Removal Characteristics of Residual Hydrogen Peroxide (H2O2) according to Application of Peroxone Process in O3/BAC Process (O3/BAC 공정에서 Peroxone 공정 적용에 따른 잔류 과산화수소 제거 특성)

  • Advanced Oxidation Processes (AOP) have been interested for removing micropollutants in water. Most of water treatment plants (WTPs) located along the lower part of Nakdong River have adopted the $O_3/BAC$ process and have interesting in peroxone process a kind of AOP. This study evaluated the removal characteristics of residual hydrogen peroxide ($H_2O_2$) combining with the biofiltration process in the next BAC process when the hydrogen peroxide is applied for the WTP operating $O_3/BAC$ process. In the experiment, changing the temperature and the concentration of $H_2O_2$ of influent, the biofiltration process showed rapidly dropped the biodegradability when the $H_2O_2$ concentration was increased and lowered water temperature while BAC process maintained relatively stable efficiency. The influent fixed at $20^{\circ}C$ and the concentration of $H_2O_2$ at 300 mg/L was continuously input for 78 hours. Most of the $H_2O_2$ in the influent did not remove at the biofiltration process controlled 5 to 15 minutes EBCT condition after 24~71 hours operating time while BAC process controlled 5 to 15 minutes EBCT showed 38~91% removal efficiency condition after 78 hours operating time. Besides, after 78 hours continuously input experiment, the biomass and activity of attached bacterial on the biofilter and BAC were $6.0{\times}10^4CFU/g$, $0.54mg{\cdot}C/m^3{\cdot}hr$ and $0.4{\times}10^8CFU/g$, $1.42mg{\cdot}C/m^3{\cdot}hr$ respectively. These biomass and activity values were decreased 99% and 72% in biofilter and 68% and 53% in BAC compared with initial condition. The biodegradation rate constant ($k_{bio}$) and half-life ($t_{1/2}$) in BAC were decreased from $1.173min^{-1}$ to $0.183min^{-1}$ and 0.591 min to 3.787 min respectively according to increasing the $H_2O_2$ concentration from 10 mg/L to 300 mg/L at $5^{\circ}C$ water temperature and the $k_{bio}$ and $t_{1/2}$ were $1.510min^{-1}$ to $0.498min^{-1}$ and 0.459 min to 1.392 min at $25^{\circ}C$ water temperature. By increasing the water temperature from $5^{\circ}C$ to $15^{\circ}C$ or $25^{\circ}C$, the $k_{bio}$ were increased 1.1~2.1 times and 1.3~4.4 times. If a water treatment plant operating $O_3/BAC$ process is considering the hydrogen peroxide for the peroxone process, post BAC could effectively decrease the residual $H_2O_2$, moreover, in case of spilling the $H_2O_2$ into the water process line, these spilled $H_2O_2$ concentration can be able to decrease by increasing the EBCT at the BAC process.

  • Present studies were carried out to investigate the distribution and formation of organic tiers contained paddy soils in Honam area characteristics to give basic informations on the effective utilization, management and improvement of the soils. The results obtained were summarized as follows; 1. The extent of organic tiers contained paddy soils in Honam area were 6.538㏊ and the amount of peat deposits were presumed about 2.41 million M/T. 2. Out of the total extent of the organic tiers contained paddy soils, about 97.6% was distributed in Honam plains (water-sheds of Mangyeong-Dongjin river), while about 1.5% in the Naju plains (water-sheds of Yeongsan river), and 0.9% in the Wando and Yeocheon areas. 3. The period of peat formation was presumed to be about the early of Seung Moon period (B.C. 4,250), and the Gongdeog series and the Bongnam series were formed in the bog conditions close to the valley mouth of near rolling and hill with small steram channels, and the Gimje series was formed in the out-skirts plains of the Gongdeog and Bongnam soils. 4. In the casue of peat formation, it was presumed to be the Gimje series that accumulated the fibrous peat out of the autochthonous peat such as reeds and grasses etc, to be the Gongdeog and Bongnam series that accumulated the autochtonous peat and the xylem and fibrous peat out of first allochthonous peat. 5. In the Organic horizons of these soils, the range of muck and peat horizons were in 62-68cm and 68-137cm of soil profile in the Gongdeog series, 52-84cm and 84-113cm in the Bongnam series respectively, one of muck horizon was in 46-71cm in the Gimje series. 6. The marks of soil horizons of the soils were expressed that the lower soils than the horizon of muck and peat were formed Cg, Aag for the muck horizon, 0 for the peat horizon, 0 of peat horizon were distingushed with Oag and Oig according to Organic forms. 7. The depthe occurred the muck and peat horizons were positively correlated with the width of local in the Gongdeog series ($r=0.881^{**}$, $r=0.827^{**}$), but not in the Bongnam series and Gimje series.

