시호가 PTU (6-n-propyl-2-thiouracil)로 유발된 Rat 갑상샘 기능저하증에 미치는 영향 (Effects of Bupleuri Radix on the Rat Hypothyroidism Induced by PTU (6-n-propyl-2-thiouracil))

  • The object of this study was to evaluate the effect of Bupleuri Radix, aqueous extracts of the root part of Bupleurum falcatum on the 6-n-propyl-2-thiouracil (PTU)-induced rat hypothyroidism. Aqueous extracts of Bupleuri Radix (BR; yield = 11.73%) were administered, once day for 42 days from 2 weeks before start of PTU treatment as an oral dose of 300 and 150 mg/kg (body weight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10 mg/kg for 28 days. The changes on the body weight, thyroid gland weights, serum thyroid hormone - thyroid stimulating hormone (TSH), triiodothyronine ($T_3$) and thyroxine ($T_4$), serum lipid profiles - total cholesterol, low density lipoprotein (LDL), high density lipoprotein (HDL) and triglyceride, liver antioxidant defense system - lipid peroxidation, $H_2O_2$, superoxide dismutase (SOD) and catalase (CAT), serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were observed with histopathology of thyroid glands. Results were compared with $LevoT_4$ 0.5 mg/kg treated rats. As results of PTU treatment, marked decreases of body weights, triglyceride contents, liver CAT activities and changes of serum thyroid hormone levels were observed with increases of serum AST, HDL contents, liver $H_2O_2$ and SOD activities and thyroid gland weight. In addition, marked hyperplasia of follicular cells with decreases of follicular colloid contents and sizes were demonstrated at histopathological inspections. However, these PTU induced hypothyroidism were dose-dependently inhibited by treatment of BR extracts, and BR extracts effectively regulated the hypothyroidism related changes on the antioxidant defense system. The results obtained in this study suggest that BR extracts have favorable effects on the thyroid hormone productions with beneficial effects on the hypothyroidism mediated by the modulatory effects on the antioxidant defense system.

PTU로 유발된 흰쥐의 갑상선 기능저하증에 미치는 십전대보탕(十全大補湯)의 효과 (Effects of Sipjeondaebotang on the white rat Hypothyroidism Induced by PTU(6-n-propyl-2-thiouracil))

  • Objectives : This study was to evaluate Sibjeondaebo-tang’s(Shiquan dabutang) effect to rat hypothyroidism which induced by 6-n-propyl-2-thiouracil(PTU)Methods : Aqueous extracts of Sibjeondaebo-tang(Shiquan dabutang)(21.29%) were administered by oral dose of 300 and 150 mg/kg(rat’s body weight) for 42 days from 2 weeks before start of PTU treatment. During this moment the body weight, thyroid gland weights, liver weight, serum thyroid hormone, lipid content and liver antioxidant defense system were measured with histopathological changes of thyroid glands, Liver and these result were compared with LevoT4 0.5mg/kg treated ratsResults : As a result of PTU treatment, marked decreases of body weight, liver weight, serum thyroid hormone levels of T3, T4, triglyceride content and liver CAT activation were observed with marked increases of thyroid gland weight, thyroid gland hormone TSH, activate liver H2O2 and SOD, serum Total cholesterol, HDL and AST amount were admitted. In addition, marked hyperplasia of follicular cells with reduce of follicular colloid contents and sizes were showed at histopathological inspections. These PTU-induces histopathological changes related to hypothyroidism were significantly reduced by treatment of both different dosages of Sibjeondaebo-tang(Shiquan dabutang).Conclusions: This study suggest that Sibjeondaebo-tang(Shiquan dabutang) extracts have favorable effect on the thyroid hormone productions with beneficial effect on hypothyroidism medicated by modulatory effects on antioxidant defense system.

