Production of Rapamycin in Streptomyces hygroscopicus from Glycerol-Based Media Optimized by Systemic Methodology

  • Rapamycin, produced by the soil bacterium Streptomyces hygroscopicus, has the ability to suppress the immune system and is used as an antifungal, anti-inflammatory, antitumor, and immunosuppressive agent. In an attempt to increase the productivity of rapamycin, mutagenesis of wild-type Streptomyces hygroscopicus was performed using ultraviolet radiation, and the medium composition was optimized using glycerol (which is one of the cheapest starting substrates) by applying Plackett-Burman design and response surface methodology. Plackett-Burman design was used to analyze 14 medium constituents: M100 (maltodextrin), glycerol, soybean meal, soytone, yeast extract, $(NH_4)_2SO_4$, $\small{L}$-lysine, $KH_2PO_4$, $K_2HPO_4$, NaCl, $FeSO_4{cdot}7H_2O$, $CaCO_3$, 2-(N-morpholino) ethanesulfonic acid, and the initial pH level. Glycerol, soytone, yeast extract, and $CaCO_3$ were analyzed to evaluate their effect on rapamycin production. The individual and interaction effects of the four selected variables were determined by Box-Behnken design, suggesting $CaCO_3$, soytone, and yeast extract have negative effects, but glycerol was a positive factor to determine rapamycin productivity. Medium optimization using statistical design resulted in a 45% ($220.7{\pm}5.7mg/l$) increase in rapamycin production for the Streptomyces hygroscopicus mutant, compared with the unoptimized production medium ($151.9{\pm}22.6mg/l$), and nearly 588% compared with wild-type Streptomyces hygroscopicus ($37.5{\pm}2.8mg/l$). The change in pH showed that $CaCO_3$ is a critical and negative factor for rapamycin production.

Software Development for Optimal Productivity and Service Level Management in Ports (항만에서 최적 생산성 및 서비스 수준 관리를 위한 소프트웨어 개발)

  • Port service level is a metric of competitiveness among ports for the operating/managing bodies such as the terminal operation company (TOC), Port Authority, or the government, and is used as an important indicator for shipping companies and freight haulers when selecting a port. Considering the importance of metrics, we developed software to objectively define and manage six important service indicators exclusive to container and bulk terminals including: berth occupancy rate, ship's waiting ratio, berth throughput, number of berths, average number of vessels waiting, and average waiting time. We computed the six service indicators utilizing berth 1 through berth 5 in the container terminals and berth 1 through berth 4 in the bulk terminals. The software model allows easy computation of expected ship's waiting ratio over berth occupancy rate, berth throughput, counts of berth, average number of vessels waiting and average waiting time. Further, the software allows prediction of yearly throughput by utilizing a ship's waiting ratio and other productivity indicators and making calculations based on arrival patterns of ship traffic. As a result, a TOC is able to make strategic decisions on the trade-offs in the optimal operating level of the facility with better predictors of the service factors (ship's waiting ratio) and productivity factors (yearly throughput). Successful implementation of the software would attract more shipping companies and shippers and maximize TOC profits.

Development of Machine Learning-Based Platform for Distillation Column (증류탑을 위한 머신러닝 기반 플랫폼 개발)

  • This study developed a software platform using machine learning of artificial intelligence to optimize the distillation column system. The distillation column is representative and core process in the petrochemical industry. Process stabilization is difficult due to various operating conditions and continuous process characteristics, and differences in process efficiency occur depending on operator skill. The process control based on the theoretical simulation was used to overcome this problem, but it has a limitation which it can't apply to complex processes and real-time systems. This study aims to develop an empirical simulation model based on machine learning and to suggest an optimal process operation method. The development of empirical simulations involves collecting big data from the actual process, feature extraction through data mining, and representative algorithm for the chemical process. Finally, the platform for the distillation column was developed with verification through a developed model and field tests. Through the developed platform, it is possible to predict the operating parameters and provided optimal operating conditions to achieve efficient process control. This study is the basic study applying the artificial intelligence machine learning technique for the chemical process. After application on a wide variety of processes and it can be utilized to the cornerstone of the smart factory of the industry 4.0.

