종양 표지 인자로서 혈장 Transforming Growth Factor-β1에 대한 연구 (Study of plasma transforming growth factor-β1 level as a useful tumor marker in various cancers)

  • Background : Many investigators have found transforming growth factor-${\beta}1$ (TGF-${\beta}1$) to be elevated in tumors. Changes in responsiveness to TGF-${\beta}1$ have been linked to malignant transformation, tumor progression and tumor regression. Many malignant cell lines of epithelial or hematopoietic origin are refractory to the antiproliferative effects of TGF-${\beta}1$. However, a little is known about the association of TGF-${\beta}1$ with progression of malignant tumor. Methods : In this study, we measured the plasma level of TGF-${\beta}1$ in various cancer patients and evaluated the utility of plasma TGF-${\beta}1$ as a possible tumor marker. Plasma TGF-${\beta}1$ levels were measured using enzyme-linked immunosorbent assay in cancer patients and normal controls. Carcinoembryonic antigen (CEA) and alpha-fetoprotein (AFP) as tumor marker were compared with TGF-${\beta}1$ in the aspects of sensitivity and specificity. Results : The mean of plasma TGF-${\beta}1$ levels was $1.219{\pm}0.834ng/ml$ in normal controls, $5.491{\pm}3.598ng/ml$ in breast cancer, $12.670{\pm}10.386ng/ml$ in lung cancer, $5.747{\pm}3.228ng/ml$ in hepatocellular carcinoma and $10.854{\pm}7.996ng/ml$ in cervical cancer. In comparison with CEA and AFP, TGF-${\beta}1$ is more sensitive. Conclusion : We conclude that the high levels of TGF-${\beta}1$ are common in the plasma of cancer patients. These results suggest that the plasma TGF-${\beta}1$ level can be a potent tumor marker in various cancer patients.

Epidermal Growth Factor(EGF)와 Transforming Growth Factor-$\alpha$(TGF-$\alpha$)가 돼지 난포란의 체외성숙에 미치는 영향 (Effect of Epidermal Growth Factor and Transforming Growth Factor-$\alpha$ on In Vitro Maturation of Porcine Oocytes)

  • The present study examined the effects of epidermal growth factor(EGF) and transforming growth factor-$\alpha$(TGF-$\alpha$) on in vitro maturation of porcine follicular oocytes. Basic medium used TCM-HEPES, and oocytes cultured for 42 hours in vitro. The results obtained are as follows; 1. The nuclear maturation rates of EGF-treated groups(10ng/ml, 75.9% ; 30ng/ml, 69.2% ; 50ng/ml, 67.2% ; 100ng/ml, 71.0%) on the porcine oocytes cultured in medium without pFF in vitro were significantly(P<0.01) higher than those of non-treated group(57.1%). When the oocytes were cultured in media with 10%(v/v) pFF, the nuclear maturation rates of 30ng EGF/ml(77.1%) treated group were significantly(P<0.01) higher than those of non-(59.2%) and EGF-treated groups(10ng/ml, 65.4% ; 50ng/ml, 65.5% ; 100ng/ml, 70.4%). 2. The nuclear maturation rates of 30ng TGF-$\alpha$/ml treated group(71.9%) in media without pFF in vitro were significatnly(P<0.01) higher than those of non-(57.1%) and TGF-$\alpha$ treated groups(10ng/ml, 60.4% ; 50ng/ml, 65.4% ; 100ng/ml, 60.0%). When the oocytes were cultured in media with 10%(v/v) pFF, the nuclear maturation rates of 30 and 50ng TGF-$\alpha$/ml(77.4% and 79.6%) treated groups(10ng/ml, 64.2% ; 100ng/ml, 61.6%). 3. On the effect of EGF(30ng/ml) and/or TGF-$\alpha$(30ng/ml) treated groups in medium without pFF in vitro, the nuclear maturation rates indicated 57.3, 60.4, 75.9 and 79.7% in media with no EGF & TFG-$\alpha$, TGF-$\alpha$ only, EGF only nad EGF+TGF-$\alpha$ treated groups, respectively. The nuclear maturation rates in medium with EGF only or EGF+TGF-$\alpha$ were significantly(P<0.01) higher than those non- and TGF-$\alpha$ treated groups. When the oocytes were cultured in media with 10%(v/v) pFF, the nuclear maturation ratesof EGF+TGF-$\alpha$ treated group(75.9%) were significantly(P<0.01) higher than those of non-(59.2%), TGF-$\alpha$ only (64.2%) and EGF only(69.4%) treated groups.

