Effects of Preheated Additives on the Fermentation Quality of Napiergrass Silage

  • Hydrolysis of plant protein to non-protein nitrogen (N) or ammonia can reduce quality of silage crops. Heating or non-enzymatic browning is a treatment to inhibit this hydrolysis. This experiment was conducted to examine the effects of pre-heated soybean meal and molasses on the fermentation quality of napiergrass silage. The initial growth of napiergrass was harvested at 85 days of age and immediately chopped into about 1 cm length. About 700 g of the grass was ensiled into a laboratory silo (1.0 liter polyethylene container) and incubated for 30 days at room temperature ($28^{\circ}C$). No additives (control), molasses, soybean meal and molasses + soybean meal treatments were prepared. All additives were non-heated or heated in an oven at $150^{\circ}C$ for 30 minutes before ensiling. Molasses was added at 3% on the fresh weight basis and soybean meal was added at 0.5% N, respectively. After opening the silo, pH, total nitrogen (TN), volatile basic nitrogen (VBN), lactic acid (LA), acetic acid (AA), butyric acid (BA) and dry matter (DM) contents were determined. The data were analyzed statistically by analysis of variance. Compared with control, molasses addition significantly decreased pH value, VBN/TN, AA and BA and increased LA production. Soybean meal addition significantly increased TN and VBN/TN of silage. Both molasses and soybean meal addition significantly reduced pH value, AA, and BA and increased DM and LA contents of silage. The heating of additives was only effective to reduce VBN/TN production compared with non-heated additives in soybean meal and soybean meal with molasses addition.

Effect of Long Term Fertilization on Soil Carbon and Nitrogen Pools in Paddy Soil

  • Fertilizer management has the potential to promote the storage of carbon and nitrogen in agricultural soils and thus may contribute to crop sustainability and mitigation of global warming. In this study, the effects of fertilizer practices [no fertilizer (Control), chemical fertilizer (NPK), Compost, and chemical fertilizer plus compost] on soil total carbon (TC) and total nitrogen (TN) contents in inner soil profiles of paddy soil at 0-60 cm depth were examined by using long-term field experimental site at $42^{nd}$ years after installation. TC and TN concentrations of the treatments which included N input (NPK, Compost, NPK+Compost) in plow layer (0-15 cm) ranged from 19.0 to 26.4 g $kg^{-1}$ and 2.15 to 2.53 g $kg^{-1}$, respectively. Compared with control treatment, SOC (soil organic C) and TN concentrations were increased by 24.1 and 31.0%, 57.6 and 49.7%, and 72.2 and 54.5% for NPK, Compost, and NPK+Compost, respectively. However, long term fertilization significantly influenced TC concentration and pools to 30 cm depth. TC and TN pools for NPK, Compost, NPK+Compost in 0-30 cm depth ranged from 44.8 to 56.8 Mg $ha^{-1}$ and 5.78 to 6.49 Mg $ha^{-1}$, respectively. TC and TN pools were greater by 10.5 and 21.4%, 30.3 and 29.6%, and 39.9 and 36.3% in N input treatments (NPK, Compost, NPK+Compost) than in control treatment. These resulted from the formation and stability of aggregate in paddy soil with continuous mono rice cultivation. Therefore, fertilization practice could contribute to the storage of C and N in paddy soil, especially, organic amendments with chemical fertilizers may be alternative practices to sequester carbon and nitrogen in agricultural soil.

Primary Production by Epiphytic Algae Attached on the Reed in Constructed Wetlands for Water Treatment (수처리용 인공습지에서 갈대부착조류의 유기물생산력)

  • To estimate the contribution of epiphytic algae attached on reed to organic matter production in constructed wetland, primary productivity by epiphytic algae was investigated in two sub-wetlands (Banweol and Donhwa wetlands) of the Sihwa Constructed Wetland (CW) with different chemistry of inflows. Chlorophyll a concentration of epiphytic algae was higher in the Banweol wetland (range:37~3,581 mgChl.a/$m^2$surface stem, average:655 mgChl.a/$m^2$surface stem) than the Donhwa wetland (range:87~2,093 mgChl.a/$m^2$surface stem, average:527 mgChl.a/$m^2$surface stem). In contrast, assimilation number (AN) representing photosynthetic activity was higher in the Donhwa wetland with low TN/TP ratio than the Banweol wetland. A negative correlation (r=0.46) was observed between TN/TP ratios of inflows and AN in two wetlands, implying that high photosynthetic activity of epiphytic algae may be related with low TN/TP ratio. The areal primary productivity ranged from 307 to 2,473 mgC/$m^2$/day in the Banweol wetland and from 756 to 2,096 mgC/$m^2$/day in the Donghwa wetland, showing high productivity in summer. Average primary production was lower in the Banweol wetland (1,166 mgC/$m^2$/day) than the Donghwa wetland (1,467 mgC/$m^2$/day), although the standing crop (as chlorophyll a concentration) was high in the Banweol wetland. This result may be due to the low photosynthetic activity of epiphytic algae in the Banweol wetland with high TN/TP ratio. The annual primary production (300 tonC/year) of epiphytic algae contributed 33% of the total production in the Sihwa CW. An excessive organic matter production in constructed wetland can negatively affect the efficiency of water treatment. Therefore, the role of epiphytic algae should be considered in management of constructed wetland for water treatment.