여성 성전환수술을 받은 환자의 호르몬 정량분석 (Hormonal Analysis of Female Transgender Patients Performed Gender Reassignment Operation)

  • Transgender is the severe type of gender identity disorder. The prevalence rate of transgender is reported to occur to about 1 out of 50,000 men, and about 1 out of 10,000 women. As for Korea, it is estimated to have about 1400 transgender patients. Lately, not only the numbers of them are increasing but also they are influencing our society increasingly. As for female transgender patients, they take female hormone for a long term before and even after the operation to maintain their physical identity of female. We have analyzed insulin like growth factor-1(IGF-1), insulin like growth factor protein binding-3(IGFBP-3), female hormone, male hormone and thyroid hormone in female transgender patients who have undergone the gender reassignment operation. We examined the changes of hormone level due to having female hormone steadily, and also examined how the steady use of the hormone could affect body organs. As for IGF-1, it showed significantly low in the female transgender group compared to control ($319.30{\pm}37.4$ vs $539{\pm}55.0$, p<0.05). As for IGFBP-3, there was no significant difference ($2859{\pm}200.3$ vs $2607{\pm}262.5$, p>0.05). As for female hormone, there was no significant differences in FSH($13.42{\pm}13.8$ vs $8.95{\pm}3.5$, p>0.05), estradiol($104.41{\pm}97.1$ vs $121.68{\pm}60.2$, p>0.05), and LH($7.62{\pm}5.6$ vs $7.4{\pm}3.3$, p>0.05). Even in comparison of testosterone, there was no significant differences($0.23{\pm}0.09$ vs $0.33{\pm}1.33$, p>0.05). As for thyroid hormone, there was no significant differences in TSH and free T4($1.34{\pm}0.94$ vs $1.71{\pm}0.12$, $1.4{\pm}0.37$ vs $1.46{\pm}0.17$, p>0.05). Therefore, this study concludes that apart from the decreased level of IGF-1, the possible endocrine side-effect problem due to female hormone seems to be low because there was no differences of female, male, and thyroid hormone level compared with normal female. Further study will be required in metabolic change including bone metabolism occurred by decrease level of IGF-I.

  • Rhizoma Rehmanniae Preparata is a Chinese herbal tonic. The hemopoietic effects of Rhizoma Rehmanniae Preparata(4-time steamed Jihwang, 4-time steamed double dose Jihwang, 8-time steamed Jihwang, 8-time steamed double dose Jihwang) were examined using in vitro rat(SD) model. Cyclophosphamide(150 mg/kg) was injected into experimental groups and control group to induce bone marrow suppression. Oral administration of Rhizoma Rehmanniae Preparata suppressed hormone levels of $T_4\;and\;T_3$. Reticulocyte count was increased by the bone marrow suppression.

Expression of Recombinant Human Growth Hormone in a Soluble Form in Escherichia coli by Slowing Down the Protein Synthesis Rate

  • Formation of inclusion bodies is usually observed when foreign proteins are overexpressed in E. coli. The formation of inclusion bodies might be prevented by lowering the rate of protein synthesis, and appropriate regulation of the protein expression rate may lead to the soluble expression. In this study, human growth hormone (rhGH) was expressed in a soluble form by slowing down the protein synthesis rate, which was controlled in the transcriptional and translational levels. The transcriptional level was controlled by the regulation of the amount of RNA polymerase specific to the promoter in front of the rhGH gene. For lowering the rate of translation, the T7 transcription terminator-deleted vector was used to synthesize the longer mRNA of the target gene because the longer mRNA is expected to reduce the availability of tree ribosomes. In both methods, the percentage of soluble expression increased when the expression rate slowed down, and more than 93% of rhGH expressed was a soluble form in the T7 transcription terminator-deleted expression system.

호르몬 대체요법에 따른 갱년기여성의 갱년증상과 삶의 질 (The Climacteric Symptoms and Quality of Life in Climacteric Women according to Hormone Replacement Therapy)

