Fruit Characteristics of Gaeryangmeoru Grapes According to Gibberellic Acid and Thidiazuron Treatments (Gibberellic acid와 thidiazuron 처리에 의한 개량머루의 과실 특성)

  • The present study was conducted to establish an effect and a proper concentration for treatment with gibberellic acid ($GA_3$) and thidiazuron (TDZ), resulting with increase berry size and yield in Gaeryangmeoru grapes. Berry size was increased by treatment with $GA_3$, and the fruit clusters obtained for the groups treated with $GA_3$ concentrations of 100 and $200mg{\cdot}L^{-1}$ were bigger. The berry number was also enhanced in $GA_3$ treated groups, but the soluble solid content and acidity was not significantly different. Damage caused by $GA_3$ treatment, such as peel pollination and berry shatter, was observed in the group with $200mg{\cdot}L^{-1}$. The berry size was larger in group treated with a high concentration of $GA_3$ and TDZ respectively than in those treated with low concentrations in the treatment mixed $GA_3$ and TDZ; however, fruit with low soluble solid content and high acidity was harvested after $GA_3$ and TDZ treatment due to delay of berry ripening. The pericarp tissue layers were not changed, but the distance from the epidermis layer to vascular bundle tissue was increased as a result of $GA_3$ and TDZ treatment. Therefore, $GA_3$ and TDZ did not affect an cell division but not cell size, resulting in an enlarged berry size. It is necessary to treat plant growth regulators 2~3 times and immediately after berry set to enhance berry set rate, because the period of berry set is short. This study suggests that the proper concentration for enhancing berry size and set were up to $100mg{\cdot}L^1$ $GA_3$ or $50mg{\cdot}L^{-1}GA_3+1.25mg{\cdot}L^{-1}$ TDZ, and it is necessary to pay attention to harvest mature fruits because of the delay of ripening caused by the usage of TDZ.

Shoot regeneration from internode sections of Ardisia pusilla DC.

  • This study was conducted to regenerate shoots from internode sections(about 1mm in thickness) of Ardicia pusilla de Candolle. Internode sections were cultured on MS medium supplemented with TDZ or both TDZ and IBA. At one month after culture, primodium, which looks like protocorm like body(PLB) of orchid, appeared around swollen internodes. And then it grew and changed into the shape similar to granule of orange at two or three months after culture. At four to five months after culture, explants covered with them became a cluster, and then multiple shoots were regenerated from them. Primodia formation was the best when internode was cultured on MS medium supplemented with 0.25 $mg{\cdot}L^{-1}$ thidiazuron(TDZ) and 0.5 $mg{\cdot}L^{-1}$ indole-3-butyric acid(IBA). That internodes were cultured on MS medium supplemented with either higher concentration of TDZ than that of IBA, or equal concentration of TDZ and IBA, or TDZ only was little effective for primodia formation.

In vitro Mass Propagation of Ardisia pusilla DC. (산호수 (Ardisia pusilla DC.)의 기내 대량번식)

  • To establish the mass proliferation system of Ardisia pusilla DC, the shoot tips of Ardisia pusilla DC were cultured on the MS and half-strength MS medium supplemented with $0{\sim}5.0$ mg/L BA or $0{\sim}0.5$ mg/L thidiazuron(TDZ), respectively. A few multiple shoot formation observed when the shoots were cultured on MS medium containing TDZ. However, the frequency of multiple shoot formation was reached up to 82.4%, when the shoots were cultured on half-strength MS medium supplemented with 0.5 mg/L BA. Also the number of shoot per explant was 7.1. To promote rooting from multiple shoot, newly formed shoots were transferred to half-strength MS medium containing 0.5 mg/L IBA or 0.5 mg/L NAA, respectively. Regenerated plantlets were grown to normal mature plants in soil.

  • Effects of light, methyl jasmonate(MJ), 6-benzylaminopurine(BA), thidiazouron(TDZ), and 2,4-dichlorophenoxyacetic acid(2,4-D) on the contents of ecdysteroids (${\beta}$-ecdysone+polypodine B) in Polypodium vulgare L. and Achyranthes japonica Nakai were studied. When the plants of P. vulgare were cultured under light control, the ecdysteroids contents in both leaves and rhizomes decreased with increasing light intensity. The ecdysteroids contents in A. japonica were also lower when cultured under light than under dark. Among the tested plant growth regulators, MJ, BA, and TDZ increased the ecdysteroids contents in both P. vulgare and A. japonica.

Influence of Growth Regulators and Potassium Humate on in Vitro Multiplication of Apple Rootstock M.26 (생장조절제 및 Potassium Humate가 사과대목 M.26 기내 증식에 미치는 영향)

  • This experiment was designed to improve the in vitro production system of apple rootstock M.26 as being influenced by the growth regulators, TDZ, BA, IAA, IBA, zeatin, and GA$_3$. Different levels of potassium humate (KH), known as cytokinin and auxin-like substance, were also supplemented to the MS basal medium along with IBA 0.6 mg/L to find out it effect on root formation in apple rootstock M.26. ID initiate and establish the in vitro multiplication of shoots byway of meristem culture, MS medium added with zeatin 1.0 mg/L was found to be the most suitable, showing the 100% of survival rate of shoot tips. A combination of thidiazuron (TDZ) 0.2 mg/L and NAA 0.5 mg/L promoted the shoot proliferation when shoot tips were used as explants. MS basal medium plus IBA 0.6 mg/L was very effective for root induction, but an addition of potassium humate (250 mg/L) to the medium containing IBA 0.6 mg/L stimulated the induction and proliferation of the rook by far the better.

