Mass Screening of Lovastatin High-yielding Mutants through Statistical Optimization of Sporulation Medium and Application of Miniaturized Fungal Cell Cultures (Lovastatin 고생산성 변이주의 신속 선별을 위해 통계적 방법을 적용한 Sporulation 배지 개발 및 Miniature 배양 방법 개발)

  • For large and rapid screening of high-yielding mutants of lovastatin produced by filamentous fungal cells of Aspergillus terreus, one of the most important stage is to test as large amounts of mutated strains as possible. For this purpose, we intended to develop a miniaturized cultivation method using $7m{\ell}$ culture tube instead of traditional $250m{\ell}$ flask (working volume $50m{\ell}$). For obtaining large amounts of conidiospores to be used as inoculums for miniaturized cultures, 4 components i.e., glucose, sucrose, yeast extract and $KH_2PO_4$ were intensively investigated, which had been observed to show positive effect on enhancement of spore production through Plackett-Burman design experimet. When optimum concentrations of these components that were determined through application of response surface method (RSM) based on central composite design (CCD) were used, maximum spore numbers amounting to $1.9\times10^{10}$ spores/plate were obtained, resulting in approximately 190 fold increase as compared to the commonly used PDA sporulation medium. Using the miniaturized cultures, intensive strain development programs were carried out for screening of lovastatin high-yielding as well as highly reproducible mutants. It was observed that, for maximum production of lovastatin, the producers should be activated through 'PaB' adaptation process during the early solid culture stage. In addition, they should be proliferated in condensed filamentous forms in miniaturized growth cultures, so that optimum amounts of highly active cells could be transferred to the production culture-tube as reproducible inoculums. Under these highly controlled fermentation conditions, compact-pelleted morphology of optimum size (less than 1 mm in diameter) was successfully induced in the miniaturized production cultures, which proved essential for maximal utilization of the producers' physiology leading to significantly enhanced production of lovastatin. As a result of continuous screening in the miniaturized cultures, lovastatin production levels of the 81% of the daughter cells derived from the high-yielding producers turned out to be in the range of 80%$\sim$120% of the lovastatin production level of the parallel flask cultures. These results demonstrate that the miniaturized cultivation method developed in this study is efficient high throughput system for large and rapid screening of highly stable and productive strains.

Development of UAV Flight Control Software using Model-Based Development(MBD) Technology (모델기반 개발기술을 적용한 무인항공기 비행제어 소프트웨어 개발)

  • This paper describes the Model-Based Development(MBD) process behind the flight control software of a close-range unmanned aerial vehicle(KUS-9). An integrated development environment was created using a commercial tool(MATLAB $Simulink^{(R)}$), which was utilized to design models for linear/nonlinear simulation, flight control law, operational logic and HILS(Hardware In the Loop Simulation) system. Software requirements were validated through flight simulations and peer reviews during the design process, whereas the models were verified through the application of a DO-178B verification tool. The integrity of automatically generated C code was verified by using a separate S/W testing tool. The finished software product was embedded on two different types of hardware and real-time operating system(uC/OS-II, VxWorks) to perform HILS and flight tests. The key findings of this study are that MBD Technology enables the development of a reusable and an extensible software product and auto-code generation technology allows the production of a highly reliable flight control software under a compressed time schedule.

A comparative study on ammonia emission inventory in livestock manure compost application through a foreign case study (국내외 가축분뇨 퇴액비 이용 분야 암모니아 배출량 인벤토리 비교 연구)

  • In Korea, more than 70% of the ammonia(NH3) released into the atmosphere is known to originate from livestock manure. The total emission (kg year-1) is calculated by multiplying the emission factor (kg head-1 yr-1) and the activity data (head). To improve the accuracy and reliability of the NH3 emission estimation process, an accurate account of livestock manure production, calculation of NH3 conversion and generation during the composting and liquefaction of manure, estimation of NH3 generation in the storage and transportation of manure and compost, and a comparative study of NH3 emission during the soil spreading process must be performed. Compared to the US and EU-28, in particular, the domestic emission factor is relatively even and the spatial/temporal scale is not broken down sufficiently to reflect the domestic situation. As a way to improve the accuracy and expertise of estimating NH3 emission factors, a 'dynamic chamber-capture system' can be utilized, which allows complex considerations of compost, liquid manure, soil, and climate characteristics. By reviewing and comparing the data related to domestic and foreign NH3 emission, we identified shortcomings in the current domestic system and the directions to be taken and suggested a chamber system that could estimate NH3 emission flux. It is also necessary to establish a methodology for mesocosm systems in the field, in addition to indoor chamber systems, to be linked with practical policies, such as the calculation of new emission factors for missing sources.

