Roasting and Cryogenic Grinding Enhance the Antioxidant Property of Sword Beans (Canavalia gladiata)

  • The objective of this study was to optimize the conditions for enhancing the antioxidant properties of sword bean (Canavalia gladiata) as a coffee substitute in two processing methods, roasting and grinding. The optimum conditions for removing off-flavor of the bean and maximizing functionality and efficiency were light roasting and cryogenic grinding (< 53 ㎛). In these conditions, extraction yield was 16.75%, total phenolic content (TPC) was 69.82 ± 0.35 mg gallic acid equivalents/g, and total flavonoid content (TFC) was 168.81 ± 1.64 mg quercetin equivalents/100 g. The antioxidant properties were 77.58 ± 0.27% for DPPH radical scavenging activity and 58.02 ± 0.76 mg Trolox equivalents/g for ABTS radical scavenging activity. The values for TFC and ABTS radical scavenging activity were significantly higher (p < 0.05) than in other conditions, and TPC and DPPH radical scavenging activity were second highest in lightly roasted beans, following raw beans. HS-SPME/GC-MS analysis confirmed that the amino acids and carbohydrates, which are the main components of sword bean, were condensed into other volatile flavor compounds, such as derivatives of furan, pyrazine, and pyrrole during roasting. Roasted and cryogenically ground (cryo-ground) sword beans showed higher functionality in terms of TFC, DPPH, and ABTS radical scavenging activities compared to those of coffee. Overall results showed that light roasting and cryogenic grinding are the most suitable processing conditions for enhancing the bioactivity of sword beans.

분말가공법에 따른 국내산 사과껍질분말의 총페놀, 총플라보노이드 및 항산화능 비교 (Comparison of Total Phenolics, Total Flavonoids Contents, and Antioxidant Capacities of an Apple Cultivar (Malus domestica cv. Fuji) Peel Powder Prepared by Different Powdering Methods)

  • 과일에는 플라보노이드, 탄닌, 카테킨 등의 폴리페놀 뿐만 아니라 여러 항산화능과 관련된 성분을 많이 함유하고 있으며, 과일의 껍질에는 과육부분보다 2-9배 이상 높은 각종 기능성 성분이 많은 것으로 보고되고 있다. 국내에서 사과는 가장 많이 생산되는 과일 중에 하나이며, 사과껍질에는 quercetin의 함량이 높은 것으로 알려져 있다. 하지만 국내에서 사과껍질의 항산화능을 분석하거나 이를 이용하여 식품에 적용한 연구는 부족한 실정이다. 따라서 국내산 사과껍질 원물 균질액, 동결건조분말, 열풍건조분말 및 초미세분말의 총페놀함량(TPC), 총플라보노이드함량(TFC), ABTS radical 소거능(ABTS), DPPH radical 소거능(DPPH)을 비교하고, 이를 통해 사과껍질을 식품 소재로 활용 시 최적 분말화 및 추출조건을 알아보고자 했다. TPC는 동결건조분말, 열풍건조분말의 추출물에서 가장 높게 나타났고, TFC는 초미세분말 추출물에서 가장 높게 나타났다. ABTS 라디컬 소거능은 초미세분말에서, DPPH 라디컬 소거능은 동결건조분말에서 가장 높게 측정되었다. 이상의 결과를 통해, 동일 품종의 사과껍질을 사용한 경우 원물의 가공방법 및 추출조건에 따라 각 기능성의 차이가 나타났다. 이중, 초미세분말에서 TFC와 ABTS 라디컬 소거능이 가장 높은 결과를 나타낸 것을 볼 때, 분말화 방법 등 가공조건의 차이에 의해 개발된 중간소재의 여러 기능성에서 차이가 날 수 있다는 결과를 얻었다. 따라서 본 연구를 통해 기능성이 향상된 분말화된 식품중간소재 개발 시 여러 가공조건에 의해 영향을 받을 수 있으므로, 이를 고려하는 것이 중요하다는 것을 알 수 있었다.

폴리페놀 함량과 항산화력에 따른 피땅콩 겉껍질의 최적 추출 조건 확립과 항비만 기능성 평가 (Polyphenols in peanut shells and their antioxidant activity: optimal extraction conditions and the evaluation of anti-obesity effects)