4-O-Methylhonokiol Protects HaCaT Cells from TGF-β1-Induced Cell Cycle Arrest by Regulating Canonical and Non-Canonical Pathways of TGF-β Signaling

  • 4-O-methylhonokiol, a neolignan compound from Magnolia Officinalis, has been reported to have various biological activities including hair growth promoting effect. However, although transforming growth factor-${\beta}$ (TGF-${\beta}$) signal pathway has an essential role in the regression induction of hair growth, the effect of 4-O-methylhonokiol on the TGF-${\beta}$ signal pathway has not yet been elucidated. We thus examined the effect of 4-O-methylhonokiol on TGF-${\beta}$-induced canonical and noncanonical pathways in HaCaT human keratinocytes. When HaCaT cells were pretreated with 4-O-methylhonokiol, TGF-${\beta}1$-induced G1/G0 phase arrest and TGF-${\beta}1$-induced p21 expression were decreased. Moreover, 4-O-methylhonokiol inhibited nuclear translocation of Smad2/3, Smad4 and Sp1 in TGF-${\beta}1$-induced canonical pathway. We observed that ERK phosphorylation by TGF-${\beta}1$ was significantly attenuated by treatment with 4-O-methylhonokiol. 4-O-methylhonokiol inhibited TGF-${\beta}1$-induced reactive oxygen species (ROS) production and reduced the increase of NADPH oxidase 4 (NOX4) mRNA level in TGF-${\beta}1$-induced noncanonical pathway. These results indicate that 4-O-methylhonokiol could inhibit TGF-${\beta}1$-induced cell cycle arrest through inhibition of canonical and noncanonical pathways in human keratinocyte HaCaT cell and that 4-O-methylhonokiol might have protective action on TGF-${\beta}1$-induced cell cycle arrest.

근육세포 분화에 대한 TGF-β1과 OP-1의 억제 효과 (The Inhibitory Effect of TGF-β1 and OP-1 onto the Myogenic Differentiation)

  • In order to investigate the effect of Transforming growth factor ${\beta}1$(below TGF-${\beta}1$) and osteogenic protein-1(below Op-1) onto the myogenic differentiation, C2C12 satellite myoblastic cell line was cultured and treated with both growth factors. At first morphological changes with microscopical examination were examined, and isolated total RNA to analyse mRNA expression of bone marker proteins, muscle regulatory proteins, TGF-${\beta}$ receptor and their ligands by Northern blot analysis. And cellular proliferative inducibility of both growth factors was also tested to C2C12 cells. Incubating the cell with $5ng/m{\ell}$ of TGF-${\beta}1$ until 4 days almost inhibited multinucleated myotube formation expressing muscular regulatory proteins, and induced decreasing Id proteins. However, no osteoblastic phenotypes was induced by TGF-${\beta}1$ in C2C12 cells. The mRNA expression of TGF-${\beta}$ receptors with TGF-${\beta}1$ was conversed after 48 hours cultured. Type I TGF-${\beta}$ receptor was seemed to play a role in negative signalling for inhibition of myogenic differentiation. OP-1 dose dependently induced ALP activity, osteopontine production and bone sialoprotein production at concentrations above $100ng/m{\ell}$ and osteocalcin production at concentrations above $300ng/m{\ell}$. The concentration of OP-1 required to induce these osteoblastic phenotypes was the same as that required to almost completely inhibit myotube formation. Incubation with above $100ng/m{\ell}$ OP-1 suppressed the expression of mRNA for muscular egulatory proteins from 2 days after incubation. Expression of Id-1, 2, 3 mRNA were stimulated by OP-1 at concentration above $300ng/m{\ell}$. When C2C12 cells were treated with both growth factors, TGF-${\beta}1$ potentiated the inhibitory effect of OP-1 on myotube formation and expression of mRNA for myogenin at 12 days. And TGF-${\beta}1$ reduced osteocalcin and bone sialoprotein production induced by OP-1 at 12 days in C2C12 cells. Both growth factor had no mitogenic effect. These results indicate that OP-1 converts the differentiation pathway of C2C12 myoblasts into that of osteoblastic lineage cells and it's not heritable, but TGF-${\beta}1$ does not and has reversible inhibitory activity on the myogenic differentiation. TGF-${\beta}1$ and OP-1 play a role in myogenic differentiation via different mechanism between them.

