The aim of this study was to provide basic information to allow improved feeding management for velvet antler production by investigating the effects of dietary protein level on production and chemical composition of velvet antler in spotted deer(Cervus nippon). Eighteen male spotted deer of 5~6 years old were assigned to 3 unreplicated groups by production record of the previous year. Deer were fed on experimental diets at different protein level of 10%(T1), 15%(T2) and 20%(T3). Dry matter intake, crude protein intake and daily gain were increased with increasing the protein level in diets and there were significant differences(P<0.05) between groups. Although not significant, mean length of main beam tended to be longer in either left or right beam with increasing the protein level, and in girth at part of base of main beam, it was a similar pattern to the result of length. The velvet antler production was affected by dietary protein level and thus that was the smallest in T1 and the largest in T3(P<0.05). The content of crude protein in velvet antler was higher in T1 than in T3, whereas the content of crude ash in velvet antler was contrast to that of crude protein, however, there were no significant differences.
Seventeen extracellular protein producing bacteria were isolated from soil samples, among which T219 strain having a strong capability of producing the protein was selected and identified for investigation of biological characteristics. The factors which affect the protein production were investigated and the results are summarized as follows. T219 strain which produces the most extracellular protein was identified as Bacillus sp. Optimum temperature and pH for production of the extracellular protein by T219 strain were $25^{\circ}C$ and 7.5 respectively. Almost no activities of protease and amylase were observed in the protein produced by the protein producing bacteria. In the medium containing yeast extract, the cell growth was moderately high, but almost no accumulation of protein was observed. However, polypeptone had significant effects on both the cell growth and the protein accumulation. The addition of glycine and L-isoleucine to the medium containing polypeptone, yeast extract and meat extract had a great effect on the protein production; 4mg/ml of protein accumulation was observed.

Background: Cytotoxic T lymphocytes (CTLs) appear to play an important role in the control and prevention of human cytomegalovirus (HCMV) infection. The pp65 antigen is a structural protein, which has been defined as a potential target for effective immunity against HCMV infection. Incorporation of an 11 amino acid region of the HIV TAT protein transduction domain (Tat) into protein facilitates rapid, efficient entry into cells. Methods: To establish a strategy for the generation of HCMV-specific CTLs in vitro, recombinant truncated N- and C-terminal pp65 protein (pp65 N&C) and N- and C-terminal pp65 protein fused with Tat (Tat/pp65 N&C) was produced in E.coli system. Peripheral blood mononuclear cells were stimulated with dendritic cells (DCs) pulsed with pp65 N&C or Tat/pp65 N&C protein and immune responses induced was examined using IFN-${\gamma}$ ELISPOT assay, cytotoxicity assay and tetramer staining. Results: DCs pulsed with Tat/pp65N&C protein could induce higher T-cell responses in vitro compared with pp65N&C. Moreover, the DCs pulsed with Tat/pp65 N&C could stimulate both of $CD8^+$ and $CD4^+$ T-cell responses. The T cells induced by DCs pulsed with Tat/pp65 N&C showed higher cytotoxicity than that of pp65-pulsed DCs against autologous lymphoblastoid B-cell line (LCL) expressing the HCMV-pp65 antigen. Conclusion: Our results suggest that DCs pulsed with Tat/pp65 N&C protein effectively induced pp65-specific CTL in vitro. Tat fusion recombinant protein may be useful for the development of adoptive T-cell immunotherapy and DC-based vaccines.
Isozyme and protein electrophoresis data from mycelial extracts of 27 isolates of Trichoderma harzianum, 10 isolates of T. aureoviride, and 10 isolates of T. longibrachiatum from Southern Peninsular Malaysia were investigated. The eight enzyme and a single protein pattern systems were analyzed. Three isozyme and total protein patterns were shown to be useful for the detection of three Trichoderma species. The isozyme and protein data were analyzed using the Nei and Li Dice similarity coefficient for pairwise comparison between individual isolates, species isolate group, and for generating a distance matrix. The UPGMA cluster analysis showed a higher degree of relationship between T. harzianum and T. aureoviride than to T. longibrachiatum. These results suggested that the T. harzianum isolates had high levels of genetic variation compared with the other isolates of Trichoderma species.

