Influence of Physiological and Environmental Factors on Growth and Sporulation of an Antagonistic Strain of Trichoderma viride RSR 7

  • Influence of physiological and environmental factors on an antagonistic strain of Trichoderma viride RSR7 were studied optimize its biocontrol potential. The growth and sporulation of T. viride was greatly influenced by various carbon and nitrogen sources, and the environmental factors such as pH and temperature. The best growth and sporulation of T. viride was observed when sucrose, peptone and trehalose were supplemented in the medium as sole carbon sources. Rhamnose, pyruvic acid and sorbitol also supported a good growth. However, with these carbon sources the sporulation was poor. Growth and sporulation was also affected by various nitrogen sources. Growth and sporulation both were favoured by ammonium forms of nitrogen compared to nitrite or nitrate forms. Urea did not support either growth or sporulation. Among amino acids, glutamic acid, asparagine, leucine, aspartic acid, glutamic acid and alanine supported good growth as well as sporulation. T. viride was able to utilize large number of amino acids as sole nitrogen source. Proline was good for growth, but not for sporulation. Maximum growth and sporulation of T. viride was between pH 4.5 to 5.5. Temperatures between $20^{\circ}C\;and\;37^{\circ}C$ were good for both growth and sporulation of T. viride. At lower temperatures(i.e. below $20^{\circ}C$) growth and sporulation were inhibited. Based on the present study it may be concluded that T. viride RSR7 is capable of growing and sporulating with varied nutritional and environmental conditions and, therefore, this strain of T. viride may be useful as a biocontrol agent under diverse physiological and environmental conditions.

Combined Application of Pseudomonas fluorescens and Trichoderma viride has an Improved Biocontrol Activity Against Stem Rot in Groundnut

  • In an attempt to develop effective biocontrol system for management of stem rot disease in groundnut, 57 bacterial isolates and 13 isolates of Trichoderma spp. were evaluated for their antagonistic activity against Sclerotium rolfsii. The antagonists were selected based on their ability to inhibit the external growth of S. rolfsii from infected groundnut seeds. Four isolates of Pseudomonas fluorescens, GB 4, GB 8, GB 10 and GB 27, and T. viride pq 1 were identified as potent antagonists of S. rolfsii. T. viride pq 1 produced extracellular chitinase and parasitized the mycelium of S. rolfsii. Under controlled environment conditions, P. fluorescens GB 10, GB 27, T. viride pq 1 and the systemic fungicide Thiram(equation omitted) reduced the mortality of S. rolfsii inoculated to groundnut seedlings by 58.0%, 55.9%, 70.0% and 25.9%, respectively compared to control. In vitro growth of P. fluorescens GB 10 and GB 27 was compatible with T. viride pq 1 and Thiram(equation omitted). Integrated use of these two bacterial isolates with T. viride pq 1 or Thiram(equation omitted) improved their biocontrol efficacy. Combined application of either GB 10 or GB 27 with T. viride pq 1 was significantly effective than that with Thiram(equation omitted) in protecting groundnut seedlings from stem rot infection.

Trichoderma viride 및 Trichoderma reesei 유래 Cellulase의 키토산 분해 특성 (Chitosanolytic Characteristics of Cellulases from Trichoderma viride and Trichoderma reesei)