  • The purpose of this study is to find the difference of the climacteric symptoms and quality of life according to hormone replacement therapy (HRT) in climacteric women. The research design was a descriptive survey with questionaries. A convenience sample of 181 climacteric women aged 45-65 who had climacteric symptoms were selected in Kwangju city, Korea. Sixty-six climacteric women among those subjects were receiving HRT at K university hospital in Kwangju city. Data were collected by the interview with questionaries from Feb 3. 1999 to March 25, 1999. Climacteric symptoms were measured using scores developed by Neugarten(1965) and modified by Park(1989), and the quality of life scale related to climacteric symptoms developed by Hildtich(1996) and modified Kim(1998). Data were analyzed by $x^2$-test, t-test, ANOVA and Pearson's correlation procedures, using SAS/win PC+. The results were as follows ; 1. No statistically significant differences were found between the receiving HRT group and not receiving HRT group in demographic, obstetrical and health related characteristics. 2. There were no significant difference on the climacteric symptoms between the group of women receiving HRT (score 2.4) and the group of women who were not (score 2.6) except psychological area (t= 2.407, p= 0.017). 3. The quality of life of the group of women receiving HRT was significantly higher than that the group of women who were not (t=2.151, p=.032). 4. The quality of life of the group of women receiving HRT were high scores There were significantly high of the quality of life in the vaso-motor change area (t=2.634, p= 0.009), psycho-social change area (t=3.239, p=0.001), and physical change area (t=2.031, p=0,043) in the group of receiving HRT compared to not receiving HRT group. 5. The variables showed significantly differences on the degree of climacteric symptoms of the group of women not receiving HRT were the subjective health state (t=15.81, p=0.000), age (t=7.50, p=0.007), feeling of menopause (t=30.88, p=0,000) and climacteric periods (t=8.66, p=0.003), and receiving HRT were number of para (t=3.95, p=0.050) and feeling of menopause (t=3.94, p=0.050). 6. The variables showed significantly difference on the quality of life of the group of women not receiving HRT were the subjective health state (t=4.14, p=0.044) and feeling of menopause (t=10.86, p=0.001). 7. There were significantly positive correlations between the climacteric symptoms and the quality of life in climacteric women (r=0.512, p=0.000).

산란노계에서 강제환우가 갑상선, 난소, 부신 및 호르몬 수준에 미치는 영향 (Effect of Induced molting on the Relative Weights and Hormone Levels of Thyroid, Ovary, and Adrenal Glands in Spent Laying Hens)

  • 본 실험은 산란노계에서 강제환우가 갑상선, 난소, 부신의 무게 및 이들 기관에서 분비되는 호르몬 수준의 변화에 미치는 영향을 구명하고자 시행하였다. 77주령 산란계 360수를 10수씩 36개의 펜에 배치하여 7주간 강제환우를 시행하였다. 사료는 단백질 수준이 16%이며 대사에너지가 2,800kca1/kg 함유된 시판용으로 84주령부터 126주까지 급여하였다. 강제환우전, 산란율 5%, 산란피크 및 실험 종료 시에 각각 36수씩 희생시켜 채혈을 하였으며, 갑상선, 난소, 부신의 무게를 측정하였고 혈청에서 T3, T4, cortisol 및 estradiol 함량을 측정하였다. 갑상선의 무게는 산란율이 5%에 도달하였을 때 제일 높게 나타났다 난소의 무게는 강제환우 후에 현저하게 감소되었지만 그 후에 지속적으로 증대되어 실험 종료 시에 극대화 되었다(P<0.05). 산란율이 5%에 도달한 시기에 혈청의 T3, T4 및 cortisol 수준은 다른 시기에 비하여 높게 나타났지만 실험 종료 시에 제일 낮게 나타났다. 혈청의 estradiol은 산란율이 5%에 도달한 시기에 제일 낮았다. 본 실험에서 강제환우 과정에서 환우의 유기에 주요 역할을 하는 내분비 기관 및 이들 기관에서 분비되는 T3, T4, cortisol, estradiol 수준이 복합적으로 관련될 수 있음을 시사하였다.

임신 중 바이러스성 감염요인과 갑상선 호르몬의 상관성 (Relationship between the Thyroid Hormone and Viral Infections in Pregnancy)