  • The Yeongsan river estuary has a serious water quality problem due to the water stagnation and it is imperative to predict the changes of water quality for mitigating water pollution. EFDC(Environmental Fluid Dynamics Code) model was mainly utilized to predict the changes of water quality for the estuary. The EFDC modeling normally accompanies the large volume of modeling output. For checking the spatial distribution of the modeling results, post-processing for converting of the output is prerequisite and mainly post-processing program is EFDC_Explorer. However, EFDC_Explorer only shows the spatial distribution of the time series and this doesn't support overlay function with other thematic maps. This means the impossible to the connection analysis with a various GIS data and high dimensional analysis. Therefore, this study aims to develop a post-processing system of a EFDC output to use them as GIS layers. For achieving this purpose, a editing module for main input files, and a module for converting binary format into an ASCII format, and a module for converting it into a layer format to use in a GIS based environment, and a module for visualizing the reconfigured model result efficiently were developed. Using the developed system, result file is possible to automatically convert the GIS based layer and it is possible to utilize for water quality management.

  • The Total Maximum Daily Load(TMDL) Act just implemented as a new tool of watershed based water quality management, in Korea. Thus, there are a number of pending questions to resolve for successful settlement of the TMDL. The allocation of pollution source is a exceedingly sensitive issue on local development planning. The simple area-based allocation (SAA) is conventional method to allocate the administrational pollution information to watershed based information. The SAA has a limitation that it can't consider the characteristics of spatial distribution of pollution source and it has caused more uncertainty of TMDL. This study was performed to reduce the uncertainty of watershed-based pollution information using the spatial distribution-based allocation(SDA). In the specific area where pollution source is concentrated such as urbanized region, it has been certified that SDA could reduce a tolerance of pollution information dramatically. As a result of study, SDA is expected a effective tool for TMDL and to solve the conflict between development and protection.

A Study on the Design Development and Construction Process in British Airways London Eye (런던아이의 디자인 전개와 건립과정에 관한 연구)

  • This study aims to identify and analyze the factors that influenced the construction of London Eye which was led by private groups, including characteristics in progress, differentiated value in design, the combination of technology and design, and construction costs. The results of this study were the followings. 1) London Eye provides a new construction method for a landmark in the downtown in that the architects became the clients of the project which might have been hoarded, procuring the construction costs and completing it by themselves. 2) London Eye presents a space utilization method where undeveloped spaces in the downtown are used in three-dimensions by erecting a structure on the river with a minimum land. 3) The process of design, production and erection is propelled the realization of technical intention design with one system and from at the initial stage, the participation of the engineer where the designer is equal with you must accompany. 4) London Eye is endowed with strong shape by a circle in design concept, which is the simplest geometrical figure, and it provides organic relationship between the past and the present by utilizing historical elements in various ways. 5) The construction was completed through four phases including idea phase, promotion phase, turn-key contract phase, and design-build phase. The most critical factor to the success is derived from the direct contact with Involved companies from the production phase. 6) In a project facing many challenges in terms of technology, the design-build method proves to be a more effective method than turn-key contract in that it may more allocate risks and enable coherent implementation of the core concept in design. The organization for the design-build method was composed of three phases including general affairs, responsible contracting by sector, and cooperative system by sector. Since that was a new concept structure, high-level contractors who hadn't had existing cooperative relationships with one another formed new cooperative relationship, while collaborating companies who had long cooperated led the responsible contracting and sub-cooperative system, which resulted in reduction of risks and time. 7) The major factors to change design were the design characteristics, derived from a technically great combination, and the opening time. 8) A new erection method was applied to London Eye, a structure that overcame the limitation in land and period, and the process of installation demonstrated that it is an important value for the construction of a landmark in the city. 9) In spite of many visitors and tremendous operational income, the factors that London Eye fails to free itself from debts include the construction costs expended double than expected, and interest burden to the investments, which occurred in the course of procuring the costs. Absolute limit in time increased the construction costs, resulting in being a great obstacle in the course of operation.

