발된 랫트의 위장관 운동장애에 대한 태음조위탕의 효과 (Beneficial Effect of Taeumjowi-tang on the Cisplatin-Induced Gastrointestinal Dysfunctions in Rats)

  • Objectives This study aimed to observe the effect of Taeumjowi-tang on the cisplatin-induced gastrointestinal dysfunctions in rats. Methods Four groups, each of 8 rats per group, were used in this study. Saline and distilled water treated control rats were intact vehicle control group. Delayed gastrointestinal motility was induced by intraperitoneal treatment of cisplatin 2mg/kg, once a week for 5 weeks(Cisplatin control group). Taeumjowi-tang aqueous extracts(TJ) were orally administered in a volume of 5ml/kg, once a day for 14 days from 4th cisplatin treatment(TJ group). Ondansetron 1mg/kg was subcutaneously treated, in a volume of 1ml/kg, as same as TJ(ondansetron group). We measured the body weights, intestinal charcoal transit ratio, fecal parameters, fundus MDA(malondialdehyde), GSH(glutathione) contents and SOD(superoxide dismutase), CAT(catalase) activities, TPH(tryptophanhydroxylase) and MAO(monoamine oxidase) activities, pyloric gastrin and serotonin contents with their immunoreactive cells, colonic serotonin-immunoreactive cells, the histopathology of pylorus, fundus mucosa and colon. Results 1) The body weight gains, the small intestinal charcoal transfer rates, the fecal parameters(numbers, weights and water contents) were increased in TJ, ondansetron group. 2) The inhibit of fundus antioxidant defense systems by cisplatin were decreased in TJ, ondansetron group. 3) The pyloric TPH activities were increased and the pyloric MAO activities were decreased in TJ group. 4) The pyloric gastric contents and the gastrin-immunoreactive cells were increased and the pyloric serotonin contents and the pyloric and colonic serotonin-immunoreactive cells were decreased in TJ group. 5) The pylorus atrophic changes and the gastric surface erosive damage regions by cisplatin were favorably inhibited by treatment of TJ group. Conclusions The results obtained in this study suggest that TJ favorably retarded the cisplatin related GI(gastrointestinal) dysfunctions and constipation through modulations of GI enterochromaffin cells, serotonin and gastrin-producing cells and antioxidative systems.

태음조위탕(太陰調胃湯) 추출액이 ICR mouse에서의 경구 투여 급성독성에 미치는 영향 (Acute Toxicity Study on Taeeumjowi-tang in Mice)

  • 1. Objectives: The aim of this study is to investigate the acute toxicity and safety of Taeeumjowi-tang. 2. Methods: We investigated the acute toxicity for water-extracted Taeeumjowi-tang. 25 male and 25 female mice were observed for 14 days after one day oral administration of Taeeumjowi-tang at the respective doses of 0(control group), 2560, 3200, 4000 and 5000 mg/kg. 3. Results: We observed survival rates, general toxicity, change of body weight and autopsy. 4. Conclusions: The data confirmed that Taeeumjowi-tang is free from the toxicity and safety problems in oral route respectively. Compared with the control group, we could not find any toxic alteration in all treated groups(2560, 3200, 4000 and 5000 mg/kg). In conclusion, LD50 of Taeeumjowi-tang was over 5000 mg/kg and it is very safe to mice.

태음조위탕(太陰調胃湯)의 마우스 단회투여 독성시험 (Single Oral Dose Toxicity Test of Taeumjowi-tang in ICR Mice)

  • The object of this study was to evaluate the single dose toxicity of Taeumjowui-tang(TUJWT), a polyherbal formula have been traditionally used as prevention or treatment agent for obesity, in male and female mice. Aqueous extracts of TUJWT (Yield = 10.5%) was administered to female and male ICR mice as an oral dose of 2000, 1000 and 500 mg/kg (body wt.) according to the recommendation of KFDA Guidelines. Animals were monitored for the mortality and changes in body weight, clinical signs and gross observation during 14 days after dosing, upon necropsy, organ weight and histopathology of 12 principle organs were examined. As results, we could not find any mortality, clinical signs, and changes in the body and organ weight except for diarrhea restricted in TUJWT 2000 mg/kg treated one male mouse at 1 day after administration, and increases of popliteal lymph node weights in all TUJWT administered groups. In addition, no TUJWT-treatment related abnormal gross findings and changes in histopathology of principle organs were detected except for increases of the frequency of hypertrophy and hyperplasia of lymphoid cells in the popliteal lymph nodes in all TUJWT treatment groups with some sporadic accidental findings. These increases of popliteal lymph node weights with hypertrophy and hyperplasia of lymphoid cells were considered that related to the immune modulate effect of TUJWT not toxicological signs. The results obtained in this study suggest that the TUJWT does not cause any toxicological signs. The 50% lethal dose and approximate lethal dose of TUJWT aqueous extracts in both female and male mice were considered as over 2000 mg/kg.