  • 임신은 갑상선 기능 검사의 중요한 해석을 필요로 하며 임신 중 갑상선 기능 이상과 외부 바이러스성 감염 인자들의 항체의 존재는 태아 및 산모의 건강에 영향을 미치기에 임신에서 갑상선 기능의 선별적 평가가 요구된다. 본 연구에서는 임신기간 동안 정상 산모들의 선택적 산전 감염인자 검사 항목 중에서 갑상선 관련 인자와 바이러스성 감염 인자의 임신시기별 상호 연관성을 알아보고자 하는 후향적 단편 실태조사이다. 분석한 결과를 살펴보면, T3는 나이가 증가함에 따라 감소하고, 특히 HCV가 양성인 그룹에서 양의 유의성을 보였다(P<0.01). 또한 HIV가 음성이지만 임계치에 근접하거나 쌍둥이 임산부에서는 FT4가 유의한 증가를 나타냈다(P<0.05). TSH는 30대 연령에서 높게 분포하였으며, 다른 바이러스성 감염인자와는 통계적 유의성이 나타나지 않았다. 또한, TSH의 결과 값을 삼분위로 나누어 분석한 결과, FT4와 T3은 양의 상관성을 보였으나 TSH와는 음의 상관성을 보였다(P<0.05). 따라서 본 연구를 통해서 임신 중 산전검사인 갑상선 검사와 바이러스성 감염인자의 검사를 통한 임신 중 평가는 임신 경과시간, 감염인자의 노출상태 및 정량적 수치의 상태를 반영하여 이루어져야 할 것이며, 갑상선 관련 내분비 인자에 대한 산전검사의 유용성에 대한 평가의 보완이 이루어져야 할 것으로 사료된다.

마치현 추출물이 3T3-L1 지방세포에서 지방 분해 및 Hormone Sensitive Lipase (HSL) 유전자 발현에 미치는 효과 (Effects of Portulaca Oleracea L. Extract on Lipolysis and Hormone Sensitive Lipase (HSL) Gene Expression in 3T3-L1 Adipocytes)

  • This study was performed to investigate the lipolytic effects of Portulaca oleracea L. extract in 3T3-L1 adipocytes. The Portulaca oleracea L. was extracted with extrusion method using twin-screw extruder under $58{\sim}60rpm$ screw speed, $4{\sim}5kg/hr$ feed rate, $140^{\circ}C$ extrusion temperature. The lipolytic action of Portulaca oleracea L. extract was estimated by measuring the amount of glycerol and free fatty acids (FFA) released from 3T3-L1 adipocytes and by measuring the cellular lipid content in 3T3-L1 adipocytes. The hormone sensitive lipase (HSL) mRNA level was analyzed using quantitative real-time PCR. The Portulaca oleracea L. extract at 1 to $100{\mu}g/ml$ suppressed lipid accumulation. The release of glycerol and FFA into the medium, and the mRNA level of HSL were significantly increased by the addition of Portulaca oleracea L. extract at dose-dependent manner. In conclusion, the Portulaca oleracea L. extract was suggested to have the lipolytic effect through release of lipolytic products (FFA and glycerol) of triacylglyceride to the culture medium and suppression of lipid accumulation via up-regulation of HSL gene expression in 3T3-L1 adipocytes.

Aqueous extracts of Lonicerae Flos (LF yield = 23.80%) were administered, once day for 42 days from 2 weeks before start of PTU treatment as an oral dose of 500 and 250 mg/kg (body weight), and hypothyroidism was induced by daily subcutaneous treatment of PTU 10 mg/kg for 28 days. The changes on the body weight, thyroid gland weights, serum thyroid hormone - thyroid stimulating hormone (TSH), tri-iodothyronine ($T_3$) and thyroxine ($T_4$), serum lipid profiles - total cholesterol, low density lipoprotein (LDL), high density lipoprotein (HDL) and triglyceride were observed with liver antioxidant defense system - lipid peroxidation, $H_2O_2$, superoxide dismutase (SOD) and catalase (CAT) and serum asparte aminotransferase (AST) and alanine aminotransferase (ALT) analysis. Results were compared with Levothyroxine ($LT_4$) 0.5 mg/kg treated rats. As results of PTU treatment, marked decreases of body weights, serum thyroid hormone levels and triglyceride contents, liver $H_2O_2_3$ and SOD activities were observed with increases of serum AST and HDL contents, liver CAT activities, thyroid gland weight. However, these PTU induced hypothyroidism were dose-dependently inhibited by treatment of LF extracts, and LF extracts effectively regulated the hypothyroidism related changes on the antioxidant defense system. The results obtained in this study suggest that LF extracts have favorable effects on the thyroid hormone productions with beneficial effects on the hypothyroidism mediated by the modulatory effects on the antioxidant defense system.
