돌나물(Sedum sarmentosum) 에탄올 추출물의 항균, 항산화, 항노화 효과와 수중유적형 크림의 제조 (Antibacterial, Antioxidant, and Antiaging Effects of the Ethanol Extract of Dolnamul (Sedum sarmentosum) and the Production of the Oil in Water Cream)

  • 돌나물(Sedum sarmentosum)의 기능성과 화장품 소재로서의 특성을 파악하기 위하여 본 연구를 수행하였다. 돌나물을 동결건조하고 분쇄하여 에탄올로 추출한 후 $-20^{\circ}C$ 에 저장하면서 실험에 사용하였다. 돌나물의 에탄올 추출물(SE) 내에 포함된 총폴리페놀화합물(TPC)의 함량은 $27.98{\pm}0.34g/kg$ (건조중량)이었고, epicatechin ($162.14{\pm}5.07mg/kg$), epigallocatechin ($55.99{\pm}2.49mg/kg$) 및 kaempferol ($47.96{\pm}3.02mg/kg$)순으로 많이 함유되어 있는 것으로 분석되었다. SE는 organic radical 소거효과($78.43{\pm}1.08%$)와 금속이온환원능(FRAP value $2.54{\pm}0.12$)을 나타내었다. 또한 SE는 지질과산화 반응의 초기 단계(FTC assay $62.03{\pm}0.38%$)와 최종단계(TBARS assay $55.36{\pm}2.05%$)를 저해하였다. SE (5 mg/mL 건조중량)는 Propionibacterium acne에 대해 항균효과를 나타내었다. SE의 elastase 저해율은 $38.94{\pm}7.09%$이었고, collagenase 저해율은 $78.94{\pm}2.49%$이었다. SE 처리구는 Col3A1 발현양이 증가하였고 collagen 생성율은 대조구보다 $58.11{\pm}1.07%$ 높았다. 0.5% SE가 함유된 수중유적형 크림은 pH 6.88, 밀도 1.47 g/mL이었고, 다양한 온도($-20-45^{\circ}C$)에 4주간 저장하는 동안 크림은 안정한 유화상태를 유지하였다. 따라서 SE는 항균, 항산화, 항노화 활성을 지닌 것으로 사료되었다.

  • 발효식품 시료로부터 분리된 세균을 $DPPH({\alpha},{\alpha}'-diphenyl-{\beta}-picrylhydrazyl)$ 전자공여능으로 항산화 활성을 측정하여 가장 활성이 강한 균주를 선별하여 형태학적, 생화학적, 생리학적 특성 및 165 rRNA 염기서열을 조사한결과 Bacillus sp.으로 판명되어 FF-7로 명명하였다. DPPH 전자공여능법에 의한 Bacillus sp. FF-7이 생산하는 항산화물질의 최적 생산 배지조건은 탄소원 2% galactose와 질소원 1% tryptone 첨가였다. Bacillus sp. FF-7에 의해 생성된 항산화 물질의 활성을 DPPH 전자공여능, 흰쥐 각 조직 microsomal 실험계 및 linoleic acid 과 산화지질 실험계에서 malondialdehyde를 thiobarbituric acid(TBA)방법으로 측정하였다. 흰쥐 각 조직 microsomal 실험계에서 지질과산화에 대한 항산화 효과는 뇌(97.50%) >심장(79.95%) >신장(77.84%) >비장(77.47%) >고환(69.96%) >간장(62.45%) 순이였다. Linoleic acid의 과산화지질를 TBA법으로 측정한 결과 반응 4일째까지 억제 효과가 강하게 나타났으며, 동시에 대조구로 사용한 0.05% BHT 첨가구에서도 실험종료시까지 항산화 활성이 강하게 나타났다.

