Objective : To investigate the effects of Pyeongwi-san-gamibang on atopic dermatitis, this study measured TEWL(transepidermal water loss), observed scratching behaviors and checked levels of Total IgE, IL-4, IFN-${\gamma}$ and conducted skin biopsy on NC/Nga mice with atopic dermatitis. Methods : NC/Nga mice were challenged with DNCB during 5 weeks to develop atopic dermatitis-like skin lesions. The NC/Nga mice with atopic dermatitis were divided into three groups of control group, PW-d, PW-e group. Once a day for 22 days, Pyeongwi-san-GamiBang extract with water was administered for the PW-d group and the extract with 80% ethanol was administered for PW-e group compared with saline solution for control group. During drug administration, sensitization by DNCB had lasted for three times per week. Results : 1. TEWL had no statistical difference among 3 groups. 2. The scratching behaviors had no statistical difference among 3 groups. 3. The levels of Total IgE in PW-d, PW-e group had a statistically significantly higher than that of the control group although difference between the control group and the PW-d, PW-e group were similar. 4. The level of IL-4 had no statistical difference among 3 groups. 5. The level of IFN-${\gamma}$ had no statistical difference among 3 groups. 6. As the observation of toluidine blue stained lesion, both PW-d and PW-e group had lower level of histamine releasement compared with the control group. Conclusion : Result based on these experiments, Pyeongwi-san-GamiBang on atopic dermatitis-like skin inflammation in NC/Nga mice is not effective. But, as the study showed significantly individual differece, we need to repeat these study after supplementing the object number and modified indicator of clinical severity.
The purpose of this study is to investigate whether the effects of topically applied petrolatum and glycerin on the barrier repair of acetone-induced skin damage in 6 beagle dogs. To confirm the effects of petrolatum and glycerin on acetone disrupted skin models, we performed to evaluate the characteristics of transepidermal water loss and SC hydration and scanning electron microscopic observations. TEWL and SC hydration measurements were carried out 3, 6, 12, 24, 48h after applying petrolatum and glycerin during recovery from acute disruption. Our results showed that there were some different effects between petrolatum and glycerin on the acetone damaged skin such as barrier function repair process and SC hydration status. The results indicate that the significant improvement could be observed in glycerin apply more than petrolatum after acetone damages, and further study will be required.
This study assessed the effects of probiotics in canine atopic dermatitis (AD). We enrolled 11 client-owned dogs with AD and randomly allocated them to two groups. The probiotics group (n = 7) was prescribed with Bifidobacterium longum while the control group (n = 4) received a placebo powder once a day for 12 weeks. In both groups, the degree of skin lesions was evaluated based on the Canine Atopic Dermatitis Extent and Severity Index (CADESI)-4. We also measured the transepidermal water loss (TEWL). We assessed pruritus severity using the pruritus visual analog scale (PVAS). Alteration of consumed drug doses was converted into medication scores. All the evaluation indices were surveyed every 4 weeks. In the probiotics group, there was a significant decrease in the CADESI-4 score at 4, 8, and 12 weeks compared to that of the baseline score (p < 0.05). There was no significant difference in TEWL, PVAS, and medication score at each time point and between groups. Although these results showed that Bifidobacterium longum did not reduce pruritus, TEWL, and the dosage of drugs for canine AD, it was effective in improving skin lesions, therefore, probiotics could be considered in canine AD with severe skin symptoms.