Why Gabor Frames? Two Fundamental Measures of Coherence and Their Role in Model Selection

  • The problem of model selection arises in a number of contexts, such as subset selection in linear regression, estimation of structures in graphical models, and signal denoising. This paper studies non-asymptotic model selection for the general case of arbitrary (random or deterministic) design matrices and arbitrary nonzero entries of the signal. In this regard, it generalizes the notion of incoherence in the existing literature on model selection and introduces two fundamental measures of coherence-termed as the worst-case coherence and the average coherence-among the columns of a design matrix. It utilizes these two measures of coherence to provide an in-depth analysis of a simple, model-order agnostic one-step thresholding (OST) algorithm for model selection and proves that OST is feasible for exact as well as partial model selection as long as the design matrix obeys an easily verifiable property, which is termed as the coherence property. One of the key insights offered by the ensuing analysis in this regard is that OST can successfully carry out model selection even when methods based on convex optimization such as the lasso fail due to the rank deficiency of the submatrices of the design matrix. In addition, the paper establishes that if the design matrix has reasonably small worst-case and average coherence then OST performs near-optimally when either (i) the energy of any nonzero entry of the signal is close to the average signal energy per nonzero entry or (ii) the signal-to-noise ratio in the measurement system is not too high. Finally, two other key contributions of the paper are that (i) it provides bounds on the average coherence of Gaussian matrices and Gabor frames, and (ii) it extends the results on model selection using OST to low-complexity, model-order agnostic recovery of sparse signals with arbitrary nonzero entries. In particular, this part of the analysis in the paper implies that an Alltop Gabor frame together with OST can successfully carry out model selection and recovery of sparse signals irrespective of the phases of the nonzero entries even if the number of nonzero entries scales almost linearly with the number of rows of the Alltop Gabor frame.

Analysis of Short-term Runoff Characteristics of CAT-PEST Connected Model using Different Infiltration Analysis Methods (CAT-PEST 연계 모형의 침투 해석 방법에 따른 단기 유출 특성 분석)

  • Catchment Hydrologic Cycle Assess Tool (CAT) is a model for hydrologic cycle assessment based on physical parameters. In this study, CAT was applied for short-term runoff simulation and connected with model-independent parameter estimation (PEST) for auto-calibrating parameters. The model performance was compared with HEC-HMS, which is widely used for short-term runoff simulation. The study area is the Pangyo Watershed ($22.9km^2$), which includes the Unjung-Cheon and Geumto-Cheon tributaries of the Tan-Cheon stream. Simulation periods were selected from six rainfall events of a two-year period (2006-2007). For the runoff simulation, CAT was applied using three types of infiltration methods (excess rainfall, Green and Ampt and Horton). Sensitivity analysis was carried out to select the parameters and then CAT was optimized using PEST. The model performance of HEC-HMS and CAT-PEST for the rainfall events were within an acceptable limit with Nash Sutcliffe efficiencies (NSE) of 0.63-0.91 and 0.42-0.93, respectively. The simulation results of HEC-HMS have high accuracy in the case of rainfall events that have a sensitive relationship between initial soil moisture conditions and runoff characteristics. The results of CAT-PEST indicated the possibility of reflecting a real runoff system using various physical parameters.

Incident Angle Dependence of Quantum Efficiency in c-Si Solar Cell or a-Si Thin Film Solar Cell in BIPV System (광 입사각이 BIPV에 적용되는 단결정 또는 비정질 실리콘 태양전지의 양자효율에 미치는 영향)

  • The conversion efficiency of solar cells depending on incident angle of light is important for building-integrated photovoltaics (BIPV) applications. The quantum efficiency is the ratio of the number of charge carriers collected by the solar cell to the number of photons of a given energy shining on the solar cell. The analysis of angle dependence of quantum efficiencies give more information upon the variation of power output of a solar cell by the incident angle of light. The variations in power output of solar cells with increasing angle of incidence is different for the type of cell structures. In this study we present the results of the quantum efficiency measurement of single-crystalline silicon solar cells and a-Si:H thin-film solar cells with the angle of incidence of light. As a result, as the angle of incidence increases in single-crystalline silicon solar cells, quantum efficiency at all wavelength (300~1,100 nm) of light were reduced. But in case of a-Si:H thin-film solar cells, quantum efficiency was increased or maintained at the angle of incidence from 0 degree to about 40 degrees and dramatically decrease at more than 40 degrees in the range of visible light. This results of quantum efficiency with increasing incident angle were caused by haze and interference effects in thin-film structure. Thus, the structural optimization considering incident angle dependence of solar cells is expected to benefit BIPV.

