Though bile acids have been well known as digestive juice, recent studies have demonstrated that bile acids bind to their endogenous receptors, including Farnesoid X receptor (FXR) and G protein-coupled bile acid receptor 1 (GPBAR1; TGR5) and serve as hormone to control various biological processes, including cholesterol/bile acid metabolism, glucose/lipid metabolism, immune responses, and energy metabolism. Deficiency of those bile acid receptors has been reported to induce diverse metabolic syndromes such as obesity, hyperlipidemia, hyperglycemia, and insulin resistance. As consistent, numerous studies have reported alteration of bile acid signaling pathways in type II diabetes patients. Interestingly, bile acids have shown to activate TGR5 in intestinal L cells and enhance secretion of glucagon-like peptide 1 (GLP-1) to potentiate insulin secretion in response to glucose. Moreover, FXR has been shown to crosstalk with TGR5 to control GLP-1 secretion. Altogether, bile acid receptors, FXR and TGR5 are potent therapeutic targets for the treatment of metabolic diseases, including type II diabetes.
Background: Incretin impairment, characterized by insufficient secretion of L-cell-derived glucagon-like peptide-1 (GLP-1), is a defining step of type 2 diabetes mellitus (T2DM). Ginsenoside compound K (CK) can stimulate GLP-1 secretion; however, the potential mechanism underlying this effect has not been established. Methods: CK (40 mg/kg) was administered orally to male db/db mice for 4 weeks. The body weight, oral glucose tolerance, GLP-1 secretion, gut microbiota sequencing, bile acid (BA) profiles, and BA synthesis markers of each subject were then analyzed. Moreover, TGR5 expression was evaluated by immunoblotting and immunofluorescence, and L-cell lineage markers involved in L-cell abundance were analyzed. Results: CK ameliorated obesity and impaired glucose tolerance in db/db mice by altering the gut microbiota, especially Ruminococcaceae family, and this changed microbe was positively correlated with secondary BA synthesis. Additionally, CK treatment resulted in the up-regulation of CYP7B1 and CYP27A1 and the down-regulation of CYP8B1, thereby shifting BA biosynthesis from the classical pathway to the alternative pathway. CK altered the BA pool by mainly increasing LCA and DCA. Furthermore, CK induced L-cell number expansion leading to enhanced GLP-1 release through TGR5 activation. These increases were supported by the upregulation of genes governing GLP-1 secretion and L-cell differentiation. Conclusions: The results indicate that CK improves glucose homeostasis by increasing L-cell numbers, which enhances GLP-1 release through a mechanism partially mediated by the gut microbiota-BA-TGR5 pathway. Therefore, that therapeutic attempts with CK might be useful for patients with T2DM.

Objective : Comparing the effects of magnetically controlled growing rod (MCGR) and traditional growing rod (TGR) techniques on the sagittal plane in the treatment of early-onset scoliosis (EOS). Methods : Twelve patients were operated using dual MCGR technique in one center, while 15 patients were operated using dual TGR technique for EOS in another center. Patients' demographic characteristics, complications and radiological measurements such as cobb angle, thoracic kyphosis, lumbar lordosis, T1-S1 range (mm), proximal junctional angle, distal junctional angle, sagittal balance, coronal balance, pelvic incidence, sacral slope and pelvic tilt were assessed and compared in preoperative, postoperative and last follow-up period. Results : Age and sex distributions were similar in both groups. The mean number of lengthening in the MCGR group was 12 (8-15) and 4.8 (3-7) in the TGR group. Two techniques were shown to be effective in controlling the curvature and in the increase of T1-S1 distance. In TGR group, four patients had rod fractures, six patients had screw pull-out and four patients had an infection, whereas three patients had screw pull-out and one patient had infection complications in the MCGR group. Conclusion : There was no significant difference between the two groups in terms of cobb angle, coronal and sagittal balance and sagittal pelvic parameters. MCGR can cause hypokyphosis and proximal junctional kyphosis in a minimum 2-year follow-up period. The implant-related complications were less in the MCGR group. However, larger case groups and longer follow-up periods are required for the better understanding of the superiority of one method on other in terms of complications.

