MSCs-Derived miR-150-5p-Expressing Exosomes Promote Skin Wound Healing by Activating PI3K/AKT Pathway through PTEN

  • Background and Objectives: The goal of this study was to investigate the mechanism of mesenchymal stem cell (MSC)-derived microRNA (miR)-150-5p-expressing exosomes in promoting skin wound healing through activating PI3K/AKT pathway by PTEN. Methods and Results: Human umbilical cord (HUC)-MSCs were infected with miR-150-5p overexpression and its control lentivirus, and HUC-MSCs-derived exosomes (MSCs-Exos) with stable expression of miR-150-5p were obtained. HaCaT cells were induced by H2O2 to establish a cellular model of skin injury, in which the expression of miR-150-5p and PTEN and the phosphorylation of PI3K and AKT were evaluated. HaCaT cells were transfected with pcDNA3.1-PTEN or pcDNA3.1 and then cultured with normal exosomes or exosomes stably expressing miR-150-5p. Cell proliferation was inspected by CCK-8. Cell migration was detected by scratch test and cell apoptosis by flow cytometry. The starBase tool was used to predict the binding site of miR-150-5p to PTEN. Dual-luciferase reporter assay and RIP assay were applied to assess the interaction between miR-150-5p and PTEN. In H2O2-induced HaCaT cells, the miR-150-5p expression decreased, and PTEN expression increased in a concentration-dependent manner. MSCs-Exos promoted the growth and migration of H2O2-induced HaCaT cells and inhibited their apoptosis. In addition, overexpression of exosomal miR-150-5p enhanced the protective effect of MSCs-Exos on H2O2-induced HaCaT cells; PTEN overexpression in HaCaT cells partially restrained miR-150-5p-mediated inhibition on H2O2-induced injury in HaCaT cells. PTEN was a target gene of miR-150-5p. MiR-150-5p regulated PI3K/AKT pathway through PTEN. Conclusions: MSCs-derived miR-150-5p-expressing exosomes promote skin wound healing by activating PI3K/AKT pathway through PTEN.

Effects of the Educational Program for Patients of T.K.R.A. (슬관절 치환술 환자의 교육 프로그램 효과에 관한 연구)

  • This study is aimed to evaluate the effects of the educational program that is designed to help the patients of T.K.R.A. to improve their self-nursing ability. The research will show how well this program can contribute to raising of their adaptability of everyday life by decreasing the difficulties that the patients had before in their daily life. The sample persons for this research were 40 adult patients who had T.K.R.A. in our hospital, and the data were collected through the questionnaires which were given to them. For the surveying tool, Jette's scale was applied to measure the degree of uncomforting that the patients experienced in their daily life. The materials of report had been gathered from May 1st to December 10th. The collected data were analyzed with real numbers, percentage, average, the standard deviation, TorF test and Pearsons correlation. The results of this study can be summarized like follows: The survey of self-caring exercise part were processed with two groups containing experiment group and contrast group. The exercise which took the highest points in experiment group was repetition of stretching and bending knees with laid down position while taking a walk, repetition of stretching and bending knees with seated on chair position and repetition of stretching and bending knees with laid down position got high points in contrast group. There were distinctive differences in statistics between repetition of stretching and bending knees in laid down position and taking a walk. There was no significant statistical gap between the degrees of inconveniences of daily life that each two groups experienced when we compared the average of the two groups, but 12 out of 20 questions got considerable differences. According to common characters of self-caring, there was a distinctive gap in the group who left the hospital in 14days and the ones who spend more than 30 minutes with exercise. The patient group who left the hospital in fourteen days after T.K.R.A. spent more time doing self-caring exercise. In the relationship between common characters and the degree of difficulties of daily life, Exercising is effective and Spending more that 30 minutes for exercise showed visible differences. The patients who answered that exercise are effective and put more time for it experienced less pain in their everyday life. The data proved by Pearson's correlation showed the relationship between self- caring and the degree of pain of daily life. According to the result, the increasing of self-caring activities helped to down the degree of inconveniences of daily life. This data show that T.K.R.A. reduced self-caring exercise ability and daily life activities. Therefore, the medical staffs who would take care the T.K.R.A. patients should offer stable nursing through well scheduled and education materials for patients. It is also important to encourage the patients to participate to the exercise by showing them how exercise after T.K.R.A. The education program is expecting to guarantee not only the right of patients to get enough information but also effective result to decrease uncomfortable of daily life.