Use of Dried Stoned Olive Pomace in the Feeding of Lactating Buffaloes: Effect on the Quantity and Quality of the Milk Produced

  • Dried stoned olive pomace (DSOP) was administered to dairy water buffaloes, and their productive performance and milk composition were analysed. Sixteen pluriparous lactating buffaloes were divided into two uniform groups (control and experimental), taking into consideration the following parameters: milk production (2,192 and 2,102 kg) and duration of lactation (254 and 252 d) of the previous year, distance from calving (51 and 43 d), milk production (9.71 and 10.18 kg/d), body condition score (BCS) (6.44 and 6.31) and weight (617 and 653 kg) at the beginning of the trial. Both diets had the same formulation: second cut alfalfa hay 20%, corn silage 42%, concentrate 38% but the two concentrates differed in their formulation, the experimental one contained 15.50% of DSOP as fed. The employed DSOP showed high amounts of secoiridoids, such as 3,4-dihydroxyphenylethanol (3,4-DHPEA) (1.2 g/kg DM), 3,4-dihydroxyphenylethanol-elenolic acid di-aldehyde (3,4-DHPEA-EDA) (12.6 g/kg DM), p-hydroxyphenylethanol-elenolic acid di-aldehyde (p-HPEA-EDA) (5.6 g/kg DM) and lignans, which are known to be powerful bioactive compounds. The control diet had an energy-protein content of 0.86 Milk FU/kg DM and 143.3 g/kg DM of crude protein, whereas the experimental diet of 0.87 Milk FU/kg DM and 146.6 g/kg DM of crude protein. Each animal of the two groups received 17 kg DM/d and each buffalo of the experimental group, by way of the concentrate, ingested 1.05 kg DM/d of DSOP. The trial lasted 40 days. No significant difference was found between the BCS (6.41 and 6.53), live weight (625.93 and 662.50 kg) and milk production (9.69 and 10.08 kg/d) of the two groups, as was the case for fat, protein, lactose, pH and coagulating parameters of the two milks. The milk fat of the experimental group had a significantly higher content of total tocopherols (10.45 vs $8.60{\mu}g/g$, p<0.01) and retinol (3.17 vs $2.54{\mu}g/g$, p<0.01). The content of the reactive substances with tiobarbituric acid (TBARs) was significantly lower in the milk fat of the experimental group (12.09 vs $15.05{\mu}g$ MDA/g, p<0.01). The acid content of the milk fat of the experimental group had a significantly higher content (p<0.05) of C18:0 and of $C18:3{\omega}6$. LC-MS/MS analysis showed the presence of 3,4-DHPEA ($36.0{\mu}g/L$) in the milk of the DSOP-fed buffaloes, while other phenols were not found. DSOP, in the quantity utilized, can be used in the feeding of the lactating buffalo; the dietetic-nutritional characteristics of the milk are improved due to a greater contribution of tocopherols, retinol and the presence of hydroxytyrosol.

원주지역 남은 음식물의 계절별 성분 함량 및 비육돈에 대한 건조 남은 음식물 급여 효과 (Seasonal Variations in Chemical Composition of Dried Food Waste in Wonjusi and Its Feeding Effects in Finishing Pigs)

  • 본 연구는 남은 음식물(FW)의 성분변이를 조사하고, 그것의 급여시 비육돈의 성장과 육질에 미치는 영향을 조사하기 위하여 실시하였다. 사료화를 위해 기초자료로 활용하고져 FW를 1년간 (월 6회) 수집하였다. 8주간의 사양시험을 위해 삼원교잡종(L${\times}$Y${\times}$D) 비육돈 $\pm$ 4.60kg) 117두를 공시하였다 (3처리 3반복, 반복당 13두). 처리는 대조구 (옥수수-대두박 위주사료), 건조 방법에 따른 단순 건조 (simple dry: SD) 및 진공발효 (vacuum fermentation: VF)로서 구분하였다. FW의 건물기준으로 에너지, 조단백질, 조지방, 회분, 칼슘 및 인의 4계절 평균치는 각각 5,111kcal/kg, 22.92%, 14.31%, 15.48%, 2.7% 및 1.05%였다. 에너지와 단백질 함량은 각각 겨울과 여름에 가장 높았다 (p<0.05). 건조한 FW의 유산균 함량은 SD와 VF 처리간에 차이가 없었다. 사양성적에서는 ADG에서 대조구가 FW이 첨가된 사료에 비해 유의적으로 높았으나 (p< 0.05), FCR에서는 처리간에 차이가 없었다. SD와 FD와의 생산성 비교에서는 유의차가 인정되지 않았다. 지육율, 등지방두께, 그리고 기타 육질(색깔, drip loss 및 TBARS)에서 처리간에 차이가 없었다. 전 시험기간의 증체 kg당 사료비를 살펴보면, FW가 포함된 사료급여군이 대조구에 비해 낮았다. 이상의 결과를 요약해 보면, 남은 음식물을 건조한 후 사료에 20% 이내를 첨가하여 펠렛사료로 급여할 경우 비육돈의 사료비를 절감할수 있을 것으로 사료된다. 그러나 FW의 가공시 FD공정은 필요치 않을 것으로 사료된다.

