개량된 이탄과 유기성 슬러지를 이용한 유류오염토양 분해제 개발 (Development of Degradation Agent for Oil Contaminated Soil using Modified Peat Moss and Organic Sludge)

  • Oil degradation agent was developed with organic sludge and modified peat moss (MPM) to recover oil contaminated soil. Waste sludge discharged from wastewater treatment plant of chemical plant in Ulsan National Industrial Park was used as organic sludge, and MPM was purchased. Organic sludge was adequate to use as growth medium for microorganism, the surface of MPM had porous structure which could enhance the cultivation condition of oil degradation microorganisms. Water contents and TPH variation with time were observed to investigate the degradation capacity of developed degradation agent. Water contents were rapidly decreased with higher contents of MPM, however, in case of TPH, high MPM content decreased the degradation capacity. Therefore, it was recommended that the content of MPM was controlled to below 10% in degradation agent as mixing organic sludge with MPM.

염소계유기용제 및 유류물질에 의한 산업단지 지하수 오염특성

  • 토양에 누적된 오염물질은 지하수 유동과 연계하여 점차 주변지역으로 확산된다. 본 조사는 오염의 개연성이 높은 산업단지를 선정하고 그 중에서도 유해화학물질의 사용이 많은 지역을 대상으로 토양과 지하수의 오염도조사를 수행하였다. 토양오염도 조사는 160개 지점에서 토양시료를 채취하여 오염물질을 분석하였으며 분석결과 TPH물질은 최고 23,151mg/kg, BTEX는 최고 1,657mg/kg로 검출되었다. 오염특성은 유류(TPH, BTEX) 물질 이외에도 TCE, PCE◎물질과같은 염소계 유기용제에 의한 오염도 확인할 수 있었으나 중금속 물질에 의한 오염은 우수 방류구와 같은 일부 지역에서 검출되었다. 지하수 수질의 경우 페놀, 비소, 납, BTEX, TCE, PCE 물질들이 복합적으로 오염되어있는 것으로 나타났다.

Enhanced Biodegradation of Total Petroleum Hydrocarbons (TPHs) in Contaminated Soil using Biocatalyst

  • Biocatalytic degradation of total petroleum hydrocarbons (TPHs) in contaminated soil by hemoglobin and hydrogen peroxide is an effective soil remediation method. This study used a laboratory soil reactor experiment to evaluate the effectiveness of a nonspecific biocatalytic reaction with hemoglobin and H2O2 for treating TPH-contaminated soil. We also quantified changes in the soil microbial community using real-time PCR analysis during the experimental treatment. The results show that the measured rate constant for the reaction with added hemoglobin was 0.051/day, about 3.5 times higher than the constant for the reaction with only H2O2 (0.014/day). After four weeks of treatment, 76% of the initial soil TPH concentration was removed with hemoglobin and hydrogen peroxide treatment. The removal of initial soil TPH concentration was 26% when only hydrogen peroxide was used. The soil microbial community, based on 16S rRNA gene copy number, was higher (7.1 × 106 copy number/g of bacteria, and 7.4 × 105 copy number/g of Archaea, respectively) in the hemoglobin catalyzed treatment. Our results show that TPH treatment in contaminated soil using hemoglobin catalyzed oxidation led to the enhanced removal effectiveness and was non-toxic to the native soil microbial community in the initial soil.

The Removal of Petroleum Hydrocarbon from Fine Soil in Soil Washing Water using Advanced Oxidation Processes

  • This study was performed to test the applicability of the ozone/hydroxy radical reaction system, which applied advanced oxidation processes, to remove total petroleum hydrocarbon (TPH) from the fine soil in washing water of the soil washing process. Removal efficiency was tested on 40 L of washing water in a pilot reaction tank. Fine soil contaminated with $800mg\;kg^{-1}$ TPH was prepared at 5% and 10% suspended solids. Testing conditions included ozone/hydroxy radical flow rates of 40, 80, and $120L\;min^{-1}$, and processing time of 2 to 12 hours. The removal efficiency of petroleum hydrocarbon from water waster by ozone/hydroxy radical was increased with higher flow rates and lower percentages of suspended solids. Optimal efficiency was achieved at $80L\;min^{-1}$ flow rate for 4 hours for the 5% suspended solids, and $120L\;min^{-1}$ for 6 hours for the 10% suspended solids. These results verified the efficiency of hydroxy radical in removing TPH and the applicability of the ozone/hydroxy radical reaction system in the field.

