Application of Images and Data of Satellite to a Conceptual Model for Heavy Rainfall Analysis (호우사례 분석을 위한 개념모델 구성에 위성영상과 위성자료의 활용 연구)

  • This study establishes a conceptual model to analyze heavy rainfall events in Korea using multi-functional transport satellite-1R satellite images. Three heavy rainfall episodes in two major synoptic types, such as synoptic low (SL) type and synoptic flow convergence (SC) type, are analyzed through a conceptual model procedure which proceeds on two steps: 1) conveyer belt model analysis to detect convective area, and 2) cloud top temperature analysis from black body temperature (TBB) data to distinguish convective cloud from stratiform cloud, and eventually estimate heavy rainfall area and intensity. Major synoptic patterns causing heavy rainfall are Changma, synoptic low approach, upper level low in the SL type, and upper level low, indirect effect of typhoon, convergence of tropical air in the SC type. The relationship between rainfall and TBBs in overall well resolved areas of heavy rainfall. The SC type tended to underestimate the intensity of heavy rainfall, but the analysis with the use of water vapor channel has improved the performance. The conceptual model improved a concrete utilization of images and data of satellite, as summarizing characteristics of major synoptic type causing heavy rainfall and composing an algorism to assess the area and intensity of heavy rainfall. The further assessment with various cases is required for the operational use.

  • Attrition is the loss of tooth hard tissue due to contact between teeth, and in severe cases, dentin is exposed, accompanied by selective corrosion and excessive wear of teeth, which is called cupping. If these lesions are left untreated, the size of the lesion gradually increases, breaking the unsupported enamel, resulting in a decrease in aesthetics and chewing function. In this case report, patients with cupping and enamel fracture due to severe attrition were directly restored using a resin with soft properties containing organic fillers. In the follow-up observation six years later, most of the filling of the occlusal surface was eliminated, but the filling on the buccal surfaces remained relatively intact, and it was confirmed that this type of resin was suitable for the area where the occlusal force was relatively weak rather than the area where the occlusal force was greatly applied.

  • Supercritical fluid chromatography (SFC) was considered for separating racemic ibuprofen. The chromatographic column (3.9 ${\times}$ 150 mm) was packed with Kromasil$^{\circledR}$ CHI-TBB, and the mobile phase was supercritical carbon dioxide with modifier of IPA. The experimental variables were the content of IPA, and temperature and pressure of supercritical mobile phase. To determine the separation condition, the empirical equation of retention factor and resolution was proposed. In the case of retention factor, the empirical equation was in the form, $k\;=\;a{\rho}\;+b/F\;+\;c\;({\rho}/F)\;+\;d$. The empirical equation for resolution was proposed as a linear form, $R\;=\;a{\rho}\;+\;bF\;+\;c$.

CK2 phosphorylates AP-2α and increases its transcriptional activity

  • Transcription factor AP-$2{\alpha}$ involves in the process of mammalian embryonic development and tumorigenesis. Many studies have shown that AP-$2{\alpha}$ functions in association with other interacting proteins. In a two-hybrid screening, the regulatory subunit ${\beta}$ of protein casein kinase 2 ($CK2{\beta}$) was identified as an interacting protein of AP-$2{\alpha}$; we confirmed this interaction using in-vitro GST pull-down and in-vivo co-immunoprecipitation assays; in an endogenous co-immunoprecipitation experiment, we further found the catalytic subunit ${\alpha}$ of protein casein kinase 2 ($CK2{\alpha}$) also exists in the complex. Phosphorylation analysis revealed that AP-$2{\alpha}$ was phosphorylated by CK2 kinase majorly at the site of Ser429, and such phosphorylation could be blocked by CK2 specific inhibitor 4,5,6,7-tetrabromobenzotriazole (TBB) in a dose-dependent manner. Luciferase assays demonstrated that both $CK2{\alpha}$ and $CK2{\beta}$ enhanced the transcription activity of AP-$2{\alpha}$; moreover, $CK2{\beta}$ increased the stability of AP-$2{\alpha}$. Our data suggest a novel cellular function of CK-2 as a transcriptional co-activator of AP-$2{\alpha}$.

Separation Characteristics of Mandelic Acid in Kromasil HPLC Column (Kromasil HPLC 칼럼에서 Mandelic Acid의 분리특성)

  • Chiral separation of racemic mandelic acid was achieved on a Kromasil KR100-5CHI-TBB column. Some chromatographic parameters (resolution, number of theoretical plates, capacity factor) are calculated under different separation conditions such as changes of mobile phase compositions (hexane/t-BME = 85/15 - 10/90) as well as formic acid concentrations for adjusting pH (0.1, 0.5, 1.0 v/v%). Flow rate versus number of theoretical plates was compared to evaluate column efficiency. To determine the adsorption isotherms, PIM (Pulse Input Method) was carried out. At the concentrations of racemic mandelic acid between 0.1 and 0.3 mg/ml, L- and D-mandelic acids have the same retention times of 8.8 and 9.4 min respectively. Mandelic acid isotherms show a linear form under the concentrations of 0.3 mg/ml with eluent (hexane/t-BME = 75/25). As the concentrations of mandelic acids increase, nonlinear Langmuir isotherms were observed as $C_{S,L}=3.358C_{M,L}/(1+0.0897C_{M,L})$ for L-mandelic acid and, $C_{S,D}=3.692C_{M,D}/(1+0.1457C_{M,D})$ for D-mandelic acid.

