Effects of TongBiEum(TBE) on Hyperlipidemia Induced by a Sucrose-rich Diet

  • Objectives : This study aimed to elucidate the effects of TBE on hyperlipidemia. Methods : We studied the effects of TBE on hyperlipidemia through gene expressions related with lipid metabolism and serum triglyceride as well as total and HDL-cholesterol levels, and perceived histological changes. Results : The present studies demonstrate that TBE can reduce the rise in plasma cholesterol and TG levels induced by a high-cholesterol diet and also reverse pre-established hypercholesterolemia and hypertriglycemia. In the TBE group total cholesterol levels decreased, TG levels decreased, but HDL-cholesterol levels also decreased. In the analysis of absolute and relative liver weight, TBE inhibited the weight gain induced by a high-cholesterol diet. In the histological observations, lipid droplet and apoptotic change in the TBE treated group were less compared with the control group. In the serum biochemical analysis, a difference of serum AST and ALT changes among groups was not shown, but TG and total cholesterol levels were less and HDL level decreased compared with the control group. In the gene expression related with TG and cholesterol metabolism, DGAT decreased slightly but ACAT decreased more as compared with control and Lipidil groups. Conclusion : From this study, we can infer that TBE possesses a hypolipidemic effect by inhibiting the intestinal absorption and storage of exogenous and endogenous cholesterol.

Comparison of the anesthetic effects of 2,2,2-tribromoethanol on ICR mice derived from three different sources

  • This study was conducted to compare the anesthetic effects of 2,2,2-tribromoethanol (TBE, $Avertin^{(R)}$) in ICR mice obtained from three different sources. TBE (2.5%) was intraperitoneally injected at three doses: high-dose group (500 mg/kg), intermediate-dose group (250 mg/kg), and low-dose group (125 mg/kg). Anesthesia time, recovery time, end-tidal peak $CO_2$ ($ETCO_2$), mean arterial blood pressure, heart rate, oxygen saturation ($SpO_2$), body temperature, pH, $PCO_2$, and $PO_2$ of the arterial blood were measured. Stable anesthesia was induced by all doses of TBE and the anesthesia time was maintained exhibited dose dependency. No significant differences in anesthetic duration were found among the three different strains. However, the anesthesia time was longer in female than in male mice, and the duration of anesthesia was significantly longer in female than in male mice in the high-dose group. The recovery time was significantly longer for female than male mice in the intermediate- and high-dose groups. In the ICR strains tested, there were no significant differences in the mean arterial blood pressure, $SPO_2$, arterial blood $PCO_2$, and $PO_2$, which decreased after TBE anesthesia, or in heart rate and $ETCO_2$, which increased after TBE anesthesia. In addition, body temperature, blood biochemical markers, and histopathological changes of the liver, kidney, and lung were not significantly changed by TBE anesthesia. These results suggested that ICR mice from different sources exhibited similar overall responses to a single exposure to TBE anesthesia. In conclusion, TBE is a useful drug that can induce similar anesthetic effects in three different strains of ICR mice.

Application of TBM/TBE to Mechanical Excavation in Rock (암반기계굴착공법의 적용연구)

  • As tunnel becomes longer and larger, TBM has become one of the most popular methods of excavatio in rock. This paper describes the degree of operation the degree of availability and penetration rate of TBM and TBE applied in Namsan roadway tunnelling site. Net penetration rate was 1.62m/hr for TBM and 0.72m/hr for TBE. Net penetration rate showed no direct relation to daily advance or penetration time, but the lower bound of penetration rate could be obtained from the relation with daily advance. For both of TBM and TBE, the degree of operation and the degree of availability were 33.8% and 68.6% respectively. Life time of normal cutter was $310m^3$ for TBM and $194m^3$ for TBE, while that of center and gauge cutter was about $50m^3$. When the two machines were compared, TBM showed 80% higher penetration rate, and 40% shorter life time of cutter.

The conidial $\beta$-glucosidase of aspergillus nidulans (Aspergillus nidulans의 분생포자에 존재하는 $\beta$-Glucosidase에 관하여)

  • Two kind of ${\beta}-glucosidase$, tightly-bound enzyme(TBE) and loosely-bound enzyme(LBE), were obtained from the conidia of Aspergillus nidulans. The existence of enzymes in conidia was conformed by the fact that these enzyme activities were proportional to the number of conidia. The levels of enzyme activities were independent of aging of the conidia. Enzymes were characterized partially. In spite of the physical and chemical treatments of conidia, there was no significant change in TBE activity. The optimum pH and temperature was 6.0 and $55^{\circ}C$, respectively. Thermostability of the TBE was remarkably higher than that of mycelial ${\beta}-glucosidase$. The electrophoretic pattern of LBE was identical to that of mycelial ${\beta}-glucosidase$. These results suggest that conidial ${\beta}-glucosidase$ are involved in adaptation process of the conidia to variable environments.

  • In this study, the cutting efficiency of TBE (Tunnel Boring Extender) was evaluated by using rotary cutting tester. In the rotary cutting test, a specimen which has a drilled hollow hole at the center was made of rock-like material. The specimen was cut by UDC (undercutting disc cutter) with spiral cutting path to simulate the cutting process of TBE. The cutting forces and specific energy were evaluated under different cutting conditions. The results indicated that the cutter forces of UDC linearly increased with the vertical and radial penetration depths. Among the three directional cutter forces, the normal force is larger than other force components. While the specific energy decreased with the two penetration depths, in particular, it was presumed that the specific energy was minimized at a certain value of the ratio of radial to vertical penetration depth.

