Biotransformation of Panax ginseng extract by rat intestinal microflora: identification and quantification of metabolites using liquid chromatography-tandem mass spectrometry

  • Background: In general, after Panax ginseng is administered orally, intestinal microbes play a crucial role in its degradation and metabolization process. Studies on the metabolism of P. ginseng by microflora are important for obtaining a better understanding of their biological effects. Methods: In vitro biotransformation of P. ginseng extract by rat intestinal microflora was investigated at $37^{\circ}C$ for 24 h, and the simultaneous determination of the metabolites and metabolic profile of P. ginseng saponins by rat intestinal microflora was achieved using LC-MS/MS. Results: A total of seven ginsenosides were detected in the P. ginseng extract, including ginsenosides Rg1, Re, Rf, Rb1, Rc, Rb2, and Rd. In the transformed P. ginseng samples, considerable amounts of deglycosylated metabolite compound K and Rh1 were detected. In addition, minimal amounts of deglycosylated metabolites (ginsenosides Rg2, F1, F2, Rg3, and protopanaxatriol-type ginsenosides) and untransformed ginsenosides Re, Rg1, and Rd were detected at 24 h. The results indicated that the primary metabolites are compound K and Rh1, and the protopanaxadiol-type ginsenosides were more easily metabolized than protopanaxatriol-type ginsenosides. Conclusion: This is the first report of the identification and quantification of the metabolism and metabolic profile of P. ginseng extract in rat intestinal microflora using LC-MS/MS. The current study provided new insights for studying the metabolism and active metabolites of P. ginseng.

국내 박물과 환경에 분포하는 미생물의 분리 (Distribution of Microorganisms in Domestic Museum Environments)

  • We isolated and identified microorganisms from the aerial environment of domestic museums. The fungi, Penicillium spp., Alternaria spp., and Cladosporium spp. were isolated in many museums. It seems that these fungi are related to biological degradation of textile remains. A total of 14 kinds of bacterial strains were isolated: Acinetobacter spp., Pseudomonas spp., Neisseria spp., Alcaligenes spp., Shigella spp., Klebsiella spp., Corynebacterium spp., Aerococcus spp., Bacillus spp., Micrococcus spp., Citrobacter spp., Erwinia spp., Salmonella spp., and Providencia spp. Acinetobacter spp., Pseudomonas spp., Neisseria spp., and Alcaligenes spp. were the predominate bacteria found in samples with a variety of bacteria. This suggests that there is a relationship between bacteria and the damage of textile remains. In the museum, we isolated Alternaria spp, Geotrichum spp., Penicillium spp. Acinetobacter spp., Pseudomonas spp., Alcaligenes spp. from the entrance, exhibit hall and storage, but they were found in smaller number and species in the exhibit cases and paulownia cases. We concluded that paulownia cases were not influenced by the microorganisms because of quality of care provided by the museum staff. Corynebacterium spp., and Bacillus spp. were not detected at the entrance and exhibit hall but were detected in paulownia cases. It is presumed that those bacteria did not flow in from outside, but resulted from contaminants in paulownia cases. In the distribution of microorganisms associated with textile remains, more fungi were detected than bacteria. Acinetobacter spp., Pseudomonas spp., and Neisseria spp., were isolated from silk items. Penicillium spp. and Cladosporium spp. were isolated in the silk and hump items. Aspergillus spp. and Penicillium spp. were isolated from the cotton items. On the other hands, there were no fungi strains in the wool items. Most of the isolated strains from textile remains were aerial microorganisms from the museum environment. These results suggest that textile remains were apt to contaminated by contact with the air.

Pseudoalteromonas sp. 배양액으로부터 아가레이즈 분리를 위한 음이온교환 크로마토그래피 최적화 (Optimization of Anion-exchange Chromatography for the Separation of Agarase from Culture Broth of Pseudoalteromonas sp.)

