Identification of the protease inhibitory domain of Trichinella spiralis novel cystatin (TsCstN)

  • The Trichinella spiralis novel cystatin (TsCstN) inhibits cathepsin L (CatL) activity and inflammation of macrophages during lipopolysaccharide (LPS) induction. To identify the protease inhibitory region, this study applied an in silico modeling approach to simulate truncation sites of TsCstN (Ts01), which created four truncated forms, including TsCstN∆1-39 (Ts02), TsCstN∆1-71 (Ts03), TsCstN∆1-20, ∆73-117 (Ts04), and TsCstN∆1-20, ∆42-117 (Ts05). The superimposition of these truncates modeled with AlphaFold Colab indicated that their structures were more akin to Ts01 than those modeled with I-TASSER. Moreover, Ts04 exhibited the closest resemblance to the structure of Ts01. The recombinant Ts01 (rTs01) and truncated proteins (rTs02, rTs03, and rTs04) were successfully expressed in a prokaryotic expression system while Ts05 was synthesized, with sizes of approximately 14, 12, 8, 10, and 2.5 kDa, respectively. When determining the inhibition of CatL activity, both rTs01 and rTs04 effectively reduced CatL activity in vitro. Thus, the combination of the α1 and L1 regions may be sufficient to inhibit CatL. This study provides comprehensive insights into TsCstN, particularly regarding its protein function and inhibitory domains against CatL.

Efficacy and Safety Profile of TS-1 or TS-1/CDDP in Patients with Advanced Gastric Cancer (진행성 위암환자에 있어 TS-1 또는 TS-1/CDDP의 항암효과 및 안정성)

  • Purpose: Although several chemotherapy regimens used against advanced gastric cancer (AGC) have been studied extensively in an attempt to further improve the prognosis of patients, to date, no standard chemo-therapeutic regimens have been established. The aim of this study was to determine the anti-tumor efficacy and safety of TS-1 or TS-1 plus cisplatin (CDDP). Material and Methods: We treated 78 patients with AGC either with $80\;mg/m^{2}$ of TS-1 for 28 days, which was followed by a 2-week rest, or with $80\;mg/m^{2}$ of TS-1 for 21 days and $80\;mg/m^{2}$ of CDDP on day 8 every 5 weeks. Results: Tumor response rates in the neoadjuvant chemotherapy group and in the recurrent or post-palliative surgery group were 87.5% and 32.4%, respectively, and they were 28.6% and 48.4%, respectively, in the TS-1 group and the TS-1 plus CDDP group. The survival rates in the recurrent and the post-palliative surgery group were significantly different according to the degree of tumor response (P=0.0016), but the one-year survival rates according to the kinds of regimens (TS-1 or TS-1/CDDP group) were not significantly different. The incidences of grade 3 or 4 adverse effects in the TS-1 and the TS-1/CDDP groups were 14.3% and 36.8%, respectively. Conclusion: The anti-tumor efficacy and safety of TS-1 and TS-1 plus CDDP in Korean patients with AGC seemed to be high with modest adverse effects, thus suggesting the possible use of this regimen as a standard chemotherapy for gastric cancer.

  • Catalytic activity such as conversion and selectivity on the phenol hydroxylation over TS-1 prepared by hydrothermal method and microwave heating method, respectively, was compared and discussed for understanding the dependence of solvent such as water, methanol, acetone, respectively, during phenol hydroxylation, with hydrogen peroxide. Basic physical properties such as XRD, EDS, SEM and $N_{2}$ adsorption/desorption were determined and compared. The relationship between catalytic activity and physical properties of TS-1 was explained.

  • A microporous zeolite-type tianosilicate (TS-1), new catalysis elements, was synthesized by differents of the reactant solution pH. The range of reactant solution pH has from 10.0 to 12.4 TS-1 Zeolite (ETS-10), having a large pore (8~10 ${\AA}$), was synthesized at 10.4 of pH, since TS-1 Zeolite (ETS-4), having a small pore (3~5 ${\AA}$), was synthesized at 11.5 of pH. Also the two materials simultaneously existed at the intermediate pH. Crystallization, physico chemical characteristics of synthesized TS-1 Zeolite were investigated by XRD, XRF, SEM and FT-IR techniques.

  • The flagella antigenic variation of nine Bacillus thuringiensis serovar kurstaki temperature-sensitive mutants grown at the permissive temperature (3$0^{\circ}C$) was detected by a serological agglutination between H-antigen and antiserum. The flagella antigens were injected to rabbits to prepared their antisera, and then their homologous and heterologous titers of the antisera were measured. The homologous titers were ranged from 1:6,400 to 1:12,800, but the heterologous titers were very low. The H-antigen of the wild type strain was not agglutinated to 4 heterologous antisera, ts-U23 not to 7, ts-U3l not 5, ts-U32 not to 4, ts-U33 not to 7, ts-U7l not to 4, ts-U73 not to 6, ts-U74 not to 6, ts-U91 not to 4 and ts-U603 not to 4 antisera.

Rapid detection of Theileria sergenti by polymerase chain reaction (중합효소연쇄반응을 이용한 Theileria sergenti의 신속한 검출)

  • Four separate pairs of oligonucleotide primers within the coding region in a T sergenti 33-kDa surface protein gene were selected to detect T. sergenti by PCR. The specificity of PCR-amplified DNA was examined by digestion with restriction enzyme 3nd Southern blot hybridization using T. sergenti p33 DNA probe. PCR appears to be specific for T. sergenti, without detectable signals from uninfected erythrocytes, uninfected bovine leukocytes and other hemoparasites, including A. morginnle and 3. ouata. Although 46 of 71 specimens (64.8%) from grazing cattle were microscopically positive. PCR in this study showed that 64 specimens (88.7%) were positive. Therefore, PCR proves a useful diagnostic tool for detecting T sergenti-infected cattle. In addition, it is also revealed that PCR was significantly more sensitive than traditional microscopic examination using Giemsa's stain.

