Deficiencies in the early ADMET(absorption, distribution, metabolism, elimination and toxicity) information on drug candidate extract a significant economic penalty on pharmaceutical firms. Microscale cell culture analogue-microscale gastrointestinal(${\mu}CCA-{\mu}GI$) device using Caco 2, L2 and HEp G2/C3A cells, which mimic metabolic process after absorption occurring in humans was used to investigate the toxicity of the model chemical, acetaminophen(AAP). The toxicity of acetaminophen determined after induction of CYP 1A1/2 in Caco 2 cells was not significant. In a coculture system, although no significant reduction in viability of HEp G2/C3A and L2 cells was found, approximately 5 fold increase in the CYP 1A1/2 activity was observed. These results appear to be related to organ-organ interaction. The oral administration of a drug requires addition of the absorption process through small intestine to the current ${\mu}CCA$ device. Therefore, a perfusion coculture system was employed for the evaluation of the absolution across the small intestine and resulting toxicity in the liver and lung. This system give comprehensive and physiologic information on oral uptake and resulting toxicity as in the body. Bioluminescence is being used as a prevailing reporter of gene expression in microorganisms and mammalian cells. Bacterial bioluminescence draws special attention from environmental biotechnologists since it has many advantageous characteristics, such as no requirement of extra substractes, highly sensitive, and on-line measurability. Using bacterial bioluminescence as a reporter of toxicity has replaced the classical toxicity monitoring technology of using fish or daphnia with a cutting-edge technology. Fusion of bacterial stress promoters, which control the transcription of stress genes corresponding to heat-shock, DNA-, or oxidative-damaging stress, to the bacterial lux operon has resulted in the development of novel toxicity biosensors with a short measurement time, enhanced sensitivity, and ease and convenient usage. Therefore, these recombinant bioluminescent bacteria are expected to induce bacterial bioluminescence when the cells are exposed to stressful conditions, including toxic chemicals. We have used these recombinant bioluminescent bacteria in order to develop toxicity biosensors in a continuous, portable, or in-situ measurement from for air, water, and soil environments. All the data obtained from these toxicity biosensors for these environments were found to be repeatable and reproducible, and the minimum detection level of toxicity was found to be ppb (part per billion) levels for specific chemicals.
Glutamate (Glu) receptor-mediated excitoxicity has been implicated in many acute and chronic types of neurological disorders. Exposure of mature rat cortical neurons (15-18 days in culture) to the various concentrations of Glu resulted in a marked neuronal death, whereas immature rat cortical neurons (4∼5 days in culture) were resistant to the Glu-induced toxicity. Glu receptor subtype-specific agonists showed differential extent of toxicity in the immature neurons. The neurons treated with NMDA or kainate (KA) did not exhibit damage. However, quisqualate (QA) treatment induced a considerable cell death (36.1%) in immature enurons. The non-NMDA antagonist DNQX did not reduce this response. Interestingly, the QA-induced toxicity was potentiated by spermine in a concentration-dependent manner. Again, the spermine-enhanced damage was not altered by the polyamine antagonist ifenprodil. Taken together, unlike NMDA or KA, QA can induce neurotoxicity in immature rat cortical neurons and the QA-induced toxicity was potentiated by spermine. The lack of antagonizing effects of DNQX and ifenprodil on QA-induced toxicity and the potentiated toxicity by spermine, respectively, implies that both QA receptor and the polyamine site of NMDA receptor may not mediate the neurotoxicity observed in this study, and that a distinct mechanism(s) may be involved in excitotoxicity in immature neurons.
1. Objectives This study was designed to andyze the acute toxicity and safety of Yeoldahanso-tang 2. Methods We investigated the acute toxicity for water-extracted Yeoldahanso-tang. 25 male and 25 female mice were observed for 14 days after one day oral administration of Yeoldahanso-tang at the respective doses of 0(control group), 2560, 3200, 4000 and 5000 mg/kg. 3. Results We observed survival rates, general toxicity, change of body weight and autopsy. 4. Conclusions The data confirmed that Yeoldahanso-tang is free from the toxicity and safety problems in oral route respectively. Compared with the control group, we could not find any toxic alteration in all treated groups(2560, 3200, 4000 and 5000 mg/kg). In conclusion, LD50 of Yeoldahanso-tang was over 5000 mg/kg and it is very safe to mice.

Textile and pigment wastewater samples collected from an industrial complex showed toxic effect on Daphnia magna. For textile wastewater, 48 h acute toxicity of effluent was not detected while toxic unit (TU) of influent was 1.79. The toxicity of influent was completely disappeared by gamma-ray treatment at 10 kGy or by suspended solids (SS) removal. In case of pigment wastewater, both influent and effluent were toxic to D. magna though the effluent satisfied current water quality standards. Gamma-ray treatment had little effect on the toxicity reduction of pigment wastewater since the toxicity was mainly caused by metal ions, in particular, Cr(VI). This work suggests the bioassay technique for monitoring adverse effects of wastewater should be introduced, and also shows the usefulness of gamma-rays as an advanced treatment technique for textile wastewater.