소인제(Aeonium sedifolium) 잎 유래 phenolic 성분의 항산화, 피부주름생성 억제, 항염증 및 모공 수축 효과 (Anti-oxidation, anti-inflammation, anti-wrinkle, and pore-tightening effects of phenolic compounds from Aeonium sedifolium leaves)

  • 소인제(Aeonium sedifolium) 잎의 total phenolics compounds 함량은 열수 및 ethanol 추출물에서 각각 34.49 mg/g, 61.64 mg/g으로 나타났다. 소인제 잎의 solid에 비해 phenolics의 DPPH, ABTS 효과가 더 우수한 결과를 나타내어 생리활성은 phenolic에 의해 발생하는 것으로 확인되었다. 소인제 잎의 항산화 효과는 DPPH가 100 ㎍/mL TPC 농도에서 85% 이상의 높은 소거 활성을 나타내었고, ABTS radical cation decolorization은 열수와 ethanol 추출물에서 모두 100%에 근접한 항산화 활성을 나타내었다. Antioxidant protection factor는 100 ㎍/mL phenolic 농도에서 열수와 ethanol 추출물에서 각각 1.07 PF와 1.22 PF를 나타내었으며, TBARs 저해 효과는 열수와 ethanol 추출물 모두 77% 이상의 활성을 나타내었다. 소인제 잎 추출물의 elastase 저해 활성은 phenolic 100 ㎍/mL의 ethanol 추출물에서 69.03%로, positive control인 ursolic acid의 35.83%보다 2배의 억제효과를 나타내었다. Collagenase 저해활성은 phenolic 농도 100 ㎍/mL에서 열수추출물이 29.82%, ethanol 추출물이 54.76%의 저해 효과를 나타내어 주름개선효과가 우수한 것으로 확인되었다. 소인제 잎의 HAase 저해활성은 400 ㎍/mL phenolic 농도의 열수 및 에탄올 추출물에서 각각 85%, 99.12%의 높은 저해활성을 나타내었다. 소인제잎 추출물의 모공수축 효과는 phenolic 농도 400 ㎍/mL에서 100% 가까운 매우 우수한 효과를 나타내었다. 이상의 결과를 종합해보면 소인제 잎 추출물은 항노화에 관여하는 높은 항산화 활성을 나타내었으며, 피부 주름 생성에 관여하는 elastase와 collagenase 저해활성과 염증반응에 관여하는 hyaluronidase 저해활성이 우수하였으며, 모공수축 효과 또한 매우 우수한 것으로 확인되었다.

Effects of Lacquer (Rhus verniciflua) Meal on Carcass Traits, Fatty Acid Composition and Meat Quality of Finishing Pigs

  • This experiment was conducted to evaluate the effects of feeding different levels of lacquer (Rhus verniciflua Stokes) meal on the growth performance, carcass traits, fatty acid profile and meat quality of longissmuss dorsi (LD) muscle in finishing pigs. Pigs (n = 117; Landrace$\times$Yorkshire$\times$Duroc; initial body weight $80{\pm}0.4kg$) were allotted to three dietary treatments and fed lacquer at 0, 2 and 4% of the diet for five weeks. Inclusion of lacquer meal in the diets of pigs had no influence on their growth performance, carcass yield, loin eye area and fat free lean; however, pigs fed lacquer diets had lower backfat (linear, p = 0.006; quadratic, p = 0.004). Pigs fed increasing levels of lacquer meal had lower moisture (linear, p<0.001; quadratic, p = 0.008), crude fat (linear, p<0.001) and crude protein (linear, p<0.001; quadratic, p = 0.002) in LD muscle. The LD muscle of pigs fed lacquer meal had lower pH (linear and quadratic, p<0.05) at 6, 8 and 10 days, and linearly lower thio-barbituric acid reactive substances (TBARS, p<0.01) at 8 and 10 days and water holding capacity (WHC, p<0.05) at 3, 6, 8 and 10 days. The fatty acid composition of LD muscle revealed linearly lower stearic (p = 0.034) and total saturated fatty acid (p = 0.049) with increasing dietary lacquer meal levels. In general, higher lightness, redness and yellowness values were observed in LD muscle of pigs fed 2% lacquer meal on day 0 and subsequently on 3, 6, 8 and 10 days of refrigerated storage. The results of the current study suggest that lacquer meal can be incorporated up to 4% in the diet of finishing pigs without any adverse effects on performance; moreover, improvements in the meat quality during refrigerated storage can be obtained by inclusion of lacquer meal in the diet of finishing pigs.