  • 본 연구에서는 초음파를 이용하여 농업 부산물인 피땅콩 겉껍질의 추출 조건을 최적화하고 추출물의 항산화능과 항비만 기능성을 측정하였다. 실험설계는 CCD를 이용하였고 독립 변수인 추출 시간, 추출 온도, 에탄올 농도에 따라 TPC, TFC와 RSA를 종속 변수로 설정하여 이들의 값을 최대로 하는 공통 조건을 예측하였다. 각각의 모델식의 R2값은 0.85 이상으로 적합하였으며, 추출 조건으로 추출 온도와 에탄올 농도가 TPC 및 TFC에 유의적인 영향을 주는 것을 알 수 있었다. 반면 RSA는 에탄올 농도에 영향을 많이 받는데, 에탄올 농도가 높을수록 RSA가 높아지다가 최고점을 보인 후 다시 감소하는 경향을 나타내었다. 최적 추출 조건인 추출 시간 35.8분, 추출 온도 82.7℃와 에탄올 96.0%에서 제조한 추출물의 TPC, TFC와 RSA의 실측치는 예측치와 유사한 값을 나타내어 회귀 방정식이 신뢰할 수 있음을 확인하였다. 추출물의 항비만 기능성 평가를 위한 GIA와 LIA 실험 결과는 각각 86.4%와 78.5%로 열수 추출물보다 높게 나타나 초음파 추출에 의한 생리활성 물질 추출이 효과적임을 알 수 있었다. 이로써 농업 부산물인 피땅콩 겉껍질로부터 고부가가치 생리활성 물질 생산 가능성을 확인하였고 식품, 화장품 및 의약품용 항산화와 항비만 기능성 소재 생산에 있어 원료 공급비용 절감과 효율적인 추출 공정 확보로 산업 전반에서 가격 경쟁력 확보와 환경문제 해결에 기여할 것으로 사료된다.

  • MB-OFDM UWB시스템은 TFC(Time-Frequency Code)를 이용하여 시간/주파수 호핑을 수행한다. 이를 통해 주파수 영역에서 다이버시티 효과를 갖는 동시에 사용되는 다른 piconet간의 간섭을 최소할 수 있다. 송신단에서는 프리앰블을 전송할 때부터 해당 TFC에 맞게 호핑을 하면서 전송을 하기 때문에 수신단에서는 신호의 시작을 추정하는 패킷 검출은 매우 중요한 과제이다. 따라서 본 논문에서는 MB-OFDM UWB시스템을 위한 간단하고 효율적인 패킷 검출 방법을 제안하였다. 시뮬레이션 수행 결과 제안된 방법은 기존 방법보다 약 0.5 dB의 SNR 개선 결과가 있는 것으로 밝혀졌다.

Fouling resistant membrane tailored by polyethylene glycol in oxidative environment for desalination

  • Surface modification is very efficient and scalable approach to achieve improved membrane performance. We treated Reverse Osmosis Thin Film Composite (TFC RO) membrane with various concentrations of Polyethylene Glycol (PEG), a hydrophilic polymer after activation with sodium hypochlorite. This treatment resulted in an increment of the water flux by 43% and the salt rejection by 2.36% for the 3000 mg/l PEG-treated membrane. Further, these PEG-treated membranes were exposed to a mixture of 3000 mg/l PEG and 1000 mg/l sodium hypochlorite for 1 hour. Further modification of this membrane by PEG and sodium hypochlorite mixture increased the water permeance up to 133% when compared with the virgin TFC RO membrane. We characterized the treated membranes to understand the changes in wettability by contact angle analysis, changes in surface morphology and roughness by scanning electron microscope (SEM) and atomic force microscope (AFM) analysis.

Nutritional value and the kaempferol and quercetin contents of quinoa (Chenopodium quinoa Willd.) from different regions

  • This study compared the nutritional value of quinoa cultivated in different regions, i.e., Peru (PQ), United States (UQ), and Korea (KQ), focusing on their proximate and nutrient compositions and functional components. Moisture, protein, lipid, and ash contents were highest in KQ, and the carbohydrate content was the highest in UQ. KQ had the highest amount of total amino acids, especially lysine. KQ had the lowest levels of Na but the highest levels of K, P, Fe, Mg, Zn, and Mn. The antioxidant compounds, quercetin and kaempferol were not detected in KQ, which consequently had the lowest total phenolic and total flavonoid contents (TPC and TFC, respectively). These values were comparatively higher in UQ. Meanwhile, PQ had the highest TPC and TFC values as well as kaempferol content, but lacking quercetin. These results demonstrate that the nutritional value of quinoa varies according to the region in which it is cultivated.

Cytotoxicity of Cratoxylum Formosum Subsp. Pruniflorum Gogel Extracts in Oral Cancer Cell Lines

  • Background: Oral cancer is a health problem in Thailand. Cratoxylum formosum subsp. pruniflorum Gogel (Teawdang), normally consumed in northeast Thailand, has proven cytotoxic to cervical cancer cell lines including HeLa, SiHa and C-33A. Recently, Asian oral cancer cell lines, ORL-48 and ORL-136, were established. Therefore, we aimed to study cytotoxicity of Teawdang in these. Total phenolic (TPC) and flavonoid content (TFC), and antioxidant activity of Teawdang were also determined. Materials and Methods: Teawdang was purchased from Khon Kaen market during June-October 2013. Hexane (CHE), ethyl acetate (CEE) and methanol (CME) extracts of its edible part were analyzed for TPC by the folin-ciocalteau method and for TFC by an aluminium colorimetric method. Antioxidant activity and cytotoxicity in normal Vero cells and oral cancer cells were investigated. Cell viability was assessed using 3-(4,5-dimethylthiazol-2yl)-2,5-diphenyltetrazolium bromide (MTT) assays. Results: CME and CEE had higher TPC and TFC and antioxidant activity than CHE. Both CME and CEE, at $200{\mu}g$ dry wt/mL, were cytotoxic to the studied oral cancer cell lines. However, CME was cytotoxic to Vero cells whereas CEE was not. Compared to Vero cells, CEE significantly inhibited ORL-48 and ORL-136 growth (p=0.03 and p=0.02, respectively). Conclusions: CEE exhibited cytotoxic effects on the studied oral cancer cell lines but not normal Vero cells. The bioactive compounds in CEE should be further purified and elucidated for their mechanisms of action for development as anticancer agents.

