Current status of natural product industry and its commercial application to health functional foods

  • Natural product substances have historically served as the most significant also be prepared by source of new leads for pharmaceutical development. They can chemical synthesis(both semisynthesis and total synthesis) and have played a important role in the field of organic chemistry by providing synthetic targets. Rcently, they have also been extended for commercial purpose to refer to medicinal products, health functional foods, dietary supplements and cosmetics from natural sources. A large number of currently prescribed drugs have been either directly derived from or inspired by natural products. However, with the advent of robotics, bioinformatics, high throughput screening(HTS), molecular biology-biotechnology, combinatorial chemistry, in silico(molecular modeling) and other methodologies, the pharmaceutical industry has largely moved away from plant derived natural products as a source for leads and prospective drug candidates. The strategy for natural prduct industry is now changing from drug approaches to health foods by identifying effective natural products as preparations. In Korea, a lot of development of natural product based drugs have been done, but very few on health functional foods. The concept of natural product based health foods is not active components as lead compounds but standardized extracts or preparation mixed with other medicinal plants. The representative material has been recently known to be a standardized ginseng extract "Ginsana G 115" developed by Swiss Pharmaton company. The purpose of this presentation is to underline how natural products research continues to make significant contributions in the domain of discovery and development of new health functional foods. It is proposed to present the development of high value added health food or health functional foods through scientific investigation on efficacy and standardization of new materials form natural products.

Phytochemical Constituents of Capsella bursa-pastoris and Their Anti-inflammatory Activity

  • Phytochemical investigation of 80% MeOH extract of the aerial parts of Capsella bursa-pastoris yielded fourteen compounds (1 - 14). The structures of the compounds were elucidated by spectroscopic methods to be methyl-1-thio-${\beta}$-D-glucopyranosyl disulfide (1), 10-methylsulphinyl-decanenitrile (2), 11-methyl-sulphinyl-undecanenitrile (3), 1-O-(lauroyl)glycerol (4), phytene-1, 2-diol (5), (3S,5R,6S,7E)-5,6-epoxy-3-hydroxy-7-megastigmen-9-one (6), loliolide (7), ${\beta}$-sitosterol (8), 3-hydroxy-1-(4-hydroxy-3-methoxyphenyl)-1-propanone (9), 1-feruloyl-${\beta}$-D-glucopyranoside (10), pinoresinol-4'-O-${\beta}$-D-glucopyranoside (11), luteolin (12), quercetin-3-O-${\beta}$-D-glucopyranoside (13), and luteolin 6-C-${\beta}$-glucopyranoside (14). Although compound 1 was reported as synthetic compound, 1 was first isolated from natural source. NMR spectral data assignments of 1, 2 and 3 were reported for the first time, and compounds 1 - 14 were for the first time reported from this plant source. The anti-inflammatory effects of 1 - 14 were evaluated in lipopolysaccharide (LPS)-stimulated murine microglia BV-2 cells. Compounds 12 exhibited strong inhibitory effects on nitric oxide production in LPS-activated BV-2 cells with $IC_{50}$ values of $9.70{\mu}M$.

고속액체크로마토그라피에 의한 계란중의 합성항균제 잔류량 (Residue of Synthetic Antimicrobial Agent in Eggs by High Performance Liquid Chromatography)

  • 고속액체크로마토그라피를 이용하여 계란 중의 합성항균제 설파메라진, 설파메타진, 설파디메톡신, 후라졸리돈, 죠렌 등의 잔류량에 관한 신속하고 간편한 동시분서법을 검토하였는데, 각 합성항균제 표준용액을 첨가하여 나타난 회수율은 설파메라진 81.2%, 설파메타진 87.6%, 설파디메톡신 92.5%, 후라졸리돈 86.1%, 죠렌 73.9%로 나타났으며, 검출한계는 설파메라진, 설파메타진, 설파디메톡신이 0.2ppb 수준으로 나타났고, 후라졸리돈과 죠렌 0.5ppb 수준으로 나타났다. 본 분석방법으로 대구지역 시판계란 84개에 대한 잔류량을 조사한 결과, 설파메타진 3개 계란, 설파디메톡신이 4개 계란에서 각각 0.005-0.008ppm과 0.012-0.019ppm 정도 검출되었으며, 설파메라진, 후라졸리돈, 죠렌은 모든 시료에서 검출되지 않았다. 이상의 결과로 추정해 볼 때 설파메라진, 설파메타진, 설파디메톡신, 후라졸리돈, 죠렌의 계란내 잔류정도는 비교적 안전한 수준으로 나타났다.