위암조직과 정상조직에서의 표피성장인자 수용체와 변환성장인자의 규명 (Identification of Epidermal Growth Factor Receptor(EGF-R) and Transforming Growth $Factor-{\alpha}(TGF-{\alpha})$ in both Malignant Gastric Adenocarcinoma and Adjacent Non-malignant Gastric Mucosa)

  • 원발성 위암환자로 확진받은 환자들의 암조직과 암조직 주위의 정상점막 조직을 대조군으로 사용하여, $TGF-{\alpha}$와 이에 대한 결합력을 갖고 있는 EGF-Receptor에 대한 mRNA를 면역세포화학적 방법과 in situ hybridization방법을 결합하여 규명하였다. 성장한 세포에서 발견되지 않는 $TGF-{\alpha}$가 위암환자의 조직학적으로는 정상적으로 간주되는 위점막 조직에서 발견된 점으로 미루어 $TGF-{\alpha}$가 암의 분화에 적극적으로 개입하고 있다는 증거가 된다. EMB-11 항체를 사용한 면역세포 화학적 방법에 의해 macrophage를 발견하고, macrophage cell에서 $TGF-{\alpha}$와 EGF-R mRNA가 발현됨을 규명할 수 있었다. 또한 단클론 항체를 이용해 EGF-R에 해당하는 단백질을 발견하였다. CEA를 이용한 면역세포화학 실험에서 정상으로 간주되는 위점막 조직에서 암 세포를 규명하였다. 특히, macrophage cell의 활동이 암의 증식과 더불어 증가하고 있다는 점을 관찰할 수 있었다. 위암과 검사 방법으로서 본 실험에서 사용된 면역세포화학적 기법과 in situ hybridization방법을 사용하여 생검을 통한 조직을 대상으로 성장인자에 대한 검사를 함으로써 정확한 위암의 발생과 진행에 대한 판단을 내리는데 이용할 수 있고 실용성이 있다고 사료된다.

  • Transforming growth factor (TGF)-$\beta$, a hormonally active polypeptide found in normal and transformed tissue, is a potent regulator of cell growth and differentiation. In this study, we examined the effect of TGF-$\beta$ on invasion and motility of MCF10A human breast epithelial cells. TGF-$\beta$-induced migration and invasive phenotype of the parental MCF10A cells in a dose-dependent manner. Activity of MMP-2 promoter was increased by TGF-b, suggesting that the TGF-$\beta$-induced invasive phenotype may possibly be mediated by MMP-2 rather than MMP-9. (omitted)

  • 본 연구의 목적은 시험관내에서 TGF-${\beta}$가 치주인대세포의 증식에 미치는 영향을 관찰하고, 치주 인대내에 TGF-${\beta}$ 를 투여하여 치주 인대조직에 대한 TGF-${\beta}$의 생물학적 역할을 알아보고자 시험관내에서 배양된 사람의 치주인대세포에 0.1, 1, 5, 10ng/ml농도의 TGF-${\beta}$를 투여하여 1,2,3일간 배양한후 MTT방법으로 세포의 활성을 관찰하였고, 백서의 실험적 이동시 압박측 및 견인측의 치주인대내에 TGF-${\beta}$를 투여하여 치주인대조직의 조직병리학적 관찰을 하여 다음과 같은 결과를 얻었다. 1. 0.1ng/ml 농도의 TGF-${\beta}$를 치주인대세포에 투여한 경우, 배양 1, 2, 3일째에 대조군과 유의한 차이가 없었다. 2. 1ng, 5ng/ml 농도의 TGF-${\beta}$를 치주인대세포에 투여한 경우, 배 양 1, 2일째에는 대조군과 유의한 차이가 없었으나 3일째에는 유의하게 증가되었다. 3. 10ng/m1 농도의 TGF-${\beta}$를 치주인대세포에 투여한 경우, 배양 1일째에는 대조군과 유의한 차이가 없었으나 2, 3일째에는 유의하게 증가하였다. 4. 실험적 치아이동시, TGF-${\beta}$ 투여군에서 3일째까지는 압박측에서의 초자양변성이 대조군에 비해 적었으나 7일째 이후에는 군 간의 차이가 없었고, 파골세포 출현 및 모세혈관 증식은 7일째까지 대조군보다 많이 관찰되었다. 5. 실험적 치아이동시, TGF-${\beta}$ 투여군에서 3일째부터 14일째까지 견인측의 골모세포 활성 및 신생골 형성이 증가되었던 점이 대조군과 구별되었다.