A Quantitative Analysis of the Effect of Ocean Emissions on the Simulated Ozone Concentration in South Korea (국내 오존 모의 농도에 대한 해양 배출량의 영향 정량 분석)

  • In this study, we quantitatively analyze the effect of ocean emission sources on the simulated O3 concentrations in South Korea using the community multi-scale air quality (CMAQ) model. To analyze changes in O3 concentrations by ocean emissions, two different CMAQ simulations considering ocean emissions (OE case) and without considering ocean emissions (NE case) were conducted during the Korea-United States air quality (KORUS-AQ) campaign period (May-June 2016). The changes in the simulated O3 concentrations due to the effect of ocean emissions (OE case-NE case) appeared mostly in the ocean areas (+1.201 ppbv). The effect of ocean emissions was positive during the daytime (+1.813 ppbv), but negative during the nighttime (-0.612 ppbv). Analysis using the integrated process rate (IPR) confirmed that the increase or decrease in O3 concentration by ocean emissions was mainly due to chemical processes. Further analysis using the integrated reaction rate (IRR) showed that the daytime increase in O3 concentration was mainly attributable to the increased O3 production via O + O2 + M → O3 + M reaction as photolysis of NO2 increased due to the added ocean emissions. The nighttime decrease in O3 concentration was mainly due to the increased O3 titration by NO (NO + O3 → O2 + NO2) due to the increased NO emission. These results indicate that the changes in the concentration O3 in the sea area by the effect of ocean emissions are mainly due to increased NOx emissions. However, there could be a number of uncertainties in ocean emissions data used in this study, thus continuous comparative research using the most updated data will need to be carried out in the future.

A Study on Application Methodology of SPDL Based on IEC 62443 Applicable to SME Environment (중소기업환경에서 적용 가능한 IEC 62443 기반의 개발 보안 생애주기 프로세스 적용 방안 연구)

  • In a smart factory environment in a small and medium-sized enterprise (SME) environment, sensors and actuators operating on actual manufacturing lines, programmable logic controllers (PLCs) to manage them, human-machine interface (HMI) to control and manage such PLCs, and consists of operational technology server to manage PLCs and HMI again. PLC and HMI, which are in charge of control automation, perform direct connection with OT servers, application systems for factory operation, robots for on-site automation, and production facilities, so the development of security technology in a smart factory environment is demanded. However, smart factories in the SME environment are often composed of systems that used to operate in closed environments in the past, so there exist a vulnerable part to security in the current environment where they operate in conjunction with the outside through the Internet. In order to achieve the internalization of smart factory security in this SME environment, it is necessary to establish a process according to the IEC 62443-4-1 Secure Product Development Life cycle at the stage of smart factory SW and HW development. In addition, it is necessary to introduce a suitable development methodology that considers IEC 62443-4-2 Component security requirements and IEC 62443-3 System security requirements. Therefore, this paper proposes an application plan for the IEC 62443 based development security process to provide security internalization to smart factories in an SME environment.

Hybrid Offloading Technique Based on Auction Theory and Reinforcement Learning in MEC Industrial IoT Environment (MEC 산업용 IoT 환경에서 경매 이론과 강화 학습 기반의 하이브리드 오프로딩 기법)

  • Industrial Internet of Things (IIoT) is an important factor in increasing production efficiency in industrial sectors, along with data collection, exchange and analysis through large-scale connectivity. However, as traffic increases explosively due to the recent spread of IIoT, an allocation method that can efficiently process traffic is required. In this thesis, I propose a two-stage task offloading decision method to increase successful task throughput in an IIoT environment. In addition, I consider a hybrid offloading system that can offload compute-intensive tasks to a mobile edge computing server via a cellular link or to a nearby IIoT device via a Device to Device (D2D) link. The first stage is to design an incentive mechanism to prevent devices participating in task offloading from acting selfishly and giving difficulties in improving task throughput. Among the mechanism design, McAfee's mechanism is used to control the selfish behavior of the devices that process the task and to increase the overall system throughput. After that, in stage 2, I propose a multi-armed bandit (MAB)-based task offloading decision method in a non-stationary environment by considering the irregular movement of the IIoT device. Experimental results show that the proposed method can obtain better performance in terms of overall system throughput, communication failure rate and regret compared to other existing methods.

