Butein, a tetrahydroxychalcone, suppresses pro-inflammatory responses in HaCaT keratinocytes

  • Up-regulation of cell adhesion molecules and proinflammatory cytokines contributes to enhanced monocyte adhesiveness and infiltration into the skin, during the pathogenesis of various inflammatory skin diseases, including atopic dermatitis. In this study, we examined the anti-inflammatory effects of butein, a tetrahydroxychalcone, and its action mechanisms using TNF-α-stimulated keratinocytes. Butein significantly inhibited TNF-α-induced ICAM-I expression and monocyte adhesion in human keratinocyte cell line HaCaT. Butein also decreased TNF-α-induced pro-inflammatory mediators, such as IL-6, IP-10 and MCP-1, in HaCaT cells. Butein decreased TNF-α-induced ROS generation in a dose-dependent manner in HaCaT cells. In addition, treatment of HaCaT cells with butein suppressed TNF-α-induced MAPK activation. Furthermore, butein suppressed TNF-α-induced NF-kappaB activation. Overall, our results indicate that butein has immunomodulatory activities by inhibiting expression of proinflammatory mediators in keratinocytes. Therefore, butein may be used as a therapeutic agent for the treatment of inflammatory skin diseases. [BMB Reports 2015; 48(9): 495-500]

콩의 초기 흡습속도의 온도의존성 (Temperature Dependence of Initial Adsorption Rate of Soybean)

  • 콩을 상대습도 57-86%, 온도 $16^{\circ}-34^{\circ}C$에서 100시간 저장할 때, 콩의 초기홉습은 다음 식으로 표시되었다. log dw/dt=b log t+log a. 여기에서 w는 수분함량(%), t는 시간이다. 식으로부터 계산한 저장시간(t)에 따른 수분함량(w)은 실측치와 잘 일치하였다. 초기 흡습속도의 활성화에너지는 상대습도에 상관없이 15,500cal/g-mole 정도이었고 시료간에 큰 차이를 보이지 않았다. 저장온도 $16^{\circ}-28^{\circ}C$에서의 콩의 흡습속도는 다음과 같이 표시될 수 있었다. log(log dw/dt)=-15,500/2.303 RT.

  • 위치지정 변이기법을 이용하여 Bacillus thuringiensis var. kurstaki NRD 6의 내독소 유전자의 N말단 독성부위를 변이시켜 Bacillus thuringiensis var. kurstaki NRD 6-Stu 1을 육종하였다. 내독소에 대한 염기서열 조사결과, 변이주의 내독소 유전자부위에 Stu 1 인식부위가 생성되었고 특히, 이부위의 177번 염기 A가 C로 치환된 silent mutation이 일어났음을 확인하였다. 변이주의 내독소 유전자의 항원성질은 친주와 차이가 없었고 Choristoneura femiferana-1에 대한 독성도는 0.015~0.030ng으로 친주의 0.01~0.024ng과 유사하였으나 다른 친주계열인 B.t.kurstaki NRD 5의 500~1000ng과 B.t.kurstaki NRD 4(무독성)보다 강하였다.

COPRA MEAL AS A SUPPLEMENT TO CATTLE OFFERED A LOW QUALITY NATIVE PASTURE HAY

  • Twenty-four Hereford steers, 22 months old and a mean liveweight (${\pm}\;s.e.$) of $250\;{\pm}\;7\;kg$ were used in an experiment to evaluate over 42 days two rates of copra meal supplementation to cattle on a low N ($8.6\;{\pm}\;0.9$ g N/kg dry matter (DM)), low digestible ($45\;{\pm}\;5.2%$ DM) native pasture hay. Steers given the two rates (500, 1000 g/steer/day; i.e. 500C, 1000C) were compared to steers on a non-supplemental diet and to the effects on steers of supplemental urea (30g/steer/day; 30U) or with copra meal (500 g/steer/day; 500C.U), or of cottonseed meal (500 g/steer/day; 500S). Liveweight change was increased (P<0.01) by all of the supplements except by supplemental urea. The most effective treatment, 1000C, increased significantly (P<0.01) liveweight change (946 g/day) in steers above all supplements except those steers given 500C.U (718 g/day). Hay intake per unit liveweight was increased (P<0.05) by 7% by the 30U and 500C.U treatment, and by 9% by 500C; this group having the highest supplements, being greatest (P<0.05) for the 1000C group (6.0 g feed intake/g gain) and least for the 500S supplemented group (11.5 g/g gain). Efficiency was lowest (18.6 g/g gain) for the non-supplemented steers on the basal hay diet. Copra meal N was less degradable (i.e. 29%) in nylon bags over 15 hours in the rumen than was cottonseed meal N (37%), and rumen ammonia concentrations were lower (P<0.05) in cattle supplemented with copra meal (25, 27 mg N/L) than in cattle given urea (36 mg N/L) or cottonseed meal (39 mg N/L). It is concluded that copra meal at a daily rate of 500 g/head, and with rumen soluble nitrogen from urea, is an effective supplement for improving growth of cattle on a low quality forage.