  • 아가로오스 분해산물은 생리활성이 우수하여 의약품 및 기능성 화장품 원료로 사용될 뿐만 아니라 바이오에탄올 생산을 위한 효모 발효용 기질로 검토되고 있어 상당한 주목을 받고 있다. 이에 따라 고성능 아가레이즈 탐색에 대한 연구가 활발히 진행되고 있다. 최근 본 연구팀에서는 남해안에서 신규 아가레이즈를 생산하는 미생물인 Pseudoalteromonas sp.를 분리하였다. 본 연구에서는 음이온교환수지를 이용하여 미생물 배양액으로부터 아가레이즈를 분리 정제하기 위한 크로마토그래피 조건을 최적화 하였다. 황산암모늄을 첨가하여 배양 상등액으로부터 침전된 단백질을 회수하고 이를 투석하여 조효소액을 얻었다. 음이온 교환수지인 DEAE-Sepharose 레진이 충진된 칼럼에 조효소액을 로딩하여 아가레이즈를 분리하였다. 410 ${\mu}g$의 단백질 로딩 시 흡착에 적합한 평형 pH는 7.5~8.0 이었고, 적정한 resin의 부피는 3 mL였다. 등용매 용출에서 용출액 중의 NaCl 농도가 증가함에 따라 용출되는 단백질 양이 증가하여 400 mM의 NaCl에서 최대에 달하였다. 최종적으로, NaCl 농도를 1,000 mM까지 선형적으로 증가시키며 아가레이즈를 분리하였다. Lugol 용액을 이용한 염색법으로 용리액 중의 아가레이즈의 존재를 확인하였다.

시호계지탕(柴胡桂枝湯)의 임상치험(臨床治驗)에 관(關)한 소고(小考) (Clinical Test on Application of ShiHo-GuizhiTang)

  • Precise and detailed clinical research and evaluation based on objective standards are imperative factors in securing reliability of a clinical test. Built on this principle, this clinical test has been conducted during the period between March 1999 and March 2000 dealing with 31 outpatients treated with ShiHo-GuizhiTang or ShiHo-GuizhiTang modify and ShiHo-GuizhiTang mixed prescriptions at the Sung-bo ORIENTAL MEDICAL CLINIC. A variety of information relating to those patients has been collected and analyzed under such criteria as precise diagnoses and their clinical effectiveness. The analysis was duly based on "The theory of cold syndrome". Then, the 31 patients' individual clinical information was compared one another by breaking down the results into sub-categories including gender, age, disease, main symptom, blood pressure, pulse beats, syndrome of abdomen, treatment period, modify and mixed prescription, and evaluation. The result of this clinical test can be summarized as follows: First, ShiHo-GuizhiTang appears to be more frequently prescribed in the case of female patients than in the case of male patients despite the fact that it does not necessarily need to be applied only to female. In addition, the numbers of prescriptions of ShiHo-GuizhiTang in the two age groups consisting of elementary school children and economically active adults respectively were the highest among other age groups. Second, ShiHo-GuizhiTang proved to be most effective in treatment for respiratory ailments and arthritis. Third, ShiHo-GuizhiTang brought down blood pressure of hypertension patients and at the same time benefited patients with normal or lower-than-normal blood pressure who were vulnerable to diseases due to low disease-resistance. Fourth, ShiHo-GuizhiTang was effective in the case of frequent pulse(rapid pulse) and thereby indicating the fact it carries Taiyang superficies syndrome. 지맥 arises from suppressed immune responses owing to adrenocortical hormones. ShiHo-GuizhiTang controls and revitalizes those suppressed immune systems which stem from slow pulse and, as a consequence of that process, helps them return to normal condition of pulse. Fifth, from the standpoint of syndrome of abdomen, feeling of obstruction in the epigastrium serves as an important standard in the process of diagnosing diseases and evaluating effectiveness of treatments. Sixth, according to the results of the clinical test with the 31 patients, a total of 81 percent of test subjects benefited from the treatment. The figure is the sum of 52 percent of I-class (both main symptom and accompanying symptoms had been eliminated) and 29 percent of II-class (part of main symptom and accompanying symptoms had been eliminated) respectively. All told it is safe to say that ShiHo-GuizhiTang can elect to be a viable clinical treatment. In conclusion, it is estimated that this clinical study has drawn up guidelines for objective diagnostic standards and evaluation on specific treatments' effectiveness. This will lead to more general application of ShiHo-GuizhiTang. On top of that, this study could also provide an opportunity to stress the significance of ShiHo-GuizhiTang and ShiHo-GuizhiTang modify and ShiHo-GuizhiTang mixed prescriptions as an alternative treatment for collagen disease which comes from environmental degradation and pollution.