Methods Four-weeks-old NC/Nga mice were divided into four groups: control group (Ctrl, n=10), lipid barrier eliminated group (LBE, n=10), Dexamethasone treatment after lipid barrier elimination group (DxT, n=10), and Baekho-tang extract treatment group after lipid barrier elimination group (BHTT, n=10). Baeko-tang extract was administered for 3 days after removal of the skin fat barrier in BHTT group. Then, we identified changes in external symptoms of the skin, factors affecting skin barrier such as potential of hydrogen (pH), filaggrin (FLG), transepidermal water loss (TEWL) and Th2 differentiation factors like Interleukin (IL)-4, Kallikrein Related Peptidase 7 (KLK7) and protease activated receptor 2 (PAR-2) through our immunohistochemistry. Results After lipid barrier elimination, the reduction of morphological skin inflammations was less in BHTT group than in LBE group and DxT group. Also, pH and TEWL were significantly decreased with BHTT group. However, FLG was significantly increased in BHTT group compared to LBE, DxT, and Ctrl group. All kinds of Th2 differentiation factors (IL-4, KLK7 and PAR-2) were also decreased in BHTT compared to the LBE and DxT. Conclusions As a result of this study, BHT administration decreased pH, TEWL, and increased FLG, thus participating in recovering damaged skin barrier. Since Th2 differentiation factors were decreased as well, BHT's regulatory effect in sequential immune reactions may be a possible explanation of how it enhances recovery of the damaged lipid barrier.

인체피부에서 화장품이나 화장품 원료의 안전성시험은 대체로 육안평가로 이루어져왔다. 하지만, 피부반응 초기의 미세한 변화에 대해서 육안으로는 감지하지 못하는 경우가 많다. 따라서, 본 연구에서는 수종의 화장품 원료에 의해 유발되는 홍반반응에 대해 육안평가뿐 아니라, 레이저도플러혈류이미지(LDPI) 측정장비를 이용한 혈류변화 측정, $Vapometer^{(R)}$를 이용한 경피수분손실량(TEWL) 측정, 분광광도계를 이용한 피부색 측정과 같은 기기평가를 병행하여 비교하였다. 30명의 건강한 여성 피험자를 대상으로 오일류 7종, 계면활성제류 6종, 보습제류 5종을 피험자의 등 부위에24 h 폐쇄첩포(D0)하여 피부반응을 유도하였고 첩포 제거 후 30 min (D1), 24 h (D2)에 육안평가와 기기평가를 각각 실시하였다. 육안평가 결과, 프로필렌글라이콜을 제외한 모든 오일류와 보습제류는 저자극 수준의 피부반응(반응도 0+ - 2.9+)을 보인 반면, 프로필렌글라이콜과 모든 계면활성제류는 중자극에서 강자극 수준의 피부반응(반응도 3+ - 5+)을 보였다. 기기측정 결과, 육안평가에서 저자극 범주의 피부반응에 대해서 혈류량이나 피부색보다 경피수분손실량이 가장 민감하게 피부변화를 감지하였다.
본 연구는 UVB 조사로 인해 손상된 피부에 있어서 자초추출물의 유효성을 검증하기 위해 시도되었다. 생후 6주령된 제모한 C57BL/6 마우스를 대상으로 대조군, UVB 조사군(UVB군), UVB조사 후 자초추출물처치군(UVB+Le군)으로 구분하여 24시간, 48시간, 72시간, 120시간, 168시간의 시간대별로 관찰하였다. 경표피수분손실량을 측정한 결과, UVB+Le군이 UVB군 보다 시간이 경과함에 따라 TEWL이 감소하였다. 특히 168시간군에서 유의하게 낮게 나타났다(p<0.05). 멜라닌 양 측정 결과, UVB+Le군이 UVB군 보다 낮게 나타났으나 통계학적으로 유의성은 없게 나타났다(p>0.05). 홍반 지수 측정 결과, UVB+Le group 24시간, 48시간, 72시간군에서 UVB group 보다 유의하게 낮게 나타났다(p<0.05). 주사전자현미경적 관찰 결과, UVB group 24시간군에서는 UVB group보다 팽윤현상이 완화되었다. 48시간군에서는 가피형성, 72시간군에서는 규칙적인 판상구조, 120시간군에서 새로운 각질세포 생성, 168시간군에서는 얇은 섬유망으로 덮혀 있는 것이 관찰되었다. 투과전자현미경적 관찰 결과, UVB+Le group은 층판소체의 증가와 층판소체의 재형성이 UVB group보다 모든 군에서 촉진되었다. 특히 165시간군에서는 지질이중막의 구조가 거의 회복되어졌다. 이상의 모든 실험결과를 통해 자초추출물이 UVB 조사로 손상된 생쥐 피부를 회복시키는 효과가 있는 것으로 사료된다.