Three-dimensional thermal-hydraulics/neutronics coupling analysis on the full-scale module of helium-cooled tritium-breeding blanket

  • Blanket is of vital importance for engineering application of the fusion reactor. Nuclear heat deposition in materials is the main heat source in blanket structure. In this paper, the three-dimensional method for thermal-hydraulics/neutronics coupling analysis is developed and applied for the full-scale module of the helium-cooled ceramic breeder tritium breeding blanket (HCCB TBB) designed for China Fusion Engineering Test Reactor (CFETR). The explicit coupling scheme is used to support data transfer for coupling analysis based on cell-to-cell mapping method. The coupling algorithm is realized by the user-defined function compiled in Fluent. The three-dimensional model is established, and then the coupling analysis is performed using the paralleled Coupling Analysis of Thermal-hydraulics and Neutronics Interface Code (CATNIC). The results reveal the relatively small influence of the coupling analysis compared to the traditional method using the radial fitting function of internal heat source. However, the coupling analysis method is quite important considering the nonuniform distribution of the neutron wall loading (NWL) along the poloidal direction. Finally, the structure optimization of the blanket is carried out using the coupling method to satisfy the thermal requirement of all materials. The nonlinear effect between thermal-hydraulics and neutronics is found during the blanket structure optimization, and the tritium production performance is slightly reduced after optimization. Such an adverse effect should be thoroughly evaluated in the future work.

Elucidation of the Vulcanization Structures of Filled cis-1,4-Polybutadiene Rubber by Solid State Carbon-13 NMR Spectroscopy (고체상태 NMR을 이용한 cis-1,4-polybutadiene 충진고무의 가황가교 구조 규명)

  • Using solid state $^{13}C$ NMR, polybutadiene rubber vulcanizates were qualitatively and quantitatively analyzed. In the filled conventional system of BR vulcanizate accelerated with TBBS, addition to the olefinic double bond and substitution in the $\alpha$ position to the double bond occurred simultaneously. Also the latter $\alpha$ substitution reaction was faster than the former addition reaction at initial reaction time. In addition, it was suggested that double bond-addition-polysulfide structures might be modified into 5-membered and 6-membered cyclic structures in overcure time. These chain modifications were correlated with the decrease in the chemical crosslink density in overcure time.

Real-time Fluorescence Lifetime Imaging Microscopy Implementation by Analog Mean-Delay Method through Parallel Data Processing

  • Fluorescence lifetime imaging microscopy (FLIM) has been considered an effective technique to investigate chemical properties of the specimens, especially of biological samples. Despite of this advantageous trait, researchers in this field have had difficulties applying FLIM to their systems because acquiring an image using FLIM consumes too much time. Although analog mean-delay (AMD) method was introduced to enhance the imaging speed of commonly used FLIM based on time-correlated single photon counting (TCSPC), a real-time image reconstruction using AMD method has not been implemented due to its data processing obstacles. In this paper, we introduce a real-time image restoration of AMD-FLIM through fast parallel data processing by using Threading Building Blocks (TBB; Intel) and octa-core processor (i7-5960x; Intel). Frame rate of 3.8 frames per second was achieved in $1,024{\times}1,024$ resolution with over 4 million lifetime determinations per second and measurement error within 10%. This image acquisition speed is 184 times faster than that of single-channel TCSPC and 9.2 times faster than that of 8-channel TCSPC (state-of-art photon counting rate of 80 million counts per second) with the same lifetime accuracy of 10% and the same pixel resolution.

  • To realize a zero-energy building, a technology that minimizes the energy loss due to thermal bridges by preventing their formation is emerging as an important design factor. In this study, we develop a thermal bridge breaker to prevent thermal bridging in a metal panel roof and attempt to analyze the effects of its application. To this end, we fabricated a thermal bridge breaker and analyzed it in terms of its strength and heat-transfer characteristics, in addition to conducting a load simulation. The thermal bridge prevention effect of the developed thermal bridge breaker improved the insulation performance of the metal panel roof, and the results of a cooling/heating peak load simulation performed by applying the heat transmission resistance test results to a building proved the existence of this effect.

Development of Cloud Amount Calculation Algorithm using MTSAT-1R Satellite Data (MTSAT-1R 정지기상위성 자료를 이용한 전운량 산출 알고리즘 개발)

  • Cloud amount calculation algorithm was developed using MTSAT-1R satellite data. The cloud amount is retrieved at 5 km ${\times}$ 5 km over the Korean Peninsula and adjacent sea area. The algorithm consists of three steps that are cloud detection, cloud type classification, and cloud amount calculation. At the first step, dynamic thresholds method was applied for detecting cloud pixels. For using objective thresholds in the algorithm, sensitivity test was performed for TBB and Albedo variation with temporal and spatial change. Detected cloud cover was classified into 3 cloud types (low-level cloud, cirrus or uncertain cloud, and cumulonimbus type high-level cloud) in second step. Finally, cloud amount was calculated by the integration method of the steradian angle of each cloud pixel over $3^{\circ}$ elevation. Calculated cloud amount was compared with measured cloud amount with eye at surface observatory for the validation. Bias, RMSE, and correlation coefficient were 0.4, 1.8, and 0.8, respectively. Validation results indicated that calculated cloud amount was a little higher than measured cloud amount but correlation was considerably high. Since calculated cloud amount has 5km ${\times}$ 5km resolution over Korean Peninsula and adjacent sea area, the satellite-driven cloud amount could show the possibility which overcomes the temporal and spatial limitation of measured cloud amount with eye at surface observatory.