The aim of this study was to provide different levels of dietary protein for optimum growth performance in calves and to investigate the impact on the concentration of insulin-like growth factor-1 (IGF-1) in serum. Female Sahiwal calves (n = 12, body weight (BW) = $40{\pm}4.3kg$ and age = $60{\pm}10days$) were selected and divided into three groups with 4 animals in each group. Three diets of T0, T1 and T2 with 18, 20 and 22% of crude protein levels, respectively, were given to the calves to assess the post-weaning growth performance. Calves fed the T2 diet showed a higher (p < 0.05) dry matter (DM) intake than those fed the T0 and T1 diets. The feed conversion ratio (FCR) for the calves fed the T2 diet was lower (p < 0.05) than those of the calves fed the T0 and T1 diets. The T2 group had the highest BW (p < 0.05) compared with the other groups. The concentration of IGF-1 in serum increased (121.9, 143.3, and 152.9 ng/ml for T0, T1, and T2, respectively) as the crude protein (CP) level increased. Overall, the results of this study suggest that post weaning diets T1 and T2 with 20 and 22% crude protein in Sahiwal female calves had significantly increased the BW and serum IGF-1 concentration. The IGF-1 estimation might be a physiological indicator for growth performance.

Genus specific antigenicity of the 10 kDa protein in cyst fluid (CF) of Taenia solium metacestodes was demonstrated by comparative immunoblot analysis. When CFs from taeniid metacestodes of T. saginata, T. solium, T. taeniaeformis and T. crassiceps were probed with specific monoclonal antibody (mAb) raised against 150 kDa protein of T. solium metacestodes, specific antibody reactions were observed in 7 and 10 kDa proteins of T. solium and in 7/8 kDa of T. saginata, T. taeniaeformis and T. crasiceps. The mAb did not react with any protein in hydatid fluid of Echinococcus granulosus and E. multilocularis. This result revealed that the 10 kDa peptide of T. solium metacestodes and its equivalent proteins of different Taenia metacestodes are genus specific antigens that are shared among different Taenia species.
Background: Cytotoxic function of killer cells is inhibited by specific recognition of class I MHC molecules on target cells by inhibitory killer Ig-like receptors (KIR) expressed on NK cells and some cytotoxic T cells. The inhibitory effect of KIR is accomplished by recruitment of SH2-containing protein tyrosine phosphatase (SHP) to the phosphotyrosine residues in the cytoplasmic tail. Methods: By in vitro coprecipitation experiments and transfection analysis, we investigated the association of KIR with an adaptor protein Shc in Jurkat T cells. Results: The cytoplasmic tail of KIR appeared to associate with an adaptor protein Shc in Jurkat T celilysates. Similar in vitro experiments showed that phosphorylated KIR cytoplasmic tail bound SHP-1 and Shc in Jurkat T cell lysates. The association of KIR with Shc was further confirmed by transfection analysis in 293T cells. Interestingly, however, Shc appeared to be replaced by SHP-2 upon engagement of KIR in 293T cells. Conclusion: Our data indicate that KIR associate with an adaptor protein Shc in Jurkat T cells, and suggest that KIR might have an additional role which is mediated by this adaptor protein.