  • 연구배경 : 탄광부 진폐증은 석탄 분진의 흡입에 의한 폐조직의 손상에 대하여 부적절하고 과도한 염증반응이 일어나 폐섬유화를 유발하여 발병하는 것으로 생각되어지고 있다. 이 반응에는 대식세포를 비롯한 많은 염증세포들과 그 세포들에서 분비되는 매개물질들이 중요한 역할을 한다. TGF-$\beta$는 특발성 폐섬유화증, 규폐증 및 석면증의 폐섬유화 과정에 관여한다고 알려져 있다. 그러나 비슷한 기전에 의해 폐 섬유화가 진행되는 것으로 생각되는 탄광부 진폐증에서는 TGF-$\beta$의 관여여부에 대한 보고가 거의 없다. 본 연구는 탄광부 진폐증 환자에서 혈청 TGF-${\beta}_1$을 측정하여 섬유화의 정도에 따른 그 활성도의 변화를 비교하여 탄광부 진폐증의 폐섬유화 과정에 TGF-${\beta}_1$이 관여하는지 알아 보고자 하였다. 방법 : 직업력과 방사선학적 소견 상 탄광부 진폐증으로 진단 된 환자 중 단순 탄광부 진폐증 20예와 복잡성 탄광부 진폐증 20예를 대상으로 하였다. 정상소견인 자 10명을 대조군으로 설정하였으며, 각 대상을 human TGF-${\beta}_1$ immunoassay kit (R&D system, Minneapolis, MN)을 이용하여 혈장 내 TGF-${\beta}_1$을 측정하였다. 결과 : 단순 탄광부 진폐증($0.64{\pm}0.17$ ng/mL)과 정상 대조군($0.63{\pm}0.18$ ng/mL)보다 복잡성 탄광부 진폐증 ($0.79{\pm}0.18$ ng/mL)의 혈중 TGF-${\beta}_1$의 농도가 의미 있게 높았다(p<0.05). 결론 : 탄광부 진폐증의 섬유화 진행 과정에 TGF-${\beta}_1$이 관여함을 알 수 있었다. 따라서 단순 탄광부 진폐증과 복잡성 탄광부 진폐증의 감별진단과 경과 예측인자로서 혈장 TGF-${\beta}_1$이 유용하리라 생각되며, TGF-${\beta}$의 생성을 억제한다면 탄광부 진폐증의 섬유화 진행을 저지함으로써 탄광부 진폐증의 치료에 있어 중요한 역할을 할 수 있을 것으로 기대된다.

Effect of ion Pairing on the Cellular Transport of Antisense Oligonucleotide

  • Antisense oligonucleotide represents an interesting tool for selective inhibition of gene expression. However, their low efficiency of introduction within intact cells remains to be overcome. Antisense-$TGF{\beta}$ (25 mer) and antisense-$TGF{\beta}$ (18 mer) were used to study the cellular transport and biological function of antisense oligonucleotide in vitro. Since TGF and TNF play on important role in regulating the nitric oxide production from macrophages, the action of the above antisense oligonucleotides was easily monitored by the determination of nitrite. Poly-L-lysine, benzalkonium chloride and tetraphenylphosphonium chloride were used as polycations, which neutralize the negative charge of antisense oligonucleotide. The production of nitric oxide mediated by .gamma.-IFN in mouse peritoneal macrophage was increased by antisense-TGF.betha. in a dose-dependent manner. Antisense-$TGF{\beta}$ reduced the nitric oxide release from activated RAW 264.7 cells. Significant enhancement in the nitric oxide production was investigated by the cotreatment of poly-L-lysine with antisense-$TGF{\beta}$On the meanwhile, inhibition effect of antisense-$TGF{\beta}$ is not changed by the addition of poly-L-lysine. These results demonstrate that control of expression of $TGF{\beta}$ and TNF.alpha. gene is achieved using antisense technology and the cellular uptake of antisense oligonucleotide could be enhanced by ion-pairing.

Tiul1 and TGIF are Involved in Downregulation of $TGF{\beta}1$-induced IgA Isotype Expression

  • [ $TGF-{\beta}1$ ]is well known to induce Ig germ-line ${\alpha}$ ($GL{\alpha}$) transcription and subsequent IgA isotype class switching recombination (CSR). Homeodomain protein TG-interacting factor (TGIF) and E3-ubiquitin ligases TGIF interacting ubiquitin ligase 1 (Tiul1) are implicated in the negative regulation of $TGF-{\beta}$ signaling. In the present study, we investigated the roles of Tiul1 and TGIF in $TGF{\beta}1$-induced IgA CSR. We found that over-expression of Tiul1 decreased $TGF{\beta}1$-induced $GL{\alpha}$ promoter activity and strengthened the inhibitory effect of Smad7 on the promoter activity. Likewise, overexpression of TGIF also diminished $GL{\alpha}$ promoter activity and further strengthened the inhibitory effect of Tiul1, suggesting that Tiul1 and TGIF can down-regulate $TGF{\beta}1$-induced $GL{\alpha}$ expression. In parallel, overexpression of Tiul1 decreased the expression of endogenous IgA CSR-predicitive transcripts ($GLT_{\alpha},\;PST_{\alpha},\;and\;CT_{\alpha}$) and $TGF{\beta}1$-induced IgA secretion, but not $GLT_{\gamma3}$ and IgG3 secretion. Here, over-expressed TGIF further strengthened the inhibitory effect of Tiul1. These results suggest that Tiul1 and TGIF act as negatively regulators in $TGF{\beta}1$-induced IgA isotype expression.