Effects of Anion Supplementation on Growth Performance, Nutrient Digestibility, Meat Quality and Fecal Noxious Gas Content in Growing-finishing Pigs

  • Forty-eight ((Duroc${\times}$Yorkshire)${\times}$Landrace) pigs with an average initial body weight (BW) of $48.47{\pm}1.13\;kg$ were used in a 12-week growth trial to investigate the influence of Anion (silicate) supplementation on growth performance, nutrient digestibility, meat quality and fecal noxious gas content in growing-finishing pigs. Pigs were allotted into three dietary treatments in a randomized complete block design according to sex and initial BW. Each dietary treatment consisted of four replications with four pigs per pen. Dietary treatments included: i) CON (basal diet), ii) HCI (basal diet+3 g/kg Anion), iii) HCII (basal diet+6 g/kg Anion). No significant difference (p>0.05) was detected for average daily gain (ADG), average daily feed intake (ADFI) and gain/feed ratio (G/F) throughout the experiment, although dietary supplementation of Anion numerically increased these characteristics compared with CON. The dietary HCI group significantly (p<0.05) increased the coefficient of total tract apparent digestibility (CTTAD) of dry matter (DM), nitrogen (N) and energy compared with the CON group (p<0.05). No significant difference was observed in meat quality except that meat firmness was linearly (p<0.05) increased by the Anion supplementation, while an increased tendency in meat color and a decreased tendency in 2-thiobarbituric acid reactive substances (TBARS) was also observed (p<0.10). Anion supplementation linearly (p<0.05) decreased the fecal $NH_3$ compared with the CON group. However, dietary Anion supplementation at 3 g/kg decreased the $H_2S$ concentration compared with CON, while no significant difference was detected in the HCII group, although the $H_2S$ emission was numerically decreased compared with CON. In conclusion, supplementation of the diet with 3 g/kg Anion was found to exert a beneficial effect on nutrient digestibility and meat quality of growing-finishing pigs, and concomitantly decreased the noxious gas emission without negative effect on growth performance.

Effects of Hydrated Potato Starch on the Quality of Low-fat Ttoekgalbi (Korean Traditional Patty) Packaged in Modified Atmosphere Conditions during Storage

  • This study was carried out to investigate the effects of hydrated potato starch on the quality of low-fat ttoekgalbi (Korean traditional patty) packaged in modified atmosphere conditions during storage. The ttoekgalbi was prepared from 53.2% lean beef, 13.9% lean pork, 9.3% pork fat, and 23.6% other ingredients. Two low-fat ttoekgalbi treatments were prepared by substituting pork fat with hydrated potato starch; either by 50% fat replacement (50% FR) or 100% fat replacement (100% FR). Both 50% and 100% FR increased the moisture, crude protein, and decreased fat content, cooking loss, and hardness. For MAP studies, 200 g of ttoekgalbi were placed on the tray and filled with gas composed of 70% $O_2$: 30% $CO_2$ (70% $O_2$-MAP) and 30% $CO_2$: 70% $N_2$ (70% $N_2$-MAP), and were stored at $5^{\circ}C$ for 12 d. During the storage time, both 50% and 100% FR showed higher protein deterioration, while no differences were found in CIE $a^*$, CIE $L^*$, lipid oxidation, and bacterial counts in comparison to control. The ttoekgalbi with 70% $O_2$-MAP was more red, lighter in color, and showed higher TBARS values compared with 70% $N_2$-MAP. The meat with 70% $N_2$-MAP showed lower aerobic bacterial counts in control than those with 70% $O_2$-MAP. The lower anaerobic bacterial counts were observed only in 50% FR and 100% FR packed with 70% $N_2$-MAP in comparison with 70% $O_2$-MAP. In conclusion, the fat replacement with hydrated potato starch showed no negative effects on the quality of low fat ttoekgalbi during storage and 70% $N_2$-MAP was better than 70% $O_2$-MAP for low-fat ttoekgalbi packaging.