Development of mcyB-specific Ultra-Rapid Real-time PCR for Quantitative Detection of Microcystis aeruginosa (Microcystis aeruginosa의 정량을 위한 mcyB 특이 초고속 실시간 유전자 증폭법의 개발)

  • A mcyB-specific Ultra-Rapid quantitative PCR was developed for the quantitative detection of Microcystis aeruginosa, which is often a dominant species in green tide. McyB-specific UR-qPCR was optimized under extremely short times of each step in thermal cycles, based on the specific primers deduced from the mcyB in microcystin synthetase of M. aeruginosa. The M. aeruginosa strain KG07 was used as a standard for quantification, after the microscopic counting and calculation by mcyB-specific UR-qPCR. The water samples from the river water with the Microcystis outbreak were also measured by using both methods. The $1.0{\times}10^8$ molecules of mcyB-specific DNA was recognized inner 4 minutes after beginning of UR-qPCR, while $1.0{\times}10^4$ molecules of mcyB-specific templates was detected inner 7 minutes with quantitative manner. From the range of $1.0{\times}10^2$ to $1.0{\times}10^8$ initial molecules, quantification was well established based on $C_T$ using mcyB-specific UR-qPCR (Regression coefficiency, $R^2=0.9977$). Between the numbers of M. aeruginosa cell counting under microscope and calculated numbers using mcyB-specific UR-qPCR, some differences were often found. The reasons for these differences were discussed; therefore, easy compensation method was proposed that was dependent on the numbers of the cell counting. Additionally, to easily extract the genomic DNA (gDNA) from the samples, a freeze-fracturing of water-sample using liquid nitrogen was tested, by excluding the conventional gDNA extraction method. It was also verified that there were no significant differences using the UR-qPCR with both gDNAs. In conclusion, the mcyB-specific UR-qPCR that we proposed would be expected to be a useful tool for rapid quantification and easy monitoring of M. aeruginosa in environmental water.

Complete Mitochondrial Genome of Haplorchis taichui and Comparative Analysis with Other Trematodes

  • Mitochondrial genomes have been extensively studied for phylogenetic purposes and to investigate intra- and interspecific genetic variations. In recent years, numerous groups have undertaken sequencing of platyhelminth mitochondrial genomes. Haplorchis taichui (family Heterophyidae) is a trematode that infects humans and animals mainly in Asia, including the Mekong River basin. We sequenced and determined the organization of the complete mitochondrial genome of H. taichui. The mitochondrial genome is 15,130 bp long, containing 12 protein-coding genes, 2 ribosomal RNAs (rRNAs, a small and a large subunit), and 22 transfer RNAs (tRNAs). Like other trematodes, it does not encode the atp8 gene. All genes are transcribed from the same strand. The ATG initiation codon is used for 9 protein-coding genes, and GTG for the remaining 3 (nad1, nad4, and nad5). The mitochondrial genome of H. taichui has a single long non-coding region between trnE and trnG. H. taichui has evolved as being more closely related to Opisthorchiidae than other trematode groups with maximal support in the phylogenetic analysis. Our results could provide a resource for the comparative mitochondrial genome analysis of trematodes, and may yield genetic markers for molecular epidemiological investigations into intestinal flukes.