Enhanced Crystallinity of Piezoelectric Polymer via Flash Lamp Annealing (플래시광 열처리를 통한 압전 고분자의 결정성 향상 연구)

  • The polymer crystallization process, promoting the formation of ferroelectric β-phase, is essential for developing polyvinylidene fluoride (PVDF)-based high-performance piezoelectric energy harvesters. However, traditional high-temperature annealing is unsuitable for the manufacture of flexible piezoelectric devices due to the thermal damage to plastic components that occurs during the long processing times. In this study, we investigated the feasibility of introducing a flash lamp annealing that can rapidly induce the β-phase in the PVDF layer while avoiding device damage through selective heating. The flash light-irradiated PVDF films achieved a maximum β-phase content of 76.52% under an applied voltage of 300 V and an on-time of 1.5 ms, a higher fraction than that obtained through thermal annealing. The PVDF-based piezoelectric energy harvester with the optimized irradiation condition generates a stable output voltage of 0.23 V and a current of 102 nA under repeated bendings. These results demonstrate that flash lamp annealing can be an effective process for realizing the mass production of PVDF-based flexible electronics.

  • With the increasing use of transparent displays and flexible devices, polymer substrates offering excellent flexibility and strength are in demand. Since polymers are sensitive to heat, precise temperature control during the process is necessary. The study proposes a temperature measurement system for the laser processing area within the polymer base, aiming to address the drawbacks of using these polymer bases in laser-based selective processing technology. It presents the possibility of optimizing the process conditions of the polymer substrate through local temperature change measurements in the laser processing area. We developed and implemented the PDPT (Photodiode-based Planck Thermometry) to measure temperature in the laser-processing area. PDPT is a non-destructive, contact-free system capable of real-time measurement of local temperature increases. We monitored the temperature fluctuations during the laser processing of the polymer substrate. The study shows that the proposed laser-based temperature measurement technology can measure real-time temperature during laser processing, facilitating optimal production conditions. Furthermore, we anticipate the application of this technology in various laser-based processes, including essential micro-laser processing and 3D printing.

Study on the Application of Multi-skilled labors to Factory Production Process for Securing Economic Feasibility of Modular Unit (모듈러 경제성 확보를 위한 공장생산 프로세스의 다기능공 적용 방안)

  • The Construction industry is a labor-intensive industry that its labor cost takes up about 30~40% out of the whole construction cost. However, due to a stereotype that on-site work is a 3D job there is a shortage of the labor forces. Modular construction method is to produce modular units in the plant so that workers could work stably. Also, after delivering the module from plant to the site, there will be only installment to be required that shortens construction duration. Even though the modular market is currently expanding based on military facilities in Korea, its best strengths are not demonstrated well which are shortened construction period and low cost. It also causes labor problem of production due to minimum utilization of the modular construction method. Multi-skilled labor means a technician that is able to perform more than two kinds of work with more than two techniques. Multi-skilled labor can proceed smoothly by figuring out the connectivity between the precedent and following operations. Therefore, this research is to apply the concept of Multi-skilled labors, suggest solutions and allocate manpowers efficiently. As a result, it helps to decrease idle manpowers during the operation and the total labor forces can be saved. Low cost is the original strength of the modular which can stand out so the modular market is expected to expand.

Urban alienation and the just city (도시적 소외와 정의로운 도시)

  • This paper is to reconsider some characteristics of philosophical discussion on alienation, especially focusing on Lefebvre's concept of alienation, and then to conceptualize a number of features of alienation in both industrial and postindustrial capitalist cities. The construction and development of modern city in industrial capitalism has brought about alienation from nature and from land(i.e. means of production), and in these contexts, has generated alienated labour of urban labourers, which has been deepened through development of modern technologies and divisions of labour. The transformation from industrial to postindustrial society can be seen not as alleviating but as further intensifying and expanding process of alienation. Urban alienation in postindustrial society has been spatially and temporally extended through processes of glocalization and of financialization with the development of credit system. It also has been widened to fields of consumption and leisure and to spheres of non-material production, and has get more deeply involved in capital circulation through built environment and landscape(or spectacles) of cities. Finally this paper is to re-examine briefly theoretical discussions on dealienation in order to conceptualize the just city for dealienation of labour and of urban space, in particular considering the concept of 'the right to the city' as practical strategy of urban dealienation, and to suggest further three kinds of justice for the just city, that is, justice for distribution, for production and for recognition.