Effects of Supplemental Levels of Bazhen on Growth Performances, Serum Traits, Immunity, Meat Quality and Antioxidant Activity of Taiwan Country Chickens

  • One hundred and sixty Taiwan country chickens (d-old chicks) were randomly assigned into four groups with four replicates and equal sex. Basal diets were supplemented with 0, 0.5, 1 and 2% of Bazhen powder, a traditional Chinese herbal medicine complex. The study was conducted for 14 wks. Experimental results indicated that Bazhen supplement did not influence feed intake, body weight gain and feed:gain ratio. Compared with control group, the percentage of serum HDL (high-density lipoprotein) linearly increased (p<0.03) and that of VLDL+LDL (very low-density+low-density lipoprotein) linearly decreased (p<0.03) in Bazhen supplemented groups, that 2% Bazhen was significantly different with control group (p<0.05). Chickens fed diets containing 2% Bazhen displayed reduced (p<0.05) serum GOT (glutamic-oxaloacetic transaminase) levels. The IgG, ${\gamma}$-globulin levels and PHA (phytohemagglutinin) skin challenge results in 1% Bazhan supplemented group were higher (p<0.05) than in the control group, the SRBC (sheep red blood cell) and ND (newcastle disease) titers in Bazhen supplemented groups were linear higher (p<0.05) than in the control group. The liver catalase activity and the capacity of scavenging DPPH (${\alpha}$-${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl) radical were linearly increased (p<0.03) in Bazhen supplemented groups, and the 1 and 2% groups were different from the control group (p<0.05). Liver TBARS (thiobarbituric acid-reactive substances) levels in all Bazhen supplemented groups and total glutathione level in the 2% group were reduced (p<0.05) compared to the control group and displayed a linear response (p<0.05). The TBA (thiobarbituric acid) and pH value of the breast muscle after 24 h post-mortem in the Bazhen supplemented groups was linear lower (p<0.05) than in the control group. Results from this study demonstrated that Bazhen supplement in chicken had several beneficial effects, including increased SRBC and ND titers, HDL and IgG, ${\gamma}$-globulin levels, PHA skin challenge result, decreased VLDL+LDL and GOT levels, and displayed antioxidation effects in serum and carcass meat parameters.

Effects of dietary lycopene on the protection against oxidation of muscle and hepatic tissue in finishing pigs

  • Objective: The objective of this study was to evaluate the effect of different levels of lycopene supplementation on the carcass traits, meat quality, concentration of lipid oxidation products and antioxidant potential in the meat and liver of finishing barrows and gilts. Methods: A total of 40 barrows and 40 gilts were allotted in a completely randomized block design, arranged in a 2×5 factorial scheme, consisting of two sexes (barrows and gilts) and five dietary levels of lycopene (0, 12.5, 25.0, 37.5, and 50.0 mg/kg). In addition, four storage times (0, 24, 48, and 72 h), at 4℃, were added to the model to evaluate the longissimus lumborum muscle. Results: An interaction (p = 0.010) was observed between storage periods and dietary lycopene levels. The unfolding of the interaction (lycopene×period) showed a decreasing concentration of malondialdehyde concentration as the dietary lycopene increased, at all storage periods. No interactions (p>0.050) were observed for the 2,2 diphenyl 1 picrylhydrazyl (DPPH) in the pork. However, the percentage of DPPH radical inhibition reduced (p = 0.001) up to 72 h. Additionally, there was a linear increase (p = 0.001) in the capture of DPPH radicals by antioxidants, as the dietary lycopene increased. No interactions were observed (p>0.05) between the evaluated factors in liver. However, lipid oxidation was reduced by supplementing lycopene in pig diets. The capture of the DPPH radical, resulted increase in the antioxidant power exerted by lycopene in the liver (p = 0.001). The concentrations of the thiobarbituric acid reactive substances and DPPH in the liver were affected by sex (p = 0.001). Conclusion: Dietary supplementation of lycopene reduced the water loss during thawing and was effective in protecting against oxidation of the longissimus lumborum muscle and liver until 72 hours of storage, and the best results were obtained by supplementing with 50.0 mg of lycopene/kg of diet.