  • A simple system to measure the correlated color temperature ($T_c$) and color rendering index ($R_a$) oflight sources was developed. And the T, and R, of incandescent lamps (I/L) and various fluorescent lamps(F/L) were measured with varying source voltage and power frequency. As results, $T_c$ variations of I/L and F/L with varying source voltage were relatively large, but $R_a$, variations were small. And $T_c$, $R_a$, variations of F/L under 5OkHz of power frequency were small, but over 50kHz, it is supposed that $T_c$, and $R_a$, will vary significantly.

Gluconacetobacter persimmonis sp. nov., Isolated from Korean Traditional Persimmon Vinegar

  • Screening was performed to isolate cellulose-producing microorganisms from the Korean traditional fermented persimmon vinegar. The resulting strain, KJ $145^{T}$, was then taxonomically investigated by phenotypic characterization, particularly chemotaxonomic, and by phylogenetic inference based on a 16S rDNA sequence analysis including other related taxa. Strain KJ $145^{T}$ was found to grow rapidly and form pale white colonies with smooth to rough surfaces on a GYC agar. Strain KJ $145^T$ also produced acetate from ethanol, and was tolerable to 10% ethanol in SM medium. In a static culture, a thick cellulose pellicle was produced, and in GYC broth, the strain grew at temperatures ranging from 28 to $40^\circ{C}$ with an optimum pH of 4.0. The genomic DNA G+C content of strain KJ $145^T$ was 61.9 mol%, and the predominant ubiquinone was Q 10 as the major quinone and Q9 as the minor quinone. The major cellular fatty acids were $C_{16:0}$ and the sum in feature 7 ($C_{18:1}$ w9c, w12t and/or w7c). A 16S rRNA-targeted oligonucleotide probe specific for strain KJ $145^T$was constructed, and the phylogenetic position of the new species was derived from a 16S rDNA-based tree. When comparing the 16S rDNA nucleotide sequences, strain KJ $145^T$ was found to be most closely related to G. hansenii LMG $1527^T$ (99.2%), although KJ $145^T$ was still distinct from G. hansenii LMG $l527^T$ and G. xylinus LMG $1515^T$ in certain phenotypic characteristics. Therefore, on the basis of 16S rDNA sequences and taxonomic characteristics, it is proposed that strain KJ $145^T$ should be placed in the genus Gluconacetobacter as a new species, Gluconacetobacter persimmonis sp. nov., under the type-strain KJ $145^T$ (=KCTC =$10175BP^T$=KCCM=$10354^T$).

  • We determine the largest integer i such that $0 and the coefficient of $t^{i}$ is odd in the polynomial $(1+t+t^{2}+{\cdots}+t^{n})^{n+1}$. We apply this to prove that the co-index of the tangent bundle over $FP^{n}$ is stable if $2^{r}{\leq}n<2^{r}+\frac{1}{3}(2^{r}-2)$ for some integer r.

  • Algebraic matrix Riccati equations of the form, FP+PF$^{T}$ -PRP+Q=0. are analyzed with reference to the stability of closed-loop system F-PR. Here F, R and Q are n * n real matrices with R=R$^{T}$ and Q=Q$^{T}$ .geq.0 (nonnegative-definite). Such equations have been playing key roles in optimal control and filtering problems with R .geq. 0. and also in the solutions of in H$_{\infty}$ control problems with R taking the form R=H$_{1}$$^{T}$ H$_{1}$-H$_{2}$$^{T}$ H$_{2}$. In both cases an existence of stabilizing solution, i.e. the solution yielding asymptotically stable closed-loop system, is an important problem. First, we briefly review the typical results when R is of definite form, namely either R .geq. 0 as in LQG problems or R .leq. 0. They constitute two extrence cases of Riccati to the cases H$_{2}$=0 and H$_{1}$=0. Necessary and sufficient conditions are shown for the existence of nonnegative-definite or positive-definite stabilizing solution. Secondly, we focus our attention on more general case where R is only assumed to be symmetric, which obviously includes the case for H$_{\infty}$ control problems. Here, necessary conditions are established for the existence of nonnegative-definite or positive-definite stabilizing solutions. The results are established by employing consistently the so-called algebraic method based on an eigenvalue problem of a Hamiltonian matrix.x.ix.x.

