Utilization of Piper betle L. Extract for Inactivating Foodborne Bacterial Biofilms on Pitted and Smooth Stainless Steel Surfaces

  • Biofilms are a significant concern in the food industry. The utilization of plant-derived compounds to inactivate biofilms on food contact surfaces has not been widely reported. Also, the increasing negative perception of consumers against synthetic sanitizers has encouraged the hunt for natural compounds as alternatives. Therefore, in this study we evaluated the antimicrobial activities of ethanol extracts, acetone extracts, and essential oils (EOs) of seven culinary herbs against Salmonella enterica serotype Typhimurium and Listeria innocua using the broth microdilution assay. Among all tested extracts and EOs, the ethanol extract of Piper betle L. exhibited the most efficient antimicrobial activities. To evaluate the biofilm inactivation effect, S. Typhimurium and L. innocua biofilms on pitted and smooth stainless steel (SS) coupons were exposed to P. betle ethanol extract (12.5 mg/ml), sodium hypochlorite (NaClO; 200 ppm), hydrogen peroxide (HP; 1100 ppm), and benzalkonium chloride (BKC; 400 ppm) for 15 min. Results showed that, for the untreated controls, higher sessile cell counts were observed on pitted SS versus smooth SS coupons. Overall, biofilm inactivation efficacies of the tested sanitizers followed the trend of P. betle extract ≥ BKC > NaClO > HP. The surface condition of SS did not affect the biofilm inactivation effect of each tested sanitizer. The contact angle results revealed P. betle ethanol extract could increase the surface wettability of SS coupons. This research suggests P. betle extract might be utilized as an alternative sanitizer in food processing facilities.

Efficacy and Reusability of Commercial Adsorbent for Isolation of Proanthocyanidins from Hot Water Extract of Pinus radiata Bark

  • This study was carried out to investigate the feasibility and usefulness of a commercial synthetic adsorbent, Diaion HP 20, for the isolation of proanthocyanidins (PAs) from hot water extract (HWE) prepared from Pinus radiata bark. Most ultraviolet (UV) absorbing materials in HWE were adsorbed onto Diaion HP 20 and easily recovered by simple ethanol (EtOH) washing. More than 50% of the UV absorbing materials were adsorbed within 20 minutes, and the adsorption equilibrium was reached within 3 h. The recovered materials from Diaion HP 20 were mostly composed of PAs, but some monomeric flavonoids such as taxifolin and unidentified hydrophobic materials were also adsorbed. The impurities such as carbohydrates and inorganic materials contained in HWE were mostly removed by Diaion HP 20. The adsorption ability of the UV absorbing materials onto Diaion HP 20 was almost the same as the first cycle even after 14 times of repeating cycles of adsorption and desorption. The radical scavenging ability of the recovered materials from the adsorbent was slightly higher than that of the pure PAs prepared by Sephadex LH 20 from the same HWE.

Efficient Recovery of Lignocellulolytic Enzymes of Spent Mushroom Compost from Oyster Mushrooms, Pleurotus spp., and Potential Use in Dye Decolorization

  • This study was conducted in order to perform efficient extraction of lignocellulolytic enzymes amylase (EC 3.2.1.1), cellulase (EC 3.2.1.4), laccase (EC 1.10.3.2), and xylanase (EC 3.2.1.8) from spent mushroom compost (SMC) of Pleurotus ostreatus, P. eryngii, and P. cornucopiae. Optimal enzyme recovery was achieved when SMCs were extracted with 50 mM sodium citrate (pH 4.5) buffer at $4^{\circ}C$ for 2 hr. Amylase, cellulase, and xylanase activities showed high values in extracts from P. ostreatus SMC, with 2.97 U/g, 1.67 U/g, and 91.56 U/g, respectively, whereas laccase activity and filter paper degradation ability were highest in extracts from P. eryngii SMC, with values of 9.01 U/g and 0.21 U/g, respectively. Enzymatic activities varied according to the SMCs released from different mushroom farms. The synthetic dyes remazol brilliant blue R and Congo red were decolorized completely by the SMC extract of P. eryngii within 120 min, and the decolorization ability of the extract was comparable to that of 0.3 U of commercial laccase. In addition, laccase activity of the SMC extract from P. eryngii was compared to that of commercial enzymes or its industrial application in decolorization.

Green Synthesis of Nanoparticles Using Extract of Ecklonia Cava and Catalytic Activity for Synthetic Dyes

  • The green synthesis of inorganic nanoparticles (NPs) using biomaterials has garnered considerable attention in recent years because of its eco-friendly, non-toxic, simple, and low-cost nature. In this study, we synthesized NPs of noble metals, such as Ag and Au using an aqueous extract of a marine seaweed, Ecklonia cava. The formation of AgNPs and AuNPs was confirmed by the presence of surface plasmon resonance peaks in UV-Vis absorption spectra at approximately 430 and 530 nm, respectively. Various properties of the NPs were evaluated using characterization techniques, such as dynamic light scattering, transmission electron microscopy, energy-dispersive X-ray spectroscopy, Fourier transform infrared spectroscopy, and X-ray diffraction analysis. Phytochemicals in the seaweed extract, such as phlorotannins, acted as both reducing and stabilizing agents for the growth of the NPs. The green-synthesized AgNPs and AuNPs were found to exhibit high catalytic activity for the decomposition of organic dyes, including azo dyes, methylene blue, rhodamine B, and methyl orange.