간섬유화 동물에서 D-페니실라민의 항섬유화 효과 검색 (The Antifibrotic Effects of D-penicillamine in Liver Fibrosis Animal)

  • One of therapeutics in liver disease (morbus wilson) is D-penicillamin (D-pen: D-3-mercapto-valin). Especially the cross-linking of collagen molecules could be inhibited by D-pe n in extracellular space. In this study we investigated the antifibrotic effects of D-pen in rats that were induced the liver fibrosis by bile duct ligation and scission (BDL/S). Rats were treated for 4 weeks with D-pen after BDL/S operation or sham operation. The balance between fibrogenesis-marker (PNIIIP) and the fibrolysis-maker (PNIVP) were observed in sera by RIA (radioimmunoassay), and the parameter of collagen deposition in liver tissue (hydroxyproline: HYP) was measured by colorimetry. The weight of liver in BDL/S operated group was increased significantly in compared with sham operation group (15.2g${\pm}$1.1, vs 11.9g${\pm}$3.9: p<0.005, p<0.05). The rats group treated by D-pen showed the lower level of PNIIIP (6.7ng/ml${\pm}$1.5, vs 9.5ng/ml${\pm}$2.8) and the higher value of PIVCP (14.0ng/ml${\pm}$1.9, vs 7.9ng/ml${\pm}$1.5) in sera that compared to untreated rats. The content of HYP was decreased by 141% in BDL/S with D-pen treated group than that of it in BDL/S group. No correlation was revealed between collagen parameters in sera and HYP in liver tissue of BDL/S operated and D-pen treated rats. The group treated with D-pen showed the lower value of clinical biochemistry parameters (GOT: glutamate oxalacetate transaminase, Total-Bilirubin) in compared with only BDL/S operated rats, but the value of GPT (glutamate pyruvate transaminase) and Alkaline phosphatase in two BDL/S groups was nearly same. In the histological finding, we observed mild bile duct proliferation, weak inflammation and fibrosis in BDL/S with D-pen treated group, but BDL/S operated group showed the formation of septum (island of hepatocytes), massive bile duct proliferation. This result represents that the BDL/S operation induces liver fibrosis (cirrhosis) in 4 weeks, and D-pen inhibits the synthesis of collagen weakly and stimulates the degradation of collagen in the extracellular space. We conclude that the monitoring of PNIIIP, PIVCP in sera is useful parameter for screening of antifibrotic effect, and D-pen delay the liver fibrosis.

헤어리스마우스 피부 국소에 적용된 $^{125}I-rhEGF$의 피부흡수 및 체내 분포 (Dermal Absorption and Body Distribution of $^{125}I-rhEGF$ in Hairless Mice)

  • Distribution of rhEGF in the skin, plasma and several organ tissues following topical application of $^{125}I-rhEGF$ (0.4${\mu}$Ci) solution in 25% Pluronic F-127 on 154$mm^2$ normal and damaged (burned and stripped) skins of hairless mice was examined. The radioactivity in the stripped skin tissues increased as a function of time, and was 10-20 times higher than that in the normal and burned skins. The fractions of intact drug in the skin tissues were 40-60% for the normal and burned skins, and 60-80% for the stripped skin. It indicates that the stratum corneum layer behaves as a barrier for the dermal penetration of the drug. The radioactivity in the plasma was much higher for the stripped skin than for the normal and burned skins. However, the concentration of intact drug in the stripped skin was comparable to those in the normal and burned skins indicating most severe degradation (or metabolism) of the drug in the stripped skin. As a result, the fraction of intact drug in the plasma was lowest for the stripped skin (<10%). Body organ distribution of the drug was much higher for the stripped skin. The concentration in the stomach. Both in total radioactivity and intact drug, showed more than 10-times higher value than in the other organs (liver, kidney and spleen). The fraction of intact drug in each organ tissue was below 10-20%. And generally lowest for the stripped skin. The lowest fraction of the drug for the stripped skin could not be explained by the activity of the aminopeptidases in the skin since it was lower for the stripped skin than for the normal skin. Thereover, the fraction of intact drug appears to be determined by the balance between dermal uptake and systemic elimination of the drug, for example. The mechanism of dermal uptake of rhEGF was examined by topical applying 200${\mu}$l of 25% Pluronic F-127 solution containing 0.4 ${\mu}$Ci of $^{125}I-rhEGF$ and 0.14${\mu}$Ci of $^{14}C$-inulin (a marker of passive diffusion). The radioactivity of $^{125}I-rhEGF$ at each sampling time point (0.5, 1, 2, 4 and 8hr) was correlated (p<0.05) with the corresponding radioactivity of $^{14}C$-inulin. It appears to indicate the rhEGF may be uptaken into the skins mainly by the passive diffusion. This hypothesis was supported by the constant specific binding of EGF to the skin homogenates regardless of the skin models. Receptor mediated endocytosis (RME) appears to contribute negligibly, if any, to the overall uptake process.