  • 키토산 올리고당 생산에 이용 가능한 경제성 있는 키토산 분해 효소를 탐색하였다. Cellulase, ${\beta}-1,4-glucosidase,\;protease$ 및 상업용 효소류를 대상으로 조제 키토산에 대한 분해력을 시험한 결과 Trichoderma viride, T. reesei 유래의 cellulase 및 상업용 효소인 Celluclast (T. reesei 유래 cellulase)가 우수한 키토산 분해능력을 나타내었다. 키토산에 대한 반응성은 T, reesei 유래 cellulase가 T. viride 유래 cellulase 보다 강하였으며 이들의 적정 키토산 분해조건은 pH 5.0이었고 적정 온도는 T. viride 유래 celluclast는 $45^{\circ}C$, T. reesei 유래 cellulase 및 Celluclast는 $55^{\circ}C$로 나타났다. 또한 dose reponse시험에서 이들 효소 모두 enz./chitosan비=0.1이 적절하였으며 T. reesei 유래 효소 및 Celluclast는 키토산 농도가 3%일 때 최대의 활성을 나타내었다. 한편, 상업용 효소인 Celluclast를 이용한 키토산 올리고당의 생산 가능성을 시험하기 위하여 3% 키토산 용액(310cp)에 Celluclast를 1% 첨가하고 $pH\;5.0,\;55^{\circ}C$에서 시간별로 반응시켰을 때, 점도는 초기30분경에 5.51cp로 98% 이상 감소하였으며 50% ethanol 가용물의 수율은 분해 15시간에 70%로 최대를 보였다. 총환원당의 함량은 분해시간이 경과함에 따라 증가하였고 반응 2시간부터 13.5% 내외의 수준을 유지하였다. 올리고당의 조성은 15시간 분해물에서 균일한 분포와 높은 함량을 보였으며 항종 양활성을 나타내는 것으로 알려진 6량체 키토산 올리고당의 생성량은 기존의 산 분해방법의 약 4배에 해당되는 8.0%로 나타났다. 따라서 T. viride 및 T. ressei 유래의 cellulase는 99% 이상의 탈아세틸화도를 가지는 키토산에 대하여 분해활성을 나타내고 특히 T. reesei유래의 상업용효소는 지금까지 효소가격이 너무 높아 활용되지 못한 chitosanase를 대신하여 키토산 올리고당을 경제성 높게 생산하는 데 이용될 수 있을 것으로 기대되었다.

Trichoderma viride와 Penicillium funiculosum Cellulase 성분효소 간의 상승작용에 관한 연구 (Cross-Synergistic Interactions between Trichoderma viride and Penicillium funiculosum Cellulase)

  • Penicillium funiculosum과 Trichoderma viride cellulase의 정제효소들을 사용하여 cross-synergism을 조사하였다. Exo-exo형 상승작용이 Avicel을 분해하는데 가장 효과적이었으며 exo-endo형은 다소 효과가 떨어졌다. 정제효소성분과 이종의 조효소를 혼합하여 70시간 이상 Avicel을 가수분해하며 생성된 total sugar를 측정한 결과, P. funiculosum cellulase에서 분리정제한 $\beta$-glucosidase 성분효소는 T. viride cellulase와 상승작용을 크게 나타내며 Avicel을 가수분해하였다. 또한, T. viride cellulase에서 분리정제한 exoglucanase 성분효소도 P. funiculosum cellulase의 가수분해능을 크게 향상시켰다. 이와같은 결과로 미루어 볼 때, 이종의 cellulase로 부터 부족한 성분효소를 보충시켜 효소성분비율을 변화시킴으로써 Avicel의 가수분해도를 향상시킬 수 있을 것으로 사료된다. 이 때 최대한 높은 가수분해를 얻기 위하여 여러 형식의 상승작용이 같이 이루어져야할 것이다.

Cellulose Utilization and Protein Productivity of Some Cellulolytic Fungal Co-cultures

  • Protein productivity by the cellulolytic fungi, Trichoderma viride(MTCC 800), Chaetomium globosum and Aspergillus terreus was compared in co-culture and mixed culture fermentations of cashewnut bran. Co-cultures were more effective in substrate saccharification, which ranged between $85{\sim}88%$ compared to the $62{\sim}67%$ saccharification shown by the monocultures. Maximum saccharification was induced by T. viride and C. globosum co-culture resulting in the highest 34% release of reducing sugars. The maximum 16.4% biomass protein and the highest protein productivity(0.58%) were shown by T. viride and A. terreus co-culture. A. terreus performed better in co-culture in the presence of T. viride rather than with C. globosum. Among the cellulolytic enzymes, FPase(Filter Paper Cellulase) activity was significantly higher in all the co-cultures and in the mixed culture than in their respective monocultures. Mixed culture fermentation involving all the three fungi was not effective in increasing the per cent saccharification or the biomass protein content over the co-cultures.