본 연구에서는 피부 상재균에 대한 참죽나무 새순 추출물의 항균 작용과 참죽나무 새순 추출물을 함유하는 크림을 제조하여 인체 피부를 대상으로 보습 효능을 측정하였다. 참죽나무 새순 추출물의 네 가지 피부 상재균에 대한 MIC를 측정한 결과, P. acnes과 S. aureus에 대한 MIC는 각각 0.13 %, 0.03 %로 비교 물질인 methyl paraben보다 더 우수한 항균 활성을 나타내었고, P. ovale과 E. coli에 대한 MIC는 모두 0.25 %로 methyl paraben 보다 낮은 MIC를 나타내었다. 피부 보습 효능을 측정하기 위해, 참죽나무 새순 추출물의 ethyl acetate 분획을 0.25 % 함유하는 크림을 제조하고, 이를 14명의 피실험자의 피부에 도포한 후 피부 수분 보유랑과 경표피 수분 손실량을 각각 30 min, 60 min 간격으로 180 min 동안 측정하였다. 피부 수분 보유량은 placebo 크림에 비하여 참죽나무 새순 추출물 함유 크림이 10 ~ 15 % 정도 더 큰 수분 보유량 증가율을 나타내었다. 경표피 수분 손실량을 측정하였을 때, 무도포(control) 상태의 경표피 수분 손실량은 8.9 g/$m^2h$, 추출물 함유 크림은 6.7 g/$m^2h$으로, 추출물을 함유한 크림이 피부 표면의 수분 손실을 감소시킴을 알 수 있었다. 참죽나무 새순 추출물의 ethyl acetate 분획의 항균 활성과 피부 보습효과에 대한 결과로부터 화장품 원료로서의 가능성을 확인하였다.
피부장벽 기능은 중요한 피부 기능 중 하나로 각질층과 표피층은 다른 피부층 보다 이러한 기능에 있어 큰 역할을 담당하고 있는 것으로 여겨지고 있다. 각질층은 표피 기저층에서부터 표피세포가 위층으로 올라오면서 분화의 과정을 거쳐 형성되게 된다. 이러한 분화 과정은 피부 표면에서 완성되게 되며 지속적으로 올라오는 새로운 각질로 대체되게 되어 건강한 방어막을 형성하게 된다. 본 연구는 tape stripping에 따른 깊이별 여러 인자들의 변화를 살펴봄으로써 각질층의 각 부분의 역할에 대해 살펴보고자 하였다. 피부 표면 수분량에 있어서는 각질층 중 위쪽에 위치하고 있는 uppermost stratum corneum (SC)가 중요한 역할을 하고 있는 것으로 나타났으며 수분증발을 막는 장벽으로써의 역할은 lower SC가 주로 역할을 하는 것으로 나타났다. 그리고 일반적으로 pH는 각질의 산성 pH에서 표피 쪽으로 진행하면서 중성의 pH가 보여지는 것으로 보고되고 있는데 각질층만을 살펴봤을 경우, 아래층으로 내려가면서 오히려 다소 산도가 높아짐을 관찰할 수 있었다. 또한 tape stripping을 통한 각질량 분석에서는 stripping을 거듭할수록 채취되는 각질량도 감소되는 양상을 보여주었다. 본 연구를 통해 수 ${\sim}$ 수십 층으로 구성되는 각질층 중 윗부분은 수분량에, 아래 부분은 장벽으로서의 기능에 있어 역할을 하고 있음을 알 수 있었고 각질층의 깊이가 증가할수록 pH와 각질량은 감소함을 알 수 있었다. 이러한 각질층 내의 위치 차이에 따른 역할에 대한 결과를 통해 화장품 개발에 있어서 다양한 접근이 가능할 것으로 기대되어진다. 또한 본 연구를 통해 각질층도 그 위치에 따라 다른 역할을 담당하고 있음을 간접적으로 알 수 있었고 향후 이러한 점에 초점을 맞춘 특화된 각질 연구도 필요할 것으로 생각되어진다.