Flavonoid in Clover Honey Exerts a Hypnotic Effect via Positive Allosteric Modulation of the GABAA-BZD Receptor in Mice

  • There is a growing demand for natural sleep aids due to various side effects of long-term administration of pharmacological treatments for insomnia. Honey has been reported to exhibit numerous potential health benefits, and it is hypothesized that honey may favorably affect insomnia treatment. Therefore, this study was performed to investigate the possible hypnotic effect of clover honey (CH) and to determine its in vivo mechanism. The total flavonoid content (TFC) of CH and fractions extracted with ethylacetate (EtOAc) and $H_2O$ was measured. The pentobarbital-induced sleep test using $GABA_A$-benzodiazepine (BZD) agonists and antagonists was conducted to evaluate the potential mechanism of action behind the sedative-hypnotic activity of CH in mice. The results showed that administration of 500 and 1,000 mg/kg of CH significantly (p<0.01) reduced the sleep latency to a level similar to that of diazepam (DZP, 2 mg/kg), and 1,000 mg/kg of CH significantly (p<0.01) prolonged the sleep duration, which was comparable to that of DZP (2 mg/kg). Administration of the EtOAc fraction with a higher TFC significantly reduced the sleep latency at 50 to 200 mg/kg and prolonged the sleep duration at 100 to 200 mg/kg, which were comparable to those after administration of DZP (2 mg/kg). However, co-administration of CH and EtOAc with flumazenil, a specific $GABA_A-BZD$ receptor antagonist, blocked the hypnotic effect. Our findings suggest that the hypnotic activity of CH may be attributed to allosteric modulation of $GABA_A-BZD$ receptors. The TFC of CH is expected to be a key factor that contributes to its hypnotic effect.

지지층 구조가 다른 복합 정삼투막의 기공구조와 분리 특성 (Pore Structure and Separation Properties of Thin Film Composite Forward Osmosis Membrane with Different Support Structures)

  • 본 연구에서는 아세틸화된 메틸 셀룰로스를 복합박막 정삼투막의 지지층으로 사용하였다. 계면중합법을 이용하여 선택성이 우수한 폴리아미드 활성층을 다양한 지지층 위에 코팅하였다. 아세틸화된 메틸 셀룰로스 지지층 위에 코팅된 복합박막 정삼투막의 구조와 성능을 다른 지지층 위에 코팅된 복합박막 정삼투막과 비교하였다. 실험적 결과는 아세틸화된 메틸 셀룰로스 지지층 위에 코팅된 복합박막 정삼투막의 성능이 다른 정삼투막들에 비해서 우수하였으며 이것은 구조적인 특성과 염의 낮은 역확산속도 때문인 것으로 사료된다.

고삼 수집종의 생육특성, 주요성분 및 항산화 활성 (Growth Characteristics, Main Constituents and Antioxidant Activities in Local Accessions of Sophora flavescens AIT.)

  • Background: Sophora flavescens AIT root is used as a herbal medicine in Asian culture. This study was aimed at selecting the best cultivars, by comparing growth characteristics, active ingredients, and antioxidant activities. Methods and Results: A total twenty nine accessions of S. flavescens were collected from five different regions in Korea. Plant height, leaf length, peduncle length, fresh root weight, and seed weight were the growth characteristics selected. Antioxidant activity was determined by measuring the total polyphenol (TPC) and total flavonoid contents (TFC) as well as the radical scavenging activity of DPPH, ABTS and FRAP. Oxymatrine and matrine, active ingredients in S. flavescens were analyzed by HPLC. Results indicated that accession YS-11 had the highest plant height (152.5 cm) and peduncle length (54.0 cm). The fresh root weight was highest in JA-01 at 4.9 kg, while the highest total seed weight was recorded in accession BH-04 at 77.7 g. The YS-03 accession contained the highest oxymatrine and matrine total contents (0.9616% and 0.9638%, respectively). Accessions BH-02, YS-11, YS-05, and MJ-03 had the highest levels of antioxidant activity (p < 0.05). Oxymatrine showed a high correlation with TPC and FRAP. TPC showed high correlations with TFC, DPPH, ABTS and FRAP. TFC showed a high correlation with DPPH. Conclusions: The superior lines can be selected for use as standard variety breeding material by comparing the growth characteristics of the accessions.