Thidiazuron-induced Shoot Formation of Hibiscus syriacus L. 'Honghwarang' by Suspension Culture (Thidiazuron이 무궁화 '홍화랑' 품종 액체 현탁 배양시 신초형성에 미치는 영향)

  • This study was conducted to determine the optimum cultural condition and method for in vitro mass production of Hibiscus syriacus L. 'Honghwarang'. When callus induced in MS solid medium supplemented with 0.01 mg/L TDZ was cultured in liquid medium containing 0.01mg/L TDZ, callus growth and shoot primordia formation was most effective. Formed shoot primordia were regenerated into shoot in MS or 1/2 MS medium of growth regulator-free condition. Effects of mesh size, shaking speed on callus and shoot primordia formation were examined after 5 weeks. Callus and shoot primordia formation was formed most effectively at 10 mesh and 80 rpm shaking speed in liquid medium.

  • In this study, a plant tissue culture system was established for Atractylodes spp., an economically valuable medicinal crop in Korea that has low domestic production and is increasingly imported. In particular, Atractylodes ovata was treated with four types of cytokinins, 6-benzylaminopurine (BA), zeatin, kinetin, and thidiazuron (TDZ), in two different concentrations (0.5 and 1.0 mg/L). Among the four types of cytokinins, the BA treatment was effective for the shoot and root growth of A. ovata. Both the 0.5 mg/L and 1.0 mg/L concentrations of BA showed similar results; however, the 1.0 mg/L concentration of BA was more effective in promoting shoot and root growth. The treatments showed that the TDZ treatment was not effective for the shoot and root growth, except for the number of shoots and the fresh weight (FW) of the root; therefore, it was unsuitable for this species. In this study, we established a mass production system of A. ovata. Our results showed that direct in vitro regeneration may make a significant contribution to improving the cultivation of the medicinal plant A. ovata.

Comparison of in vitro propagation and occurrence of morphological and genetic variation in strawberry tissue culture with various plant hormone treatments (딸기 조직배양 시 여러가지 식물호르몬 처리에 따른 기내 증식 및 형태적, 유전적 변이 발생 비교)

  • The objective of this study was to carry out treatment of various plant hormones in order to determine morphological and genetic variation degree of tissue-cultured strawberry. The cultivar used in this experiment was 'Goha' and 'Seolhyang', the plant hormones used for experiment were benzyladenine (BA), N-(2-Chloro-4 pyridyl)-N'-phenylurea (CPPU) and thidiazuron (TDZ), and the concentrations were 0.5, 1.0, 2.0, $4.0mg{\cdot}L^{-1}$ with each hormone. The BA treatment of the proliferation efficiency of tissue-cultured strawberry 'Goha' and 'Seolhyang' was the highest. When processing BA, CPPU and TDZ, morphological variation and genetic variation happened in strawberry 'Goha' and 'Seolhyang', especially, the variations appeared highly in CPPU treatment. The genetic variation in 'Goha' appeared at the concentration more than BA $0.5mg{\cdot}L^{-1}$ as 1.1%, appeared at the concentration of CPPU $0.5mg{\cdot}L^{-1}$ as 15.3%, and at the concentration of TDZ $2.0mg{\cdot}L^{-1}$ as 1.2%. The genetic variation in 'Seolhyang' appeared at the concentration of BA $4.0mg{\cdot}L^{-1}$ as 2.3%, and at the concentration of CPPU $0.5mg{\cdot}L^{-1}$ as 14.3%. Therefore, CPPU should not be treated during strawberry tissue culture, and BA and TDZ should be treated at low concentration.

Effects of Precursor and Thidiazuron on Triterpene Saponin Biosynthesis in Centella asiatica (L.) Urban

  • Plants have been known to accumulate a very diverse range of triterpene saponins. We have investigated the regulation of saponin biosynthesis in higher plants using Centella asiatica (L.) Urban as a model plant. Effects of a feeding precursor on asiaticoside production from leaves and on the level of two-type OSCs mRNA were investigated. As a feeding precursor, squalene negatively affected the levels of CYS and bAS mRNA, but it also decreased the production of asiaticoside from whole plants. Plant hormones regulate secondary metabolism, and in plant tissue cultures they could affect both culture growth and secondary metabolite production. Although enhancement of asiaticoside production from whole plant cultures by addition of TDZ (thidiazuron) has been reported, the positive effect of TDZ on the levels of OSCs transcripts was not observed.

Micropropagation of Philodendron wend-imbe through Adventitious Multi-bud Cluster Formation (다아체 형성에 의한 Philodendron Wend-imbe의 대량번식)

  • In order to micropropagate uniform plantlets of Philodendron wend-imbe, the shoot tips were cultured on media supplemented with 0.5-10.0mg/L BA or 0.01-1.0 mg/L thidiazuron (TDZ). The multi-bud clusters from basal part of shoots formed vigorously on media containing 5.0-10.0 mg/L BA or 0.05-0.1 mg/L TDZ. Shoot formation from the bud cluster sections (5-7mm) was achieved favorably on medium with 5.0mg/L BA and 20 g/L sucrose. Lowering of sucrose in medium to 20 g/L was effective for the inhibition of callus growth from basal part of shoots. Growth of shoots and their rooting were favorable on media containing 1.0-2.0 mg/L IBA or 0.1mg/L NAA. The rooted plantlets were acclimatizated effectively in soil mixed with perlite 1: peat moss 1 or peat moss alone.