Introduction of a New Method for Total Organic Carbon and Total Nitrogen Stable Isotope Analysis of Dissolved Organic Matter in Aquatic Environments (수환경 내 용존성 유기물질의 총 유기탄소 및 총 질소 안정동위원소 신규 분석법 소개)

  • Dissolved organic matter (DOM) is a key component in the biogeochemical cycling in freshwater ecosystem. However, it has been rarely explored, particularly complex river watershed dominated by natural and anthropogenic sources, such as various effluent facility and livestock. The current research developed a new analytical method for TOC/TN (Total Organic Carbon/Total Nitrogen) stable isotope ratio, and distinguish DOM source using stable isotope value (δ13C-DOC) and spectroscopic indices (fluorescence index [FI] and biological index [BIX]). The TOC/TN-IR/MS analytical system was optimized and precision and accuracy were secured using two international standards (IAEA-600 Caffein, IAEA-CH-6 Sucrose). As a result of controlling the instrumental conditions to enable TOC stable isotope analysis even in low-concentration environmental samples (<1 mgC L-1), the minimum detection limit was improved. The 12 potential DOM source were collected from watershed, which includes top-soils, groundwater, plant group (fallen leaves, riparian plants, suspended algae) and effluent group (pig and cow livestock, agricultural land, urban, industry facility, swine facility and wastewater treatment facilities). As a result of comparing characteristics between 12 sources using spectroscopic indices and δ13C-DOC values, it were divided into four groups according to their characteristics as a respective DOM sources. The current study established the TOC/TN stable isotope analyses system for the first time in Korea, and found that spectroscopic indices and δ13C-DOC are very useful tool to trace the origin of organic matter in the aquatic environments through library database.

Characterization of cadC and cadR Mutants in Mediating the Expression of the Salmonella typhimurium cadBA Operon (Salmonella typhimurium cadBA 오페론의 발현에 관여하는 돌연변이체의 선별 및 그 특성)

  • It has been well known that the expression of S. typhimurium cadBA operon requires at least two extracellular signals: low pH and high concentration of lysine. To better understand the nature of pH-dependent and lysine dependent signal transduction, mutants were isolated in JF2238(cadA-lacZ) by Tn10 insertion, spontaneous mutagenesis, and EMS treatment. Two mutants were isolated from JF2238, expressed as a cadA-lacZ operon fusion in various growth conditions, and analyzed to have mutations in cadC, a gene encoding a function necessary for transcriptional activation of cadBA. One isolate (cadC6) conferred pH-independent and lysine-independent cadBA expression and the other(cadC4) showed pH-independent and lysine-dependent cadBA expression. cadR::Tn10 and cadR4 mutants were expressed in the absence of exogenously added lysine. They were also resistant to thiosine and complemented by lysP clone from E. coli. Thus, in the absence of exogenous lysine, cadR is a negative regulator of cadBA expression. Cadaverine, the product of lysine decarboxylation, was shown to inhibit expression of cadA-lacZ fusion in cad $C^+$ cell.

  • We studied the nematic liquid crystal (NLC) alignment capability with a new alignment method utilizing an inkjet printed polyimide (PI) layer. A good, uniform LC alignment was achieved by the good PI printing using a new alignment method. The pretilt angle generated on the printed PI layer using the alignment method was almost the same as that on printed PI layer using rubbing alignment method. In addition, the good electro-optical performances of the new aligned twisted nematic (TN) cell with printed PI surface was obtained

Variation of Nitrogen Removal Efficiency and Microbial Communities Depending on Operating Conditions of a CANON Process (CANON 공정에서 운전조건에 따른 질소 제거효율 및 미생물군집 변화)