최적의 야외유인을 위한 복숭아명나방(Dichocrocis punctiferalis)의 성페로몬 조성 (Sex Pheromone Composition for Field-trapping of Dichocrocis punctiferalis (Lepidoptera: Pyralidae) Males)

  • 사과, 복숭아와 밤 열매 등을 가해하는 주요해충 중의 하나인 복수아명나방(Dichocrcis punctiferalis)을 성페로몬을 이용하여 발생예찰하기 위해 성페로몬 성분분석과 행동관찰, 성페로몬 조성별 야외포획실험을 수행하였다. 처녀암컷의 교미행동은 16L:8D의 광주기 $26\pm$1$^{\circ}C$ 온도조건에서 소등 후 4시간째에, 수컷의 가는털뭉치 돌출행동은 5시간째에 최고를 나타내었다. 이시간대의 암컷 복부 끝 헥산추출물에서는 E10-hexadecenal과 Z10-hexadecenal이 가스크로마토그래피 분석에 의해 85:15의 비율로 검출되었다. 야외에서 수컷 성충을 유인하기 위해 두 성분의 합성화합물을 이용하여 사과 및 복수아와 밤 과수원에서 포획실험이 수행되었는데 E10-hexadecenal과 Z10-hexadecenal이 70:30~80:20의 비율에서 가장 많이 포획되었고, 이와 유사한 조성에서 수컷의 가는털뭉치 돌출반응과 상승비행반응이 관찰되었다.

Resveratrol-Enriched Rice Down-Regulates Melanin Synthesis in UVB-Induced Guinea Pigs Epidermal Skin Tissue

  • Synthetic compounds that are used in the clinic to regulate skin hyperpigmentation, such as arbutin, hydroquinone, and kojic acid, are only moderately effective. But, their use is limited by side effects. As part of an effort to overcome the limitations, we developed resveratrol-enriched rice (RR) using genetic engineering technique. Each of resveratrol and rice has been reported to produce anti-melanogenic effects. Therefore, we hypothesized that RR would show more anti-melanogenic effects than those of resveratrol or rice alone. Anti-melanogenic effect of RR was done by using melan-a mouse melanocytes. The depigmenting efficacy was then observed following topical application of the RR to UVB-stimulated hyperpigmented dorsal skin of guinea pigs. Treatment with RR extract resulted a $21.4{\pm}0.7%$ decrease in tyrosinase expression at melan-a cells. Colorimetric analysis showed a significantly lower depigmenting value by day 9 following treatment with RR in UVB-irradiated guinea pigs the dorsal skin (p<0.01), indicating that RR produced a depigmentation effect. By staining with Fontana-Masson stain, we found that the RR-treated group had more effect histopathologically in epidermal melanin production than resveratrol or rice alone-treated group. RR was associated with reduction in the levels of microphthalmia-associated transcription factor (MITF), and downregulation of tyrosinase and tyrosinase-related protein (TRP-2) expression, leading to inhibit epidermal melanin production by western blot analysis. This study suggests that the resveratrol-enriched rice may be a promising candidate in regulating skin pigmentation with UVB exposure.

영상의 깊이정보 추출을 위한 weighted cost aggregation 기반의 스테레오 정합 기법 (Weighted cost aggregation approach for depth extraction of stereo images)

  • 스테레오 비전 시스템(stereo vision system)은 2차원 영상정보를 이용하여 3차원 깊이 정보를 획득하는 데 유용한 방법으로, 그동안 많은 연구가 진행되었다. 3차원 깊이 정보를 획득하기 위해서는 영상의 대응점을 찾아야 하는데, 속도와 정확성을 동시에 만족시키기가 어렵다. 이러한 문제를 해결하기 위해, 본 논문에서는 적응적 가중치(weight)를 적용한 cost aggregation 기반의 스테레오 정합 기법을 제안한다. 이 방법은 스테레오 영상의 특징을 이용하여 가중치를 획득하고, 색상정보, 밝기정보에 거리정보를 이용하여 가중치를 적용한 후, 이를 이용하여 대응점을 찾아 깊이 정보를 추출한다. 제안된 방법의 성능을 평가하기 위하여 ground truth가 존재하는 다양한 스테레오 영상을 이용하여 실험하였으며, 실험 결과 다양한 영상에서도 향상된 결과를 보였다.

탈지참깨박 중 페놀산의 대두유에 대한 항산화 효과(III) (Antioxidative Effectiveness of Phenolic Acids in Defatted Sesame Meal on the Soybean oil)

  • 본 연구에서는 탈지참깨박의 기능성을 검토하기 위하여 탈지참깨박으로부터 페놀산을 추출하여 이들 추출물의 항산화 효과를 합성항산화제인 BHA, AP 및 TBHQ의 항산화 효과와 비교하였다. 탈지참깨박 중 페놀산 추출물은 유리형, 에스터형, 불용성 페놀산이 각각 246.8, 109.6, 85.8mg/100g으로 유리형이 가장 높았으며, 유리형 페놀산에는 6종류의 페놀산이 다량 함유되어 있었고 에스터형과 불용성 페놀산에는 각각 3종류의 페놀산이 함유되어 있었다. 탈지참깨박 중의 3가지 형태의 페놀산의 항산화력은 유리형이 가장 컸고 에스터형, 불용성 순으로 나타났는데 이것은 유리형 페놀산 group의 종류와 함량이 가장 많은 것에 기인하는 것으로 생각된다. 탈지참깨박 페놀산 추출물의 항산화 효과는 BHA나 AP(0.02%)보다 강한 것으로 나타났으며, 유리형 페놀산의 경우는 합성 항산화제 중 가장 우수한 것으로 나타난 TBHQ의 항산화력과 거의 유사할 정도로 높았다.