Removal Efficiency of the Heavy Metals Zn(II), Pb(II) and Cd(II) by Saprolegnia delica and Trichoderma viride at Different pH Values and Temperature Degrees

  • The removal efficiency of the heavy metals Zn, Pb and Cd by the zoosporic fungal species Saprolegnia delica and the terrestrial fungus Trichoderma viride, isolated from polluted water drainages in the Delta of Nile in Egypt, as affected by various ranges of pH values and different temperature degrees, was extensively investigated. The maximum removal efficiency of S. delica for Zn(II) and Cd(II) was obtained at pH 8 and for Pb(II) was at pH 6 whilst the removal efficiency of T. viride was found to be optimum at pH 6 for the three applied heavy metals. Regardless the median lethal doses of the three heavy metals, Zn recorded the highest bioaccumulation potency by S. delica at all pH values except at pH 4, followed by Pb whereas Cd showed the lowest removal potency by the fungal species and vice versa in case of T. viride. The optimum bio-mass dry weight production by S. delica was found when the fungus was grown in the medium treated with the heavy metal Pb at pH 6, followed by Zn at pH 8 and Cd at pH 8. The optimum biomass dry weight yield by T. viride amended with Zn, Pb and Cd was obtained at pH 6 for the three heavy metals with the maximum value at Zn. The highest yield of biomass dry weight was found when T. viride treated with Cd at all different pH values followed by Pb whilst Zn output was the lowest and this result was reversed in case of S. delica. The maximum removal efficiency and the biomass dry weight production for the three tested heavy metals was obtained at the incubation temperature $20^{\circ}C$ in case of S. delica while it was $25^{\circ}C$ for T. viride. Incubation of T. viride at higher temperatures ($30^{\circ}C\;and\;35^{\circ}C$) enhanced the removal efficiency of Pb and Cd than low temperatures ($15^{\circ}C\;and\;20^{\circ}C$) and vice versa in case of Zn removal. At all tested incubation temperatures, the maximum yield of biomass dry weight was attained at Zn treatment by the two tested fungal species. The bioaccumulation potency of S. delica for Zn was higher than that for Pb at all temperature degrees of incubation and Cd bioaccumulation was the lowest whereas T. viride showed the highest removal efficiency for Pb followed by Cd and Zn was the minor of the heavy metals.

청피(靑皮)가 천식유발 백서에서 VEGF에 미치는 영향 (Effects of Citro Reticulatae Viride Pericarpium on the Vascular Endothelial Growth Factor(VEGF) in the bronchial asthma mouse model)

  • Objective : Author evaluate the effects of Citri Reticulatae Viride Pericarpium for the bronchial asthma using assesment of the vascular endothelial growth factor after Citri Reticulatae Viride Peicarpium was intravenously administered Ovalbumine(OVA)-sensitized and -challenged mice. Material and Methods : Twenty-four female mice, 8-10 weeks old and free of murine specific pathogens, were sensitized by intraperitoneal injection of OVA. Of the sensitized mice, ten mice didn't administrate Citri Reticulatae Viride Pericarpium and the other ten mice administrated Citri Reticulatae Viride Pericarpium. Mice were sensitized on the first and fourteen days introspectional injection of 20 ${\mu}g$ OVA. After$21^{st}\;22^{th}\;and\;23^{rd}$, the initial sensitization, the mice were challenged for 30 minutes with an aerosol of 1% OVA in saline. Citri Reticulatae Viride Pericarpium administered 200mg/kg in the tail of the mouse, one a day, for 7 days and beginning 14 days after first sensitization. Bronchoalveolar lavage was performed 72 hours after the last challenge, and total cell numbers in the BAL fluid were count. Also, lever of VEGF in the BAL fluid was measured by Enzyme immunoassays and Western blot analysis. Results: Total cell numbers in BAL fluid were significantly greater than from 72 hrs after OVA inhalation compared with cell numbers in the control group. However, there was no difference of the total cell numbers between OVA-challenge groups without Citri Reticulatae Viride Pericarpium, and OVA-challenge with Citri Reticulatae Viride Pericarpium. Enzyme immunoassay revealed that VEGF levels in the BAL fluids were significantly increased 72 hrs after OVA inhalation compared with levels in the control group. After administration of the Citri Reticulatae Viride Pericarpiu, the levels of the VEGF in BAL fluids 72 hrs after OVA inhalation reduced dramatically. Western blot analysis revealed that VEGF protein levels were increased in the all mice which were challenge with OVA, without administered Citri Reticulatae Viride Pericarpium, compared the normal mouse. However, in the groups of the administered Citri Reticulatae Viride Pericarpium, the VEGF protein level markedly decreased. Conclusion: Citri Reticulatae Viride Pericarpium might affect the treatment of the bronchial ashma as a inhibition of the VEGF.