Objective : We investigated the pattern of glucose uptake in meningiomas using $^{18}F$-fluoro-2-deoxy-D-glucose[FDG] PET/CT. It was hypothesized that the degree of glucose uptake in each tumor could predict the histologic grade. Methods : In 19 patients with meningiomas, the Ki-67 proliferative index, standardized uptake values[SUV] of FDG uptake, tumor to contralateral gray matter ratio[TGR] of SUV, tumor size, edema grade, vascular endothelial growth factor[VEGF] expression, histopathologic grade and the blood supply pattern were assessed. Results : Of the 19 meningiomas, 8 were meningothelial, 1 fibrous, 2 transitional, 1 psammomatous, 2 angiomatous, and 5 atypical. The tumor proliferative index of Ki-67, tumor size, and peritumoral edema were larger in the histopathologic grade-2 meninigiomas than in the grade-1 meningioma group. There were no significant differences in SUV and TGR between two groups. Tumor size and peritumoral edema were significantly larger in VEGF-positive tumors than in negative tumors. Conventional angiography was performed in 12 patients. Dural supply was noted predominantly in 2 patients. Four patients had mainly pial cortical supply patterns. In tumors with more pial supply, VEGF was more frequently positive. There was a significant relation between SUV and Ki-67 and between SUV and peritumoral edema. Conclusion : We found FOG uptake in meningiomas is associated with proliferative potential, however, no clear limits of SUV and TGR can be set to distinguish between grade-1 and grade-2 meningiomas, which makes the assessment of malignancy grade using PET scan metabolic imaging difficult in individual cases.
We investigated the effect of Chenodeoxycholic acid (CDCA) on the proliferation of osteoclast precursor cells. CDCA decreased the proliferation of osteoclast precursor cells through the control of cell cycle regulators such as cyclin D1, p21 and p27. When we checked the signaling pathway, CDCA decreased Erk activation in osteoclast precursor cells. Furthermore, two bile acid receptors, FXR and TGR5, were involved in the suppressive effect of CDCA. Taken together, this study suggested that bile acid plays an important role in the proliferation of osteoclast precursor cells.
The objective of this study was to examine the impact of pruning (P treatment) and transplanting (T treatment) of Acer palmatum on cambial growth and compartmentalization of pruning wounds for one year after treatments. Changes of cambial electrical resistance (CER), sizes of pruning wounds, cambial growth of trunks and stems near the wounds, and total phenols at branch unions during the period were examined using a total of 49 trees. After harvesting, areas of discolored wood behind the wounds, relative proportions of extractives, holocellulose and lignin at branch unions were also determined. CER and the cambial growth of trunk at 30 cm above the ground (TGR) were inversely correlated, and differences of CER and TGR among three treatments were significant. TGRs of control, P treatment and P+T treatment after the treatments were 112.2%, 72.4% and 52.5% of the annual growth for the year before the treatments, respectively. The cambial growth rate of stem (SGR) at 1.5 cm above the branch bark ridge and the closure rate of pruning wound (WCR) for one year after treatments were positively correlated, and WCR of P treatment of 39.8% was significantly higher than that of P+T treatment of 31.8%. Wounds of P+T treatment formed greater discolored area per unit area of pruning wound (D/W Ratio) than those of P treatment significantly. Lower WCR and higher D/W Ratio of P+T treatment suggested less ability of compartmentalizing the wounds than P treatment. Total phenols at branch core of pruning wound for both treatments heightened a month after treatment, and then lowered. The contents at below core of the wound were higher than those at control ones continuously, while they became similar each other at above core. Relatively high phenol contents of the extractives at P+T treatment implied that trees with P+T treatment allocated more energy to compartmentalize their wounds. Holocellulose and lignin contents at the branch core of treated branch unions of both treatments were lower and higher, respectively, than at the same part of the union with living branch, as results of the tree reaction to protection from wounding and microbial invasion.
A bisdesmosidic cholestane glycoside was isolated from the rhizomes of Polygonatum sibiricum and the structure was elucidated by spectroscopic methods and acid hydrolysis as (22S)-cholest-5-ene-$1{\beta}$,$3{\beta}$,$16{\beta}$,22-tetrol 1-O-${\alpha}$-L-rhamnopyranosyl 16-O-${\beta}$-D-glucopyranoside. This compound exhibited weak cytotoxic activity with the $IC_{50}$ value, $63.6\;{\mu}M$ in human MCF-7 breast cancer line, whereas it failed to show agonistic activity at $100{\mu}M$ in TGR5 assay with Chinese hamster ovary (CHO) cells. This is the first report of a bisdesmosidic cholestane glycoside from Polygonatum species and the full assignments of $^1H$, $^{13}C$ NMR by HMBC, TOCSY and NOESY experiments were provided.
최근 3년간 세계 식품 3D프린팅 시장은 연 평균 31.5%의 높은 성장률과 약 94억 6000만 달러에 이르는 산업규모를 보이고 있다. 식품 3D 프린팅 기술은 기존 식품을 자유롭게 디자인함으로써 개인의 취향과 목적에 따라 식품을 제작할 수 있어 다양한 범위로 활용이 가능하다는 장점을 지니고 있다. 세계 여러 국가에서는 식품 3D프린터를 제작하여 식품 3D 프린팅의 장점들을 활용한 음식과 같은 시제품들을 내놓고 있으며 전투식량, 우주식량, 음식점, 유동식, 노인식, 환자식, 유아식 등의 여러 분야에서까지 활용하고자 시도 중에 있다. 성장가능성이 높고 산업 규모가 지속적으로 확대될 전망인 3D 프린팅 시장은 블루오션이 될 가능성이 다분한 반면 우리나라는 식품 3D 프린팅 기술뿐만 아니라 전반적인 3D프린팅 활용률이나 산업 규모 자체가 작다. 이는 선진국에 비해 미흡한 제도화와 표준화된 국산 소재 개발이 지연 등의 문제가 존재하기 때문으로 따라서 본고에서는 식품 3D프린팅의 필요성에 대해 알리고 우리나라의 식품 3D 프린팅 시장 활성화라는 부가 효과를 얻고자 식품 3D프린팅 기술과 식품소재에 관해 기술하고자 한다.