Development and Validation of Reservoir Operation Rules for Integrated Water Resources Management in the Geum River Basin (금강유역의 유역통합수자원관리를 위한 저수지 운영률 개발 및 적용성 검토)

  • In recent, the integrated water resources management should consider not only existing management objects such as water supply, power generation, and instream flows but also new management objects such as water quantity, water quality, and water habitats which management system is large and complex. Moreover, integrated basin plan or operation are needed for solving conflicts problems between basins and between water usages and to maximize water resources usages. To increase use of optimization method for actual operation and apply various objects, a reservoir operation rule was developed and the KModSim's hydrologic states for integrated water resources management were tested in this study. The simulation results show that the developed operation rules applied in hydrologic states good represent the actual storages of both the Yongdam and the Daecheong Reservoirs so, it is possible to improve the water allocation method usually used in the basin management and manage the integrated basin water resources if new operating rules are applied in optimized programming.

Calibration and Validation of the Hargreaves Equation for the Reference Evapotranspiration Estimation in Gyeonggi Bay Watershed (경기만 유역의 기준 증발산량 산정을 위한 Hargreaves 공식의 보정 및 검정)

  • It is essential to locally adjust the Hargreaves parameter for estimating reference evapotranspiration with short data as a substitute of Penman-Monteith equation. In this study, evaluation of daily-based reference evapotranspiration is computed with Hargreaves equation. in Gyeonggi bay area including Ganghwa, Incheon, Suwon, Seosan, and Cheonan station for the time period of 1997-2004. Hargreaves coefficient is adjusted to give the best fit with Penman-Monteith evapotranspiration, being regarded as a reference. Then, the preferred parameters are validated for the same stations for the time period of 2005-2006. The optimization-based correction in calibration for 1997-2004 shows improved performance of the Hargreaves equation, giving 0.68-0.77 to 0.92-0.98 in Nash-Sutcliffe coefficient of efficiency (NSC) and 14.63-23.30 to 5.23-11.75 in RMSE. The validation for 2005-2006 shows improved performance of the Hargreaves equation, giving 0.43-0.85 to 0.93-0.97 in NSC and 14.43-26.81 to 6.48-9.09 in RMSE.

GA-Based Optimal Design for Vibration Control of Adjacent Structures with Linear Viscous Damping System (선형 점성 감쇠기가 장착된 인접구조물의 진동제어를 위한 유전자 알고리즘 기반 최적설계)

  • This paper proposes an optimal design method of distribution and capacities of linear viscous dampers for vibration control of two adjacent buildings. The previous researches have dealt with suboptimal design problem under the assumption that linear viscous dampers are distributed uniformly or proportionally to the sensitivity of the modal damping ratio according to floors, whereas this study deals with global optimization problem in which the damping capacities of each floor are independently selected as design parameters. For this purpose, genetic algorithm to effectively search multiple design variables in large searching domains is adopted and objective function leading to the global optimal solutions is established through the comparison of several optimal design values obtained from different objective functions with control performance and damping capacity. The effectiveness of the proposed method is investigated by comparing the control performance and total damping capacity designed by the proposed method with those of the previous method. In addition, the time history analyses are performed by using three historical earthquakes with different frequency contents, and the simulation results demonstrate that the proposed method is an effective seismic design method for the vibration control of the adjacent structures.

Measurement of Solubilities in the Ternary System NaCl + CaCl2 + H2O and KCl + CaCl2 + H2O at 50℃ (NaCl + CaCl2 + H2O 및 KCl + CaCl2 + H2O 삼성분계에 대한 50℃에서의 용해도 측정)

  • The solubility and the physicochemical property (refractive index) in the NaCl-$CaCl_2$-$H_2O$ and KCl-$CaCl_2$-$H_2O$ systems were determined at $50^{\circ}C$ and the phase diagrams and the diagrams of physicochemical property vs composition were plotted. One invariant point, two univariant curves, and two crystallization zones, corresponding to sodium Chloride (or potassium chloride), dihydrate ($CaCl_2{\cdot}2H_2O$) showed up in the phase diagrams of the ternary systems. The mixing parameters ${\theta}_{M,Ca}$ and ${\Psi}_{M,Ca,Cl}$ (M = Na or K) and equilibrium constant $K_{sp}$ were evaluated in NaCl-$CaCl_2-H_2O$ and KCl-$CaCl_2-H_2O$ systems by least-squares optimization procedure, in which the single-salt Pitzer parameters of NaCl, KCl and $CaCl_2$ ${\beta}^{(0)}$, ${\beta}^{(1)}$, ${\beta}^{(2)}$ and $C^{\Phi}$ were directly calculated from the literature. The results obtained were in good agreement with the experimental data.