Markable Green Synthesis of Gold Nanoparticles Used As Efficacious Catalyst for the Reduction of 4-Nitrophenol

  • The biocompatibility and plasmonic properties of Au nanoparticles make them useful for photothermal therapy, drug delivery, imaging, and many other fields. This study demonstrated a novel, facile, economic, and green synthetic method to produce gold nanoparticles. Gold nanoparticles (AuNPs) with spherical and triangular shapes were effectively synthesized using only Schisandra chenesis fruit extract as the capping and reducing agent. The shape of the AuNPs could be engineered simply by adjusting the molar concentration of HAuCl4 in the reaction mixture. The as-synthesized AuNPs were characterized using UV-VIS spectroscopy, transmission electron microscopy (TEM), X-ray diffraction (XRD), dynamic light scattering (DLS), and energy dispersive X-ray analysis (EDXA). This study revealed that by using the HAuCl4 concentration in the AuNP synthesis, the shape and size of the AuNPs could be controlled by the concentration of HAuCl4 and Schisandra chinensis fruit extract as a surfactant. The as-synthesized AuNPs samples had sufficient colloidal stability without noticeable aggregation and showed the predominant growth of the (111) plane of face-centered cubic gold during the crystal growth. The catalytic efficiency of the AuNPs synthesized using Schisandra chenesis fruit extract was examined by monitoring the catalytic reduction of 4-nitrophenol to 4-aminophenol using Ultraviolet-visible spectroscopy (UV-Vis spectroscopy). The synthesized AuNPs showed good catalytic activity to reduce 4-nitrophenol to 4-aminophenol, revealing their practical usefulness.

대마줄기껍질 추출 섬유를 활용한 항균 양말이 발 건강 개선에 미치는 영향 (Effects of Antimicrobial Socks Using Hemp Stem Bark Extract Fibers on Foot Health Improvement)

  • With increasing global interest in environmentally friendly materials and the consequent rise in demand, there is a growing need for alternatives to synthetic fibers, which can cause skin diseases and other side effects. The fashion industry is emphasizing material sustainability owing to concerns about increasing carbon emissions. Moreover, consumers express a strong desire for ecofriendly and sustainable materials. Therefore, clothing brand companies are developing eco-friendly products to enhance their corporate image. Hemp fibers are recognized for their functionality and are utilized as crucial materials in the development of eco-friendly products by global fashion companies. In this study, we produced socks that effectively improve foot health using hemp stem bark extract fibers and demonstrated the positive efficacy of natural fibers through functional and wearability evaluations. Hemp stem bark extract fibers showed 99.9% antimicrobial effectiveness against bacteria responsible for sweat-induced bacterial proliferation and odor, when blended with lyocell fibers and woven into fabric to manufacture socks. Wearability evaluations of these terry cloth socks confirmed a reduction in foot odor and fatigue among the participants with a consumer satisfaction of 4.63/5. These findings confirm the effectiveness and positive impact of the natural antimicrobial properties of hemp fibers and terry cloth structure in improving foot health.

아위버섯의 생리적 특성 (The physiological characteristics of Pleurotus Ferulae Lanzi.)

  • 아위버섯(Pleurotus Ferulae Lanzi)의 온도별 균사생장도를 조사한 결과, 균사생육온도는 4$^{\circ}C$~35$^{\circ}C$로 나타났으며, 최적 균사생장 온도는 $25^{\circ}C$였고, $25^{\circ}C$~32$^{\circ}C$에서도 비교적 균사생장이 양호하였다. pH별 균사생장도를 조사한 결과는 pH가 4.0, 5.0, 6.0, 7.0, 8.0으로 조절된 MYPA(Yeast Malt Peptone Arar)배지를 기본배지로 하여 균사생장을 조사한 결과, 최적 pH는 6.0이었고, 5.0, 7.0순으로 균사생장이 양호하였던 반면, 4.0에서는 균사생장이 극히 불량하였다. 아위버섯의 균사생장을 위한 최적 배지조성은 탄소원별 균사생장도를 조사한 결과, starch, maltose 첨가구에서 균사생장이 가장 양호하였다. 또한, fructose, glucose, mannitol, lactose, xylose 순으로 균사생장이 비교적 양호하였던 반면, 다른 탄소원에서는 불량하였다. 질소원별 균사 생장량을 조사한 결과 yeast 첨가구에서 균사생장이 가장 양호하였고, 반면, 다른 질소원에서는 불량하였다. 위의 내용을 종합해보면 아위버섯의 최적온도는 25~32$^{\circ}C$, 최적 pH는 5.0~6.0으로 나타났다 또한 적정 영양원은 기본배지로 선발된 MYPA에 탄소원은 starch, maltose, 질소원은 yeast extract을 첨가하는 것이 가장 좋게 나타났다.