(${\beta}-glucosidase$의 고생산을 위한 복합균주 개발

  • 본 연구의 목적은 기존의 단일 균주보다 두 종의 서로 다른 균주를 복합시켰을 때 두 균 주간의 상호 보완적인 면을 이용하여 효소생산을 극대화하기 위함이며, 다음과 같은 결론을 얻었다. 실험에서 사용된 T. viride와 FB01는 최적 pH와 온도가 비슷하여 혼합배양이 가능 하였으며, 이들의 복합균주를 개발하는데 성공하였다. 두 균주간의 상호작용으로 인해 단일 균주일 때보다 CMCase, ${\beta}-glucosidase$ 및 avicelase의 활성이 우수했으며 또한 계대배양 및 pH 조절을 통해 ${\beta}-glucosidase$이 활성이 최고 3.2배까지 증가함을 알 수 있었다. 탄소원으로 섬유소 폐기물중 특히 볏짚을 이용했을 때 복합균주에 의한 효소생산이 효과적이었으므로 앞으로 이 조건에서 복합균주를 장기적으로 계대배양하면서 효소의 생산성 향상과 복합균주의 안정성을 관찰할 필요가 있다고 사료된다.

  • 버섯 재배상(栽培床에)서 Trichoderma속(屬)을 분리, 동정하고 아울러 Trichoderma속(屬)이 생산하는 항생물질(抗生物質)이 느타리버섯균의 생육에 미치는 영향을 검토하여 다음과 같은 결과를 얻었다. 1) 분리된 Trichoderma속(屬)은 Rifai의 검색표에 따라 동정한 결과 Trichoderma hamatum, Trichoderma viride, Trichoderma koningii로 확인되었다. 2) 분리 동정된 Trichoderma속(屬) 중에서 Trichoderma viride(No. 10)이 느타리 버섯균 생육(生育)에 대한 저해율이 제일 높았다. 3) 각종 버섯균 중에서 Trichoderma viride(No. 10)에 의한 저해는 Pleurotus ostreatus와 Pleurotus sajor-caju가 제일 심했고 그 다음으로는 Lentinus edodes, Flammulina velutipes, Auricularicula auricula순이었다. 4) 추출된 백색 침상 결정 항균력(抗菌力)은 각각 1/10의 농도와 500ppm의 농도에서 cycloheximide $100{\sim}200ppm$의 항균력(抗菌力)에 해당하였다.

Mycelial response and ligninolytic enzyme production during interspecific interaction of wood-rotting fungi

  • To evaluate effects of ligninolytic enzyme type on the mycelial response and ligninolytic enzyme production during interspecific interactions among wood-rotting fungi, 4 fungal strains, Trichophyton rubrum LKY-7, Trichophyton rubrum LSK-27, Pycnoporus cinnabarinus, and Trichoderma viride, were selected. Regarding ligninolytic enzyme production, LKY-7 secreted laccase and manganese peroxidase (MnP), P. cinnabarinus secreted only laccase, and LSK-27 secreted only MnP in glucose-peptone medium, while T. viride did not produce any ligninolytic enzymes. In the co-culture of LKY-7 with P. cinnabarinus, the formation of aerial mycelium was observed and the enhancement of laccase activity owing to interspecific interaction appeared to be very low. In the co-culture of LKY-7 and P. cinnabarinus with LSK-27, a hypha-free clear zone was observed, which resulted in deadlock, and increased laccase or MnP activity was detected at the interaction zone. The interaction responses of LKY-7, P. cinnabarinus, and LSK-27 with T. viride were characterized by the formation of mycelial barrages along the interface. As mycelial barrages were observed at the T. viride territory and no brownish pigment was observed in the mycelial barrages, it is suggested that laccase and MnP are released as part of an offensive response, not as a defensive response. The co-culture of P. cinnabarinus with T. viride lead to the highest enhancement in laccase activity, yielding more than 14-fold increase in laccase activity with respect to the mono-culture of P. cinnabarinus. MnP activities secreted by LKY-7 or LSK-27 was generally low in interspecific interactions.