  • This work is concerned with oscillation of the second order sublinear neutral delay dynamic equations of the form $$\(r(t)\;\((y(t)+p(t)y(a(t)))^{\Delta}\)^{\gamma}\)^{\Delta}+q(t)y^{\gamma}({\beta}(t))=0$$ on a time scale $\mathcal{T}$ by means of Riccati transformation technique, under the assumptions $\int^{\infty}_{t_0}\(\frac{1}{r(t)}\)^{\frac{1}{\gamma}}$ ${\Delta}t={\infty}$ and $\int^{\infty}_{t_0}\(\frac{1}{r(t)}\)^{\frac{1}{\gamma}}$ ${\Delta}t$ < ${\infty}$, where 0 < ${\gamma}{\leq}1$ is a quotient of odd positive integers.

The Molecular Weight Dependance of Paramagnetic Gd-chelates on T1 and T2 Relaxation Times (상자성 복합체의 분자량에 따른 T1 및 T2 자기이완시간에 관한 연구)

  • To evaluate the T1, T2 magnetic relaxation properties of water molecule according to molecular weight of paramagnetic complex. 4-aminomethyicyclohexane carboxylic acid (0.63 g, 4 mmol) was mixed with the suspension solution of DMF (15 ml) and DTPA-bis-anhydride (0.71 g, 2 mmol) to synthesize the ligand. The ligand was then mixed with $Gd_2O_3$ (0.18 g, 0.5 mmol) to synthesize Gd-chelate. For the measurement of magnetic relaxivity of paramagnetic compounds, the compounds were diluted to 1 mM and then the relaxation times were measured at 1.57 (64 MHz). Inversion-recovery pulse sequence was employed for T1 relaxation measurement and CPMG (Carr-Purcell-Meiboon-Gill) pulse sequence was employed for T2 relaxation measurement. In case of inversion recovery sequence, total 35 images with different inversion time(T1)s ranging from 50 msec to 1,750 msec. To estimate the relaxation times, the signal intensity of each sample was measured using region of Interest (ROI) and then fitted by non-linear least square method to yield T1, T2 relaxation times and also R1 and R2. Compared to T1=($205.1{\pm}2.57$) msec and T2=($209.4{\pm}4.28$) msec of Omniscan (Gadodiamide), which is commercially available paramagnetic MR agent, T1 and T2 values of new paramagnetic complexes were reduced along with their molecular weight. That is, T1 value was ranged from $(96.35{\pm}2.04)\;to\;(79.38{\pm}1.55)$ msec and T2 value was ranged from $(91.02{\pm}2.08)\;to\;(76.66{\pm}1.84)$ msec. Among new paramagnetic complexes, there is a tendency that the R1 and R2 increase as the molecular weight is increases. As molecular weight of paramagnetic complex increases, T1 and T2 relaxation times reduce and thus the increase of relaxivity (R1 and R2) Is proportional to molecular weight.

Determination of Rheological Properties of Surimi Gels and Imitation Crab-leg Products by Stress-Relaxation Test (시판 어묵 및 게맛살의 변형력완화 실험을 통한 유변학적 특성)

  • The purpose of this study was to investigate the rheological properties of surimi gels and imitation crab-leg products by stress-relaxation test and to examine the correlations between stress-relaxation parameters and T.P.A. parameters. The linear viscoelasticity of surimi gels and imitation crab-leg products was observed in the range of the strain of $5{\sim}20%$ at cross-head speed 2.4 mm/sec. The average tensile forces of surimi gels and imitation crab-leg products were similar, 370.4 g and 436.4 g, respectively, but surimi gels showed higher relaxation time and viscous component (17256.1 sec, $1.357{\times}10^{10}$ poise) than those of imitation crab-leg products (6110 sec, $0.519^{\ast}10^{10}$ poise). Estimated tensile force of each exponential term in relaxation test was highly related with hardness, gumminess and chewiness of T.P.A (r=0.93, 0.93, 0.95, p<0.01), the relaxation time of each exponential term was rrelated with cohesiveness (r=0.89, p<0.01) of T.P,A. and the elastic component of exponential term with gumminess, chewiness and hardness (r=0.92, 0.94, 0.93. p<0.01) of T.P.A.. The viscous component of exponential term was related with cohesiveness (r=0.83, p<0.05) of T.P.A.. The degree of texturization was negatively related with the relaxation time and viscous component (r=-0.92, -0.96, p<0.01).