박판 자성 재료를 이용한 전력 케이블 인근의 자기장 차폐 (Magnetic Shielding with Thin Magnetic Materials near Power Cables)

  • 본 연구에서는 자기장 차폐를 위하여 3상 전력 케이블을 얇은 자성 판재로 둘러싸는 방법을 제안한다. 두꺼운 상용 뮤-메탈, 방향성 및 무방향성 규소 강판을 출발 재료로 하여 두께 0.1 mm의 차폐재 3종류를 제조하였다. 3상 전류일 때, 차폐재 위치의 자기장이 100 ${\mu}T$ 정도이면 뮤-메탈이 (SF<0.1) 가장 효과적이었고, 500 ${\mu}T$ 이상 이면 규소 강판이 (SF 0.3${\sim}$0.4) 더 효과적이었다. 또한, 안쪽에 방향성 규소 강판, 바깥쪽에 뮤-메탈을 함께 둘러쌀 경우 500 ${\mu}T$까지도 SF 를 0.1 이하로 할 수 있었다. 한편, 단상 전류에서는 고 투자율 소재의 적용은 오히려 자기장을 증가시키는 결과를 보였다. 이상의 결과는 자기장 강도 H의 크기에 따라 각 소재의 투자율 우열이 서로 다른 점과 이로 인해 차폐재 내에 유도되는 자기장 벡터와 원래의 자기장 벡터의 상호 상쇄 및 중첩 작용으로 설명할 수 있었다.

  • $25{\circ}$에서 $[Co(en)]_3Cl_3,\;[Co(NH_3)_6Cl_3$수용액의 열역학적 해리상수를 1~2000bar의 압력범위에서 전도도법으로 측정하였다. 착물이 해리할 때 하전을 띤 이온이 생성하여 부피가 감소하므로 압력이 증가함에 따라 해리상수($K^T$)는 커졌다. 즉 $[Co(en)]_3Cl_3$에 대한 $pK^T$값은 1bar에서 2.16, 500bar에서 2.08, 1000bar에서 2.08, 1500bar에서 2.05, 2000bar에서 2.03이었고, $[Co(NH_3)_6Cl_3$$P^K^T$는 1bar에서 2.02, 500bar에서 1.96, 1000bar에서 1.90, 1500bar에서 1.88, 2000bar에서는 1.87이었다. 각 압력에서 Stokes반경과 해리상수($K^T$)값을 비교 분석하여 착물의 이온쌍 형성에는 정전기적 인력 이외에 Internal Conjugate Base(ICB)효과도 영향을 미쳤으며 이효과는 압력이 증가할 수록 커졌다.

순기제습방(順氣除濕方)이 고지방식이(高脂肪食餌)로 유발된 비만(肥滿) 백서(白鼠)에 미치는 영향 (The inhibitory effects of Soongijeseub-bang(順氣除濕方) on the obese-mice caused by high-fat diet)

  • Objective : In order to investigate the effects of Soongijeseub-bang(here in after referred to SJB) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high fat diet Methods : C57BL/6 mice were divided into three groups(normal, high fat diet with control, high fat diet with SJB extract) and fed for 15 weeks. And measured body weight change, the weight change of the adipocytes in body, the levels of ALT, AST, creatinine, total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, NEFA(non-esterified fat acid), glucose in serum and the expression of ${\beta}3AR$, leptin, and TNF- ${\alpha}$ gene in primary adipocytes and adipocytes tissue. Results : The following results were obtained in this study. 1. SJB 500 mg/kg extract group showed significant decrease in final weight. 2. SJB 500 mg/kg extract group showed significant decrease in the amount of adipocyte in weight. 3. All experimental group showed significant decrease in ALT and AST levels. 4. All experimetal group did not show significant decrease in glucose and creatinine levels. 5. All experimental group showed significant decrease in total cholesterol, LDL-cholesterol, triglyceride and NEFA levels. HDL-cholesterol were increase significantly in SJB 500 mg/kg extract group. 6. SJB 500 mg/kg extract group showed significant decrease in leptin levels. 7. All experimental group showed significantly increased expressions of fJ 3AR in primary adipose cell and 3T3-Ll cell, and those of leptin in primary adipose cell and 3T3-Ll cell were decreased significantly. SJB $100{\mu}l/ml$ extract group showed significant decrease in the expression of TNF-${\alpha}$. 8. SJB 500 mg/kg extract group showed decrease in the size of adipocyte in adipocytes tissue. 9. All experimental group showed significant decrease in adipose vacuoles in liver tissue.

식물 추출 정유가 어류의 병인체에 미치는 항미생물 효과 (Effect of Plant Extracted Essential Oil on Antimicrobial Activity for Fish Phathogens)

  • Antimicrobial activity of the essential oil extracted from plants Artemisia princeps var. orientalis, Thuja orientalis and Chamaecyparis obtusa were tested against selected pathogenic bacteria and fungi of fish. At the concentrations above 500ppm, ingibitory effect of the oil of A. princeps var. orientalis was at its peak against Aeromonas hydrophila ATCC 14715, A. hydrophila CF-2, A. salmonicida ATCC 14174 and A. salmonicida EL-1 but the bacteria Edwardsiella tarda KBF-1, Vibrio anguillarum ATCC19264, V. ordalii ATCC33509 and Streptococcus sp. were insensitive. Likewise, the oil extract of T. orientalis showed the highest inhibitory activity against V. ordalii ATCC33509, E. tarda ECK-1, and E. tarda KBF-1 at 300ppm; however the activity was highest at 500ppm or A. hydrophila ATCC14715, A. hydrophila CF-2, A. salomonicida ATCC14174, A. salmonicida EL-1 and Streptococcus sp. SF-1. With increasing dose of C. obtusa oil, the inhibitory activity became more and more effective against A. hydrophila CF-2, A. salomonicida ATCC14174, E. tarda ECK-1 and Streptococcus sp. SF-1, but A. hydrophila ATCC14174, A. salmonicida EL-1, E. tarda KBF-1, V. anguillarum ATCC19264, V. ardalii ATCC33509 and gram positive bacteria (Streptococcus sp.) were somewhat resistant. A. princeps var. orientalis, T. orientalis and C. obtusa were also tested against Saprolegnia sp. at the oil concentrations of 10, 100, 500, 1,000, 1,500 and 2,000ppm. The inhibitory effect of the oil on the inhibit the mycelial growth of Saprolegnia sp. at 10ppm and completely inhibited at over 500ppm.