분심기음가미방(分心氣飮加味方)의 항산화능과 serotonin 대사에 미치는 영향 (The Effects of BunSimGiEumGami-Bang(Fenxinqiyinjiameifang) on Serotonin of P815 cell)

  • Objectives : This experiment was designed to investigate the effect of the BSGE hot water extract on serotonin biosynthesis of depression model. Methods : The cytotoxicity of the BSGE hot water extract was analyzed by MTT assay on P815 cell. The antioxidant activity was measured by DPPH free radical-scavenging activity and SOD activity on P815 cell. The quantity of 5-HT and expression of TPH-1, AAADC and MAOa mRNA was measured by of HPLC profle Analysis on P815 cell. Results : 1. The BSGE hot water extract increased DPPH free radical-scavenging activity and SOD activity on P815 cell. 2. The BSGE hot water extract increased significantly the quantity of 5-HT. 3. The BSGE hot water extract increased the expression of TPH-1 mRNA. Conclusions : This experiment shows that the BSGE hot water extract might be effective for the prevention and treatment of depression. Investigation into the clinical use of the BSGE hot water extract for depression is suggested for future research.

Application of Bioremediation to Soil Contaminated by Lubricants Around Railroad Turnouts

  • In this study, the feasibility of using bioremediation to treat lubricant-contaminated soil around railroad turnouts was investigated. Lubricants used during the maintenance of railroad turnouts can drip onto the ground causing soil contamination. In the laboratory experiments, the residual TPH (Total Petroleum Hydrocarbons) concentration in soil gradually decreased after microorganisms degrading the lubricants were added. Generally, the soil around railroad turnouts is covered by a layer of ballasts. In the column experiments that were designed considering field sites, the removal efficiency of TPH was about 11% after 60 days of cultivation time. In the field experiments, microorganisms were added into the soil periodically, and finally the residual TPH concentrations were reduced to less than 1,700 mg/kg-soil on average. These results indicate that the lubricant in the contaminated soil around railroad turnouts could be efficiently removed through bioremediation method.

  • Landfarming that supplies aerobic biodegradation condition to indigenous microbes in soils is a biological remediation technology. In this research, volatilization and biodegradation rate by indigenous microbes in the soil contaminated with total petroleum hydrocarbons (TPH) were measured. Soils were contaminated with diesel artificially and divided into two parts. One was sterilized by autoclave to remove indigenous microorganism and the other was used as it was. Various moisture contents and number of tillings were applied to the soil to find out proper condition to minimize volatilization and enhance bioremediation. Volatilization of TPH was inhibited and biodegradation was enhanced by increase on moisture content. Tilling was usually used to supply air for microbes, but tillings did not affect the growth of microbes in our study. Enough moisture content and proper aeration are important to control volatilization in landfarming. Also, TPH degradation was a function of the microbe counts (x1), numbers of tilling (x2), and moisture content (x3) from the application of the response surface methodology. Statistical results showed the order of significance of the independent variables to be microbe counts > numbers of tilling > moisture content.