This experiment was conducted to study the sparing effects of animal proteins on weight gain, nutrients utilizability and economic analysis of broiler. Experiment diet contained different ratio of animal and plant protein and were supplemented different levels of methionine and lysine for the respective protein levels. A total of 264 broiler chicks were fed four diets of control, $T_1$, $T_2$ and $T_3$ for 42 days from April 7, 1990 to May 19, 1990. Dietary protein levels of control, $T_1$, $T_2$ and $T_3$ for starter and finisher were 20~18, 18~16, 18~16 and 22~20%, respectively. Methionine and lysine levels of control, $T_1$, $T_2$ and $T_3$ were 0.4~1.1, 0.44~1.21, 0.48~1.32 and 0.48~1.32% for starter diet, respectively, and were 0.32~0.90. 0.35~0.99, 0.38~1.08 and 0.38~1.08% for finisher diet, respectively. The results obtained were summarized as follows. 1. The birds fed control diet gained most for overall period. $T_3$ treatment which was high in protein, methionine and lysine levels gained most for finisher period. 2 The birds fed control diet consumed most feed, and the birds fed T$_3$ diet consumed least feed. for overall period. Feed conversion during 1~4 weeks was better in $T_1$ (1.51) and $T_2$ (1.53) than in control (1.61) and $T_3$ (1.63) . During 4~6weeks, feed conversion was better in $T_3$ (1.37) and control(1.58) than T, (2.05) and T, (2.16) (P<0.01) 3. Dry matter, crude fiber and NFE utilizability were increased for 1~4 weeks and decreased for 4~6 weeks as methionine and lysine levels increased and crude protein utilizability tended to be increased as protein levels increased. 4. Abdominal fat content was lowest in bird fed control diet and was high in birds fed low protein diet Carcass percentage was highest at control and the abdominal fat content was higher in bird fed lower protein diet than bird fed other protein diets(P<0.05). 5. Feed cost per kg weight gain was lowest at $T_3$ which contained more soybean oil meal than other feeds and next was control. According to the results of this experiment, it was revealed that optimum protein, methionine and lysine levels for starter and finisher broiler diet were 20~18, 0.4~0.32 and 1.1~0.9%, respectively.
Toxoplasma gondii infections occur throughout the world, and efforts are needed to develop various vaccine candidates expressing recombinant protein antigens. In this study, influenza matrix protein (M1) virus-like particles (VLPs) consisting of T. gondii rhoptry antigen 4 (ROP4 protein) were generated using baculovirus (rBV) expression system. Recombinant ROP4 protein with influenza M1 were cloned and expressed in rBV. SF9 insect cells were coinfected with recombinant rBVs expressing T. gondii ROP4 and influenza M1. As the results, influenza M1 VLPs showed spherical shapes, and T. gondii ROP4 protein exhibited as spikes on VLP surface under transmission electron microscopy (TEM). The M1 VLPs resemble virions in morphology and size. We found that M1 VLPs reacted with antibody from T. gondii-infected mice by western blot and ELISA. This study demonstrated that T. gondii ROP4 protein can be expressed on the surface of influenza M1 VLPs and the M1 VLPs containing T. gondii ROP4 reacted with T. gondii-infected sera, indicating the possibility that M1 VLPs could be used as a coating antigen for diagnostic and/or vaccine candidate against T. gondii infection.

Bacillus subtilis spores can be used for protein display to engineer protein properties. This method overcomes viability and protein-folding concerns associated with traditional protein display methods. Spores remain viable under extreme conditions and the genotype/phenotype connection remains intact. In addition, the natural sporulation process eliminates protein-folding concerns that are coupled to the target protein traveling through cell membranes. Furthermore, ATP-dependent chaperones are present to assist in protein folding. CotA was optimized as a whole-cell biocatalyst immobilized in an inert matrix of the spore. In general, proteins that are immobilized have advantages in biocatalysis. For example, the protein can be easily removed from the reaction and it is more stable. The aim is to improve the pH stability using spore display. The maximum activity of CotA is between pH 4 and 5 for the substrate ABTS (ABTS = diammonium 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonate). However, the activity dramatically decreases at pH 4. The activity is not significantly altered at pH 5. A library of approximately 3,000 clones was screened. A E498G variant was identified to have a half-life of inactivation ($t_{1/2}$) at pH 4 that was 24.8 times greater compared with wt-CotA. In a previous investigation, a CotA library was screened for organic solvent resistance and a T480A mutant was found. Consequently, T480A/E498G-CotA was constructed and the $t_{1/2}$ was 62.1 times greater than wt-CotA. Finally, E498G-CotA and T480A/E498G-CotA yielded 3.7- and 5.3-fold more product than did wt-CotA after recycling the biocatalyst seven times over 42 h.