Desalination of Tuna Boiled Extract by Electrodialysis (전기투석에 의한 참치 자숙액의 탈염특성)

  • The optimum conditions for selective .elimination of salt from tuna boiled extract (TBE) by electrodialysis were determined. The desalination conditions of TBE were determined at various pH's, concentrations and volumes of TBE. The ion-exchange membrane with a molecular weight cut off 100Da was used for desalting of TBE. The desalination times on $1\%$ and $10\%$ of TBE concentrations were 40 min and 240 min, respectively. The electrodialysis process could removed above $95\%$ of the initial salt content in $1\%$ and $10\%$ of TBE concentrations. The desalination of TBE at pH 4.0 was $14\%$ higher than that at pH 9.0 The amount of water transferred by the electrodialysis was determined. The electrodialysis process could remove above $90\%$ of the initial salt content in $5\%$ TBE for 80 min. The initial volume and the permeate did not have significant effects on desalination time and ratio. The key parameters for the desalination of TBE were pH and concentration of TBA.

  • The purpose of this study was to determine the optimum hydrolysis conditions for the production of enzymatic hydrolysate from tuna boiled extract (TBE) using membrane (molecular weight cut off 10,000Da) reator. The tuna pyloric caeca crude enzyme (TPCCE) was identified as the most suitable enzymes for the hydrolysis of TBE. The optimum hydrolysis conditions of TBE in the batch reactor were $40^{\circ}C$, pH 9 and substrate to TPCCE ratio 50 (w/w). For 6hr under the above conditions, $70\%$ of the total amount of initial TBE was hydrolysed. The optimum hydrolysis conditions of TBE in the membrane reactor were $40^{\circ}C$, pH 9, enzyme 0,1 g/$\ell$, volume 1$\ell$ and substrate to enzyme ratio 100(w/w). The degree of hydrolysis of TBE was above $60\%$ for 3 hr. The TBE hydrolysate were prepared with $5\%$ TBE solution under the optimum hydrolytic conditions in the membrane reactor

Comparison of Electricity Generation Efficiencies depending on the Reactor Configurations in Microbial Fuel Cells (미생물 연료 전지의 반응조 형상에 따른 전기 생산효율 비교)

  • Two different MFC designs were evaluated in batch mode: single compartment combined membrane-electrodes (SCME) design and twin-compartment brush-type anode electrodes (TBE) design (single chamber with two air cathodes and brush anodes at each side of the reactor). In SCME MFC, carbon anode and cathode electrodes were assembled with a proton exchange membrane (PEM). TBE MFC was consisted of brush-type anode and carbon cloth cathode electrodes without the PEM. A brush-type anode was fabricated with carbon fibers and was placed close to the cathode electrode to reduce the internal resistance. Substrates used in this study were glucose, leachate from cattle manure, or sucrose at different concentrations with phosphate buffer solution (PBS) of 200 mM to increase the conductivity thereby reduce the internal resistance. Hydrogen generating bacteria (HGB) were only inoculated in TBE MFC. The peak power densities ($P_{peak}$) produced from the SCME systems fed with glucose and leachate were 18.8 and $28.7mW/m^2$ at external loads of 1000 ohms, respectively. And the $P_{peak}$ produced from TBE MFC were 40.1 and $18.3mW/m^2$ at sucrose concentration of 5 g/L and external loads of 470 ohms, with a mediator (2-hydroxy-1, 4-naphthoquinone) and without the mediator, respectively. The maximum power density ($P_{max}$) produced from mediator present TBE MFC was $115.3mW/m^2$ at 47 ohms of an external resistor.

Harmonic Peak Picking-based MVF Estimation for Improvement of HMM-based Speech Synthesis System Using TBE Model (TBE 모델을 사용하는 HMM 기반 음성합성기 성능 향상을 위한 하모닉 선택에 기반한 MVF 예측 방법)

  • In the two-band excitation (TBE) model, maximum voiced frequency (MVF) is the most important feature of the excitation parameter because the synthetic speech quality depends on MVF. Thus, this paper proposes an enhanced MVF estimation scheme based on the peak picking method. In the proposed scheme, the local peak and the peak lobe are picked from the spectrum of a linear predictive residual signal. The normalized distance between neighboring peak lobes is calculated and utilized as a feature to estimate MVF. Experimental results of both objective and subjective tests show that the proposed scheme improves synthetic speech quality compared with that of the conventional one.

Development of Natural Seasoning using Desalinated Tuna Boiled Extract (탈염된 참치 자숙액을 이용한 천연조미료 개발)

  • The hydrolysate of desalinated tuna boiled extract (TBE) were prepared by continuous hydrolysis of TBE using a membrane reactor. TBE and tuna boiled extract hydrolysate (TBEH) were isolated depending on molecular weights. The major molecular weight distributions of TBEH-l0K, TBEH-5K and TBEH-lK were 9,800Da, 3,000Da and 990Da, respectively. The amounts of nucleotides and their related compounds of TBE were 3.47 $\mu$mole/g AMP, 23.75 $\mu$mole/g IMP, 9.07 $\mu$mole/g inosine and 1.89 $\mu$mole/g hypoxanthine. Total content of amino acids having desirable taste (glycine, glutamic acid, alanine, proline, aspartic acid, serine) was about $63\%$ of total amino acid from TBE and about $62\%$ from TBEH. The natural seasoninings were prepared with TBE and TBEH. From the results of sensory evaluations, complex seasoning containing TBEH-1K was almost equal to the shellfish complex seasoning obtained from the market. The mixed sauce which was made by mixing of $50\%$ TBEH sauce and $50\%$ fermented soy sauce was similar to the tradition soybean sauce in product quality and it showed the possibility to be used for the substitute product for acid hydrolyzed soysauce.