Numerical Modeling of Water Transfer among Precipitation, Surface Water, Soil Moisture and Groundwater

  • In the processes of hydrological cycle, when precipitation reaches the ground surface, water may become surface runoff or infiltrate into soil and then possibly further percolate into groundwater aquifer. A part of the water is returned to the atmosphere through evaporation and transpiration. Soil moisture dynamics driven climate fluctuations plays a key role in the simulation of water transfer among ground surface, unsaturated zone and aquifer. In this study, a one-layer canopy and a four-layer soil representation is used for a coupled soil-vegetation modeling scheme. A non-zero hydraulic diffusivity between the deepest soil layer modeled and groundwater table is used to couple the numerical equations of soil moisture and groundwater dynamics. Simulation of runoff generation is based on the mechanism of both infiltration excess overland flow and saturation overland flow nested in a numerical model of soil moisture dynamics. Thus, a comprehensive hydrological model integrating canopy, soil zone and aquifer has been developed to evaluate water resources in the plain region of Huaihe River basin in East China and simulate water transfer among precipitation, surface water, soil moisture and groundwater. The newly developed model is capable of calculating hydrological components of surface runoff, evapotranpiration from soil and aquifer, and groundwater recharge from precipitation and discharge into rivers. Regional parameterization is made by using two approaches. One is to determine most parameters representing specific physical values on the basis of characterization of soil properties in unsaturated zone and aquifer, and vegetations. The other is to calibrate the remaining few parameters on the basis of comparison between measured and simulated streamflow and groundwater tables. The integrated modeling system was successfully used in the Linhuanji catchment of Huaihe plain region. Study results demonstrate that (1) on the average 14.2% of precipitation becomes surface runoff and baseflow during a ten-year period from 1986 to 1995 and this figure fluctuates between only 3.0% in drought years of 1986, 1988, 1993 and 1994 to 24.0% in wet year of 1991; (2) groundwater directly deriving from precipitation recharge is about 15.0% t of the precipitation amount, and (3) about half of the groundwater recharge flows into rivers and loses through evaporation.

Distributions of the Temperature and Salinity in Kamak Bay (가막만의 수온과 염분의 분포)

  • The distributions of the temperature and salinity in Kamak bay which has two channels and three sea bottom topographic parts were studied by taking the detailed hydrographic data at the ebb and flood during four seasons from May, 1988 to Feb., 1989. The general pattern of the distributions of characteristics which the temperature and salinity has in Kamak bay is basically formed by the topography and sea water movement of the bay. The changes of these distributions by seasons mainly come from the heating and cooling of the sea surface and the increase of the run-off. The bay has three remarkable water masses and the their general characteristics are follows: the inner bay water has a stagnation character influenced by the inland and the concave of the sea bottom in the north west, Yosu harbor water has an estuary character of the low salinity caused by the run-off of Somjin river and Yon Tung brooklet in the north east, and the outer bay water has an out-sea character, as it is located near by the big mouth in the south of the bay. The distributions of those water masses at the ebb and flood show some different features due to the flow patterns, and the daily changes of oceanic conditions at the vicinity of Hangdae-ri are so big that it may influence the habitation and production of the living things in the bay.

Influence of Density and Feeding Frequency on Early Life History and Cannibalism of River Puffer, Takifugu obscurus (황복, Takifugu obscurus의 초기 생활사와 공식 행위에 있어 밀도와 먹이 공급 횟수의 영향)

  • To investigate the influence of density and feeding frequency (food amount) on growth, cannibal dynamics and survival of puffer Takifugu obscurus (TL $25.0{\pm}0.5$ mm, BW $0.53{\pm}0.03$ g) during early life history, we conducted one experiment composed with 6 density groups set up as 1.43 inds./${\ell}$, 2.14 inds./${\ell}$, 2.86 inds./${\ell}$, 3.57 inds./${\ell}$, 4.29 inds./${\ell}$ and 7.14 inds./${\ell}$. The experiment was triplicately performed in 1 ton FRP tank for 30 days. Daily food intake (DFI), feed efficiency (FE), growth, cannibal frequency, and survival rates were examined. Although not find any significant relation of density to DFI and FE, we found that growth decreased significantly with the increasing of density under 3.57 inds./${\ell}$, but significantly increased with the increasing of density over 4.29 inds./${\ell}$. Cannibal frequency and daily mortality rate was significantly increased with increasing density and decreasing feeding frequency, inducing the most poor survival rate in the highest density group and low feeding frequency. Therefore, we concluded that the density and the feeding frequencies affect sibling cannibal dynamics and survival of puffer stock.