We investigated methiozolin acute toxicity using with Sprague-Dawley rats. The results of acute oral toxicity using rats showed $LD_{50}$ value of over 2,000 mg/kg bw for methiozolin. The calculate acute dermal $LD_{50}$ value of methiozolin was over 4,000 mg/kg. The skin irritation test showed moderately irritation and weak response of eye irritation test was observed in this experimental condition. According to these results, We concluded that methiozolin was Category IV in GHS chemical classification for acute toxicity. Future, we need more chronic toxicity test for safety.
The aim of this study was to investigate the acute and subacute oral toxicity of crude antifungal compounds produced by Lactobacillus plantarum HD1 in Sprague-Dawley rats. In the acute toxicity study, the crude antifungal compounds (0.625, 1.25, 2.5, and 5.0 g/kg) did not produce mortality, significant changes in general behavior, or changes in the gross appearance of the organs. In the subacute toxicity study, the crude antifungal compounds were administered orally to rats at doses of 0, 0.5, 1.0, and 2.0 g/kg daily for 28 days. There were no test article-related deaths, abnormal clinical signs, or body weight changes. The study also showed no significant differences between the control and treated groups in hematological and serum biochemical parameters, histopathological examination, or any other findings. These results suggest that acute or subacute oral administration of crude antifungal compounds from L. plantarum HD1 is not toxic in rats.
In this study, we have developed the database of safety and health information for pesticide active ingredients used in Korea. There were 1,283 pesticide items among which 296 were found to be out of use in current. A total of 349 pesticide ingredients were being used in Korea. The database consists of 32 types of information including chemical characteristics, acute toxicity, chronic toxicity (carcinogenic and reproductive toxicity), specific symptoms by exposure route and first aid. When pesticide ingredients were assessed in terms of key properties such as color, odor, acute toxicity, carcinogenic and reproductive toxicity, they were white, colorless and odorless, in general. When ingredients were classified by category of acute toxicity, 'Non-hazardous' represented 29%, followed by 'Slightly hazardous' at 16%, 'Moderately hazardous' at 14%, 'Highly hazardous' at 5%, and 'Extremely hazardous' at 2%. 85 out of 349, or 24% of ingredients were found to be possibly carcinogenic to human. This database is expected to provide an easy access for farmers, agriculture supervisors, researchers and consumers, and it can ultimately be used as basic data on farmer's safety and health.

Endosulfan is an organochlorine pesticide that is widely used throughout the world for higher agricultural production. Its extreme toxicity, however, has caused health and environment concerns that have led to an interest in detoxification. In this study, the radiolytic degradation of endosulfan was investigated. Endosulfan in methanol solution (100 ppm) was irradiated at 0, 10, 30, and 50 kGy, and subsequent changes in immune toxicity and degradation of endosulfan were observed. The concentration of endosulfan that was used in this experiment did not affect the cell proliferation. The irradiation of endosulfan decreased the production of NO, indicating a decrease in the immune toxicity of endosulfan by irradiation. The concentration of endosulfan was significantly reduced by irradiation in a dose-dependent manner. The results suggest that gamma irradiation can degrade endosulfan and can reduce its immune toxicity.
Objectives: Effective genetic toxicology and molecular biology research techniques and strategies that are highly correlated with the carcinogenic inhalation toxicity test and related research are required. The aim of this study was to maximize the utilization of chemical substances to prevent workers' occupational diseases. Methods: We surveyed the literature, domestic and international references, and the status of relevant domestic and foreign professional organizations. Expert advisory opinions were reflected, and experts were consulted by participating in domestic and overseas academic conferences. Results: The current status of domestic and international genotoxic toxicity evaluation was examined through various documents from related organizations. Cell models for in vitro lung toxicology were investigated and summarized, and the human resources and performance results of genetic toxicity studies and pilot projects were compared and analyzed by holding an advisory meeting. We examined domestic and international genotoxicity guidelines and investigated new test methods for the development of genotoxicity and carcinogenicity. Ultimately, we described long-term future predictions, including the implementation of our researchers' recommendations and occupational genetic toxicology forecasts for future worker health protection. Conclusions: This research project aims to establish current genetic toxicology and molecular biology research techniques and strategies that can maximize the linkage with the carcinogenic inhalation toxicity test and research in the future. We expanded the study of genetic toxicity and establish a foundation forgenetic toxicity in accordance with research trends in Korea and abroad.