Cytotoxicity and Structure Activity Relationship of Dammarane-Type Triterpenoids from the Bark of Aglaia elliptica against P-388 Murine Leukemia Cells

  • Six dammarane-type triterpenoids, dammar-24-en-$3{\beta}$-ol (1), $3{\beta}$-epicabraleahydroxy lactone (2), (E)-25-hydroperoxydammar-23-en-$3{\beta}$,20-diol (3), dammar-24-en-$3{\beta}$,20-diol (4), $3{\beta}$-acetyl-20S,24S-epoxy-25-hydroxydammarane (5), and $3{\beta}$-epiocotillol (6) were isolated from the methanolic extract of the bark of Aglaia elliptica. The chemical structure were identified on the basis of spectroscopic evidence and by comparison with those spectra previously reported. Compounds 1 - 6 were isolated first time from this plant. Compounds 1 - 6, along with a known synthetic analog, cabraleone (7) were evaluated their cytotoxic activity against P-388 murine leukimia cells in vitro. Among those compounds $3{\beta}$-acetyl-20S,24S-epoxy-25-hydroxydammarane (5) showed strongest cytotoxic activity with $IC_{50}$ value of $8.02{\pm}0.06{\mu}M$.

Dendropanax morbifera Extract Inhibits Intimal Hyperplasia in Balloon-Injured Rat Carotid Arteries by Modulating Phenotypic Changes in Vascular Smooth Muscle Cells

  • The plant Dendropanax morbifera Léveille is effective folk medicines for the treatment of several conditions, such as infectious diseases, skin diseases, and other illnesses. Although the inhibitory effects of D. morbifera on the proliferation and migration of vascular smooth muscle cells (VSMCs) have been shown in our previous study, its effects in vivo remain to be elucidated. In this study, we aimed to investigate the protective effects of the extracts from D. morbifera (EDM) on neointimal hyperplasia of rat carotid artery and explore the underlying mechanisms. We observed that the ratio of intima to media thickness (I/M) was significantly decreased in the EDM-treated groups by ~80% compared to that of the control. The expression of Ki-67 and proliferating cell nuclear antigen was decreased by ~70% in the EDM-treated groups compared to that of the control. In addition, matrix metalloproteinase (MMP)2 and MMP9 significantly reduced in the neointimal layer of the EDM-treated groups. Moreover, the decreased levels of contractile phenotypic markers of VSMCs, such as α-smooth muscle actin, myocardin, and smooth muscle-myosin heavy chain, were successfully restored by EDM treatment. Furthermore, the levels of synthetic phenotypic markers, cellular retinal binding protein 1 and connexin 43 were also restored to normal levels. These results suggest that EDM inhibits vascular neointimal hyperplasia induced by balloon injury in rats via phenotypic modulation of VSMCs. Therefore, EDM may be a potential drug candidate for the prevention of restenosis.

측백엽(側柏葉) 메탄올 추출물이 멜라닌 형성에 미치는 영향 (Effect of Methanolic Extract from Biota Orientalis Folium on Melanin Synthesis)

  • Recently many efforts were focused to understand the mechanical insights of melanogenesis to develop the agents for hyper-pigmentation and hypo-pigmentation. In the melanin bio-synthetic pathway, tyrosinase is the rate limiting enzyme, and ${\alpha}$-melanocyte stimulating hormone(MSH) stimulates melanogenesis and enhances the melanin synthesis and the tyrosinase activity. The author has analyzed the effects of Biota Orientalis Folium on the basal melanogenic activities of B16 mouse melanoma cells, and on the ${\alpha}$-MSH or tyrosinase-induced melanogenesis. Biota Orientalis Folium alone markedly suppressed melanin content and tyrosinase activity in a dose-dependent manner. The decrease of cell propagation was observed in B16 cells treated with 200${\mu}$g/ml dose of Biota Orientalis Folium, indicating that Biota Orientalis Folium-induced depigmenting effect was caused by inhibition of melanin synthesis, not due to destruction of B16 cells. Pretreatment of the cells with Biota Orientalis Folium also suppressed the increase of ${\alpha}$-MSH (10 nM) induced melanin content and tyrosinase activity. Biota Orientalis Folium inhibited the revelation of ${\alpha}$-MSH induced tyrosinase protein and tyrosinase related protein and mRNA of tyrosinase in B16 melanoma cell. These results suggest that Biota Orientalis Folium inhibits melanogenesis and abrogates ${\alpha}$-MSH and tyrosinase-induced melanogenesis in B16 melanoma cells.