  • Nitrogen removal is one of the most important issues about wastewater treatment because nitrogen is a primary pollutant caused various problems such as eutrophication. We developed a CANON microbial community by using AOB and ANAMMOX bacteria as seeding sources. When 100 mg-N/L of influent ammonium was supplied, the DO above 0.4 mg/L showed a very low TN removal efficiency while the DO of 0.3 mg/L showed TN removal efficiency as high as 71.3%. When the influent ammonium concentration was reduced to 50 mg/L, TN removal efficiency drastically deceased. However, TN removal efficiency was recovered to above 70% after 14 day operation when the influent nitrogen concentration was changed again from 50 mg-N/L to 100 mg-N/L. According to the operating temperature from $37{\pm}1^{\circ}C$ to $20{\pm}1^{\circ}C$, TN removal efficiency also rapidly decreased but gradually increased again up to $70.0{\pm}2.6$%. The analysis of PCR-DGGE showed no substantial difference in microbial community structures under different operational conditions. This suggests that if CANON sludge is once successfully developed from a mixture of AOB and ANAMMOX bacteria, the microbial community can be stably maintained regardless of the changes in operational conditions.

Secretory Production of Biologically Active Human Thrombopoietin by Baculovirus Expression System

  • Human thrombopoietin (hTPO) was expressed to high levels in insect cells using the baculovirus expression system. Full-length hTPO cDNA containing a native signal peptide sequence was amplified by PCR from a human fetal liver cDNA library and cloned into the Autographa californica nuclear polyhedrosis virus (AcNPV) expression vector. Immunoblot analysis with antiserum against hTPO indicated that an approximately 55 kDa protein was produced in recombinant AcNPV infected insect cells. Recombinant hTPO was produced 4-fold higher in Trichoplusia ni (Tn5) cells than in Spodoptera frugiperda (Sf9) cells. with most of the hTPO produced in Tn5 cells secreted into the culture medium. Addition of tunicamycin in the culture medium resulted in the reduction of the size of hTPO to 35-38 kDa, and most of the protein remained within the cell. These results suggest that N-glycosylation of hTPO is required for the secretion of the protein into the culture medium in insect cells. hTPO produced in insect cells induced proliferation and maturation of megakaryocyte progenitors, indicating that it is in a biologically active form.

Radiolabelled Monoclonal Antibodies (McAb): An Alternate Approach to the Conventional Methods for the Assessment of Cardiomyocyte Damage in an Experimental Brain-Death Pig Model

  • The present study was carried out to determine the possible use of cTn-I in the cardiac myofibrillar architecture, as a potential target for in vivo radioimmunodetection of cardiac damage in a brain death pig model. Radioiodiantion of the anti-cTn-I 5F4 McAb was carried out by lactoperoxidase method. the percentage iodine incorporation achieved was 70-75%. The radioiodinated McAbs were purified on Sephadex G-25 column and characterised by Paper chromatography, Phast Gel electrophoresis and electroimmunoblotting. Radioiodinated anticTn-I 5F4 McAbs were employed alongside Pyrophosphate($Tc_{99m}$-PPi$) and $Thallium^{201}$ chloride($TI^{201}$) in 24 landrace pigs (brain-dead=18 & sham-operated=6). The percentage cardiac uptake of the radiolabelled antibody injected dose was significantly higher in the brain dead animals(0.196%) as compared to that of sham-operated animals (0.11%). Specific in vivo localization of radiolabelled McAbs in the infarcted cardiac tissue was confirmed by computer-aided reconstruction of 3-D images of the isolated heart. The preliminary results of the study revealed preferential uptake of radiolabelled antibody at the site of myocyte damage resulting from artificially induced brain death.

EO Performances of Flexible TN-LCD using in-situ Ultraviolet Exposure during Imidization of Polyimide on the Polymer Film

  • We have investigated the generation of pretilt angle for a nematic liquid crystal (NLC) alignment with in-situ photoalignment method on polyimide (PI) surfaces using polymer films. Especially, we studied in-situ photoalignment changing heating temperature from $50^{\circ}C\;to\;120^{\circ}C$ on the polymer film. The LC aligning capabilities and pretilt angle on the polymer substrates were better than those on the glass substrate using in-situ photoalignment method. It is considered that this increase in pretilt angle may be attributed to the roughness of the micro-groove substrate induced by the in-situ photoalignment. As temperature of heated subtrate and UV exposure time increase, pretilt angle of the cell used polymer film increased. It is considered that the heating temperature of substrate is attributed to generate pretilt angle. Also, electro-optical performances of the in-situ photoaligned TN cell using the polymer substrate are almost the same as that of the TN cell using the glass substrate.