치이즈 숙성(熟成)에 관(關)한 연구(硏究) 제5보(第5報) Gouda-type 치이즈의 질소화합물(窒素化合物)의 변화(變化) (Studies on Cheese Ripening Part V. Degradation of Nitrogenous Compounds During the Ripening of Gouda-type Cheese)

  • 본(本) 실험(實驗)은 Gonda-type 치이즈의 숙성중(熟成中) casein의 변화(變化)를 구명(究明)하고자 총질소(總窒素), 수용성질소(水溶性窒素), 12% TCA 가용성질소(可溶性窒素) 및 5% NaCl 가용성질소(可溶性窒素)의 양(量)을 정량(定量)하여 비교(比較)하였고 또한 DEAE-cellulose column과 polyacryl amide gel electrophoresis를 사용(使用)하여 그 변화(變化)를 실험(實驗)한 결과(結果)는 다음과 같다. 1. 총질소중(總窒素中) 수용성질소량(水溶性窒素量)은 숙성(熟成) 2개월(個月)과 4개월(個月)에 각각 19.03% 및 30.61%이였으며, 12% TCA 가용성질소량(可溶性窒素量)은 각각 7.72% 및 11.73%이였다. 2. 5% NaCl 가용성질소량(可溶性窒素量)은 총질소중(總窒素中) 제조직후(製造直後)에 10.76%에서 숙성(熟成) 2개월(個月)에는 41.13%로 증가(增加)하였으나 숙성(熟成) 4개월(個月)에는 22.86%로 감소(減少)하였다. 3. 각종(各種) 질소화합물(窒素化合物)을 분가(分加)한 바 각각 모두 5개의 주(主) peak와 $3{\sim}4$개의 minor peak가 나타났으며 각 fraction의 면적비(面積比)가 숙성기(熟成期)에 따라서 증가 또는 감소를 나타내었다. 4. 4개월(個月) 숙성(熟成)된 치이즈를 전기영동(電氣泳動)한 결과(結果) 질소화합물(窒素化合物)에 따라서 $5{\sim}8$개의 band를 나타내었다. 5. 12% TCA에서 침전(沈澱)된 부분(部分)과 가용성(可溶性) 부분(部分)을 아미노산(酸) 조성(組成)을 분석(分析)한 바 모두 16개의 아미노산(酸)으로 나타났으나 조성(組成)의 비율(比率)은 서로 다르게 나타났다.

정상혈압 폐색전증에서 위험도 평가도구로서의 D-dimer의 역할 (D-dimer as a Prognostic Tool in Patients with Normotensive Pulmonary Embolism)

  • Background: D-dimer testing is widely applied as a first step in the diagnostic work-up of pulmonary embolism (PE). Although this is the most sensitive assay for ruling out PE, the prognostic implications of D-dimer testing in patients with normotensive PE are not well known. The aim of this study was to determine if D-dimer testing on admission predicts major adverse cardiac events (MACE) in patients with normotensive PE. Methods: A total of 180 consecutive patients with normotensive PE admitted between January 2003 and June 2009 were included. The group was divided into quartiles on the basis of their D-dimer levels. We compared the frequency of MACE by quartile of D-dimer level and estimated sensitivity, specificity, and predictive values for MACE in the first and fourth quartile. Results: In the 37 (20.6%) patients with MACEs, the median D-dimer level (7.94[IQR:4.03~18.17]${\mu}g/mL$) was higher than in patients with a benign course (5.29[IQR:2.60~11.52]${\mu}g/mL$, p<0.01). The occurrence of MACEs was increased with increasing D-dimer level (p=0.017). In the first quartile (D-Dimer <$2.76{\mu}g/mL$) sensitivity, specificity, and positive and negative predictive values for predicting MACEs were, respectively, 91.9%, 29.4%, 25.2%, and 93.3%. Conclusion: Patients with D-dimer levels below $2.76{\mu}g/mL$ have a low risk of MACEs. Our study suggest that D-dimer level may be used to identify low risk patients with normotensive PE.