웅성 모래쥐의 양경동맥을 7분간 폐쇄하여(OBC7) 뇌의 허혈을 유발하여, 해마의 microdialysate내 polyamine 함량과 조직내 polyamine 생합성효소(ornithine decarboxylase: ODC와 S-adenosylmethionine decarboxylase: SAM-DC)의 활성도를 분석하고, 해마의 cresyl violet(CV) 염색과 glial fibrillary acidic protein(GFAP) 면역염색소견들을 관찰하여, 허혈성 해마의 신경손상과 polyamine대사의 연관성을 검토하였다. 1) OBC부하 후, 해마의 dialysate내 polyamine 변동에서, putrescine(PT)은 현저히 증가되었으나, spermidine과 spermine은 다소 감소되는 경향을 보였고, 이에 해마조직내 ODC활성의 현저한 상승과 SAM-DC활성도의 유의한 저학 동반되었다. 2) Difluoromethylornithine(DFMO)는 OBC에 의한 PT증가와 ODC활성도 상승을 유의하게 억제하였으나, methylglyoxal bis(guanylhydrazone)(MGBG)는 각각 다소 억제하는 경향을 보였다. 3) OBC부하 7일후에 관찰한 조직소견에서, 해마의 CA1 부위의 유의한 신경손상이 유도되었으나 CA3와 dentate gyrus 부위에는 미약한 손상만 보였으며, GFAP 양성반응도 CA1 부위에서만 유의한 증가를 보였다. 이같은 소견들은 DFMO에 의하여 크게 영향을 받지 않았으나 MGBG에 의하여 유의하게 억제되었다. 이상의 성적들은 해마 polyamine의 과도한 허혈성 증가가 허혈성 신경손상에 관여할 수 있으나, 한편으로 polyamine대사의 과도한 억제도 허혈성 신경손상을 악화시킬 수 있으며, 허혈성뇌손상에 대한 MGBG의 보호작용은 polyamine 대사보다는 다른 작용에 매개되는 것으로 사료된다. 가장 많았고 습진과 소양증이 각각 19.5%였다. 30대 환자군에서는 습진이 30.3%였고 소양증이 19.1%였다. 40대에서는 소양증이 28.8%였고 지루성 피부염이 16.0%였다. 50대에서는 습진이 26.7%였고 60세 이상의 그룹에서는 습진이 50.0%, 바이러스 질환과 소양증이 각각 15.0%였다. 이들 결과는 통계적으로 유의한 차이를 보였다($X^2\;=\;74.995^{**}$, p = .001, df : 40). 결론 : 상대적으로 50세 이상 연령충의 대상 환자 수가 많지 않은 원인은 여리 가지 분석이 가능할 것으로 생각된다. 연령에 따라 피부질환이 다른 양상을 보임을 고려할 때 노인층의 신체적, 경제적, 사회적 특성을 이해하고 이에 따른 효과적이고 사회-경제학적 부담을 최소할 수 있는 한방 치료 영역의 확대 및 개발이 필요할 것으로 사려된다.주식 및 후식의 선호도에 남녀 간에 차이를 보이고 있으므로 단체급식을 위한 식단 작성 시 이러한 차이를 고려할 필요가 있으며 대학생 및 성인을 대상으로 한 영양 교육을 실시할 때 이러한 차이를 고려한 신생활 교정프로그램을 작성할 필요가 있다고 사료된다.$은 대조군에 비해 발아에 의하여 세포독성 효과를 증가되었지만, MCF-7와 Caco-2에 대한 항암효과는 없음을 알 수 있었다.것으로 사료된다.높게 인식할수록 재방문의도 및 추천의도가 커지는 것을 알 수 있었다. 대학교 급식소 운영주체에 대한 소비자 인지도 조사결과 향후 대학교 급식소를 운영하는 위탁급식 전문업체의 경우 그들의 브랜드를 알리기 위한 홍보전략이 절실히 필요함을 알 수 있었으며, 최근고객감소로 인하여 다양한 급식운영 마케팅전략을 수립하고 있는 단체급식 운영자들은 재방문 및 추천의도의 선행요건이 급식서비스 품질에 대한 소비자의 인식임을 명심하여 가장 기본이 되는 급식서비스 품질수준을 향상시키기 위하여 노력하여야 할 것이다.