토양내 중금속 및 유류 오염농도 저감을 위한 생화학적 기작의 효율성 평가 (Assessment of Biochemical Efficiency for the Reduction of Heavy Metal and Oil Contaminants in Contaminated Soils)

  • 중금속 및 유류 오염토양 정화를 위해 효율적인 토양세척법과 공정 선정을 목적으로 최적의 오염정화 설계인자를 제시하기 위한 실험적 연구를 수행하였다. 실험 분석항목은 구리, 납, 아연을 포함하는 중금속 항목과 총석유계탄화수소(TPH)인 유류 항목에 대해 흡광광도법(absorptiometric analysis), 기체크로마토그래피(gas chromatography)법을 이용하여 단계별로 분석하였다. 실험방법은 최적 세척용매(washing solution) 결정시험, 최적 세척시간(washing time) 도출시험, 최적 진탕비(dilution ratio) 결정시험 등을 통해 얻어진 결과를 토대로 계면활성제(surfactant) 첨가량별 중금속 용출영향 분석시험, 미생물 및 과산화수소 주입시험 순으로 실시하였다. 실험결과에서 세척용매인 염산 0.1 mole, 체류시간 1시간, 진탕비 1 : 3 조건에서 오염물질의 저감효과가 우수하게 나타났으며, 이들 조건을 적용하였을 때 1%의 계면활성제를 세척 용매에 첨가하였을 경우 추가적인 오염물질의 농도저감 효과를 보이는 것으로 확인되었다. 또한 미생물과 과산화수소 주입에 따른 추가적인 TPH 농도 저감이 있는 것으로 파악되었다.

아연 또는 비소와 경유로 오염된 토양의 복합정화공법 개발 (Development of Hybrid Remediation Method for Contaminated Soils with Zinc or Arsenic and Diesel)

  • The purpose of this study was to develope the remediation method of contaminated soils with metals and petroleum. The diesel degrading strain was isolated and identified from the soil contaminated by petroleum at industrial sites. Diesel biodegradation experiment was performed by diesel degrading bacteria in both solution and soil slurry. Contaminated soils by Zn or As and diesel were treated consecutively by steam-vapor extraction, biodegradation, and acid washing. The strain was identified as Pseudomonas aeruginosa, and named as Pseudomonas aeruginosa TPH1. The optimal culture conditions of TPH1 were $20^{\circ}C$ and pH 7.0, 3% of diesel concentration. Biodegradation of diesel was performed using the separated strain in liquid medium, and 63% of diesel was degraded in 72 hours. And 52% of diesel was removed in the tested soils. In the treatment of contaminated soils with diesel and Zn or As, 29% ~ 44% of diesel was reduced by steamvapor extraction, 60% ~ 71% of diesel was removed after biodegradation. 47% of Zn and 96% of As were removed after acid(mixture of sulfuric and oxalic acids) washing. It is recommended that consecutive treatment method of steam-vapor extraction, biodegradation and acid washing is effective for remediation of complex contaminated soils with metals and petroleum.

디젤로 오염된 토양의 효과적인 Bioventing

  • In this work, cost effective venting is considered by comparing flow rates of 5$m\ell$/min, 10$m\ell$/min, and 20$m\ell$/min. Studies were performed on a soil artificially contaminated with diesel oil (the initial TPH(Total Petroleum Hydrocarbon) concentration of 7098mg/kg), and nutrient condition was C:N:P rate of 100:10:1. The soil has a sandy texture with pH of 6.8, 2.16 ~2.38% organic matter, a total porosity of 47~52% and field capacity 16.2~ 17.2%. The column experiments was made of glass column of 60cm length and 10cm I.D. at controlled temperature of 2$0^{\circ}C$($\pm$2.5$^{\circ}C$). The efficiency of continuous flow rate of 5, 10 and 20$m\ell$/min resulted in separately 61.3%, 58.1%, and 55% reduction of initial TPH concentration(7098mg/kg). Hydrocarbon utilizing microbial count and dehydrogenase activity in air flow of 5$m\ell$/min were higher than those of the others. The first order degradation rate of n-alkanes ranging from C10 to C28 was higher than that of pristane and phytane as isoprenoids. The $C_{17}$/pristane and $C_{18}$phytane ratios for monitoring the degree of biodegradation were useful only during the early stages of oil degradation. Degradation contributed from about 89% to 93% of TPH removal. Volatilization loss of diesel oil in contaminated soil was about 7% to 11%, which was significantly small compared to degradation.n.