제한된 주파수 대역에서 Link-16 웨이브폼의 멀티넷 성능 향상을 위한 그룹 기반의 주파수 도약 방식 (Group-Based Frequency Hopping Scheme for Improving Multi-Net Performance of Link-16 Waveform with Limited Frequency Band)

  • Link-16은 현재 미 공군 및 북대서양 조약기구 (NATO : North Atlantic Treaty Organization)에 의해 운용되고 있는 대표적인 전술데이터링크 (TDL : Tactical Data Link)이며 멀티넷 구조를 지원한다. 이러한 멀티넷 환경에서는 Link-16의 단말 노드들이 동시에 전체 대역에서 도약함으로써 작전을 효과적으로 수행할 수 있다. 최근 항공 교통량이 급증함에 따라 이를 수용하기 위해 새로운 항공 시스템을 도입하거나 기존의 시스템을 확충해야 하며 이러한 시스템을 운용하기 위해 할당되어야 할 주파수 대역이 부족한 상황이다. 이를 해결하기 위해 Link-16에서 사용하는 주파수 대역의 재분배 계획이 예정되어 있다. 재분배 계획이 수행되어 Link-16의 운용을 위한 주파수 대역이 제한될 경우 Link-16에서 다중 접속 간섭 발생률이 증가하게 되므로 멀티넷 성능 저하가 일어날 수 있다. 이로 인해 다양한 군사 작전을 수행하는데 어려움이 따른다. 따라서 본 논문에서는 주파수 부족 현상으로 인해 Link-16 웨이브폼의 멀티넷 성능이 저하되는 문제를 해결하기 위해 그룹 기반의 주파수 도약 방식을 제안하였다. 모의실험을 통해 제안 방식을 사용할 경우 Link-16 웨이브폼의 멀티넷 성능이 향상되는 것을 확인하였다.

한천-아크릴아마이드 미생물 고정화법에 의한 폐수 중 폴리비닐알콜의 분해 (Degradation of Polyvinyl Alcohol in Dye-Processing Wastewater by Agar-Acrylamide Microbial Immobilization Method)

  • 염색가공 폐수 중 난분해성 물질이 PYA를 처리 하기 위하여, 한천-acrylamide를 이용한 bead를 제 조한 후 air-lift 반응기에서 연속실험을 행하였다. 합성폐수의 PYA놓도가 $3,100mg/\ell$. 체류시간 24hr일 때 유출수의 농도는 $4500mg/\ell$ 이며, 제거효율은 85% 이상을 나타내었다. 실제 호발폐수의 경우 PYA 및 COD농도가 $3,253mg/\ell$, $4,500mg/\ell$일 때, 체류시간 24hr에서 유출수의 농도는 $840mg/\ell, 480mg/\ell$이며, 제거효 율은 81.3%와 85.2%로 각각 나타났다. bead의 지름이 lmm일 때는 내부의 미생물 성장 이 양호하였으나 bead의 지름이 2mm일 때는 기질 과 산소전달저항에 의하여 반지름의 48% 이상은 미 생물의 성장이 저해를 받았다. 고정화 반응기에서 전체 기질 제거속도 중 bead 내 고정화 cell의 제거 분율은 평균 70%로 나타났다. 현탁 반응기에서 희석율 $0.083hr^{-1}$ 이상에서는 기질 이용속도가 감소하였으나 고정화 반응기에서는 희석율 $0.125hr^{-1}$까지 거의 선형척으로 증가하였다. PYA 제거속도식에서 포화상수 $K_s=6.60(g PVA/\ell)$와 최대 비기질 이용속도 k=0.175(g PVA/g cell.hr)를 얻었다.