Antifungal Activity Against Colletotrichum spp. of Curcuminoids Isolated from Curcuma longa L. Rhizomes

  • Methanol extract of the rhizomes of turmeric, Curcuma longa L., effectively controlled the development of red pepper anthracnose caused by Colletotrichum coccodes. In addition three antifungal substances were identified from the methanol extract of C. longa rhizomes as curcumin, demethoxycurcumin, and bisdemethoxycurcumin using mass and $^{1}H-NMR$ spectral analyses. The curcuminoids in a range $0.4-100\;{\mu}g/ml$ effectively inhibited the mycelial growth of three red pepper anthracnose pathogens, C. coccodes, C. gloeosporioides, and C. acutatum. The three curcuminoids inhibited mycelial growth of C. coccodes and C. gloeosporioides to an extent similar to the synthetic fungicide dithianon did, but the synthetic agent was a little more effective against C. acutatum. The curcuminoids also effectively inhibited spore germination of C. coccodes, and bisdemethoxycurcumin was the most active. Among the three curcuminoids, only demethoxycurcumin was effective in a greenhouse test in suppressing red pepper anthracnose caused by C. coccodes.

합성 유전자를 이용하여 Escherichia coli에서 백신 후보의 생산 혹은 진단용 항체의 개발을 위한 인간 rotavirus VP8* 부분 단백질의 발현 (Use of the Synthetic Gene Encoding the Truncated Human Rotavirus VP8* Protein in Escherichia coli for Production of Vaccine Candidates or Development of Diagnostic Antibodies)

  • 인간 rotavirus는 영아에게 급성 설사를 일으키는 병원체의 하나이다. 본 연구에서는 Escherichia coli의 코돈 선호도를 따라서 인간 rotavirus A (serotype 1 strain WA)의 $VP8^*$ 단백질을 일부분 암호화하도록 인공적인 유전자를 합성하였다. 합성된 $VP8^*$ 유전자는 코돈을 번역틀에 일치시키고 클로닝이 용이하도록 하기 위한 NdeI 및 HindIII 제한효소 절단 부위와 친화적 정제를 위한 6-히스티딘 암호화 서열을 C-말단에 보유하고 있다. 합성된 $VP8^*$ DNA 절편을 pT7-7 발현 벡터에 삽입하여 E. coli BL21 (DE3)로 형질전환한 후에 최종 농도 0.05 mM IPTG로 생산을 유도한 결과 예상했던 대로 19.7-kDa 크기의 $VP8^*$ 단백질이 고농도로 발현되었다. SDS-PAGE에 전개된 단백질들을 대상으로 mouse anti-rotavirus capsid antibody를 사용한 Western blotting의 결과 ~20-kDa $VP8^*$ 단백질 밴드가 관찰되었다. 인공 $Vp8^*$ 단백질이 피하 주사된 토끼의 polyclonal antibody 혈장을 이용한 조사에서도 동일한 크기의 단백질 밴드를 확인할 수 있었다. 이는 합성된 유전자가 바이러스성 질환을 통제할 항원성 백신 후보의 생산 혹은 진단용 항체를 개발하기 위한 쉽고 빠른 방법을 제공할 수 있다는 의미이다.

Antimicrobial Activity of Phenolic-Rich Extracts from Mango Seed Kernel on Microorganisms

  • Replacing synthetic additives to preserve food products with natural antimicrobial compounds needs to be considered due to public health. In the present study, the phenolic extract from mango seed kernel (MSK) with the total phenolic content (TPC) value of 5300 ± 380 mg gallic acid equivalent (GAE)/L was used for evaluating the antimicrobial properties against five types of bacterial strains, including Listeria monocytogenes, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Salmonella typhimurium in vitro. This extract was also used for in situ method on the storage of Pangasianodon hypophthalmus fish fillets. The minimum inhibitory concentration (MIC) values for all tested strains were determined at an average concentration of 1325 mg GAE/L. Furthermore, the minimum bactericidal concentration (MBC) values of E. coli, S. aureus, and S. typhimurium were seen at 5300 mg GAE/L, while the extract did not show eliminations in the growth of the remaining strains. The bacterial inhibition speed of the extract illustrated that the concentration equal MIC value eliminated S. typhimurium growth after 24 h, the 4 times MIC value had remarkable effects on S. aureus growth after the 9 h of incubation, and 24 h of incubation for E. coli, L. monocytogenes, P. aeruginosa. Additionally, the MSK extract could inhibit the growth of P. aeruginosa on fish fillets in 4 days of storage. These results provide important evidence for the utilization of MSK as a natural source of antimicrobial agents in food products.