Pyrolysis kinetics and microstructure of thermal conversion products on toluene soluble component from two kinds of modified pitch

  • Modified pitch A (MPA) and modified pitch B (MPB) were prepared by oxidative polymerization and thermal polycondensation reaction with refined pitch as the raw material, respectively. The toluene soluble components (TS-1 and TS-2) were obtained by solvent extraction from MPA and MPB, separately. The Flynn-Wall-Ozawa method and Kissinger-Akahira-Sunose method were used to calculate the pyrolysis activation energy of TS. The Satava-Sestak method was used to investigate the pyrolysis kinetic parameters of TS. Moreover, the optical microstructure of the thermal conversion products (TS-1-P and TS-2-P) by calcination shows that TS-1-P has more contents of mosaic structure and lower contents of fine fiber structure than TS-2-P. The research result obtained by a combination of X-ray diffraction and the curve-fitting method revealed that the ratios of ordered carbon crystallite (Ig) in TS-1-P and TS-2-P were 0.3793 and 0.4417, respectively. The distributions of carbon crystallite on TS-1-P and TS-2-P were calculated by Raman spectrum and curve-fitting analysis. They show that the thermal conversion product of TS-2 has a better graphite crystallite structure than TS-1.

Partial Characterization of Bacillus thuringiensis var. kurstaki Temperature-sensitive Mutants (Bacillus thringiensis var. kurstaki 감온성 돌연변이주의 일부특성)

  • Partial characterization of B. thuringiensis var. kurstaki 3ab temperature-sensitive mutants was carried out through biochemical analyses, utilization tests of carbohydrate sources, antobiotic resistant test, hemolytic reaction test, growth measurement of Fructus gardenia sxtrant medium and toxicity test against mice. Six ts mutants, ts-U154, ts-U601, ts-U602, ts-U603, tsU-604, and ts-U788 could not produce urease, ts-U603 lost its motility, ts-U154 could not use salicin and cellobiose and ts-U603 not ribose. All ts mutants except ts-U154 and wild type strain were resistant to cephalothin, ampicillin, and penicillin. but ts-U154 was sensitive to the three. Four mutants, ts-U21, ts-U74, ts0U131 and ts-U154 did not form pigment colonies on the F. gardenia medium. All the mutants and wild type strain showed hemolysis reaction on the blood agar. The B. thuringiensis and mutants were not toxic to mice.

The Relation between Microsatellite Instability and the Thymidylate Synthase Expression in Gastric Cancer (위암에서 Microsatellite Instability와 Thymidylate Synthase의 상관관계)

  • Purpose: The main target of 5-fluorouracil (5-FU) is thymidylate synthase (TS). A high TS expression has been identified as promoting resistance to 5-FU. For colorectal cancers, the response to 5-FU based adjuvant chemotherapy is different according to the microsatellite instability (MSI) status. The reports on the relationship between MSI and the TS expression in colorectal cancer have been inconsistent. No data is available for gastric cancer regarding the relationship between MSI and the TS expression. Therefore, we studied the relationship between MSI and the TS expression in gastric cancer. Materials and Methods: Ninety-nine consecutive patients who underwent radical gastrectomy for gastric cancer from January 2004 to May 2006 at Bundang CHA hospital were studied. MSI was assessed for five markers (BAT25, BAT26, D2S123, D5S346, and D17S250) by PCR and with using an ABI prism 3100 Genetic analyzer. The TS expression was analyzed by immunohistochemistry with using mouse anti-thymidylate synthase monoclonal antibody to the TS 106 clone. Results: Out of the ninety-nine patients, MSS/MSI-L was detected in 92 (92.1%) cases and 7 cases (7.1%) were MSI-H. A negative TS expression was found in 46 (46.5%) cases, a low TS expression was found in 33 (33.3%) and a high TS expression was found in 20 (20.2%). Out of 92 MSS/MSI-L patients, the number of patients with negative, low and high TS expressions were 46 (50%), 30 (32.6%) and 16 (17.4%), respectively. Out of the 7 MSI-H patients, the number of patients with negative, low and high TS expressions were 0 (0%), 3 (42.9%) and 4 (57.1%), respectively. The relationship between MSI-H and a high TS expression was statistically significant. Conclusion: Gastric cancer with MSI-H showed higher levels of a TS expression compared to the gastric cancer with MSS/MSI-L.

Study on Large Diameter Steel-cased Piles Socketed in Weathered and Fresh Rocks (풍화암과 연암에 근입된 대구경 말뚝의 축하중지지 거동에 관한 연구)

  • Two large diameter steel cased piles (TS-1 and TS-2) were installed and load-tested with the maximum load of 2000 ton at Kwang-An grand bridge. One of the test piles (TS-1) was socketed into the fresh rock and the other test pile (TS-2) was socketed into the weathered rock. Most loads were carried by the weathered rock layer at the maximum applied load for the test piles. In addition, numerical studies were performed using PENTAGON 3D and ROCKET. Based on this study, the weathered rock layer provided sufficient side resistances and possibility of an alternative embedded layer if weathered rocks are deeply layered over fresh rock, which caused difficulties and cost in construction.
